Effects of Dietary Fructooligosaccharide on Digestive Enzyme Activities, Intestinal Microflora and Morphology of Growing Pigs

  • One hundred and twenty-eight growing barrows (Jiaxing Black${\times}$Duroc${\times}$Landrace) at an average BW of 20.8 kg were allocated to four treatments for 42 days, each of which was replicated four times with eight pigs per replicate and used to investigate the effects of fructooligosaccharide (FOS) on digestive enzyme activities, intestinal microflora and morphology of growing pigs. The pigs received the same basal corn-soybean meal diet and FOS was added to the basal diet at 0, 2, 4, 6 g/kg diet at the expense of corn, respectively. As compared to control, supplementation with 4 and 6 g/kg FOS significantly improved average daily gain and feed efficiency. Addition of FOS enhanced the growth of Bifidobacterium and Lactobacillus, but inhibited Clostridium and Escherichia coli in the small intestinal and proximal colonic contents. Supplementation with 4 and 6 g/kg FOS significantly improved the activities of total protease, trypsin and amylase in the small intestinal contents. However, FOS had no significant effect on the activity of lipase in the small intestinal contents as well as the digestive enzymes in pancreas. Morphological measurement of jejunal mucosa did show response to consumption of FOS. Villus height and the villus height to crypt depth ratio at the jejunal mucosa were significantly higher with 4 and 6 g/kg FOS supplementation as compared to control.

Microencapsulated basil oil (Ocimum basilicum Linn.) enhances growth performance, intestinal morphology, and antioxidant capacity of broiler chickens in the tropics

  • Objective: Microencapsulation is a technique to improve stability, bioavailability, and controlled release of active ingredients at a target site. This experiment aimed to investigate the effects of microencapsulated basil oil (MBO) on growth performance, apparent ileal digestibility (AID), jejunal histomorphology, bacterial population as well as antioxidant capacity of broiler chickens in a tropical climate. Methods: A total of 288 one-day-old female broilers (Ross 308) were randomly allocated into 4 groups (6 replicates of 12 birds), based on a completely randomized design. Dietary treatments were as follows: i) basal diet (NC), ii) basal diet with avilamycin at 10 ppm (PC), iii) basal diet with free basil oil (FBO) at 500 ppm, and iv) basal diet with MBO at 500 ppm, respectively. Results: Dietary supplementation of MBO improved average daily gain, and feed conversion ratio of broilers throughout the 42-d trial period (p<0.05), whereas MBO did not affect average daily feed intake compared with NC group. The broilers fed MBO diet exhibited a greater AID of crude protein and gross energy compared with those in other groups (p<0.05). Lactobacillus spp. and Escherichia coli populations were not affected by feeding dietary treatments. Both FBO and MBO had positive effects on jejunal villus height (VH), villus height to crypt depth ratio (VH:CD) and villus surface area of broilers compared to NC and PC groups (p<0.05). Superoxide dismutase level in the duodenal mucosa of MBO group was significantly increased (p<0.01), whereas malondialdehyde level was significantly decreased (p<0.01). Conclusion: Microencapsulation could be considered as a promising driver of the basil oil efficiency, consequently MBO at 500 ppm could be potentially used as a feed additive for improvement of intestinal integrity and nutrient utilization, leading to better performance of broiler chickens.

Effects of yeast hydrolysate supplementation on intestinal morphology, barrier, and anti-inflammatory functions of broilers

  • Objective: This study was conducted to evaluate the effects of dietary yeast hydrolysate (YH) supplementation on intestinal morphology, barrier, and anti-inflammatory functions of broilers. Methods: A total of 320 one day old male broilers were randomly allocated into four groups with eight replicates of ten broilers each. The broilers were supplemented with a basal diet (the control group) or basal diets adding 50, 100, 150 mg/kg YH, respectively. This trial lasted for 42 days. The orthogonal polynomial contrasts were used to determine the linear and quadratic effects of increasing levels of YH. Results: In our previous research, supplementing YH improved growth performance by enhancing body weight gain but decreased feed-to-gain ratio. In this study, compared with the control group, dietary YH addition linearly and quadratically decreased serum diamine oxidase activity (p<0.05). Additionally, supplementing YH linearly and/or quadratically decreased jejunal crypt depth (CD), tumor necrosis factor-alpha (TNF-α) concentration as well as mucin 2, interleukin-6 (IL-6), IL-1β, TNF-α, nuclear factor kappa B, and myeloid differentiation factor 88 gene expression levels (p<0.05). Whereas the jejunal villus height (VH), VH/CD, IL-10 concentration as well as zonula occludens-1 and IL-10 gene expression levels were linearly and/or quadratically increased by YH supplementation (p<0.05). Conclusion: Dietary YH supplementation improved intestinal morphology, barrier and anti-inflammatory functions while decreased intestinal permeability of broilers, which might be related with altering pertinent genes expression. This study provides evidence of YH as a promising feed additive for broilers.

방사선 조사 마우스에서 소장움세포의 Apoptosis 발생에 미치는 생약의 효과 (The effects of herbs on the radiation-induced apoptosis in intestinal crypt cells)

  • Apoptosis의 유발을 억제하는 한약제를 파악하여 apoptosis와 관련된 정상적 또는 질병에 대한 연구에 자료를 제공하고자 방사선에 의해 유도된 apoptosis를 지표로 한의학적 처방에서 많이 사용되는 대표적 한약제(24종)의 효과를 검증하였다. 용안육, 산조인, 원지, 인삼, 복령 목향, 천궁 백작약, 승마, 시호 및 눈꽃동충하초 투여군에서 apoptosis는 감소되었으며, 이들 생약제는 apoptosis와 관련된 질병의 예방 및 치료에 적용할 수 있을 것이다. 추후 이들 생약의 작용 기전에 대한 연구가 요구된다.

멜라토닌이 생쥐 소낭 세포 재생과 아포토시스에 미치는 영향에 대한 연구 (The Effect of Melatonin on Mouse Jejunal Crypt Cell Survival and Apoptosis)

  • 목적 :멜라토닌의 방사선 증개 방어 효과와 기전을 밝혀 방사선 치료제로서의 기초 자료를 제공하고자 하였다. 방법 : 168 마리의 생쥐를 방사선량과 멜라토닌 투약 여부에 따라 28개 군으로 나누어 실험하였다 방사선량은 0.8, 10, 12, 14, 16, 18 Gy를 조사하고 소낭의 생존 곡선을 보기 위하여 Withers와 티kind의 방법을 이용한 microcoiony survival assay를 시행하였고 아포토시스를 보기 위하여 TUNEL assay를 시행하였다. 결과 : 멜라토닌의 방사선 장애 방어 효과는 전 선량군에서 확인되었고 특히 고선량군에서 방어 효과가 많은 차이를 보였다. Apoptosis index는 8 Gy 방사선 조사 후 대조군에서 18.4$\%$, 멜라토닌 투여군에서 16.5$\%$이었고 18 Gy 방사선 조사 후 대조군에서 17.2$\%$, 멜라토닌 투여군에서 15.4$\%$로 나타나 통계적인 차이를 보이지 않았다. 결론: 멜라토닌은 생쥐 소장의 소낭 세포에서 방사선 장애 방어 효과를 나타냈으나 이의 기전은 아포토시스 억제 효과와는 관련이 없는 것으로 판단하였다.

Dietary Bovine Colostrum Increases Villus Height and Decreases Small Intestine Weight in Early-weaned Pigs

  • This experiment examined the effect of dietary spray-dried bovine colostrum on intestinal histology and organ weights in early-weaned pigs. In a randomised complete block design, twelve 14-day-old weaner pigs were offered a diet containing either 5% spray-dried bovine colostrum or no colostrum (control). Diets were formulated to contain 14.8 MJ/kg DE, 1.26% available lysine and to meet or exceed requirements for other nutrients. Piglets were offered the diets for a period of 14 days. No effect of diet on growth rate or feed intake was observed (p>0.10). Small intestine weight was reduced by 12% in piglets consuming dietary bovine colostrum (p< 0.05). Villous height and crypt depth were increased and decreased, respectively, in the proximal jejunum, mid jejunum and distal ileum of pigs consuming dietary bovine colostrum (p<0.05). Mid-jejunal lamina propria $CD4^+$ and $CD8^+$ T lymphocyte density was increased by 28 and 37%, respectively, in piglets consuming dietary bovine colostrum (p<0.05). Diet did not affect thickness of tunica muscularis externa or tunica submucosa (p>0.10). Collectively, these results suggest a positive effect of dietary bovine colostrum on intestinal morphology and immune status in early-weaned pigs.

감마선조사 한약의 방사선방호 효능 안정성 (Stability in Effects of ${\gamma}$-Irradiated Chinese Medicinal Prescriptions on Protection of Mice from Radiation)

  • 건조 농산물을 비롯한 식품에 대한 방사선 조사가 허가, 시행되고 있으며, 그 적용 범위도 확대되고 있다. 이에 따라 건조 생약재의 위생화의 대체방법으로 방사선 조사 기술의 적용이 산업계로부터 요구되고 있다. 본 연구는 감마선 조사 생약재의 효능 변화 유무를 확인하기 위한 일환으로 수행되었다. 저자 등은 사물탕, 보중익기탕 및 삼령백출산 등이 방사선 방호 효과를 보이는 것으로 이미 보고한 바 있다. 본 실험에서는 감마선 조사(10 kGy)한 이들 한약제 각각의 방사선 방호 효과를 비조사 시료와 비교 평가하였다. 효과평가를 위하여 방사선 조사 마우스에서 소장움 생존시험, 내재성 비장집락 형성시험 및 apoptosis 측정을 시행하였다. 각 한약제의 감마선 조사 시료는 3가지 시험 모두에서 비조사 시료와 유사한 효과를 나타내었으며(p<0.05), 생체내 독성도 나타내지 않았다. 이는 생약재의 여러 가지 고유 효능 중 일부의 안정성으르 확인한 것으로 생각되며, 이 결과로 미루어 보아 감마선 조사 생약재의 효능 안정성은 한의학적 고유의 효능 비교 평가를 통하여 확인될 수 있을 것으로 사료된다. 한편, 저자 등은 감마선 조사 생약재의 유효성분 안정성 및 유전 독성학적 안전성을 확인하여 보고한 바 있다. 이들 결과를 토대로 향후 감마선 조사 행약재의 고유 효능의 안정성에 관한 체계적인 연구결과를 얻는다면 생약재의 위생화 수단으로 감마선 조사기술의 이용이 실용화될 수 있을 것으로 사료된다.

  • 방사선장해(放射線障害) 예방약제(豫防藥劑)의 인체적용(人體適用)은 약제(藥劑)의 부작용(副作用)으로 아직 임상적(臨床的) 이용(利用)이 되지 못한 실정(實情)으로서 생리학적(生理學的) 및 약리학적(藥理學的) 독성(毒性), 물리학적(物理學的) 제성질(諸性質), 약제(藥劑)의 작용기전(作用機轉) 구명(究明) 및 임상적(臨床的) 기술개발(技術開發)이 미흡(未洽)하여 방사선(放射線) 장해자(障害者)에 대(對)한 치료(治療)는 대증료법(對症療法)에 불과(不過)한 실정(實情)이다. 저자(著者)들은 방사선(放射線)에 의(依)한 인체장해예방(人體障害豫防)에 필요(必要)한 기초자료(基礎資料)를 얻기 위하여 현재(現在)까지 비교적(比較的) 독성(毒性)이 적고 화학적(化學的)으로 안정(安定)하며 약효(藥效)가 우수(優秀)하다고 알려진 DDC, MEA, WR-2721을 선정(選定)하여 실험동물(實驗動物) 비근교계(非近交系) Swiss Webster(NIH-GP) 마우스에 이용(利用)한 방사선(放射線) 방호작용(防護作用)에 관(關)한 실험결과(實驗結果)는 다음과 같다. 1) 각종(各種) 방사선(放射線) 방호제(防護劑)(DDC, MEA, WR-2721)의 마우스에 대한 적정(適定) 투여량(投與量)은 각각(各各) DDC; 1,000, MEA; 200, WR-2721; 400 mg/kg 이었다. 2) 실험동물(實驗動物)의 방사선(放射線) 반치사선량(半致死線量) LD50/10에 대(對)한 약제(藥劑)의 방호효과(防護效果) DMF는 DDC; 1.2, MEA; 1.4, WR-2721; 1.9이었고 LD50/30에 대(對)한 DMF는 DDC; 1.7, MEA; 1.8, WR-2721; 2.5이었다. 3) 공장(空腸) 소낭선(小囊腺)(Jejunum crypt)의 방사선평균치사선량(放射線平均致死線量)(Do)에 대(對)한 방호효과(防護效果) DMF는 DDC: 1.07, MEA; 1.21, WR-2721; 1.76이었고, 소낭선세포(小囊腺細胞)의 방사선(放射線) 평균치사선량(平均致死線量)에 대(對)한DMF는 DDC; 1.04, MEA; 1.08, WR-2721; 1.38이었다. 4) 이상(以上)으로 상기(上記) 3종(種)의 방사선(放射線) 방호제중(放護劑中) WR-2721의 방사선(放射線) 방호효과(防護效果)가 가장 우수(優秀)하였고 이 결과(結果)는 향후(向後) 임상적(臨床的) 적용(適用)에 관(關)한 연구(硏究)의 기초자료(基礎資料)가 될 것으로 사료(思料)된다.

Growth performance, carcass traits and gut health of broiler chickens fed diets incorporated with single cell protein

  • Objective: This study was conducted to evaluate single cell protein (SCP), produced from Methylococcus species, as a protein source on the growth performance, carcass traits and gut health of broiler chickens. Methods: Ten iso-nitrogenous and iso-caloric diets containing 0 (Control), 2.5%, 5%, 7.5%, and 10% SCP replacing either soybean meal (T1 to T5) or fish meal (T6 to T10) were formulated. Each diet prepared for starter (0 to 14 days), grower (15 to 21 days), and finisher (22 to 42 days) phases was offered to four replicates of 10 chicks each (n = 400). Growth performance at different phases and carcass characteristics and intestinal morphology on 42nd day of trial were measured. Results: Body weight gain in groups fed 2.5% and 5% SCP diets were comparable to control during different phases and cumulatively, however lower (p<0.01) in 7.5% and 10% SCP diets. Feed conversion ratio was better (p<0.01) in 2.5% and 5% SCP diets. Dressing percentage, abdominal fat percentage and meat:bone ratio were not affected (p>0.05) by SCP inclusion in the diets. However, breast percentage was higher (p<0.01) in 2.5% and 5% SCP groups and thigh percentage higher in 7.5% and 10% SCP groups. Total microbial count in duodenum, jejunum and ileum were not affected (p>0.05) by SCP inclusion up to 10% in diets. Duodenal villi length and crypt depth were highest (p<0.01) in group fed 5% SCP diets and lowest in group fed 10% SCP diets. Jejunal villi length and crypt depth as well as ileal villi length were lowest (p<0.01) in group fed 10% SCP diets. Body weight gain, feed consumption, feed conversion ratio and gut health were better (p<0.01) in broilers fed fish meal based diets compared to soybean meal based diets. Conclusion: It was concluded that inclusion of SCP up to 5% replacing soybean meal in broiler diets is beneficial in improving growth rate, breast yield and gut health status.

Effects of essential oil coated with glycerol monolaurate on growth performance, intestinal morphology, and serum profiles in weaned piglets

  • Objective: This study aimed to investigate the effects of essential oil coated with glycerol monolaurate (GML) on the growth performance, intestinal morphology, and serum profiles of weaned piglets. Methods: A total of 144 weaned piglets (Duroc×[Landrace×Yorkshire], average weight 8.07±3.33 kg) were randomly assigned to three groups with six replicate pens and eight piglets per pen: i) CON: a corn-soybean basal diet; ii) LEG: with 1,000 mg/kg essential oil coated with GML; and iii) HEG: with 2,000 mg/kg essential oil coated with GML. Results: Results showed that average daily gain was increased (p<0.05) linearly by essential oil coated with GML supplementation on day 14 to 28 and day 0 to 28 compared with the CON group. Dietary supplementation with HEG increased (p<0.05) total antioxidant capacity and catalase activity on day 14, and immunoglobulin A (IgA) and IgM concentration on day 28 and tended to increase IgG on day 28. In addition, the crypt depth in the jejunum was reduced (p<0.05), and villus height and villus height/crypt depth in the ileum were increased (p<0.05) in the HEG group compared with the CON group. Moreover, lower (p<0.05) concentrations of tumor necrosis factor-α, interferon-γ, interleukin-1β (IL-1β), IL-8, and IL-10 were observed in the jejunum of piglets supplemented with HEG compared with the CON group. In addition, dietary HEG tended to decrease IL-6 level in the jejunum of piglets compared with the CON group. Conclusion: Dietary essential oil coated with GML can improve growth performance of weaned piglets. Moreover, supplementing 2,000 mg/kg essential oil coated with GML was demonstrated to improve antioxidant ability, and intestinal morphology, and reduce jejunal inflammatory factor levels.
