Cronobacter sakazakii (C. sakazakii) is a foodborne pathogen, posing a high risk of disease to infants and immunocompromised individuals. In order to develop a quick, easy, and sensitive assay for detecting C. sakazakii, a rabbit anti-C. sakazakii immunoglobulin G (IgG) was developed using sonicated cell protein from C. sakazakii. The developed anti-C. sakazakii (IgG) was of good quality and purity, as well as species-specific. The developed rabbit anti-C. sakazakii IgG was attached to the surface of a sulforhodamine B-encapsulated liposome to form an immunoliposome. A test strip was then prepared by coating goat anti-rabbit IgG onto the control line and rabbit anti-C. sakazakii IgG onto the test line, respectively, of a plastic-backed nitrocellulose membrane. A purple color signal both on the test line and the control line indicated the presence of C. sakazakii in the sample, whereas purple color only on the control line indicated the absence of C. sakazakii in the sample. This immunochromatographic strip assay could produce results in 15 min with a limit of detection of $10^7CFU/ml$ in C. sakazakii culture. The immunochromatographic strip assay also showed very good specificity without cross-reactivity with other tested Cronobacter species. Based on these results, the developed immunochromatographic strip assay is efficient for the detection of C. sakazakii and has high potential for on-site detection.
The complex roles of cell surface modification in the biofilm formation of Campylobacter jejuni, a major cause of worldwide foodborne diarrheal disease, are poorly understood. In a screen of mutants from random transposon mutagenesis, an insertional mutation in the eptC gene (cj0256) resulted in a significant decrease in C. jejuni NCTC11168 biofilm formation (<20%) on major food contact surfaces, such as polystyrene and borosilicate glass, when compared with wild-type cells (p < 0.05). In C. jejuni strain 81-176, the protein encoded by eptC modified cell surface structures, such as lipid A, the inner core of lipooligosaccharide, and the flagellar rod protein (FlgG), by attaching phosphoethanolamine. To assess the role of eptC in C. jejuni NCTC11168, adherence and motility tests were performed. In adhesion assays with glass surfaces, the eptC mutant exhibited a $0.77log\;CFU/cm^2$ decrease in adherence compared with wild-type cells during the initial 2 h of the assay (p < 0.05). These results support the hypothesis that the modification of cell surface structures by eptC affects the initial adherence in biofilm formation of C. jejuni NCTC11168. In motility tests, the eptC mutant demonstrated reduced motility when compared with wild-type cells, but wild-type cells with the transposon inserted in a gene irrelevant to biofilm formation (cj1111c) also exhibited decreased motility to a similar extent as the eptC mutant. This suggests that although eptC affects motility, it does not significantly affect biofilm formation. This study demonstrates that eptC is essential for initial adherence, and plays a significant role in the biofilm formation of C. jejuni NCTC11168.
Vibrio parahaemolyticus is recognized as one of the most important foodborne pathogens responsible for gastroenteritis in humans. The blaCARB-17 gene is an intrinsic β-lactamase gene and a novel species-specific genetic marker of V. parahaemolyticus. In this study, a loop-mediated isothermal amplification (LAMP) assay combined with a lateral flow dipstick (LFD) was developed targeting this blaCARB-17 gene. The specificity of LAMP-LFD was ascertained by detecting V. parahaemolyticus ATCC 17802 and seven other non-V. parahaemolyticus strains. Finally, the practicability of LAMP-LFD was confirmed by detection with V. parahaemolyticus-contaminated samples and natural food samples. The results showed that the optimized reaction parameters of LAMP are as follows: 2.4 mmol/l Mg2+, 0.96 mmol/l dNTPs, 4.8 U Bst DNA polymerase, and an 8:1 ratio of inner primer to outer primer, at 63℃ for 40 min. The optimized reaction time of the LFD assay is 60 min. Cross-reactivity analysis with the seven non-V. parahaemolyticus strains showed that LAMP-LFD was exclusively specific for V. parahaemolyticus. The detection limit of LAMP-LFD for V. parahaemolyticus genomic DNA was 2.1 × 10-4 ng/μl, corresponding to 630 fg/reaction and displaying a sensitivity that is 100-fold higher than that of conventional PCR. LAMP-LFD in a spiking study revealed a detection limit of approximately 6 CFU/ml, which was similar with conventional PCR. The developed LAMP-LFD specifically identified the 10 V. parahaemolyticus isolates from 30 seafood samples, suggesting that this LAMP-LFD may be a suitable diagnostic method for detecting V. parahaemolyticus in aquatic foods.
Toxoplasma gondii, an intracellular protozoan parasite that infects one-third of the world's population, has been reported to hijack host cell apoptotic machinery and promote either an anti- or proapoptotic program depending on the parasite virulence and load and the host cell type. However, little is known about the regulation of human FHs 74 small intestinal epithelial cell viability in response to T. gondii infection. Here we show that T. gondii RH strain tachyzoite infection or ESP treatment of FHs 74 Int cells induced apoptosis, mitochondrial dysfunction and ER stress in host cells. Pretreatment with 4-PBA inhibited the expression or activation of key molecules involved in ER stress. In addition, both T. gondii and ESP challenge-induced mitochondrial dysfunction and cell death were dramatically suppressed in 4-PBA pretreated cells. Our study indicates that T. gondii infection induced ER stress in FHs 74 Int cells, which induced mitochondrial dysfunction followed by apoptosis. This may constitute a potential molecular mechanism responsible for the foodborne parasitic disease caused by T. gondii.

Importance: Salmonella outbreaks linked to poultry meat have been reported continuously worldwide. Therefore, Salmonella contamination of poultry meats in slaughterhouses is one of the critical control points for reducing disease outbreaks in humans. Objective: This study examined the carry-over contamination of Salmonella species through the entire slaughtering process in South Korea. Methods: From 2018 to 2019, 1,097 samples were collected from the nine slaughterhouses distributed nationwide. One hundred and seventeen isolates of Salmonella species were identified using the invA gene-specific polymerase chain reaction, as described previously. The serotype, phylogeny, and antimicrobial resistance of isolates were examined. Results: Among the 117 isolates, 93 were serotyped into Salmonella Mbandaka (n = 36 isolates, 30.8%), Salmonella Thompson (n = 33, 28.2%), and Salmonella Infantis (n = 24, 20.5%). Interestingly, allelic profiling showed that all S. Mbandaka isolates belonged to the lineage of the sequence type (ST) 413, whereas all S. Thompson isolates were ST292. Moreover, almost all S. Thompson isolates (97.0%, 32/33 isolates) belonging to ST292 were multidrug-resistant and possessed the major virulence genes whose products are required for full virulence. Both serotypes were distributed widely throughout the slaughtering process. Pulsed-field gel electrophoretic analysis demonstrated that seven S. Infantis showed 100% identities in their phylogenetic relatedness, indicating that they were sequentially transmitted along the slaughtering processes. Conclusions and Relevance: This study provides more evidence of the carry-over transmission of Salmonella species during the slaughtering processes. ST292 S. Thompson is a potential pathogenic clone of Salmonella species possibly associated with foodborne outbreaks in South Korea.
이 연구에서는 식중독 발생건수를 원인물질별로 나눈 자료와 합한 자료를 별개로 분석하여 예측값을 유도한 후 계층구조를 만족하도록 하는 계층 시계열 예측에 대해 알아본다. 원인물질별 식중독 방생건수는 영과잉 포아송 회귀모형과 음이항 회귀모형으로 분석하고 합한 식중독 발생건수 포아송 회귀모형과 음이항 회귀모형으로 분석한다. 계층 시계열 예측을 위해 최적결합 중 하나인 Wickramasuriya 등 (2019)의 MinT 추정이 사용되었다. 계층조정 과정에서 발생한 음의 예측값은 0으로 수정하고 나머지 최하위 변수에 가중치를 곱해 계층구조를 만족시킨다. 실증분석 결과를 보면 원인물질별 예측에서는 계층조정을 한 결과와 하지 않은 결과에 차이가 거의 없었으나 주요, 기타 및 전체에 대한 예측에서는 계층조정 한 결과가 대체로 우수한 것으로 나타났다. 중요한 것은 계층조정을 하지 않으면 최하위 변수의 예측빈도가 주요나 기타의 예측빈도 보다 큰 경우도 발생하지만 제안된 방법을 적용하면 계층구조를 이루는 예측값을 얻을 수 있다.
Hepatitis E Virus (HEV) infection is a worldwide disease and the primary cause of acute viral hepatitis in the world. It can be isolated from many different species including pigs. HEV is a zoonotic pathogen and foodborne disease. The main animal reservoir is domestic pigs. It is usually asymptomatic in pig but it is a public health concern, causing acute hepatitis in humans of varying severity. This study focused on the presence of HEV in pig and pork product. One hundred feces and one hundred fifty serum samples were randomly collected from pigs in slaughterhouses in Gwangju from November in 2018 to February in 2020. In addtion, seventy-five pork products were collected from markets in Gwangju. Feces and pork product samples were examined for the presence of HEV RNA using an reverse-transcription realtime PCR (RT-qPCR) assay. Serum samples were tested for the presence of HEV-specific IgG antibodies using Enzyme-linked immunosorbent assay (ELISA). HEV antigen and antibody positive rates were 3.0% (3/100) and 19.3% (29/150), respectively, in Gwangju and nearby areas such as Jeonnam and Jeonbuk. However, HEV antigen was not detected from any of pork product in this study. In conclusion, the prevalence of HEV should be continuously monitored because HEV was sporadically detected in Gwangju and nearby areas.
임상적으로 감염 시 치명적인 뇌염이나 유산을 일으키는 병원체인 톡소포자충의 치료제로 주로 사용되고 있는 설파디아진이 사용되고 있으나, 이들 약물들은 부작용과 약물 내성에 의한 치료 실패 사례들이 자주 보고되고 있다. 이러한 이유로 톡소포자충에 대한 새로운 치료제의 개발 필요성이 대두되고 있으며, 합성 화학약물들에 비교하여 부작용이 적고 안전성이 높은 천연물들을 원료로 한 톡소포자충의 치료가 모색되고 있다. 본 연구는 약용 작물로서 보존 활용가치가 높을 것으로 예상되는 염생식물인 해홍나물 에탄올 추출물을 이용하여 톡소플라즈마 원충에 대한 항원충 효과를 알아 보고자 수행 되었다. 본 연구 결과, 해홍나물 에탄올 추출물은 톡소포자충의 치료제로 임상에서 실제 톡소포자충의 치료로 사용되고 있는 설파다이아진의 선택성 2.06 보다 해홍나물 에탄올 추출물의 선택성이 6.63으로 매우 높은 것을 알 수 있었다. 또한 해홍나물 에탄올 추출물이 용량 의존적으로 톡소포자충에 감염된 HeLa 세포수를 감소시키는 것을 확인하였다. 톡소포자충에 감염된 세포에 해홍나물 에탄올 추출물을 농도별로 처리하여 감염 세포들의 형태학적 변화를 관찰한 결과, 해홍나물 에탄올 추출물의 적용 농도가 높아질수록 톡소포자충 감염에 의한 세포의 형태적 변화를 감소시키는 것을 알 수 있었다. 이러한 결과로부터 해홍나물은 향후 톡소포자충 치료에 합성 약물인 설파다이아진을 대체할 수 있는 내성이 없는 천연물 소재로 개발될 수 있을 것으로 판단되었다.

민간요법 또는 식품가공 공정에서 그 안정성이 확보되어 생약재료 또는 식품가공재료로서 사용되고 있는 약용식물에 대하여 항균력을 검색하였고 그중 가장 항균력을 나타낸 황련을 사용하여 활성성분을 분획 추출물한 다음 이 추출물로 부터 여러 종류의 부패 미생물 및 병원성균에 대한 항균성을 검색하였다. 산사, 황련, 측백, 창출 및 석창포 에탄올 추출물은 그람 양성 또는 음성세균 모두에 대하여 강한 항균성을 나타내었으며, 토사자 에탄올추출물은 그람음성 세균에서만 항균성을 나타내었고 사군자, 숙지황 및 백제 에탄올추출물은 거의 또는 모든 시험균에 대하여 항균력을 나타내지 않았다. 황련의 에탄올추출물은 기존에 보고된 많은 한약제의 에탄올추출물과 비교하여 볼 때 적은 농도에서도 매우 다양하고 강한 항균력을 나타내었으며 특히 그람양성 또는 음성세균 모두 강한 항균력을 보였다. 한편, 황련 에탄올 추출물은 $100^{\circ}C$에서 각각 30분 또는 1시간 동안, $121^{\circ}C$에서 15분간 열처리한 후 Listeria monocytogenes균에 대한 생육저해환을 측정한 결과 상기의 모든 열처리에 의해서도 무열처리군과 비교하여 볼 때 항균력의 차이가 없어 열에 매우 안정 하였다. 또한 생육배지의 pH 변화가 Listeria monocytogenes균의 생육억제에 대한 황련 에탄올 추출물의 항균활성에 대하여 미치는 영향을 조사한 결과 배양배지의 pH 가 중성에서 알칼리성으로 갈수록 황련 에탄올 추출물의 항균활성은 더욱 증가되었고 약산성으로 갈수록 감소하는 경향을 나타내었다. 따라서 황련 에탄올추출물은 알칼리성 조건하에서 더욱 강한 항균력을 나타내었다. 황련 에탄올추출물로부터 부분정제한 물질의 항균력은 검색된 미생물중 그람양성세균, 그람음성세균, 효모 및 곰팡이균 모두에 항균력을 나타내었으나 Candida utilis, Rhizopus javanicus에는 억제효과를 나타내지 않았다. 식중독세균인 Listeria monocytogenes과 Staphylococcus aureus에 대한 부분정제물질의 생육저해 효과를 조사한 결과 두균주 모두 $100{\;}{\mu}g/mL$의 농도에서는 무첨가 대조균과 비교하여 차이가 없었으며 $500{\;}{\mu}g/mL$ 첨가구에서는 Listeria monocytogenes균은 36시간까지는 생육을 억제하였으나 그 이후부터는 급속한 생육을 하였고 Staphylococcus aureus균은 완전히 생육이 억제되었고 $1000{\;}{\mu}g/mL$ 첨가구에서는 두 균주 모두 완전히 생육이 억제되었다.

제 18 조 (손해배상)
당 사이트는 무료로 제공되는 서비스와 관련하여 회원에게 어떠한 손해가 발생하더라도 당 사이트가 고의 또는 과실로 인한 손해발생을 제외하고는 이에 대하여 책임을 부담하지 아니합니다.
제 19 조 (관할 법원)
서비스 이용으로 발생한 분쟁에 대해 소송이 제기되는 경우 민사 소송법상의 관할 법원에 제기합니다.

1. (시행일) 이 약관은 2016년 9월 5일부터 적용되며, 종전 약관은 본 약관으로 대체되며, 개정된 약관의 적용일 이전 가입자도 개정된 약관의 적용을 받습니다.
