Protective Effects of BK-1202 on the Indomethacin-induced Gastric Ulcer in Rats

  • Purpose: The object of this study is to observe the anti-ulcerative effects of BK-1202 (IGM), a mixed herbal formula consisting of 9 herbal drugs, which have been traditional Korean medicine for treating various digestive diseases, on indomethacin-induced gastric ulcer in rat. Methods: Three different doses of IGM extract (200, 100 and 50 mg/kg) were orally administered once 30 min before indomethacin treatment. Six hours after indomethacin treatment, changes in the gross lesion scores, fundic histopathology, MPO activity and antioxidant activities were observed. The results were compared with two reference groups treated with omeprazole (10 mg/kg), antioxidant and proton pump inhibitor, and DA-9601 (100 mg/kg), a standardized extract of the herb Artemisiaasiatica. Results: In all three doses of IGM extract, significantly decreased gastric damages were observed in the indomethacin-induced gastric ulcer rats, when compared with the indomethacin-treated control rats. IGM extracts also strengthened the antioxidative defense systems, decreasing the level of lipid peroxidation and catalase activity while increasing the superoxide dismutase and glutathione contents. IGM extracts showed similar anti-ulcerative effects to those shown by equal dose of DA-9601, and the effects of 50 mg/kg IGM extracts were comparable to those of 10 mg/kg omeprazole. Conclusion: The results obtained in this study suggest that IGM extract has favorable effects on the indomethacin -induced gastric damages by strengthening the antioxidative defense systems and enhancing anti-inflammatory effects.

Prevention effect of Sunkiwhajung-tang, a prescription, on the gastric ulcer induced by indomethacin in rats (순기화중탕이 Indomethacin으로 유발된 위궤양에 미치는 영향)

  • In order to evaluate the prevention effect of Sunkiwhajung-tang (SWT) which has been used as a traditional prescription for the treatment of digestive disease in Korea on the gastric ulcer induced by indomethacin in rats, the changes of number and size of ulcerative lesions, parietal, chief, Grimelius and Serotonin-positive cells in the peri-ulcerative tissues were detected with histological examinations of ulcerative and peri-ulcerative lesions after oral injections of SWT extracts (125, 250 and 500 mg/kg, respectively). SWT prevented to a great extent the expected indomethacin-induced elevation in hemorrhagic ulcerative lesions, the number and size of ulcerative lesions, and the number of parietal cell, chief cell, Grimelius-positive cells and Serotonin-positive cells in the peri-ulcerative lesions in a dose dependent manner. These results provide a story evidence that SWT produced an protective effect on gastric ulcer induced by indomethacin. Determination of the specific mechanisms involved in the protective effect of SWT on the gastric ulcer will require additional study.

Studies on the effect of Kamikuibitang on the Gastric Ulcer in Rats (가미귀비탕(加味歸脾湯)이 흰쥐의 위궤양(胃潰瘍)에 미치는 영향(影響))

  • This study was aimed to evaluate the anti-pain effect of Kamikuibitang in acetic acid method and the anti-ulceration effect of Kamikuibitang in indomethacin, aspirin and immobilization stress method in rats. The results were follows; 1. The anti-pain effects of Kuibitang and Kamikuibitang were decreased compared with those of control group. 2. In indomethacin and aspirin method, the anti-ulcerative effects of experimental groups were shown compared with those of control group. 3. In immobilization stress method, the anti-ulcerative effect of experimental groups was significantly shown compared with that of control group. 4. The serum gastrin levels of Kuibitang groups showed very significant decrease in indomethacin-induced and immobilization stress-induced ulcers. The serum gastrin levels of Kamikuibitang groups showed very significant decrease in indomethacin-induced, aspirin- induced and immobilization stress-induced ulcers. 5. The serum $V_{B12}$ levels of Kuibitang groups showed very significant increase in both indomethacin-induced and immobilization stress-induced ulcers. The serum $V_{B12}$ levels of Kamikuibitang groups showed significant increase in aspirin-induced and immobilization stress-induced ulcers whereas very significant increase in indomethacin-induced ulcer. According to the above results, it was concluded that Kamikuibitang had very significant anti-ulceration effect as well as anti-pain effect on gastric ulcer in rats.

Curative Effect of Selenium Against Indomethacin-Induced Gastric Ulcers in Rats

  • Indomethacin is a nonsteroid anti-inflammatory agent that is known to induce severe gastric mucosal lesions. In this study, we investigated the effect of selenium on gastric mucosal lesions in rats. To confirm the curative effect of selenium against indomethacin-induced gastric ulcers, gastric ulcers were induced by oral administration of 25 mg/kg indomethacin, and then different doses (10, 50, and 100 ${\mu}g$/kg of body weight) of selenium or vehicle were treated by oral gavage for 3 days. Oral administration of indomethacin clearly increased the gastric ulcer area in the stomach, whereas selenium applied for 3 days significantly decreased the gastric ulcer area in a dose-dependent manner. In addition, selenium markedly reduced the increase of lipid peroxidation induced by indomethacin in the gastric mucosa and increased activities of radical scavenging enzymes such as superoxide dismutase, catalase, and glutathione peroxidase in a dose-dependent manner. These results reveal that selenium can heal indomethacin-induced gastric ulcers through elimination of the lipid peroxides and activation of radical scavenging enzymes.

Treatment Effect of Jecksodoodangkuisan against Colonic Mucosal Ulcer Induced by Indomethacin in Mouse (Indomethacin으로 유발된 생쥐의 대장점막 손상에 대한 적소두당귀산(赤小豆當歸散)의 치료효과)

  • Objectives: This study was carried out to investigate the effects of Jecksodoodangkuisan(JD) and Jecksodoodangkuisankahyunjicho(JH) against Colonic Mucosal Ulcer induced by indomethacin in mouse. Methods: The normal group consisted of mice that were not inflammation-induced. The control group was untreated gastro-inflammation-elicited mice. The sample group mice were those administered BO, JD and JH after gastro-inflammation elicitation. Results: In the immunohistochemical change, the distribution of COX-1 in mice treated with BO, JD and JH noticeably increased than control group(P<0.05). The distributions of $NF-{\kappa}B$, p50, COX-2, ICAM-1, HSP70 and Substance P in mice treated with BO, JD and JH noticeably decreased more than control group(P<0.05). And JH was most effective treatment against Colonic Mucosal Ulcer in the immunohistochemical change. Conclusions: According to the above results, Jecksodoodangkuisan and Jeckmdoodangkuisankahyunjicho are applicable treatments for Colonic Mucosal Ulcer.

The Effects of Taheebo Extracts on Gastric Secretion and Gastric Injury in Rats (타히보 추출물의 흰쥐 위액 분비 및 위 손상에 미치는 영향)

  • The effects of methanol and water extracts of Taheebo were investigated on gastric secretion, gastric lesion and ulcer in rate. Experimental gastric lesion and ulcer was produced in rats using the following methods : HCl.aspirin-induced lesion, HCl.ethanol-induced lesion, indomethacin-induced ulcer and water-immersion stress ulcer model. In addition, the amount of gastric secretion in pylorus-ligated rats for 4 hours was determined. Water extracts of Taheebo significantly inhibited HCl.aspirin-induced gastric lesion at 1,000mg/kg, po in rats. Likewise, Water extracts of Taheebo caused significant inhibition of indomethacin-induced ulcer at oral dose of 1,000mg/kg. The lesion induced by HCl.ethanol was significantly reduced by both water and methanol extracts of Taheebo. It also showed significant antiulcer activity in water-immersion stress ulcer, respectively. In gastric secretion experiments, methanol extracts of Taheebo also showed significant inhibition of gastric juice secretion, acidity and acid output at doses 500 and 1,000mg/kg. These results may suggest that Taheebo shows antigastritic and antiulcerative action in rats in part by the inhibition of gastric juice secretion and acidity.

Studies on the Efficacy of Ganoderma lucidum in Digestive System (영지(靈芝)버섯(Ganoderma lucidum)의 소화기계(消化器系)에 대한 약효연구(藥硏究))

  • This study was attempted to investigate the effect of Ganoderma lucidum extract on digestive system in experimental animals. Ganoderma lucidum water extract (GWE) was found to be promoted the charcol transport rate in the small intestine of mice. GWE exhibited the augmentation of spontaneus movement(motility) and contractile response(tension) in the ileum and colon strips of rabbit, and these action were inhibited by atropine. GWE given intraduodenaly(i.d.) exhibited the significant increase of gastric acid secretion in pylorus-ligated rats. GWE inhibited the formation of some experimental gastric ulcers(pylorus ligation ulcer i.d., indomethacin-induced ulcer p.o., i.d. and aspirin-induced ulcer p.o.) in rats, which are considered to relate to a protective action. GWE and EtOH extract(water soluble phase) were remarkably increase of bile excretion, when administration of i.d., intravenation(i.v.) and per os (p.o.) compared with normal-control group. GWE was observed antibacterial activity aginst several intestinal microoganisms and others bacteria in vitro test.

The Effects of Jiyu-tang against Gastric and Ileac Mucosal Ulcer Induced by Indomethacin in Mouse (Indomethacin으로 유발된 생쥐의 위와 회장 점막 염증에 미치는 지유탕(地楡湯)의 효과)

  • Objectives : This study was carried out to investigate the effect of Jiyu-tang on gastric and ileac mucosal ulcer induced by indomethacin in mouse. Methods : The normal group was that no inflammation elicited mouse. Control group is that water administered mouse after gastro-inflammation elicitation. Sample group is that Jiyu-tang administered mouse after gastro-inflammation elicitation. Results : In the common morphology and histochemical change, control group were observed various injury by hemorrhagic erosion, while sample group was noticeably decreased than control group. In the immunohistochemical change, the distributions of COX-1 treated with Jiyu-tang noticeably increased than control group(p<0.05). The distributions of COX-2, MAC 387 and HSP-70, PCNA and TUNEL treated with Jiyu-tang noticeably decreased than control group(p<0.05). Conclusions : According to the above results, it is supposed that Jiyu-tang is applicable to gastric and ileac mucosal ulcer.

Inhibition of Experimental Gastric Ulcer by Potato Tubers and the Starch

  • In an attempt to establish anti-ulcerogenic activity of potato tubers, inhibitory activity against ethanol- and indomethacin-induced gastric ulcer models in rats was evaluated for the first time. From several varieties of potato tubers including Solanum tuberosum L. cv. Superior (white skin and fresh potato) and two new varieties of (Bora valley and Gogu valley), raw potato juice was prepared and the starch was obtained from each juice by filtration and drying. Upon oral administration to rats, raw potato juice showed more or less inhibitory activity. The starch showed higher and dose-dependent inhibitory activity, suggesting that the active ingredient in raw potato juice may be the starch. Particularly, the starch obtained from the tubers of new potato variety, "Bora valley," with purple color, showed the highest inhibitory activity (62.4% and 37.1% inhibition of ulcer index at 500 mg/kg), while omeprazole (proton pump inhibitor) used as a reference drug showed 74.4% and 75.7% inhibition at 20 mg/kg against ethanol- and indomethacin-induced ulcer formation, respectively. The present study provides a first evidence of anti-ulcerogenicity of raw potato juice and the starch. Especially, the starch from "Bora valley" strongly inhibited ulcer formation in rats. Considering that these are food components, they may be safely used for anti-ulcerogenic nutraceuticals.

  • This study was attempted to investigate the effect of Machili Cortex extract(Machilus thunbergii Sieb. et Zucc. ) on digestive system in experimental animals. EtOH and MeOH extracts(E.E. and M.E.) were found to inhibit the charcoal transport rate in the small intestine of mice. E.E. exhibited the inhibition of spontaneous movement(motility) and tension in the ileum and colon strips of rabbit, and these actions were inhibited by action of acetylcholine. E.E. and M.E. given intraduodenaly(i.d.) exhibitied the significant decrease of gastric acid secretion in pylorus-ligated rats. E.E. and M.E. inhibited the formation of some experimental gastric ulcers(pylorus ligation-ulcer i.d., indomethacin-induced ulcer p.o. and aspirin-induced ulcer p.o. ) in rats, which are considered to relate to a protective action. E.E. and M.E. caused remarkable increase of bile excretion, compared with normal-control group, when adminstered through i.d., i.v. and p.o. The antibacterial activity against several intestinal microorganisms and other bacteria in vitro test was observed in the administration of E.E. and M.E.
