Effect of Green Tea Extract on Lipid Synthesis in Human Sebocyte Cell Line (녹차추출물이 인체 피지선세포주에서 지질 생성에 미치는 효과)

  • The aqueous Green tea comes to be used with the Oriental medicine plant, it has the numerous health benefits, including antioxidant, anti-inflammatory, and anti- carcinogenic properties. Epidermal progenitor cells give rise to multiple skin lineages: hair follicle, sebaceous gland and the overlying interfollicular epidermis. Sebocytes are the cells of the sebaceous gland, which synthesize and accumulate lipid dropolets. In order to determine the effect of Green tea on lipid production, several experiments were performed in SZ95 cells (sebocytes). We found that Green tea increased lipid droplets compared with control in a dose-dependent manner. Human sebaceous glands produce sebum, a lipid mixture of squalene, wax esters, triglycerides, cholesterol esters, and free fatty acids that is secreted onto the skin. Therefore, to investigate the effects of Green tea on intracellular lipid levels, we treated SZ95 cells with Green tea, and then examined cholesterol and triglyceride levels. After treatment of the cells with Green tea, the cholesterol and triglyceride levels of SZ95 cells were increased significantly in a dose-dependent manner.

  • Recently. the herbal industry has expanded into the herbal dietary supplements industry, the herbal cosmetics industry, etc. But still the main form of herbal drug usage is water extraction- packaging after herb preparation in private clinics. There was no report or data of investigations about industrial herbal preparations. To search for a better plan of herbal drug improvement and popularization. we surveyed the present state of Korean herbal preparation production. We made an industrial herbal preparation list, investigated the total amount of production, the chief items of manufacture. names of prescriptions each item is based on. scales of each production company, etc., and report on them on this article.

Inhibitory Efficacy of Dioscoreae Rhizoma on MITF, TRP-1, TRP-2, Tyrosinase, PKA and ERK Expression in Melanoma Cells (B16F10) (산약의 멜라노마 세포(B16F10)에서 MITF, TRP-1, TRP-2, Tyrosinase, PKA, ERK 발현 억제 효과)

  • Objectives : The purpose of this study was to research the whitening effects and developing by cosmetics of the extract fromDioscoreae Rhizoma, which is one of the most popular health-promoting herb in herbal medications.Methods : We performed tyrosinase inhibition assay, reverse transcription-polymerase chain reaction (RT-PCR) and western blot for whitening effects. Also we measured MTT assay for cell viability.Results : The results were obtained as follows : For whitening effect, tyrosinase inhibition rate of extract fromDioscoreae Rhizomashowed more than 42.28% at 1,000 ㎍/㎖ concentration. Cell toxicity effect on melanoma cells (B16F10) of extract fromDioscoreae Rhizomashowed 81.97% with toxicity at 50 ㎍/㎖ concentration. So we were measured at a concentrations of 5, 10 and 50 ㎍/㎖ in all experiments involving cell. In addition, whitening related mRNAs including microphthalmia associated transcription factor (MITF), tyrosinase related protein-1 (TRP-1), tyrosinase related protein-2 (TRP-2), tyrosinase were reduced byDioscoreae Rhizoma. We also foundDioscoreae Rhizomatransiently decreased protein kinase A (PKA) which is known to be upstream to the down regulation of MITF and tyrosinase. But phosphorylation of extracellular signal related kinase (pERK) were increased byDioscoreae Rhizoma. These results imply thatDioscoreae Rhizomadecrease melanogenesis via ERK activation and subsequent down regulation of MITF and tyrosinase.Conclusions : Therefore, all these findings suggested the potent usage ofDioscoreae Rhizomaas materials of functional cosmetics by confirming whitening activity related with melanin content.

  • Cytotoxic activity against the P388 cell line was seen in a crude extract of Anisotome lyallii. A bioactivity guided isolation led to the isolation of a deterpene. which displayed strong Cytotoxic activity against the P388 cell line (IC50 2.3 $\mu$g/ml), as well as antimicrobial activity against Bacillus subtilis. The structure of de terpene 1 was elucidated by spectroscopic methods. (omitted)

  • Objectives : Since cosmetics have been one of the help of life, unlike medicine, natural products have been used for cosmetics, generally giving the image of safety and relief compared to synthetic products. Among them, Houttuynia cordata has been known as a useful herbal medicine with antibiotic, anti-inflammatory and anti-oxidative activities. In this study, we were to use Houttuynia cordata extract on formulation of cosmetic emulsions. Methods : The effects of Houttuynia cordata extract on emulsion stability and viscoelastic properties of emulsion were measured using turbiscan and rheometer. And we assessed the anti-oxidative and antibiotic activities of Houttuynia cordata extract. Results : 1. The results of this investigation for emulsion stability showed that the stability emulsion containing Houttuynia cordata extract was decreased depending on concentration of Houttuynia cordata extract. 2. Super oxide dismutase activity was strongly dependents on concentration of Houttuynia cordata extract. 3. Houttuynia cordata extract showed good anti-microbial activity against Staphylococcus aureus. Conclusions : Houttuynia cordata extract can be effectively used in cosmetic emulsions when the relation between natural product extracts and formulation of cosmetics is elucidated.

Study on the Benefit of Medical Herbal Cosmetics via the pattern identification about fluid-humor of skin in Traditional Korean Medicine (한방 피부 진액변증을 통한 한방화장품의 효능 평가)

  • Objectives : The aim of this experiment is to find out relationship between the effect of cosmetics and 4 types of pattern identification about fluid-humor, which are based on the general idea of traditional oriental medicine, Qi-Blood and deficiency-excess. Methods : Korean female volunteers in good health (n=25, $23.12{\pm}2.83$) participated in this experiment. Three Korean medical doctors classified them into 4 groups: Blood-deficiency: group A; Blood-excess: group B; Qi-deficiency:group C; Qi-excess:group D). Cosmetics that contains herb extract for Blood deficiency were given to all volunteers and they used the cosmetics for 4 weeks. Volunteers were assessed non-invasively with the skin measuring devices before and after using cosmetics. And we analyzed the correlation of skin physiological parameters with 4 groups. Results : Three doctors diagnosed participants and classified them into 4 groups ; group A(n=8),group B(n=7), group C(n=3), group D(n=5) as highest score. After 4 weeks, facial skin moisture showed no significant difference in comparison between 4 groups. Sebum showed significant increase in Group A and significantly decreased Group B. Measurement of facial skin elasticity tended to increase in Group A, C, D but skin elasticity was decreased significantly in Group B. Conclusions : In case of a group that pattern identification about fluid-humor corresponds to herb extract in cosmetic, skin improving effect was better than the other group that pattern identification oppose to properties of herb in cosmetic. Therefore, from the view of traditional oriental medicine, it is very important to understand user's pattern of identification or physical conditions and properties of herbs in cosmetics on the matter of safety and efficacy.

  • Melanin is a main pigment found in skin. hair and eyes, and tyrosinase plays an important role in the process of melanin polymer biosynthesis. Caesalpinia sappan L.(C. sappan) has been commonly used in Oriental folk medicines to promote blood circulation and as analgesic as well as remedy for thrombosis. Ethanol Extract of Astragalus membranaceus Bunge Induces Aquaporin-3 Expression in HaCaT Cells (HaCaT Cell에서 황기 에탄올 추출물의 Aquaporin-3 발현 효과)

  • Astragalus membranaceus Bunge is used in herbal medicine in Eastern Asian countries including Korea. In this study, we assessed the effects of A. membranaceus extract (AM) on the aquaporin-3 (AQP3) protein expression in HaCaT cells. AM did not affect viability of HaCaT cells. AQP3 expression and cell migration seem to be maximal at $100{\mu}g/mL$ concentration. Epidermal growth factor receptor (EGFR) kinase inhibitor, PD153035, blocked AM-induced AQP3 expression and cell migration. In addition, an 80% ethanol extracts of herbal prescription, SinhyoTakleesan (ST), which is composed of A. membranaceus, Angelicae gigantis, Glycyrrhiza glabra Linne, and Lonicera japonica Flos also induced AQP3 expression at $20{\mu}g/mL$ in HaCaT cells. Collectively, these results suggest that AM induce AQP3 expression via EGFR pathway.

Ponciri Fructus Extract Induces Lipogenesis through Transcription Factor SREBP-1 Activation (지실 추출물의 전사인자 SREBP-1 활성에 의한 지질 생성 촉진)

  • This study was to explore the lipogenic effect by ethanol extract of ponciri fructus (EPF) and possible molecular mechanisms in sebocyte. When SZ95 sebocyte cell line were treated with the EPF, lipid droplets were accumulated in the majority of cells. EPF increased expression of sterol regulatory element-binding protein-1 (SREBP-1) and fatty acid synthase (FAS) in the SZ95 cells. EPF augmented expression of PPAR-${\beta}$ and PPAR-${\gamma}$ but not that of PPAR-${\alpha}$. These results suggest that EPF induces lipogenesis in SZ95 cells through SREBP-1, PPAR-${\beta}$ and PPAR-${\gamma}$ activations.

Clinical Evaluation of Fermented-C. Atrati Radix Cosmetic (Whitecrow) for Skin Whitening (백미발효 화장품(Whitecrow)의 임상적 미백효과 평가)

  • This study evaluated the Skin Whitening effects of 'fermented-C.' for Atrium cosmetics using a single-arm clinical trial. Twenty female adults (mean age 42.1 ± 6.6 year) were enrolled, all participants used this cosmetic product on their face for 4 weeks. The change of brightness (L-value) was measured by spectrophotometer while subjective assessments were also obtained to find the participants opinions on whitening and their general satisfaction measured by a 5-point scale (5 for very satisfactory, 5 for satisfactory, 3 for moderate, 2 for unsatisfactory, 1 for very unsatisfactory). The participants were also questioned about any adverse effects by filling out a self-monitoring report. The average L-value increased from 62.55 ± 2.14 on day 0 to 63.22 ± 1.96 after 4 weeks corresponding to a 1.1% brightness improvement (p < 0.01). The average scores for subjective whitening and general satisfaction were better than moderate with scores of 3.6 ± 0.5 and 3.7 ± 0.7, respectively. No notable complaints were reported regarding any kind of adverse effects such as erythema, itching or burning. The clinical data collected in this study supports that fermented-C. from Atrium cosmetics has whitening potential in a safe Functional Cosmetics package.
