목화진딧물(Aphis gossypii Glover)이 참외의 생육과 당함량에 미치는 영향 (Effect of Aphis gossypii Glover on Growth and Sugar Content of Oriental Melon)

  • 목화진딧물은 여러 작물에 헤를 입히는 문제해충이다. 본 연구에서는 목화진딧물의 접종밀도를 달리했을 때 참외의 생육고 당함량에 미치는 영향과 진딧물 제거후 생육의 회복 정도를 알아보고자 수행하였다. 진딧물의 피해를 받은 참외의 증상은 잎이 오그라들고 신초부분이 위축되었다. 목화진딧물이 참외에서 20일간 흡즙한 후 생육은 진딧물을 접종하지 않은 참외에 비해 생육이 월등히 감소하였고, 진딧물의 접종밀도간에는 진딧물의 밀도가 높아질수록 생육이 부진하였다. 초장은 무접종의 1/3수준이었고, 엽면적은 무접종에 비해 12${\sim}$38% 수준이었으며 건물증은 무접종의 30${\sim}$51% 수준이었다. 진딧물을 제거한 후 참외의 생육은 10일 후에는 진딧물의 피해를 받은 잎은 거의 신장하지 못한 채로 있으나, 신초부분의 생육이 다소 회복되었다. 20일 후에는 5마리 접종에서 초장, 엽면적 건물증이 월등히 증가하여 생육이 회복되었다. 진딧물 제거후 시간이 지날수록 회복되는 속도가 빨라졌으며 접종밀도간 회복속도는 5마리 접종이 가장 빨랐다. 주당 진딧물을 접종하였을 때 참외 전체 잎의 당함량에는 차이가 없었으나, 엽당 진딧물을 접종하고 접종한 잎만 분석한 경우는 진딧물누적일수량이 증가할 수록 전체 당 함량이 현저히 감소하였다.

천년초 분말의 기능성분 분석과 물리적 특성 연구 (Functional component analysis and physical property of Cheonnyuncho (Opuntia humifusa) powder)

  • 본 연구에서는 천년초의 줄기와 열매를 동결 건조하여 분말화 한 후 기능성 성분으로 Vit. $B_1$, $B_2$, $B_3$, 루틴 및 베타카로틴 등의 비타민, 식이섬유, 조 사포닌을 분석하고, 물리적 특성으로 pH 및 색도, 보수력, 보유력, 팽윤력, 용적밀도, 점액물질 함량 및 수분흡수량을 조사하였다. 천년초 분말의 비타민을 측정한 결과 비타민 C(ascorbic acid)의 함량이 가장 많았으며 줄기가 42.14 mg, 열매가 105.21 mg으로 줄기의 약 2배 이상 더 높게 나타났다. Lutein의 함량은 줄기가 열매 보다 약 2.3배 정도 더 높게 나타났으며 ${\beta}$-carotene의 함량은 열매가 줄기보다 약 2.9배 정도 높은 함량을 나타내었다. 비타민 B의 경우 줄기와 열매의 함량이 각각 비타민 $B_1$(thiamin)는 0.16 mg 및 0.15 mg, 비타민 $B_2$(riboflavin)은 0.08 mg 및 0.12 mg으로 아주 적은양이 함유되어 있었으며 비타민 $B_3$(niacin)은 0.92 mg 및 1.01 mg으로 나타났다. 식이섬유는 줄기에서 수용성 식이섬유가 45.24%, 불용성 식이섬유가 22.15%로 열매보다 높게 나타났다. 조 사포닌은 줄기가 19.30 mg/g, 열매가 25.10 mg/g으로 나타났다. 물리적 특성의 pH는 줄기가 pH 5.34, 열매가 pH 5.07로 약산성이었고 보유력을 제외한 보수력, 용적밀도 및 팽윤력은 줄기가 높게 나타났으며 점질물질의 함량과 수분흡수량도 줄기가 열매보다 더 높게 나타났다. 이러한 결과 보아 천년초에 함유되어 있는 비타민류, 식이섬유 등은 천연 기능성 소재로 이용 가능하며 물리적 특성을 잘 이용한다면 가공적성 및 제품의 기호성을 증진시키는데 유용한 식품소재가 될 것으로 기대된다.

우리 나라 지방자치단체 식재 조례 기준의 현황 및 개선방향 (A Study on Improvements of Local Governments' Planting Regulations in Korea)

  • In 1977, Local governments' planting regulations for the development projects were established in accordance with the revised building law in Korea. A landscaped area, planting densities of trees and shrubs, a percentage of evergreen plants, minimum tree size and species were prescribed in the planting regulations. But the clauses for an excessive planting density and a high ratio of evergreen trees that the regulations includes, have been gradually in the way to a creative planting design, and raise a problem of poor growth of trees an a disordered planting landscape. Therefore, in this study the present planting regulations of 124 local governments throughout the country were analyzed and compared with 13 foreign local governments' of 4 countries ; Japan, the United States of America, Canada and Singapore. And the linitations and characteristics of the regulations are drawn as follows ; 1. The regulations focus on controlling the green spaces and plantings by quantitative methods such as controlling the number of trees and the landscaped area, which are inadequate for estimating the crown coverage of mature trees, and which areinadequate for estimating the crown coverage of mature trees, and implementing the function of polantings and the use of green spaces. 2. Minimums of tree densities are higher than those of foreign countries, especially higher about 10 times to 100 times than those of the United States of America. 3.Excessive number of evergreen trees and fruit trees should be planted under the present planting regulations, that results in constricting the creativity in planting design. 4. An article for using specific tree size, 2.0ms over in height makes it difficult to use a wide variety of different sizes of trees. And there is no incentive measures when larger trees are planted. To enhance the quality of green spaces and plantings, it is needed that the function and locating of green spaces and plantings have to be emphasized, and the planting density should be concerned about the mature tree size. The incentive measure to use various sizes of trees is also needed, and the regulations to use excessive number of evergreen trees or fruit trees should be loosened.

Detection of Xanthomonas axonopodis pv. citri on Citrus Fruits Using Enzyme-Linked Immunosorbent Assay

  • Detection of Xanthomonas axonopodis pv. citri (Xac) on citrus fruits for exporting is usually made by bacteriophage test (BPT) to demonstrate the pathogen-free status. BPT has rather time-consuming and complicate procedures for dealing with massive samples to be inspected. In this study, enzyme-linked immunosorbent assay (ELISA) was applied to detect Xac on fruits, and compared with BPT. In ELISA, positive reactions occurred in the bacterial densities of $3\times10^5$ cells/ml or more. To detect the bacterial infection on citrus fruits with a density of lower than $3\times10^5$ cells/ml, the bacterial suspensions were mixed with fruit rinse water and incubated in broth medium. Ordinary peptone sucrose broth (PSB) was not a proper medium for increasing Xac density specifically enough to be detect by ELISA. On the other hand, modified PSB (MPSP) amended with Fe-EDTA (0.25 g/$\ell$) and 2.5% potato-dextrose broth sufficed to differentiate uninfected and infected citrus fruits by ELISA after 24 h incubation of the fruit rinse water. Using various citrus samples from infected and uninfected fields, efficiencies in detecting Xac on fruits were compared between ELISA and BPT. For infected fruits samples, ELISA detected Xac by 100%, while BPT by about 44%, indicating that the detection efficiency was improved by 23.5% by ELISA, compared to BPT. In addition, ELISA has simpler procedures for testing and is less time-consuming than BPT, suggesting that ELISA may be accurate and simple method to detect Xac on citrus fruits.

통영 연안에 분포하는 거머리말의 계절변동과 생식특성 (Phenology of Zostera marina at Tongyeong in Southern Coast of Korea)

  • We investigated the variation of morphological features, density, biomass and characteristics of reproductive shoot on Zostera marina. This species was monthly monitored and collected during a year in Tongyeong (from January 2016 to December 2016). Morphological features, biomass and density showed significantly seasonal variation (p<0.001). Dimensions of Z. marina were highest in spring and early summer but lowest in winter. Biomass and leaf density of Z. marina showed highest value in May (4,700.5 g w·wt m-2) and June (858.0 leaves m-2) respectively and lowest value in November (515.9 g w·wt m-2 and 312.0 leaves m-2). Reproductive shoot was monitored from April (13℃) to June (21℃) and grew up to ca. 200 cm. Spathe length, spadix weight, numbers and weight of fruit showed significantly different on monthly. None mature flowers were monitored in April but matured flowers, fruits were founded in May and seed releasing were occurred in June.

Inhibition on LDL-oxidation by Phenolic Compounds from the Fruit Body of Phellinus linteus

  • The fruit body of Phellinus linteus was extracted with 80% aqueous MeOH, and the concentrated extract was successively partitioned using EtOAc, n-BuOH and $H_2O$. From the EtOAc and n-BuOH fractions, three phenolic compounds were isolated through repeated silica gel and ODS column chromatography. The chemical structures of these compounds were determined as 2-(3',4'-dihydroxyphenyl)-1,3-benzodioxole-5-aldehyde (1), 4-(3,4-dihydroxyphenyl)-3-buten-2-one (2), and protocatechuic acid methyl ester (3) by spectroscopic data including NMR, MS and IR. Compounds $1{\sim}3$ exhibited low density lipoprotein (LDL) antioxidant activity with $IC_{50}$ values of 1.7, 0.7, and $2.4{\mu}M$, respectively.

Effect of Chemical Fertilizer and Compost on Soil Physicochemical Properties, Leaf Mineral Content, Yield and Fruit Quality of Red Pepper (Capsicum annuum L.) in Open Field

  • Nowadays, sustainable and environment-friendly agriculture has become an important issue all around the world, and repeated applications of mineral and/or organic fertilizer will probably affect mineral nutrient dynamics in soil in the long term but only a limited number of observations are available. This study was carried out to investigate whether there is any influence of different fertilizer management for red pepper (Capsicum annuum L.) cultivation on soil physicochemical properties, leaf mineral content, yield and fruit quality in the aspect of long-term practice in open field condition. NPK, NPK+compost, compost only, and unfertilized control plot were included in the treatments. The application of chemical fertilizer and/or compost repeated annually for 17 years from 1994 to 2011. Soil organic matter content was higher in compost treatments than in no-manure treatments. Available phosphate and the yield of red pepper were highest in NPK+compost treatment followed by NPK (chemical fertilizer), compost, and control. The results indicate that in the long term, nitrogen supply is still needed for increasing red pepper yield, but reduction in the use of chemical fertilizer could be also possible with the proper application of compost.

주간 절단시기 및 생장조절제를 이용한 '후지'/M9 사과나무 수관 상단부 생장조절 (Growth Control of Upper Part in 'Fuji'/M.9 Apple Tree Canopy by Cutting Time of Trunk and Plant Growth Regulators)

  • BACKGROUND: The vigorous shoot growth in upper part of apple tree canopy leads to poor fruit quality and flower bud formation in lower part of canopy. So, this study was conducted to develop the proper control method about the shoot growth in upper part of apple tree canopy. METHODS AND RESULTS: Trunks of 'Fuji'/M9 apple trees were cut (back pruned) to 2.5 m in tree height on 11 February (dormant) or 12 April (full bloom). Naphthalene acetic acid (NAA) was applied at 2.0% to cut surface when trunk was pruned. Prohexadione-calcium (Pro-Ca) was sprayed at 250 mg/L above 2.0 m in tree height at 23 April (petal fall). The NAA or Pro-Ca application after trunk was pruned at dormant (TR-2 and TR-3) significantly reduced shoot growth in upper part of canopy compared with the control (tree was only pruned at dormant, TR-1), but the percent of shoots showing the secondary growth of TR-3 was higher over 2 times than that of TR-2. The reduction of shoot growth in upper part of canopy by TR-2 and TR-3 increased the fruit red color from the lower part in the treating year and blooming of the lower part in the following year. CONCLUSION: Applying 2.0% NAA to cut surface of pruned apple trunk at dormant was the most effective way for stabilization of the tree vigor in upper part of the canopy in a high density apple orchard.

지속적 농업을 위한 고성능 토양의 개발 연구-I. 계분 및 돈분퇴비의 연용이 방울토마토(Lycopersicum esculentum var. cerasiforme)의 생육 및 토양의 이화학성에 미치는 영향 (Study on the Improvement of Soil for High Efficient and Sustainable Agriculture-I. Effect of Repeated Application of Chicken and Pig Manure Composts on Tomato Growth and Soil Physico-chemical Properties)

  • 지속적농업을 위한 고성능토양을 개발하기 위하여 방울토마토를 공시작물로 계분 및 돈분퇴비를 3년간 연용하면서 방울토마토의 생육 및 수량, 그리고 연용후 토양이화학성을 조사하였다. 계분 및 돈분퇴비의 사용량과 시용 횟수가 증가할수록 토마토의 생육량 및 과실수량은 유의성 있게 증가하였으며, 뚜렷한 경향은 없으나 대체로 과실의 당도와 과실 개체당 중량 역시 증가하는 경향을 보였다. 그러나 계분 및 돈분퇴비만 사용할 경우 가비중과 경도는 현저히 낮고 공극율은 높았으며, 유기물과 치환성 칼륨의 함량은 높은 반면 치환성 칼슘의 함량이 낮아 일부 토양의 물리성과 화학성이 적정범위에 미달하거나 초과하는 불균형이 나타났다. 결론적으로 퇴비와 화학비료의 조화있는 활용을 통한 지력증진이 필요하다.

Effects of Ethyl Acetate Extract of Poncirus trifoliata Fruit for Glucocorticoid-Induced Osteoporosis

  • Poncirus trifoliata fruit (PTF) affects the digestive and cardiovascular systems, and kidney function. The authors studied the effects of ethyl acetate (EtOAc) extract of PTF on the activities of osteoblasts and in an animal model. The main compounds of the EtOAc extract, naringin and poncirin have been confirmed by HPLC and NMR analysis. Effects of osteoblastic differentiation were measured by alkaline phosphatase (ALP) activity, osteopontin (OPN) protein expression and osteoprotegerin (OPG) mRNA expression in MC3T3-E1 cells. Also, osteoclast differentiation was measured by multinucleated cells (MNCs) formation through tartrate resistance acid phosphatase (TRAP)-positive staining. Bone mineral density (BMD) was measured before and after treatment with EtOAc extract of PTF in prednisolone-induced osteoporotic mice. Dexamethasone (DEX) decreased OPN and OPG expression level in MC3T3-E1 cells and ALP activity was decreased by DEX dose-dependently. EtOAc extract of PTF recovered the levels of ALP activity, and the expression of OPN and OPG in MC3T3-E1 cells treated with DEX. In osteoclast differentiation, multinucleated TRAP-positive cell formation was significantly suppressed by the EtOAc extract of PTF. Total body BMD was restored by EtOAc extract of PTF in prednisolone-induced osteoporotic mice. In conclusion, EtOAc extract of PTF recovered DEX-mediated deteriorations in osteoblastic and osteoclastic functions, and increased BMD in glucocorticoid-induced osteoporosis.
