시호사물탕이 t-BHP로 유도된 간세포의 산화적 손상 및 자유기에 의한 지질과산화 반응에 미치는 영향 (Preventive Effects of Sihosamultang Extract on Lipid Peroxidation by Free Radicals and Oxidative Damage of Hepatocytes by tert-Butyl Hydroperoxide)

  • Sihosamultang(SST) has been used for the treatment of puerperal fever, liver disease in traditional medicine. The present study was carried out to evaluate the antioxidant effects of SST extract in vitro. The inhibitory effect of SST extract on lipid peroxidant was examined in the linoleic acid autoxidation system. In this test, SST extract significantly inhibited the time course of the lipid peroxidation. And SST extract showed about 73% scavenging effect on DPPH radical. And this extract inhibited not only the lipid peroxide formation induced by hydroxyl radical derived from $ H_{2}O_{2}-Fe^{2+}$ in the rat liver homogenate, but also the superoxide generation from xanthine-xanthine oxidase system in a dose-dependent manner. In addition, SST extract protected the hepatic cell death induced by tert-butyl hydroperoxide. These data indicated that SST might play a protective role against oxidative injury by free radicals.

비타민 C의 만성적 과량투여가 흰쥐의 혈액과 간의 지질성상, 과산화상태 및 혈소판 Thromboxane $A_2$ 생성에 미치는 영향 (Effects of Excess Vitamin C Feeding on Blood and Liver Lipid and its Peroxidation Levels, and Platelet Thromboxane $A_2$Formation in Rats)

  • The effect of excess L-ascorbic acid(AsA) in blood, liver lipid levels and peroxidation status were investigate . Ten Sprague-Dawley male rats weighing 150-200g were fed 300mg AsA/100g body weight/day, mixed into ground chow diet, for 4 weeks. And another set of then rats were fed only chow diet as the control. Average body weight gain was slightly lowered by AsA feeding without food intake change. The AsA group showed higher AsA levels in plasma and liver than the control group. In addition, the AsA group showed a higher plasma TBARS value. Liver TBARS seemed to be elevated in the AsA, but not significantly. The hemolysis of red cells tended to increase with excess AsA, accompanied by a raised GSH-Px activity and lowered total GSH levels. Plasma HDL-Chol level was increased while the levels of total Chol, LDL-plus VLDL-Chol , and triglyceride were unchanged . Atherogenic index decreased. Hepatic TG levels were also decreased, but the total amount of Chol increased slightly . Platelet TXA$_2$ production was inhibited by excess AsA feeding. Above results indicafe that oral feeding of excess AsA may be beneficial in reducing the risk of atherosclerosis ; however such practice may be detrimental for tissue lipid peroxidation and weight gain.

  • Hepatic lipid peroxide contents were examined in bromobenzene-treated rats firstly after the oral administration of MeOH extract of Kalopanax pictus stem bark its n-BuOH fraction, EtOAc fraction and an alkaline hydrolysate of the n-BuOH fraction, and secondly after the intraperitoneal administration of hederagenin monodesmosides and bisdesmosides. Two hederagenin monodesmosides, kalopanaxsaponin A (KPS-A) and sapindoside C, exhibited significant anti-lipid peroxidation effects after intraperitoneal administration at doses of $10-30{\;}{\mu}mole/kg$, whereas their bisdesmosides did not exhibit any significant activity. These results suggest that it is the hederagenin monodesmosides that are responsible for anti-lipid peroxidation in vivo. The activity of KPS-A was established by the observation of decreased aminopyrine N-demethylase activity and increased epoxide hydrolase activity.

사염화탄소에 의해 유발된 생쥐의 간독성에 미치는 누에 추출물의 영향 (Effects of Bombyx mori Larvae Extracts on Carbon Tetrachloride-Induced Hepatotoxicity in Mice)

  • To evaluate the protective effects of extracts of silkworm on carbon tetrachloride-induced hepatotoxicity, serum glutamic-oxaloacetic transaminase, glutamic-pyruvic transaminase and lactic dehydrogenase activities, malondialdehydes values and glutathione-S-transferase activity were measured in ICR mice. Extracts of silkworm was administered orally at 30min after the administration of CCl4 Mice were sacrificed at 24h after the administration of extracts of silkworm. The activities of serum aminotransferase and the hepatic content of lipid peroxide after carbon tetrachloride treatment were markedly increased than normal control but those levels were decreased by the treatment of butanol soluble fraction of silkworm methanol extract. Glutathione S-transferase activity was decreased by carbontetrachloride than control, but also inhibited by the treatment of butanol soluble fraction of silkworm methanol extract.

  • In order to evaluate the protective effects of extracts of Mori cortex radicis on carbon tetrachloride-induced hepatotoxicity, glutamic oxaloacetic transaminase, glutamic pyruvic transaminase, lactic dehydrogenase, malondiadehyde values and glutathione S-transferase activity were measured in ICR mice. The activities of serum aminotransferase and the hepatic content of lipid peroxide after carbon tetrachloride-treatment were markedly increased than normal control but those levels were significantly decreased by the treatment of butanol fraction of Mori cortex radicis mathanol extract. Glutathione S-transferase activity was decreased by carbon tetrachloride than control, but that also inhibited by the treatment of butanol fraction of Mori cortex radicis methanol extract. These results demonstrate a possible hepatoprotective role of extract role of extract of Mori cortex radicis against ${CCl_4}-induced$ hepatoxicity in vivo.

40주령의 생쥐 간에 미치는 ginsenoside의 항산화효과 (Effects of the Antioxidative Components to Ginsenoside in the Liver of 40-week-old Mice)

  • 40주령의 생쥐에게 홍삼 총사포닌, PD, PT ginsenoside G-Rd, 5-Re 및 C-K 등을 경구투여하여 생쥐 간에 미치는 항산화효과를 알아보기 위하여 SOD와 CAT활성도 변화와 과산화수소 함량, MDA수준의 변화를 조사하였다. SOD와CAT 활성도는 모든 홍삼 사포닌 투여군에서 증가하였지만 PD투여군에서만 유의성(p<0.01) 있는 증가를 보였다. 과산화수소 함량은 홍삼 사포닌의 투여로 모두 감소하였으며, 특히 PD, PT G-Rd(p<0.01) 및 G-Re 투여군(p<0.05)에서 유의성 있게 감소하였다. CAT 활성도는 G-Rd 투여군에서 유의성(p<0.05) 있게 증가하였다. 지질과산화의 최종산물인MDA수준은 홍삼 투여군 모두에서 감소되었으며, 특히 G-Rd투여군에서 유의성(p<0.05) 있게 감소하였다. 따라서 홍삼 사포닌 성분은 40주령의 생쥐 간에서 항산화효소의 합성증가를 유도하거나 활성을 촉매하여 활성산소들을 효율적으로 소거하는 기능과 지질과산화를 억제하는 항산화물질의 합성능력을 강화하여 산화적 손상으로부터 생체를 방어하는 효과가 있다고 생각되었다. 특히 홍삼 사포닌 중에서도 PD와G-Rd투여군은 항산화 효소활성을 증가시키는 반면 지질과산화를 .억제하였다.

진당원(珍糖元)의 고혈당 조절 작용 및 기전에 관한 연구 (Effects of Jindangwon extract in streptozotocin-induced diabetic rats)

  • Objective : Diabetes is a disease in which the body does not produce or properly use insulin. Etiological studies of diabetes and its complications showed that oxidative stress might playa major role. Therefore, many efforts have been made to regulate oxygen free radicals for treating diabetes and its complications. Because Jindangwon has been known to be effective in treatment of diabetes, the methanol extract of Jindangwon was tested for its effectiveness in reducing the oxidative stress induced by Streptozotocin. Methods : Jindangwon was washed, dried in the shade and crushed. The crushed Jindangwon was extracted 3 times, each time with 3 volumes of methyl alcohol at $60^{\circ}C$ for 24 hours. The extract was filtered and evaporated under reduced pressure using a rotary evaporator to yield 30.6 g. Jindangwon extract was oral-administered to the diabetic rats induced by streptozotocin 50 mg per 1 kg of body weight for 15 days. The efficacy of the Jindangwon extract was examined with regard to the enzymatic pathways involved in the oxygen free radical production and the glutathione balance. Results : he effects of the methanol extract of Jindangwon in streptozotocin-induced diabetics rats with regard to body weight, blood glucose level, hepatic lipid peroxide level, hepatic xanthine oxidase activity and type conversion rate, hepatic glutathione level, hepatic glutathione peroxidase activity, hepatic glutathione reductase activity, hepatic aldose reductase activity, and hepatic sorbitol dehydrogenase activity were favorable enough to suggest that it is a cure for diabetes and its complications. Conclusions :These results support Jindangwon as an effective reducing agent for oxidative stress in the tissues and organs by regulating the production of oxygen free radicals. Jindangwon, in particular, shows promising results for its use as a cure, or preventative medicine for diabetes and its complications by reducing oxidative stress in beta-cells of the pancreas.

어성초 분획물이 사염화탄소로 유발된 흰쥐의 간손상에 대한 보호효과 (Protective Effects of Houttuynia cordata Thunb on Carbon Tetrachloride-induced Hepatotoxicity in Rats)

  • This study was performed to investigate the protective effect of Houttuynia cordata Thunb on hepatotoxicity in carbon tetrachloride$(CCI_4)$ intoxicated rats. The examined effects hexane, chloroform, butanol and water fractions prepared from the Houttuynia cordata Thunb methanol exlract and rats were administrated with those orally once a day for successive 6 days, fellowed by treaoent with $CCl_4$ on the sixth day. After 6 days, the activities of aminotransferase, alkalinephosphatase, ${\gamma}-glutamyl$ transpeptidase, lactate dehydrogenase and contents of triglyceride, hepatic lipid peroxide in butanol fraction pretreated rats were significantly decreased compared to the only $CCl_4$ treated rats, also depletion glutathione content induced by treatment with $CCl_4$ was prevented by butanol fraction pretreated rats. In addition, activities of hepatic superoxide dismutase, catalase, glutathione peroxidase in butanol fiaction pretreated rats were significantly decreased compared to the only $CCl_4$ treated rats, but the activity of hepatic glutathione-S-transferase was not significantly effect. These results suggest that butanol fiuction of Houttuynia cordata Thunb methanol extract have potent hepatoprotective effect against carbon tetrachloride intoxicated rats.

Adriamycin을 투여한 흰쥐의 간 지질대사에 미치는 비타민 A와 E의 급여 효과 (Effect of Dietary Vitamin A and E on Hepatic Lipid Metabolism in Adriamycin-Treated Rats)

  • Adriamycin의 투여로 변화되는 생체내 지질과산화대사에 대하여 비타민 A및 E의 급여와 이들의 동시급여의 효과를 살펴보기 위해 이유한지 1주된 Sprague-Dawley 흰쥐를 6주간 사육하였다. 대조군을 제외한 모든 실험군에게 매주 일정시각에 ADR(2mg/kg/week)을 복강주사 한 후 비타민A와 E의 급여효과를 관찰하였으며 그 결과는 다음과 같다. 간 조직에서의 과산화지질 함량은 ADR투여로 현저한 증가로 보여주었고, 비타민 A 와 E급여가 이들의 생성을 저하시켰다. Catalase 활성도는 ADR의 투여로 증가된 반면 GSH-Px는 감소되는 경향이었다. 이에 비타민 A나 E의 급여로 ADR투여에 의한 효소 활성도의 변화를 완화시켰다. SOD의 활성은 ADR의 투여로 유의적인 변화는 나타나지 않았지만 비타민 A와 E동시 급여군에서 그 활성도가 현저히 유도외었다. 간 미토콘드리아의 지방산조정은 ADR투여로 큰 변화는 없었으나 비타민 E급여군이 비타민 A급여군 보다 방어효과가 더 좋았다. 또한 지질성분들의 변화는 ADR의 투여로 중성지질과 콜레스테롤 함량이 증가하였고 비타민 A의 급여에 의해 이들 함량이 유의적으로 감소되었다.

고혈당 흰쥐에서 제조의 혈당 조절과 항산화 작용에 관한 연구 (Effects of the Extract in Streptozotocin-induced Diabetic Rats)

Because Holotrichia has been known to be effective for the treatment of diabetes, the methanol extract of Holotrichia was tested for its effectiveness in reducing the oxidative stress induced by streptozotocin. Methods : Holotrichia was washed, dried in the shade and crushed. The crushed Holotrichia was extracted 3 times, each time with 3 volumes of methyl alcohol at $60^{\circ}C$ for 24h. The extract was filtered and evaporated under a reduced pressure using a rotary evaporator to yield 17 g. Holotrichia extract was oral-administed to the diabetic rats induced by streptozotocin 50 mg per 1 kg of body weight for 20 days. The efficacy of the Holotrichia extract was examined with regard to the enzymatic pathways involved in the oxygen free radical production and the glutathione balance. Results : The Effects of the methanol extract of Holotrichia in streptozotocin-induced diabolic rats with regards to body weight, blood glucose level, hepatic lipid peroxide level, hepatic superoxide anion radical content. hepatic xanthine oxidase activity and type conversion rate, hepatic glutathione level, hepatic aldose reductase activity, and hepatic sorbitol dehydrogenase activity were shown to be good enough to cure and prevent the diabetes and its complications. Conclusions : These results indicated that Holotrichia might reduce the oxidative stress in the tissues and organs by regulating the production of oxygen free radicals. Especially, Holotrichia might prevent and cure the diabetes and its complications by reducing the oxidative stress in the ${\beta}$-cells of pancreas. Some suggestions on biophoton experiments were made.
