Docetaxel is a taxoid antineoplastic drug, which is widely used to treat locally advanced or metastatic non-small cell lung cancer (NSCLC). Among the adverse dermatological reactions, nail disorders such as bending, onycholysis, hypoor hyperpigmentation are rare. We report a case of a 62-year-old male with advanced NSCLC (cT4N3M1, stage IV), who developed purulent discharge and onycholysis in the nail of all his fingers and the left great toe after five courses of anti-neoplastic chemotherapy, which included docetaxel (cumulative dose: $370mg/m^2$, 590 mg). Seven days after the final session of chemotherapy, the patient had become aware of discoloration and swelling of the nail beds with out pain. Three days later, greenish-yellow purulent discharge oozed out from the involved nails. Microbiologic studies revealed Pseudomonas aeruginosa. Intravenous and topical antibiotics (mupirocin) were applied. After 2 weeks, regrown nails were observed and the onycholysis had improved.
Undesirable hyperpigmentation that can arise from increased melanocyte activity may be alleviated by targeting active melanocytes for apoptosis. The role of Stathmin 1 as an important regulator of microtubule dynamics is well documented. The current study examined the potential of Stathmin 1-targeting strategies in eliminating active melanocytes. A vector to overexpress Stathmin 1 and vectors to express three distinct small hairpin RNAs to knockdown Stathmin 1 expression in normal melanocytes were produced and in cell cultures acted accordingly. Both overexpression and knockdown of Stathmin 1 led to a marked increase in melanocyte apoptosis, as indicated by the accumulation of apoptotic cells and increased levels of cleaved caspase-3. Both up- and down-regulation of Stathmin 1 expression inhibited the activity of differentiated melanocytes, as indicated by decreases in both melanin production and tyrosinase activity. Taken together, these results indicate that hyperactive melanocytes can be inhibited by altering Stathmin 1 expression.

Objectives : The objective of this study was to report the efficacy of the Crucial Incision Acupuncture Therapy(CIAT) for removing melanocytic nevi. Methods : CIAT is penetration acupuncture to remove a lesion by sticking needles like cross-shape. We used the CIAT for treating one patient with melanocytic nevi, and then we investigated efficacy and side effect for about 5 months. Results : In 16 melanocytic nevuses treated with CIAT, one of the nevuses recurred after about 3 months. And hyperpigmentation lasted for about 2-5 months was caused after the CIAT. Conclusions : In Korean medicine, the CIAT might be used for treating melanocytic nevi. But additional studies might be needed for improvement of treatment effect.

Lipoteichoic acid (LTA) is a major component of the cell wall of Gram-positive bacteria. Its effects on living organisms are different from those of lipopolysaccharide (LPS) found in Gram-negative bacteria. LTA contributes to immune regulatory effects including anti-aging. In this study, we showed that LTA isolated from Lactobacillus plantarum (pLTA) inhibited melanogenesis in B16F10 mouse melanoma cells. pLTA reduced the cellular activity of tyrosinase and the expression of tyrosinase family members in a dose-dependent manner. The expression of microphthalmia- associated transcription factor (MITF), a key factor in the synthesis of melanin, was also decreased by pLTA. Further, we showed that pLTA activated melanogenesis signaling, such as extracellular signal-regulated kinase (ERK) and phosphatidylinositol 3-kinse (PI3K)/AKT. In addition, the expression of heterogeneous nuclear ribonucleoprotein A1 (hnRNP A1) and HuR, which are important RNA-binding proteins (RBPs), was reduced. pLTA likely degrades MITF via regulation of melanogenic signaling and RNA stability of melanogenic proteins, resulting in the reduction of melanin. Thus, our data suggest that pLTA has therapeutic potential for treating hyperpigmentation disorders and can also be used as a cosmetic whitening agent.
In the previous study, we reported the inhibitory effects of Rodgersia podophylla root extract on tyrosinase activity and melanin production in melan-a cells. However, mechanism of the inhibitory activity and in vivo assay were not yet examined. This study performed the examination of the effects of Rodgersia podophylla root extract on protein expression and in vivo depigmenting activity using melan-a cells and brown guinea pigs. As the results of western immunoblotting analysis, treatment of Rodgersia podophylla root extract reduced tyrosinase expression rates in 10 and 100 ppm concentrations, dose dependently. Moreover, Rodgersia podophylla root extract exhibited depigmenting activity on UV-B induced hyperpigmentation in brown guinea pig skin. These results suggested that Rodgersia podophylla root extract could act as whitening agent for the skin via not only direct tyrosinase activity inhibition but also reducing of tyrosinase expression.
To examine the potential of Terminalia chebula as a whitening agent, we measured antioxidant activity using DPPH$\cdot$, ABTS${\cdot}^+$ assays and ferric-reducing antioxidant power (FRAP) assays, and depigmenting activity using B16F10 melanoma cells. The intracellular reactive oxygen species (ROS) level was monitored by $H_2DCFDA$ fluorescence labeling, and melanin contents in B16F10 melanoma cells by 960 $J/m^2$ dose of UVA-induced oxidative stress. The radical-scavenging activities of T. chebula extract (TCE) were measured in terms of $EC_{50}$ values using DPPH$\cdot$, ABTS${\cdot}^+$ assays and FRAP value were 280.0 ${\mu}g/mL$, 42.2 ${\mu}g/mL$ and 113.1 ${\mu}mol$$FeSO_4{\cdot}7H_2O/g$, respectively. We found that ROS and melanin concentrations were reduced by TCE treatments of 25 ${\mu}g/mL$ under UVA-induced oxidative stress. Tyrosinase activity and melanin contents in $\alpha$-melanocyte stimulating hormone (MSH)-induced melanoma cells both decreased dose-dependently in the treatment groups. TCE similarly reduced melanogenesis in B16F10 melanoma cells stimulated by $\alpha$-MSH as compared to arbutin as a positive control. T. chebula may prove to be a useful therapeutic agent for hyperpigmentation and an effective component in skin whitening and.or lightening cosmetics.

INTRODUCTION : Psoralens are planar tricyclic furocoumarins present in numerous plants and fungi found throughout the world.' Naturally occurring and synthesized psoralen derivatives(see Figure 1) are photosensitizers of UVA especially from 320 nm to 400 nm, a range at which cellular nucleic acids and proteins are weakly absorbing if any at all. Because of their skinphotosensitizing properties, these compounds have been used in the photochemotherapy of psoriasis and vitiligo. However, undesirable side effects such as carcinoma development in hairless mice as well as possible liver damage from the use of 8-methoxypsoralen(8-MOP) have been reported. The other photobiological effects include inactivation of DNA viruses, killing and mutagenesis of bacteria, inhibition of tumor transmitting capacity of various cells, and hyperpigmentation on human and guinea pig skin. PUVA(psoralen+UVA) photochemotherapy is in fact thousands of years old, having been used in Egypt and India since B.C. 1200-2000. Photochemotherapy for a common disfiguring disease, vitiligo, was practiced in the ancient world by physicians and herbalists who used boiled extracts of the fruits of certain umbelliferous plants, e.g. Ammi majus Linnaeus in Egypt or the leguminous plants, Psoralea corylifolia L. in India. It was first described by Kuske in 19388 that photosensitization of skin by plants was related to the presence of psoralen. He identified natural psoralens in plants as photosensitizers and isolated bergapten(5methoxypsoralen) from the oil of bergamot. Withania somnifera has been used in folk medicine to treat various ailments for centuries. In this studies to investigate the whitening effect of Withania somnifera extracts as an active ingredient for whitening cosmetics, the antioxidant capacity and the effects of Withania somnifera extracts on melanogenesis in B16-F10 melanoma cells were identified. Withania somnifera extracts significantly reduced both tyrosinase activity and melanin content in a concentration-dependent manner. Furthermore, it was found that Withania somnifera extracts decreased ${\alpha}-MSH$ (melanocyte-stimulating hormone)-induced tyrosinase activity and MITF(microphthalmia associated transcription factor) protein expression. These data indicate that Withania somnifera extracts attenuate ${\alpha}$-MSH-stimulated melanin synthesis by modulating MITF expression and that they may be a useful therapeutic agent for treating hyperpigmentation and an ingredient of whitening cosmetics.

This study was purposed to investigate the antioxidative effects, the enzyme activity of the alcohol metabolizing and melanin production of Maesil(Prune mume). The antioxidant activity of Maesil(Prunu mume) was analyzed by measuring thiobarbituric acid reactive substances(TBARS value) and electron donating ability. And we investigated the changes of alcohol dehydrogenase(ADH) and acetaldehyde dehydrogenase(ALDH) activity by measuring the maximum absorbency at 340nm in vitro and human study. The inhibitory effects of Maesil were investigated in vitro and in B-16mouse melanoma cells on melanin biosynthesis that is closely related to hyperpigmentation. The antioxidant activities for TBA values were 29.65% in ascorbicacid, 45.35% in BHT, 15.99% in extract of dehydrated maesil flesh(EDMF) and 25.00% in extract of dehydrated maesil juice(EDMJ). The electron donating abilities by DPPH were 96.69% in ascorbic acid, 77.82% in BHT, 34.25% in EDMF, and 42.99%in EDMJ. Electron donating abilities by DPPH in the presence of 0.02% EDMF and EDMJ were 53.21% and 59.19% respectively. Facilitating rates of ADH activity were 137.92, 131.58, 152.96, 218.70, 111.76, and 144.27% in maesil juice, 5, 10, and 15% GMT, and 0.5 and 1.0% aspartic acid, respectively. ALDH activity increased in the order of Maesil juice > ALDH > GMT > aspartic acid, and facilitating rate of ALDH activity in Maesil juice was the highest at 976.44%. Maesil extracts inhibited tyrosinase activity that converts dopa to dopachrome in the biosynthesis process. B-16 cells treated by Maesil extracts showed that the viability was over 80%. Maesil and maesil products in vitro and B-16 cells inhibited melanin production significantly.

The aim of this study was to investigate the effect of ethanol extract of Fagopyrum escuentum(FE) on the melanogenesis. To determine whether ethanol extract of FE suppress melanin synthesis in cellular level, B16F10 melanoma cells were cultured in the presence of different concentrations of FE ethanol extract. In the present study, the author examined the effects of FE ethanol extract on cell proliferation, melanin contents, tyrosinase activity. Cell proliferation was slightly increased by treatment with ethanol extract of FE (25-200 ${\mu}$g/ml). The ethanol extract of FE effectively suppressed melanin contents at a dose of 100 ${\mu}$g/ml. It was observed that the color of cell pellets was totally whitened compared with the control. The ethanol extract of FE inhibited tyrosinase activity, regulate melanin biosynthesis as the key enzyme in melanogenesis. These results suggest that the ethanol extract of FE exerts its depigmenting effects through the suppression of tyrosinase activity. And it may be a potent depigmetation agent in hyperpigmentation condition.
