Effect of chloroform extract of traditional Dicranopteris linearis leaves against paracetamol- and CCl4-induced liver toxicity in rats

  • The present study aimed to determine the hepatoprotective activity of the chloroform extract of D. linearis leaves (CEDL) using the paracetamol (PCM)- and carbon tetrachloride ($CCl_4$)-induced liver injury models in rats. The rats received $dH_2O$ (negative control), 200 mg/kg of silymarin (positive control) or CEDL (50, 250 and 500 mg/kg) orally once daily for 7 days and then were subjected to the hepatotoxic induction on the $7^{th}$ day. The samples (i.e. blood and liver) were collected and underwent biochemical and microscopical analysis, respectively. From the data obtained, both inducers caused significant (p < 0.05) increase in the levels of AST and ALT when compared to the control group, which were significantly (p < 0.05) reduced by CEDL in a generally dose-dependent manner. These biochemical findings were supported by the histopathological analysis and histological scoring. In conclusion, CEDL possesses potential hepatoprotective activity, which could be associated with its flavonoid and tannin contents with the mechanisms of hepatoprotection linked to either its antioxidant or anti-inflammtory/immunomodulating activities. Further in-depth studies are required to identify the responsible bioactive compound.

The Immunomodulating Effects of the Supplementation of Paeonia Japonica Extracts in Mice

  • Paeonia japonica var. pilosa $N_{AKAI}$, (PJ; Baek-Jak-Yak) is a medicinal plant which has been widely used as a component or blood-building decoctions. This study was performed to investigate the immunomodulative effects of PJ in mice, using in vitro and in vivo experiments. The immunomodulative effects were studied in vitro by determining the proliferation or mice splenocytes and the production of three kinds of cytokines (IL-1$\beta$, IL-6, TNF-$\alpha$) by mire peritoneal macrophages which were cultured with sequential fractions of PJ methanol extract (methanol, hexane, chloroform, ethylacetate, butanol and water). In an in vivo experiment using mice, different concentrations of PJ water extract were orally administrated every other day for two weeks. The production of cytokines (IL-1$\beta$, IL-6, TNF-$\alpha$) secreted by activated macrophages, and the proliferation of mice splenocytes, were used as indices for immunocompetence. In vitro supplementation using a hexane fraction of PJ in the range of 1 to 100 $\mu$ g/ml enhanced splenocyte proliferation by 1.8 to 12%, and by 10-15% using an aqueous fraction, compared to the control. IL-l$\beta$ production was significantly increased with the supplementation of butanol, hexane and water extracts of PJ Higher levels of IL-6 production were detected with supplementation of chloroform or water extracts. However, there were no significant differences in the production of TNF-$\alpha$ among the treated groups and the control. From the in vivo study, the highest proliferation of splenocytes was seen in the mice orally administrated with the PJ water extract at the concentration of 500 mg/kg body weight. In the case of cytosine production, IL-1-$\beta$, IL-6, and TNF-$\alpha$ released by activated peritoneal macrophages were augmented by the oral administration of a PJ water extract. These results indicate that Pl may enhance the immune function by regulating splenocyte proliferation and cytokine production capacity in mice.

황련감초 하태독법과 프로바이오틱스의 대장점막 내 면역조절 효과 비교연구 (A Comparative Study of Hataedock versus Probiotics on Immunomodulating Effect in Intestinal Mucosa)

  • Objectives Hataedock is a treatment that dispels toxic heat and meconium which has been accumulated to the fetus from a pregnant mother via orally administering herbal extracts to a newborn baby. This study was conducted to compare the efficacy of Hataedock, with using the extract of Coptis japonica & Glycyrrhiza uralensis, to the early administration of probiotics for immunomodulation in the intestinal mucosa. Methods NC/Nga mice were divided into three groups; Control group (no treatment), CGT group (3-week-old mice given the extract of Coptis japonica & Glycyrrhiza uralensis), and MBT group (3-week-old mice given a Bifidobacterium). After 2 weeks, the intestinal mucosa tissues of each group of mice were observed. Immunohistochemical staining for IL-4, IL-13, CD40, $Fc{\varepsilon}RI$, $p-I{\kappa}B$, EGF, and VEGF in the intestinal mucosa was performed. Results CGT group showed 65% decrease in IL-4, 67% decrease in IL-13, 58% decrease in CD40, 72% decrease in $Fc{\varepsilon}RI$, 76% decrease in $p-I{\kappa}B$, 41% increase in EGF and 100% increase in VEGF compared to the control group. MBT group also showed 50% decrease in IL-4, 63% decrease in IL-13, 33% decrease in CD40, 53% decrease in $Fc{\varepsilon}RI$, 46% decrease in $p-I{\kappa}B$, 23% increase in EGF and 151% increase in VEGF compared to the control group. Conclusions These results suggest that both Hataedock, with using the extract of Coptis japonica & Glycyrrhiza uralensis, and early administration of probiotics were effective in regulating Th2, relieving inflammation and developing intestinal mucosal tissues. Hataedock with extract of Coptis japonica & Glycyrrhiza uralensis may be more effective for immunomodulation in intestinal mucosa than probiotics.

식용버섯의 면역조절에 미치는 영향 (Immunomodulating Effect of Edible Mushrooms in Mice)

  • 본 연구는 식용버섯들의 면역조절기능제로서의 가치 여부를 평가하기 위하여 수행되었다. 본 연구를 위해 민자주방망이버섯, 먹물버섯, 표고버섯, 새송이버섯이 사용되었다. 버섯을 투여한 생쥐의 혈장 내 IgG1, IgG2a 수준을 측정하였고, 비장 단일 세포군을 이용하여 T 림프구 및 B 림프구 in vitro 활성화 결과 생성된 IFN-${\gamma}$와 IL-4, IgG1과 IgG2a 수준을 각각 분석하였다. 실험 결과 표고버섯 1 mg/체중 kg을 투여한 군에서 IFN${\gamma}$/IL-4의 비가 다른 군에 비해 유의하게 높았다. 또한 혈장 내 IgG2a/IgG1의 비가 표고버섯의 경우에 다른 군보다 높았다. 아울러 TNF${\alpha}$의 생성 역시 표고버섯 1 mg/체중 kg을 투여한 군에서 다른 군보다 높았다. 이는 표고버섯이 항암작용, 항바이러스 작용과 같은 type-1 반응을 촉진할 가능성이 있음을 예측케 해주는 결과라 생각된다.

Probiotic Potential of Enterococcus faecium Isolated from Chicken Cecum with Immunomodulating Activity and Promoting Longevity in Caenorhabditis elegans

  • Probiotics, including Enterococcus faecium, confer a health benefit on the host. An Enterococcus strain was isolated from healthy chicken cecum, identified as E. faecium by 16S rDNA gene sequence analysis, and designated as E. faecium L11. To evaluate the potential of E. faecium L11 as a probiotic, the gastrointestinal tolerance, immunomodulatory activity, and lifespan extension properties of the strain were assayed. E. faecium L11 showed >66% and >62% survival in artificial gastric juice (0.3% pepsin, pH 2.5) and simulated small intestinal juice (0.5% bile salt and 0.1% pancreatin), respectively. Heat-killed E. faecium L11 significantly (p < 0.05) increased immune cell proliferation compared with controls, and stimulated the production of cytokines (IL-6 and $TNF-{\alpha}$) by activated macrophages obtained from ICR mice. In addition, E. faecium L11 showed a protective effect against Salmonella Typhimurium infection in Caenorhabditis elegans. In addition, feeding E. faecium L11 significantly (p < 0.05) extended the lifespan of C. elegans compared with the control. Furthermore, genes related to aging and host defense were upregulated in E. faecium L11-fed worms. In conclusion, E. faecium L11, which prolongs the lifespan of C. elegans, may be a potent probiotic supplement for livestock.

Effect of Kp, an Antitumor Protein-Polysaccharide from Mycelial Culture of Phellinus Linteus on the Humoral lammune Response of Tumor-Bearing ICR Mice to Sheep Red Blood Cells

  • The immunomodulating activity of Kp, an antitumor protein-polysacchanide preparation from the shake-cultured mycelia of Phellinus linteus, was investigated in ICR mice subcutaneously implanted wit $1\times10^6$ cells of sarcoma 180. The mice were intraperitoneally administered with Kp at a does of 100 mg/kg once daily for five consecutive days starting from 24 hrs after the tumor implantation. Ten days after the last injection, the mice were immunized with $1\times10^7$ or $4\times10^8$ sheep red blood cells (SRBC) and five days later, the antibody-forming immune response were assessed by direct hemolytic plaque assay. To an immunization does of $1\times10^7$ SRBC, the Kp-treated mice elicied a successful humoral immune response despite the turmor-burden and produced $259\times10^3$ plaque-forming cells (PFC)/spleen, while the corresponding tumor-bearing control mice showed virtually no reponse $(2.0\times10^3$ PFC/spleen) (the stimulation index=129.5). However, to an immunization dose of $4\times10^8$ SRBC, both of the control mice and Kp-treated mice showed almost the same level of strong humoral immune response. From these data it is clear that Kp effectively restores the humoral immune response of the turmor-bearing ICR mice.

Inhibition of Osteoclast Differentiation and Promotion of Osteogenic Formation by Wolfiporia extensa Mycelium

  • Osteoporosis, Greek for "porous bone," is a bone disease characterized by a decrease in bone strength, microarchitectural changes in the bone tissues, and an increased risk of fracture. An imbalance of bone resorption and bone formation may lead to chronic metabolic diseases such as osteoporosis. Wolfiporia extensa, known as "Bokryung" in Korea, is a fungus belonging to the family Polyporaceae and has been used as a therapeutic food against various diseases. Medicinal mushrooms, mycelium and fungi, possess approximately 130 medicinal functions, including antitumor, immunomodulating, antibacterial, hepatoprotective, and antidiabetic effects, and are therefore used to improve human health. In this study, we used osteoclast and osteoblast cell cultures treated with Wolfiporia extensa mycelium water extract (WEMWE) and investigated the effect of the fungus on bone homeostasis. Subsequently, we assessed its capacity to modulate both osteoblast and osteoclast differentiation by performing osteogenic and anti-osteoclastogenic activity assays. We observed that WEMWE increased BMP-2-stimulated osteogenesis by inducing Smad-Runx2 signal pathway axis. In addition, we found that WEMWE decreased RANKL-induced osteoclastogenesis by blocking c-Fos/NFATc1 via the inhibition of ERK and JNK phosphorylation. Our results show that WEMWE can prevent and treat bone metabolic diseases, including osteoporosis, by a biphasic activity that sustains bone homeostasis. Therefore, we suggest that WEMWE can be used as a preventive and therapeutic drug.

오적산(五積散) 경구투여에 의한 면역활성과 종양 전이 억제 효과 (Activation of Immune System & Antimetastatic Effects of Ojeok-san by Oral Administration)

  • Objectives: This study was designed to investigate intestinal immune system activation and antimetastatic effect of Ojeok-san on cancer cells by oral administration. Methods: Cell viability of Ojeok-san was tested with colon 26-M3.1 carcinoma cells and Peyer's patch cells in vitro. Antimetastatic experiments were conducted in vivo mouse model by using colon 26-M3.1 carcinoma cell. To observe immunomodulating effects of Ojeok-san on Peyer's patch cells, we measured interleukin (IL)-4, GM-CSF. In addition to observing effects of Ojeok-san on hematopoiesis, we measured proliferation of bone marrow cells mediated by Peyer's patch cells in vitro. IgA induction activated in serum and intestinal content was measured to observe the effect of orally administered Ojeok-san on mucosal immune system. After administering Ovalbumin (OVA) with Ojeok-san, Proliferation of Peyer's patch cell was measured to investigate gut immunostimulatory effect. Results: in vitro cytotoxicity analysis, the inhibitory concentration $(IC)_{50}$ of the colon 26-M3.1 carcinoma cell was $890{\mu}g/ml$. $IC_{50}$ of the Peyer's patch cells with LPS was $990{\mu}g/ml$. We found that orally administered Ojeok-san significantly inhibited tumor metastasis in vivo. In addition, the amounts of IL-4 and GM-CSF in the culture supernatant of Peyer's patch cells were significantly increased compared to the control group. The proliferation of bone marrow cell was significantly up-regulated with Ojeok-san. These results indicate that oral administration of Ojeok-san enhances the secretion of hematopoietic growth factors such as GM-CSF and IL-4 from Peyer's patch cells, and these cytokines also act on modulator of bone marrow cell proliferation. After orally administering Ovalbumin (OVA) with Ojeok-san, IgA induction and Proliferation of peyer's patch cell was up-regulated with Ojeok-san. These results means orally administered Ojeok-san activates intestinal immune system and has an inhibitory effect on tumor metastasis. Conclusions: Orally administered Ojeok-san appears to have considerable activity on the anti-metastasis by activation of immune system.

장미무당버섯(Russula rosacea)의 자실체에서 추출한 조다당류의 생쥐 Sarcoma 180에 대한 면역증강 및 항암 효과 (Immuno-potentiating and Antitumor Effects against Mouse Sarcoma 180 by Crude Polysaccharides Extracted from Fruiting Body of Russula rosacea)

  • 장미무당버섯은 담자균문, 주름버섯목, 무당버섯과에 속하며 맛이 좋은 식용 버섯으로 예로부터 암의 치료에 효과가 있다고 알려져 있다. 본 연구에서는 장미무당버섯의 자실체로부터 중성염용액, 열수 및 메탄올을 이용하여 조다당류를 추출하고 ICR mice에 주사하여 항암 및 면역증강 효과를 조사하였다. Sarcoma 180, HepG2, HT-29, NIH3T3 등의 암세포에 대한 독성을 조사한 각각의 암세포는 $10{\sim}2000\;{\mu}g/ml$의 조다당류 농도에서 세포독성을 나타내지 않았다. 각각의 조다당류가 투여된 실험군은 대조군에 비해 수명이 각각 $21.4{\sim}45%$ 연장되었다. 열수로 추출한 조다당류는 B 임파구의 alkaline phosphatase 활성을 $500\;{\mu}g/ml$의 농도에서 대조군에 비해 약 6.8배의 증가시켰다. 중성염추출 조다당류를 50 mg/kg body weight의 농도로 투여한 생쥐의 총 복강 세포 수와 백혈구의 수는 대조군에 비하여 각각 6배 와 2.6배로 증가하였다. 또한 면역에 관련된 장기인 간, 비장 및 흉선의 체중이 대조군에 비하여 소폭 증가된 것으로 조사되었다. 따라서 장미무당버섯의 자실체에서 추출한 조다당류가 Sarcoma 180에 대한 항암작용을 나타내는 것은 이들 조다당류가 Sarcoma 180에 대한 세포독성효과가 아니고 면역을 증강시킨 효과에 의한 것으로 사료된다.

삼지구엽초 용매별 분획 추출물의 면역관련 활성 (Immunological Activity of Solvent Fractions from $Epimedium$ $koreanum$ Nakai)

  • 본 연구는 삼지구엽초의 기능성식품 이용성 증진을 위한 기초적인 연구로써 우리나라 산야에서 존재하며 예로부터 약용으로 이용되고 있는 삼지구엽초를 극성에 따라 용매별 분획 추출물들의 면역관련 활성을 검토하였다. 삼지구엽초 분획물별 생육촉진 활성은 B와 T cell 모두에서 에틸아세테이트와 물 분획 추출물이 대조구에 비해 활성이 높게 나타났으며, 시료의 농도가 증가할수록 생육도 증가하였다. 삼지구엽초 추출물의 돌연변이 및 항돌연변이성에 미치는 영향을 조사한 결과, 삼지구엽초 추출물뿐만 아니라 각 용매별 분획물들도 돌연변이성을 나타내지 않았다. 간접변이원 B(${\alpha}$)P에서는 TA98 균주에서 에틸아세테이트 분획 추출물이 98%, TA100 균주에서는 부탄올 분획 추출물이 84%로 가장 높은 억제효과를 보였다. 직접변이원 4NQO에서는 TA98 균주와 TA100 균주에서 클로로포름 분획 추출물을 제외한 모든 시료가 70% 이상의 높은 억제효과를 나타내었으며, 특히 TA98 균주에서는 에틸아세테이트 분획 추출물이 80%, TA100 균주에서는 부탄올 분획 추출물이 90%의 높은 억제효과를 나타내었다. 이러한 결과는 삼지구엽초를 이용한 기능성 식품 및 소재로서의 이용성 증진을 위한 기초자료로 제공될 수 있을 것으로 판단된다.
