Isozyme Patterns at Five Loci Salmonids and Their Hybrid(I) (연어류에서의 종간교배 및 Isozyme Pattern에 관한 연구)

  • 연어류의 종간교배 실험과 LDH, MDH, IDH, $\alpha$GODH, ME 등 다섯가지 isozyme에 대한 genetic marker로서의 활용 가능성을 알아보고자 1차적으로 연어, 산천어 및 무지개송어를 이용하여 종간교배를 실시하였고 이들 세종의 isozyme pattern을 비교하였다. 교배실험은 종간교배, 및 allotriploid 구간 등 12구간으로 나누어 실시하였으며 연어 암컷과 산천어 수컷의 교배결과가 초기성장 단계적에서 가장 우수하게 나타났고이들의 allotriploid도 부활율이 28.1%로 가장 우수하였다. Genetic identification을 위한 isozyme loci 분석결과 연어와 산천어는 대부분의 loci에서 거의 찾아볼 수 없었고 무지개송어는 MDH-B와 IDH에서 다형현상을 확인할 수 있었다. 특히 MDH-B loci는 b 유전자의 출현 빈도에서 세 종간의 식별이 가능하였으며, IDH pattern을 산천어와 무지개송어의 비교에 유효한 것으로 나타났다. 이들 두 loci는 hybrid의 genotype 분석시 유용한 marker로 활용할 수 있는 가능성을 보였고, 앞으로의 어류 육종에 좋은 기초 자료로 이용될 것으로 사료된다.

  • Mutation rate of M$_2$ plants that were treated with three types of double treatments of chemical mutagens(1.5mol Na$N_2$ + 0.75mol MNH, 0.75mol MNH + 0.75mol MNH and 0.5mol MNH + 0.5mol MNH) were estimated on the rate of chlorophyll mutant, changes of isozyme loci ; esterase (Est), glutamate oxaloacetate transaminase(GOT ; AAT) and leucyl aminopeptydase(LAP ; AMP). Rate of chlorophyll mutants (3.3% =no. of seedling carrying mutant / all number of M$_2$ seedlings $\times$ 100) and rate of esterase isozyme loci mutants(3.5% =no. of plant carrying mutant / all number of M$_2$ plant) in Dema were higher than one of Sacheon 6, but no significant differences in GOT, LAP. Among isozymes, most of mutants in M$_2$ plant of two varieties were found in esterase (73% of total mutants were occurred in esterase loci). Although many of null bands were found in GOT 3, these were not repeatable and no real mutants. It might be due to qualities of starch, amount of extract buffer and degradation of isozyme during electrophoresis and staining.

  • Number of loci, allele frequencies, and subunit structures of 17 kinds of isozymes were analyzed in a laboratory strain of the beet armyworm, Spodoptera exigua (Hubner) to get genetic markers. These isozymes had 30 loci with 21 polymorphic (70.0% polymorphism); effective number of alleles per locus, average heterozygosity (H,), and inbreeding coefficient (F) were 2.52, 32.8%, and 2 1.0%, respectively.

Genetic Variations of Natural and Hatchery Populations of Korean Ayu (Plecoglossus altivelis) by Isozyme Markers

  • Genetic variability and population structure of 11 natural ayu, Plecoglossus altivelis populations and one hatchery stock were assessed by starch gel electrophoretic analysis with 10 enzyme coding loci. Three loci were polymorphic (lower than 0.95 in major allele frequency) in natural populations,2 loci in hatchery stock. The average number of alleles per locus was 1.38. Observed heterozygosities ranged from 0.0235 to 0.088 (0.055 on the average) in natural population while 0.0925 in hatchery stock. The genetic distance among natural populations measured 0.000047-0.005407 and no significant differentiation was observed among them. On the other hand, a signifcant genetic distance was found between natural populations and the hatchery stock with measuring 0.002032-0.O08605. The results in this study suggest that the hatchery stock has diverged from natural populations, and also that careful to maintain sustainable and effective population size (parents number) should be made.

Isozyme Analysis on the Allotriploid between Rainbow Trout (Oncorhynchus mykiss) and Coho Salmon (O. kisutch) (무지개송어와 은연어간 잡종3배체의 부화자어에 대한 동위효소 분석)

  • For the purpose of identification of inheritance in allotriploid between rainbow trout (Oncorhynchus mykiss) and coho salmon (O. kisutch), five isozymes, lactate dehydrogenase (LDH), malate dehydrogenase (MDH), isocitrate dehydrogenase (IDH), phosphoglucose isomerase (PGI) and phosphoglucomutase (PGM) from skeletal muscle in two species and their allotriploid were analyzed. All of these loci showed differences between two species and their allotriploid except PGI. Generally, coho salmon was more monomorphic in these isozyme loci than rainbow trout. Their allotriploids showed intermediate patterns between the parental species in those isozyme loci except PGI. As a result of this study, LDH, MDH, IDH and PSM may be used as useful genetic markers in these two species, and they also be of use in studying hybrid and allotriploid in salmonids.

  • Genetic characterization and identification of two types of sea bass Lateolabrax japonicus were performed by examining electrophoretic patterns of isozymes. Twenty five loci coding for thirteen enzymes were detected in two types. Among the twenty five loci, one completely divergent loci (Pt-1) was observed between two types. Nei's genetic distance between two types was 0.12036. The estimated divergence time of these two types may have about $6.2{\times}10^5$ years ago. On the other hands, the expected average heterozygosity was 0.084 in not spotted type on the body surface and 0.067 in spotted type on the body surface. These results mean that the existance of two types of sea bass was established in present study which may have had genetic divergence.

  • A horizontal starch gel electrophoresis for enzyme proteins extracted from 2 species of Korean viviparid snails; Cipangopaludina chinensis malleata and C. japonica was carried out in order to elucidate their genetic relationships. A total of 10 enzymes were employed in three different kinds of buffer systems. Two loci from each enzyme of alcohol dehydrogenase, esterase, glucose phosphate isomerase, isocitrate dehydrogenase, iditol dehydrogenase, malate dehydrogenase and peptidase(VL); and only one locus dach from two enzymes, glycerlo-3-phosphate dehydrogenase and phosphoglucomutase were detected; but, four loci from peptidase(LGG) were observed. Most of loci in two viviparid species showed homozygous monomorphic banding patterns and some of them were specific as genetic markers between two different species. However, EST-1, MDH-1, PEP(VL)-1loci showed polymorphic banding patterns. Foru populations of C. chinensis malleata were more closely clustered in a dendrogram within the range of genetic identity values of 0.928-1.00, and these clusters were lineated with C. japonica at the value of 0.355. In summarizing the above results, two viviparid snail species dmployed in this study mostly showed monomorphic enzyme protein banding patterns, and genetic differences specific between two species.

  • For the purpose of genetic stock indentification of three species of salmonid fishs and their hybrid, lactate dehydrogenase(LDH), malate dehydrogenase(MDH), isocitrate dehydrogenase(IDH), a-gylycerophosphate dehydrogenase(a-GPDH), malic enzyme(ME), 6-phospho-gluconate dehydrogenase(6-PGD), phosphoglucose isomerase(PGI) and phospho-glucomutase(PGM) from skeletal muscle, liver, heart and gill tissues in all three species were analyzed. Chum and masu salmon showed no polymorphic patterns in all isozyme loci, however rainbow trout were found to have polymorphic patterns at MDH-B, LDH and IDH loci. Especially, significant differences were found at MDH-B loci between the three species and the IDH patterns of rainbow trout were also different from the other two species. These loci therefore can be utilized as efficient genetic markers for the identification of hybrids and improve the efficiency of fish breeding. There was no difference except PGI between diploid and triploid isozyme patterns but PGI showed some potential as a marker for triploid in masu salmon.

  • Megagametophyte tissues from seeds of 45 loblolly pine clones were subjected to horizontal starch-gel electrophoresis. The resulting gels were tested for activity of five enzyme systems (GOT, SDH, PGM, MDH, and AP). Isozymes observed were under control of 11 loci. All 45 clones could be identified with unique genotypes at above 10 loci.

  • Previously, genetic relationships among 321 silkworm races preserved in the Department of Sericulture and Entomology, NIAST, RDA were studied using six isozymes comprising 20 loci. As a part of additional studies, three additional isozymes with 7 loci [phos-phoglucomutase (PGM), glucose phosphate isomerase (GPI), malete dehydrogenase (MDH)] from hemolymphs, midgets, and eggs were employed to reevaluate the previous dendrogram (UPGMA method). Although the current study supported the recognition of the previously identified major groups, some minor changes were apparent among the selected 47 forms. The current study showed that the polymorphic loci of GPI from eggs and MDH from hemolymph appear to be responsible characters for generating a new major group in the genetic relationship. Interpreting the data with current additive isozymes may represent more robust genetic relatioships among 321 silkworm races preserved in Korea, until further evidence is available.
