Cloning and Expression of a Serine Proteinase Gene Fragment from Acanthamoeba culbertsoni

  • Serine proteinase cDNA fragment from protozoan parasite Acanthamoeba culbertsoni was amplified by the reverse transcription-polymerase chain reaction (RTPCR) using degenerate oligonucleotide primers derived from conserved serine proteinase sequences. The amplified DNA fragment was subcloned and sequenced. The sequence analysis and alignment showed significant sequence similarity to other eukaryotic serine proteinases and conservation of the His, Asp, and Ser residues that form the catalytic triad. The cDNA fragment was cloned into the pGEMEX-1 expression vector and expressed in Escherichia coli. A resulting fusion protein of 56 kDa had proteolytic activity. The fusion protein reacted with sera of mice immunized with purified serine proteinase of A. culbertsoni in Western blot. Immune recognition of the fusion protein by mouse antisera suggested that the fusion protein may be valuable as a diagnostic reagent.

  • The HANARO reactor has a vertical hole for a cold neutron source (CNS) in the heavy-water reflector tank, i.e., the CNS hole, which was considerably deformed during its welding to the horizontal cold neutron (CN) beam tube. This paper presents an investigation of the form of the CNS hole for the optimal design of the a vacuum chamber for the CNS. In addition, the installation method of the vacuum chamber into the CNS hole for minimizing the water thickness between the vacuum chamber and the nose of the CN beam tube is proposed.

Effect of fractional order on energy ratios at the boundary surface of elastic-piezothermoelastic media

  • In the present investigation reflection and transmission of plane waves at an elastic half space and piezothermoelastic solid half space with fractional order derivative is discussed. The piezothermoelastic solid half space is assumed to have 6 mm type symmetry and assumed to be loaded with an elastic half space. It is found that the amplitude ratios of various reflected and refracted waves are functions of angle of incidence, frequency of incident wave and are influenced by the piezothermoelastic properties of media. The expressions of amplitude ratios and energy ratios are obtained in closed form. The energy ratios are computed numerically using amplitude ratios for a particular model of graphite and Cadmium Selenide (CdSe). The variations of energy ratios with angle of incidence are shown graphically. The conservation of energy across the interface is verified. Some cases of interest are also deduced from the present investigation.

  • The diagrid structural system is considered to be not only the best structural system for constructing free form structures, but also a very effective system in resisting lateral load. As a newly investigated structural system, its complicated node has not yet been completely investigated and minimal experimentation of manufacturing and constructing the system have been conducted. Therefore, the constructing cost of the diagrid structural system is still comparatively high. In this paper, the cyclic performance of a diagrid node with an H-section brace will be discussed. Design details that consider productivity were proposed and their structural performances were assessed through experimental and analytical investigation.

  • Experimental results for pressure gradient of HCs refrigerants R-290. R-600a. R-1270 and HCFC refrigerant R-22 during condensing inside horizontal double pipe heat exchangers are presented. The test sections which have the tube inner diameter of 10.98mm. and the tube inner diameter of 8mm are used for this investigation. Hydrocarbon refrigerants have higher pressure drop than R-22 in both test sections with the diameters of 12.70mm and 9.52mm. Pressure drop increased with the increase of the mass flux. These results form the investigation can be used in the design of heat transfer exchanger using hydrocarbons as the refrigerant for the air-conditioning systems

  • This work is devoted to investigate heat and mass transfer effects on MHD natural convection flow past an inclined plate with ramped temperature numerically. The dimensionless governing equations for this investigation are solved by using finite element method. The effects of angle inclination, buoyancy ratio parameter, permeability parameter, magnetic parameter, Prandtl number, heat generation, thermal radiation, Eckert number, Schmidt number, chemical reaction parameter and time on velocity, temperature and concentration fields are studied and presented with the aid of figures. The effects of the pertinent parameters on skin friction, rate of heat transfer and mass transfer coefficients are presented in tabular form. The numerical results are compared graphically with previously published result as special case of the present investigation and results found to be in good agreement.

  • The present article is aimed at an investigation of stresses produced in a microstretch elastic half-space due to a moving load. The expressions of normal stress, shear stress and tangential couple stress produced in this case have been obtained in closed form. To find the displacement fields the perturbation method is applied. Significant effect of moving load on variation of stresses developed at different depths below the surface due to the depth of substrate and frictional coefficient of the rough surface of the medium has been observed. The effects of different shapes of irregularity and depth of irregularity on normal, shear and tangential couple stresses have been discussed. Some particular cases have also been deduced from the present investigation. Finally, the analytical developments have been illustrated numerically for aluminium-epoxy-like material substrate under the action of moving load.

  • The purposes of this investigation was to determine the appearance rate of nutrient canals and the aspect of alveolar bone trabecular patterns. 1949 subjects, averages 23 years old, who have normal occlusion were used. The film used in this investigation was full mouth roentgenographic survey. The obtained results were as follows: 1. In the trabecular pattern, coarse form was 35.17% and fine one was 64.14% in both jaws. 2. The appearance rate of nutrient canals was much higher in mandibular anterior segment(89.13%) than in the maxillary posterior segment(0.86%). 3. In the trabecular forms, small trabecular space and horizontal relation appeared in the mandible, and vertical relation in the maxilla.

지체부자유아동을 위한 주거용 의자설계에 관한 연구 (Resicential Chair Design for Physically Handicapped Children)

  • The purpose of this study is the investigation of the physically handicapped childrens' residentical life, using furniture and opinion of auxiliary furniture ot find out alternative forms of desirable furniture. This study consists of two parts: a survey of residential life and furniture usage and a new chair design based on the survey. Documentary research, observational investigation and questionaire survey methods were used. According to the survey, a chair was chosen as the most needed piece of furniture. The alternative chair was designed to accomodate to the individual physical needs of each child. The structural charasteristics of the chair form consist of four parts: 1) basic seat 2) supporting metal frame 3) detachable chair legs 4) detachable table top. Auxilliary options include adjustable parts such as a head rest, seat belt, arm rest and pedestal.

Stress state around cylindrical cavities in transversally isotropic rock mass

  • The present paper is dealing with the investigation of the stress field around the infinitely long cylindrical cavity, of a circular cross section, contained in the transversally isotropic elastic continuum. Investigation is based upon the determination of the stress function that satisfies the biharmonic equation, for the given boundary conditions and for rotationaly symmetric loading. The solution of the partial differential equation of the problem is given in the form of infinite series of Bessel's functions. Determination of the stress-strain field around cavities is a common requirement for estimation of safety of underground rock excavations.
