The pharmacological actions to methanol extract(PME) obtained from Plantaginis semen were examined in the rabbit. 1) PME, when administered into the vein of rabbit, produced the fall of blood pressure and stimulation of respiration. The former action was inhibited by atropine but the latter not affected by atropine. 2) PME caused contraction in both isolated intestinal and uterus strips, atropine blocked the contraction of intestinal strips while did not the uterus contraction. 3) PME decreased the heart rate of rabbit anesthetized with urethane. 4) PME elicited antidiuresis with doses ranging from 10mg/kg. The antidiuresis appeared to be related to the hemodynamic changes decreases in the renal plasma flow and glomerular filtration rate. Urinary sodium and potassium decrease in the renal plasma flow and glomerular filtration rate. Urinary sodium and potassium decrease in relation to the diminished filtration.
The cause of infertility is either fertilization failure or early embryonic death. The aetiology may involve a combination of many factors, e.g. genetic factors, abnormalities in the gametes nutritional disorders, inadequate luteal function, and delayed ovulation. This study was conducted to investigate the effect of microorganisms collected from uterus of Hanwoo cattle on early embryonic development. Microorganisms isolated from the uterus of Hanwoo cattle included Bacillus cereus (Bc), Bacillus thuringiensis (Bt), Staphylococcus warneri (Sw) and Enterococcus faecalis (Ef). When cultured with Bc, Bt, Sw, and Ef, oocytes were not developed into a blastocyst in vitro. The proportion of blastocyst was dramatically increased after reducing the number of microorganisms ($10^4CFU/ml$). Interestingly, the proportion of blastocyst was decreased by adding the Sw and Ef. These results indicate that among intrauterine microorganisms, Sw and Ef strains negatively affect theearly embryonic development, thereby aggravate the rates of implantation and pregnancy. These findings will provide useful information for association studies in other pig populations.

Bovine brucellosis is a zoonosis, long incubation period and chronic infectious disease, usually caused by Brucella abortus. This study was carried out to investigate the biotyping and biochemical characterization of B. abortus isolated from 208 farm 871 korean cattle and holstein diagnosed brucellosis by serological positive in Gyeongbuk province during the period from 2002 to 2006. B. abortus was isolated from 124 (14.2%) of 871 cattle, and isolated 110 (13.4%) of 820 Korean cattle and 14 (27.5%) of 51 holstein in breed. The uterus of korean cattle was isolated in 8 (17.8%) of 45 cattle and supramammary lymph none of holstein was isolated 11 (68.8%) of 16 cattle. 101 (12.5%) of 810 serological positive blood samples were isolated B. abortus. The isolation rate of B. abortus was correlated with antibody titers. The biochemical characterization of isolates was non-hemolytic, production of H$_2$S, oxidase-positive, catalase-positive, hydrolyzation of urea and growth of basic fuchsin dye medium. As a result, all of isolates was identified B. abortus bv 1. 124 isolates were susceptible to ampicillin, lincospectin, amikacin, gentamicin, kanamycin, neomycin, streptomycin, tetracycline, ciprofloxacin, norfloxacin and enrofloxacin.
In an attempt to investigate the effect of Beekeumsan on the hemostatic action and isolated uterine muscle, Beekeumsan was administered orally and the bleeding time in mouse tail, prothrombin time in vitro were estimated. Its activity on the isolated uterine muscle in rats were investigated. The following results were obtained; The bleeding time was not shortened, but the plasma prothrombin time in vitro was significantly shortened. The uterotonic action produced by the Beekeumsan was not inhibited by pretreatment of atropine, but was slightly inhibited by cyproheptadine and completely inhibited by pretreatment of diltiazem.
Thirty-one medicinal plants in Korea are screened for anticholinergic and oxytocic activity The extracts of Cyperus rotundus L. and Scutellaria baicalensis showed anticholinergic action and other seven kinds of extracts showed weak action. The oxytocic action is observed with Tarraxacum platycarpium and Albizzia julibrissin $D_{URZ}$.
The general and some pharmacological actions of DWP 305 were investigated in animals and the following results were obtained. In central nervous system, DWP 305 had no effects on the pentobarbital induced anaesthesia, locomotor activity, rotarod test, traction test, analgesic action in mice and body temperature in rat. DWP 305 showed no depressive action on convulsion induced by strychnine, electronic shock and pentylenetetrazole. From these results, DWP 305 was considered to have no pharmacological effect on the central nervous system. Furthermore, DWP 305 had no influences on the normal blood pressure and heart rate. In the isolated ileum of guinea pig, DWP 305 inhibited contractive effects against the acetylcholine (10$^{-6}$ g/mι), histamine (10$^{-6}$ g/mι), 5-hydroxytryptamine (10$^{-6}$ g/mι) and BaCl$_2$(10$^{-4}$ g/mι) at a concentration of 2.15$\times$10$^{-4}$ g/ml in bath. In the isolated trachea and vats deference, DWP 305 showed no effect on the contractions produced by histamine and norepinephrine, respectively. DWP 305 showed inhibitory effect on the contractions produced by acetylcholine and oxytocin at a concentration of 2.15$\times$10$^{-4}$ g/ml on the isolated nonpregnant rat uterus. DWP 305 had no effect on the isolated right atrium of guinea pig, bile excretion, urine volume, pH, gastrointestinal motility, gastric secretion and blood aggregation.
$K^+$ channel openers (KCOs) are known to have a wide range of effects by opening the $K^+$ channel in plasma membranes of various smooth muscles, cardiac muscle and pancreatic ${\beta}-cell$. In the present study, we investigated the effects of 5 types of KCOs, cromakalim, RP49356, pinacidil, nicorandil and diazoxide on the contractility of isolated rat uterus. All KCOs tested inhibited the uterine contraction induced by 0.2 nM oxytocin in a dose-dependent manner. Individual KCO and its $pD_2$ values were cromakalim 6.5, RP49356 6.3, pinacidil 5.92, nicorandil 4.43 and diazoxide 4.18. The relaxant effects of KCO were inhibited by glibenclamide (0.3, 1 and $10\;{\mu}M$) with $pA_2$ values of cromakalim 6.91, RP49356 6.59, pinacidil 6.55, nicorandil 5.97 and diazoxide 6.37. In addition, the relaxant effect of cromakalim or pinacidil was antagonised by TEA, a non-selective $K^+$ channel blocker, but not by apamin. Contractions induced by low concentration of KCI (< 40 mM) were inhibited by cromakalim $(100{\mu}M)$ and nicorandil $(300{\mu}M)$, but those evoked by higher concentration (> 40 mM) of KCI were little affected. In ovariectomized rat uterus, cromakalim dose-dependently inhibited oxytocin-induced contraction and glibenclamide $(10{\mu}M)$ inhibited the relaxant effect of cromakalim with $pD_2$ and $K_B$ values of 7.48 and $1.26{\times}10^{-7}M$, respectively. In estrogen-primed rat uterus, these values were 6.51 and $1.57{\times}10^{-7}M$, respectively, indicating that the cromakalim is less effective on the estrogen-treated uterine smooth muscle. Our results suggest that the KCO-sensitive $K^+$ channels participate in the motility of uterine smooth muscle and such channels are, at least in part, under the control of estrogen. In addition, our data Indicate that the type of $K^+$ channels activated by KCO is ATP-sensitive $K^+$ channels which is blocked by glibenclamide.
Pivotal roles of steroid hormones in uterine endometrial function are well established from the mouse models carrying the null mutation of their receptors. Literally androgen belongs to male but interestingly it also detected in female. The fluctuations of androgen levels are observed during reproductive cycle and pregnancy, and the functional androgen receptor is expressed in reproductive organs including uterus. Using high throughput methodology, the downstream genes of androgen have been isolated and revealed correlations between other steroid hormones. In androgen-deficient mice, uterine responses to exogenous gonadotropins are impaired and the number of pups per litter is reduced dramatically. As expected androgen has important role in decidual differentiation through AR. It regulates specific gene network during those cellular responses. Recently we examined the effects of steroid hormonal complex containing high level of androgen. Interestingly, on the contrary to the androgen-alone administration, the hormonal complex did not disturb the decidual reaction and the pubs did not show any morphological abnormality. It is suspected that the complexity of communication between other steroid hormone and their receptors are the reasons. In summary, androgen exists in female blood and it suggests the importance of androgen in female reproduction. However, the complex interactions with other hormones are not fully understood compared with estrogen and progesterone. The further studies to evaluate the possible role of androgen are needed and important to provide the in vivo rational for the prevention of associated pregnancy complications and help human's health.

An in vitro assay following culture of endometrial epithelial cells is essential for understanding epithelial cell function in reproduction. Several diverse techniques have been developed for isolating endometrial epithelial cells, although an optimal technique has not been identified. In this study, we describe a sedimentation-adherence (S-A) isolation technique with a high-yield cell-separating ability to isolate endometrial epithelial cells from 8-week-old female C57BL/6 mice. We analyzed total cell number, viability, morphology, and expression of cytokeratin 18 as an endometrial epithelial cell-specific marker in cells isolated using a mechanical method compared to the S-A technique. There were no significant differences in the total number, viability, or morphology of the putative endometrial epithelial cells with either method. In contrast, significantly more endometrial epithelial cells harvested using the S-A method were positively stained for cytokeratin 18 than those isolated using the mechanical method. These results confirm that the S-A method is more efficient for retrieving endometrial epithelial cells than a mechanical method.
Aspalatone [3-(2-methyl-4proponyl)]-2-acetyloxybenzoate, CAS 147249-33-0) is a compound having an antithrombotic action. General pharmacological properties of aspalatone were studied. Aspalatone had no effect on central nervous system and no anticonvulsant effect up to 1200 mg/kg p.o. However, the compound has hypothermic and analgesic effect. When administered intravenously in rabbits, aspalatone did not affect blood pressure, heat rate and respiration rate and depth, and it did not inhibit transient hypotensive effect of acetylcholine. The compound did not affect isolated guinea-pig ileum and tracheal strip at a concentration of 1${\times}$$10^{-4}$, and did not inhibit histamine-induced contraction of guinea-pig ileum. It also did not affect isolated rat stomach fundus and estrogenated rat uterus at 1${\times}$$10^{-4}$, and did not inhibit contraction produced by acetylcholine or oxytocin. The pupil size and intestinal propulsion were not influenced at a large dose of was shown. The compound showed a slight increase in urine volume and led to decreased excretion of potassium in urine of rats. The results suggest that aspalatone may have no considerable adverse effects in general pharmacological aspect.
