Gaeumannomyces graminis var. tritici에 의한 벼 엽초(葉?) 썩음병 (Sheath Rot Dieseases of Rice Seedling Caused by Gaeumannomyces graminis var. tritici in Korea)

  • Gaeumannomyces graminis var. tritici was first isolated from rice in 1981. Sheath rot of rice seedling in box culture for machine transplanting was produced by artificial inoculation of the causal organism. Especially sheath rot was developed from crown to leaf sheath above water level. Perithecia were usually formed in dead leaf sheaths and in PDA culture. They were black, globose, and immersed with obliquely erumpent neck. Asci were unitunicate with an apical ring and contained 8 spores. Ascospores were hyline, slender, $80{\sim}140\;um$ in length. Each spore showed $3{\sim}4\;septa.$ Based on morphological and cultura characteristics, the isolate from seeds was identified as Gaeumannomyces graminis (Sacc.) Arx & Olivier tritici J. Walker.

Pseudocercospora fuligena에 의한 토마토 검은잎곰팡이병 (Black Leaf Mold of Tomato Caused by Pseudocercospora fuligena in Korea)

  • 2011년 9월, 충남 보령시와 부여군의 시설재배단지에서 재배중인 토마토의 잎에 Pseudocercospora fuligena에 의한 검은잎곰팡이병이 발생하였다. 이 병의 병징은 초기에는 잎에 연한 노란색의 불규칙한 점무늬가 발생하며, 점차 연한 갈색으로 변하고 잎 뒷면의 병반부위에 병원균의 분생포자를 다량으로 형성하면서 검은색의 그을음 병징을 나타내었다. 병든 토마토 식물체로부터 원인균을 분리하였고 현미경 검경을 통하여 형태적 특징을 분석한 결과 P. fuligena로 동정되었다. 온실조건에서 인공접종을 통하여 이 균의 병원성을 증명하였다. P. fuligena에 의한 토마토 검은잎곰팡이병의 발생은 우리나라에서 최초의 보고이다.

  • 2023년 8월 청주 땅콩 재배포장에서 기보고되어 있는 점무늬 병징과는 다른 양상의 잎 반점을 발견하였다. 갈색 반점은 주로 잎 가장자리에 형성되었으며, 반점 부위에서 원인균을 분리하였다. 순수 분리한 병원균을 땅콩 유묘에 인공접종하여 병원성 검정을 수행하였다. 그 결과, 접종 5-7일 뒤에 유사한 병징을 확인할 수 있었고 병징 부위에서 동일한 균을 재분리하였다. 분리한 균주의ACT-CHS-1-GAPDH-ITS-TUB2 영역을 이용하여 동정 및 유연관계 분석을 수행한 결과 C. sojae임을 확인하였다. 따라서 본 연구는 국내 최초로C. sojae에 의해 발생한 땅콩 탄저병을 보고하고자 한다.

벼 도열병에 대한 품종의 지속저항성 발현요인에 관한 연구 3. 품종과 도열병균 레이스의 상호관계 (Factors Affecting the Expression of Durable Resistance of Rice Cultivars to Blast Caused by Pyricularia grisea Sacc)

  • Interaction of races of rice blast fungus and cultivars has been evaluated. Dongjinbyeo was infected by four KJ races and two KI races, while other varieties were susceptible to four to six KJ or KI races. Among the single spores reisolated from the leaf blast lesion of Dongjinbyeo after mixed inoculation of 10 races, 63% was race KJ-301 which produced a small number and small size lesions, while 30% was race KI-313 which produced a large number of lesions. However, 93% of single spores reisolated from Palgongbyeo was a highly virulent race KJ-105. On the other hand, all the races were equally reisolated from the susceptible cultivar Jinmibyeo. Frequency of races isolated from the naturally infected leaf blast lesions in the field was similar to that of reisolated races from the cultivars inoculated with 10 mixed races in the greenhouse. However, 30% of single spores isolated from the naturally infected Dongjinbyeo was race KI-329 ut ace KI-313 was not detected. Genetic relationship of the isolates collected from leaf and neck blast fungus of Jinmibyeo and Nakdongbyeo showed specific bands on RFLP-P64, However, their genetic similarity was 80%.

Lactobacillus plantarum으로 발효시킨 황칠나무 잎 추출물의 피부 미백 관련 효과 (Melanin Production Inhibitory Activity of the Dendropanax morbifera Leaf Extract Fermented by Lactobacillus plantarum)

  • This study was conducted to investigate the tyrosinase inhibitory and melanin production inhibitory activity of the distilled water extract of Dendropanax morbifera leaf (DMW) and the fermented extract by Lactobacillus plantarum (DMF). DMF was prepared by inoculation of L. plantarum after the extraction procedure with distilled water. Fermentation for 48 hours at $37^{\circ}C$ is the most effective condition in this study. In DPPH radical scavenging ability, $SC_{50}$ values of the fermented DMF was $37.9{\mu}g/ml$ as a result of more effective than DMW extract ($52.6{\mu}g/ml$). Moreover, tyrosinase inhibitory activity of DMF showed higher activity than DMW. In nontoxic concentration range, DMF showed strong melanin production inhibitory effect in ${\alpha}$-melanocyte stimulating hormone-stimulated B16F10 cell. As a result, the fermentation of the distilled water extract of D. morbifera leaf by L. plantarum could be applicable to functional materials production for skin-whitening agents.

  • Tomato yellow leaf curl virus (TYLCV), a member of the genus Begomovirus, is responsible for one of the most devastating viral diseases in tomato-growing countries and is becoming a serious problem in many subtropical and tropical countries. The climate in Korea is getting warmer and developing subtropical features in response to global warming. These changes are being accompanied by TYLCV, which is now becoming a large problem in the Korean tomato industry. The most effective way to reduce damage caused by TYLCV is to breed resistant varieties of tomatoes. To accomplish this, it is necessary to establish a simple inoculation technique for the efficient evaluation of resistance to TYLCV. Here, we present the rolling circle amplification (RCA) method, which employs a bacteriophage using phi-29 DNA polymerase for construction of infectious TYLCV clones. The RCA method is simple, does not require sequence information for cloning, and is less expensive and time consuming than conventional PCR based-methods. Furthermore, RCA-based construction of an infectious clone can be very useful to other emerging and unknown geminiviruses in Korea.

Seed Transmission of Tomato yellow leaf curl virus in White Soybean (Glycine max)

  • Tomato yellow leaf curl virus (TYLCV) infection of the common bean (Phaseolus vulgaris) has been reported, but soybean (Glycine max) has not previously been identified as a TYLCV host. Five cultivars of white soybean were agro-inoculated using an infectious TYLCV clone. At 30 days post-inoculation, they showed infection rates of 25% to 100%. Typical TYLCV symptoms were not observed in any inoculated plants. To examine whether TYLCV was transmitted in soybean seeds, DNA was isolated from bundles of five randomly selected seeds from TYLCV-inoculated soybean plants and amplified with a TYLCV-specific primer set. With the exception of one bundle, all bundles of seeds were verified to be TYLCV-infected. Virus dissemination was also confirmed in three of the 14 bunches. Viral replication was also identified in seeds and seedlings. This is the first report demonstrating that soybean is a TYLCV host, and that TYLCV is a seed-transmissible virus in white soybean.

벼흰빛잎마름병에 있어서 병원균의 접종 농도가 병의 발전속도에 미치는 영향 (Studies on the Relationship between Inoculum Concentration and Disease Development in Bacterial Leaf Blight of Rice)

  • 본 실험은 벼흰빛잎마름병에 대한 저항성계통선발에 필요한 인공접종법 개발을 위해 시도한 것으로서 병원균의 접종농도와 식물체의 노유가 본병의 발전에 미치는 영향에 관해 중점을 두었다. 일정한 환경조건하에서 본병에 대한 식물의 감수성은 식물의 묘령이 어릴 때 일수록 예민하게 나타났으며 식물의 생장과 더불어 점차 둔감해지는 경향이었다. 파종후 14, 37, 48 및 58일째 되는 식물에서 본병에 대한 저항성과 이병성 계통을 선별할 수 있는 병원균의 적정농도는 각각 $10^6,\;10^7,\;10^7-10^{-9}$$10^9 cells/ml.,$였다. 어린 식물에 대해 지나치게 높은 농도의 접종액은 식물의 품종에 관계없이 단시일에 고사시키는 결과를 초래하였고 묘령이 많은 식물에 대해 일정수준 이하의 세균접종농도를 적용했을 때는 식물의 품종간 특성을 구별할 수 없었다.

  • 질소고정력(窒素固定力)이 큰 두과작물(豆科作物)로 알려진 Sesbania를 Growth cabinet에서 Leonard-Jar법(法)으로 재배(栽培)한 시험결과(試驗結果)는 다음과 같다. 1. 근류균접종시(根瘤菌接種時) Sesbania의 부위별(部位別) 건물중(乾物重)은 경(莖)>엽(葉)>근(根)> Stem nodule > Root nodule순(順)으로 많으며 총건물중(總乾物重)은 근(根)+경근류균접종(莖根瘤菌接種)>근근류균(根根瘤菌) 접종(接種)> 무접종(無接種)으로 경(莖)에 근류균(根瘤菌) 접종효과(接種效果)가 인정(認定)되었다. 2. 질소고정력(窒素固定力)은 총건물중(總乾物重)과 같이 건물중(乾物重)이 많은 것이 질소고정력(窒素固定力)도 큰 경향(傾向)이며 뿌리외(外) 경(莖)에서도 질소고정력(窒素固定力)이 컸다. 3. 공기중(空氣中)에 $^{15}N$을 기준(基準)해서 자연(自然)에 존재(存在)하는 물질(物質)의 동위체(同位體)의 부위별(部位別) ${\delta}^{15}N$증가(增加)는 근(根)+경(莖)>근(根)> 무접종구(無接種區)의 순(順)이고 Root nodule 과 Stem nodule은 positive 근(根), 경(莖), 엽등(葉等)은 Negative이었다.

Responses of Guava Plants to Inoculation with Arbuscular Mycorrhizal Fungi in Soil Infested with Meloidogyne enterolobii

  • In the Northeast of Brazil, expansion of guava crops has been impaired by Meloidogyne enterolobii that causes root galls, leaf fall and plant death. Considering the fact that arbuscular mycorrhizal Fungi (AMF) improve plant growth giving protection against damages by plant pathogens, this work was carried out to select AMF efficient to increase production of guava seedlings and their tolerance to M. enterolobii. Seedlings of guava were inoculated with 200 spores of Gigaspora albida, Glomus etunicatum or Acaulospora longula and 55 days later with 4,000 eggs of M. enterolobii. The interactions between the AMF and M. enterolobii were assessed by measuring leaf number, aerial dry biomass, $CO_2$ evolution and arbuscular and total mycorrhizal colonization. In general, plant growth was improved by the treatments with A. longula or with G. albida. The presence of the nematode decreased arbuscular colonization and increased general enzymatic activity. Higher dehydrogenase activity occurred with the A. longula treatment and $CO_2$ evolution was higher in the control with the nematode. More spores and higher production of glomalin-related soil proteins were observed in the treatment with G. albida. The numbers of galls, egg masses and eggs were reduced in the presence of A. longula. Inoculation with this fungus benefitted plant growth and decreased nematode reproduction.
