Effects Evodiae Fructus on the Blood Pressure in Spontaneously Hypertensive Rats (오수유가 선천성고혈압흰쥐의 혈압에 미치는 영향)

  • The present study examined the effect of a methanol extract of Evodiae Fructus on the blood pressure in spontaneously hypertensive rats (SHR). The systolic blood pressure was measured after rats were pretreated with phentolamine, propranolol, or $N_{\omega}$-nitro-$_{L}$-arginine methyl ester(NAME) and subsequently received methanol extract of Evodiae Fructus. In SHR, intraperitoneal administration of methanol extract of Evodiae Fructus (0.5 mg/kg) produced antihypertensive effect that lasted for at least 4 hours. Antihypertensive effect of Evodiae Fructus was more stronger than that with $\alpha$-adrenergic receptor antagonist phentolamine and was not affected by $\beta$-adrenergic receptor antagonist propranolol. The antihypertensive effect of Evodiae Fructus was abolished by pretreatment of NAME. Our findings suggest Evodiae Fructus has an hypertensive effect, which may be mediated through nitric oxide synthesis.s.

  • Objective : This research was carried out to compare the effect of Gorynju and a Soju extract Psoraleae fructus on melanin synthesis of B16 melanoma cells. Methods : To investigate melanin synthesis of B16 melanoma cells, this research was measured cell survival, tyrosinase activity, melanin synthesis, western blot. Results : Both Gorynju and Soju extract Psoraleae fructus, cell toxicity depended on the density. Tyrosinase activity depended on the density of Gorynju extract Psoraleae fructus and statistic was showed significant(0.5, 1, 2, 3 ${\mu}g/ml$), in a Soju extract Psoraleae fructus, 1 ${\mu}g/ml$ were showed significant. Melanin synthesis was showed significant in a Soju extract Psoraleae fructus(3, 4 ${\mu}g/ml$). Western blot was showed to depend on the density of Gorynju and a Soju extract Psoraleae fructus. Conclusions : In a tyrosinase activity and a melanin synthesis, the intermediate alcohol of Gorynju and a Soju may be suitable to use.

  • We sometimes directed taking medicine by using decoction of Jujubae Fructus in taking herbs medicine. And I studied meaning on using decoction of Jujubae Fructus in taking herbs medicine. The obtained results were follows: 1. In taking medicine by using decoction of Jujubae Fructus, chief effect positions of that prescription were heart, spleen, stomach, and kidney. 2. In taking medicine by using decoction of Jujubae Fructus, chief treatment symptoms were heart throb, sleeplessness, reddish turbid urine, retention of phlegm and fluid, retention of fluid in the body, weakness. 3. In taking medicine by using decoction of Jujubae Fructus, chief pathogenic factors of that prescription were deficiency syndroms of the heart, retention of phlegm and fluid, retention of fluid in the body, consumptive disease, weakness.

Inhibitory Effects of Gardeniae Fructus on Apoptosis induced by 4-HNE in PC-12 Cell (치자(梔子)의 Apoptosis유발 억제효과)

  • Objectives : The purpose of this study was to observe the effects of Gardeniae Fructus on 4-HNE-induced apoptosis in PC-12 cell. Methods : A MTT assay was conducted to observe the cytotoxicity of Gardeniae Fructus on the PC-12 cell viability and the cytoprotective effects of Gardeniae Fructus on PC-12 cell against oxidative stress caused by 4-HNE. And western blot was conducted to observe the expression of Bax, Bcl-2, Caspase-3, $TNF-{\alpha}$ proteins which are involved in intrinsic and extrinsic apoptosis pathway. Results : 25, 50, 100, 200 and $400{\mu}g/m{\ell}$ of Gardeniae Fructus water extract had no cytotoxicity on PC-12 cell. $200{\mu}g/m{\ell}$ of Gardeniae Fructus water extract had significant cytoprotective effect on PC-12 cell against oxidative stress caused by 4-HNE. The expression of Bax protein in 50, 100 and $200{\mu}g/m{\ell}$ of Gardeniae Fructus was significantly decreased in PC-12 cell. The expression of Bcl-2 protein in $200{\mu}g/m{\ell}$ of Gardeniae Fructus was significantly increased in PC-12 cell. The expression of Caspase-3 protein in 100 and $200{\mu}g/m{\ell}$ of Gardeniae Fructus was significantly decreased in PC-12 cell. The expression of $TNF-{\alpha}$ protein in $50{\mu}g/m{\ell}$ of Gardeniae Fructus was significantly decreased in PC-12 cell. Conclusions : These results suggest that Gardeniae Fructus water extract is effective to protect PC-12 cell from 4-HNE induced apoptosis.

Protective Effect of Aurantii Immaturus Fructus on Hypoxia Reperfusion Induced by PC12 Cell Damage and Global Ischemia in Gerbil (PC12 손상 세포 및 전뇌허혈 유발 Gerbil에 대한 지실의 세포보호효과 연구)

  • Object : This research was performed to investigate the protective effect of Aurantii Immaturus Fructus against ischemic damage using PC12 cells and global ischemia in gerbils. Methods : To observe the protective effect of Aurantii Immaturus Fructus on ischemia damage, viability and changes in activities of superoxide dismutase (SOD), glutathione peroxidase (GPx), catalase and production of malondialdehyde (MDA) were observed after treating PC12 cells with Aurantii Immaturus Fructus during ischemic insult. Gerbils were divided into three groups : a normal group, a 5-min two-vessel occlusion (2VO) group, and an Aurantii Immaturus Fructus administered after 2VO group. The CCAs were occluded by microclip for 5 minutes. Aurantii Immaturus Fructus was administered orally for 7 days after 2VO. The histological analysis was performed at 7 days after the surgery. For histological analysis, the brain tissue was stained with 1% cresyl violet solution. Results : The results showed that 1. Aurantii Immaturus Fructus had a protective effect against ischemia in the CAI area of the gerbil hippocampus 7 days after 5-minute occlusion, 2. In the hypoxia/reperfusion model using PC12 cells, the Aurantii Immaturus Fructus had a protective effect against ischemia in the dose of $0.2{\;}\mu\textrm{g}/ml,{\;}2{\;}\mu\textrm{g}/ml{\;}and{\;}20{\;}\mu\textrm{g}/ml$ 3. Aurantii Immaturus Fructus increased the activities of glutathione peroxidase and catalase, 4. The activity of superoxide dismutase (SOD) was increased by ischemic damage, which might represent self protection. This study suggests that Aurantii Immaturus Fructus has some neuroprotective effect against neuronal damage following cerebral ischemia in vivo with a widely used experimental model of cerebral ischemia in Mongolian gerbils, and it also has protective effects on a hypoxia/reperfusion cell culture model using PCq2 cells. Conclusions : Aurantii Immaturus Fructus has protective effects against ischemic brain damage at the early stage of ischemia.

Daily Dose of Zizyphi Fructus in Treatise on Cold Damage (상한론(傷寒論)에서 대조(大棗)의 1 일 복용량)

  • Objectives : As Daily Dose of Zizyphi Fructus was designated by the numbers in Treatise on Cold Damage Disease, estimation of Daily Dose was all different among researchers. The purpose of this study was to estimate exact Daily Dose of Zizyphi Fructus. Methods : We fixed the errors in various copy of Treatise on Cold Damage Disease and considered the meaning of the Bee Zizyphi Fructus(肥大棗) and general rules of Daily Dose in Treatise on Cold Damage Disease. So we reduced Daily Dose of Zizyphi Fructus, and compared this with the standard of Zizyphy Fructus in Pharmacopoeia of several Nation and Korean Forest Service. Results : Daily Dose of Zizyphi Fructus was generally 12 pieces; less was for prescriptions which scaled down the amount of ingredients prescribed in the originals; 15, 25, and 30 pieces were used when more was required. The medicinal part was the pulp of fructus, and the dosage of 12 pieces was respectively equivalent to 3 Ryang(兩), and 19.5 g. As defined in the Korean Pharmacopoeia Ninth Edition and standards of forest products by Korea Forest Service, Zizyphi Fructus was medium-sized, and weighs about 1.625 g if properly dehydrated. Conclusions : In short, the proper Daily Dose of Zizyphi Fructus in Treatise on Cold damage Disease was 12 pieces of Zizyphi Fructus and 19.5 g of its pulp, weighing three Ryang(兩). The pulp was estimated to be 1.625 g; it was medium-sized according to the present standard.

Antioxidant and Antihemolytic Activities of Ethanol Extracts of Carpesii Fructus and Farfarae Flos (학슬 및 관동화 에탄올 추출물의 항산화 및 항용혈 효과)

  • Objectives : To develop a natural antioxidant and anti-hemolytic agents, we investigated the effects of ethanol extracts of Carpesii Fructus and Farfarae Flos. Methods : Aerial parts of Carpesii Fructus and Farfarae Flos were extracted with 80% ethanol. Antioxidant activity of Carpesii Fructus or Farfarae Flos extract was evaluated by employing three different assays, i.e., 1,1-diphenyl-2-picryl hydrazyl (DPPH), 2,2'-azino-bis (3-ethylbenzthiazoline-6-suphonic acid) diammonium (ABTS) scavenging and reducing power activities. Also, anti-hemolytic activity of Carpesii Fructus or Farfarae Flos extract was determined using [2,2'-azobis (2-methylpropionamidine) dihydrochloride (AAPH)]-induced hemolysis, glutathione (GSH) depletion and malondialdehyde (MDA) formation in normal rat red blood cells (RBC) or plasma. Results : The extracts obtained from Carpesii Fructus and Farfarae Flos dose-dependently increased the scavenging activity on DPPH- or ABTS-induced radicals and the reducing power activities. Carpesii Fructus and Farfarae Flos were similar to the scavenging activity and the reducing power of butylated hydroxy anisole effect at high concentration ($1,000{\mu}g/mL$). RBC oxidative hemolysis and plasma MDA formation induced by AAPH were significantly suppressed by the extracts of Carpesii Fructus and Farfarae Flos in a dose-dependent manner. Also, Carpesii Fructus and Farfarae Flos extracts prevented the depletion of cystosolic antioxidant GSH in RBCs. Carpesii Fructus generally had better than the free radical scavenging activity, the reducing power and anti-hemolytic effects of Farfarae Flos. Conclusions : These results suggest that Carpesii Fructus and Farfarae Flos may have value as the potential antioxidant and anti-hemolytic medicinal plant.

Antimicrobial Activity of Streptococcus mutans by Schizandrae fructus and Evodiae fructus extracts (오미자, 오수유 추출물의 Streptococcus mutans에 대한 항균효과)

  • This study was to investigate the antibacterial effect of Schizandrae fructus and Evodiae fructus extracts on S. mutans growth. The antibacterial effect of these extracts on S. mutans was good at Schizandrae fructus extract than the Evodiae fructus extract. The antibacterial activities of these extracts were measured 8.5 mm and 6.5 mm at 20 mg/ml concentration, respectively. The growth of S. mutans in control medium was the highest at 8 hrs, while the media of Schizandrae fructus and Evodiae fructus extracts added-medium(2 mg/ml) showed maximum growth at 16hrs. The pH values of the control medium was 5.08 at 8 hrs, but the media supplemented with Schizandrae fructus and Evodiae fructus extracts were 6.00, and 5.36 at 8 hrs, respectively. The amounts of total carbohydrate for the control media was 0.81 mg/ml at 8 hrs, but the media supplemented with Schizandrae and Evodiae fructus extracts were 1.32 mg/ml and 1.88 mg/ml at 8 hrs, respectively. In the change of culture medium proteins, the control culture broth and cultures supplemented with Schizandrae fructus and Evodiae fructus extracts were 8.39 mg/ml, 8.59 mg/ml and 9.97 mg/ml at 8 hrs, respectively. The S. mutans polysaccharide contents of the control medium and the media supplemented with Schizandrae and Evodiae fructus extracts were 300 mg/100 ml, 240 mg/100 ml and 280 mg/100 ml at 8 hrs, respectively.

Antidiabetic and Antioxidative Effects of Corni fructus In Streptozotocin-Induced Diabetic Rats (산수유 추출물이 Streptozotocin으로 유발된 흰쥐의 항당뇨 및 항산화 작용에 미치는 효과)

  • The extraction yield of Corni fructus was about 47.5% by extract apparatus. This study was done to investigate the antidiabetic and antioxidative effects of Corni fructus in Streptozotocin (STZ)-induced diabetic rats. The contents of serum glucose, total cholesterol and hepatic lipid peroxide, glutathione were significantly decreaed (p<0.05) in Corni fructus treated group compared to the those of STZ-control group, also content of triglyceride (TG), atherogenic index (AI) and activity of catalase were decreased, but not statistically significant. High density lipoprotein (HDL)-cholesterol and HDL-cholesterol/total cholesterol ratio (HTR) were increased in Corni fructus treated group compared to the those of STZ-control group. The activities of glutathione-S-transferase (GST) and superoxide dismutase (SOD) were significantly decreaed (p<0.05) in Corni fructus treated group compared to the those of STZ-control group. The content of hepatic glycogen and activities of glucose-phosphate dehydrogenase (G-6 PDH), glucokin-ase were significantly increased, but activity of glucose-Grphosphatase (G-6 Pase)was decreased in Corni fructus treated group compared to the those of STZ-control group. Therefore, these results indicated that ethanol extract of Corni fructus would have antidiabetic and antioxidative effects in STZ-induced diabetic rats.
