Effects of Dietary Protein and Lipid Levels on Growth and Body Composition of Sub-adult Flounder Paralichthys olivaceus During the Summer Season

  • A $3{\times}2$ factorial experiment was conducted to investigate the proper dietary protein and lipid levels for the growth of sub-adult flounder Paralichthys olivaceus reared during the summer season. Six experimental diets were formulated to contain three levels of protein (45%, 50% and 55%) and two levels of lipid (9% and 14%). Duplicate groups of fish (initial body weight of 298 g) were hand-fed to apparent satiation during the summer season ($21.8{\pm}1.7^{\circ}C$) for 60 days. Survival of each group was over 83% and there was not significant difference among all groups. Weight gain of fish fed the 45% protein diet with 14% lipid was not significantly different from that of fish fed the 50% and 55% protein diets with 9% and 14% lipids, but weight gain of fish fed the 45% protein diet with 9% lipid was significantly lower than that of fish fed the 55% protein diets with 14% lipid. Feed efficiency tended to increase with increasing dietary lipid level at each protein levels, although no significant differences were observed at 50% and 55% protein levels. Protein efficiency ratio, daily feed intake, condition factor and hepatosomatic index were not significantly affected by dietary protein and lipid levels. Crude lipid content of the liver tended to increase with increasing dietary lipid level at the same protein levels, but the opposite appearance was found for moisture content. The contents of moisture, crude protein and crude lipid of the dorsal muscle were not significantly affected by dietary protein and lipid levels. Based on data obtained form this study, inclusion of dietary protein at level of 45% appears sufficient to support optimal growth, and an increase of dietary lipid level from 9% to 14% has beneficial effects on feed utilization of sub-adult flounder during the summer season.

Effects of supplemental undaria powder, herb and wasabi in the diets on growth, body composition, blood chemistry and non-specific immune response of juvenile flounder, Paralichthys olivaceus

  • This study was conducted to investigate the effects of several additives(macroalgae, wasabi, and herb) in formulated diets on the growth, body composition, blood chemistry and non-specific immune response of juvenile flounder. Three replicates of juveniles (average weight 8.4 g) in flow-through aquarium system were fed one of six isonitrogenous (45%) and isolipidic (8%) diets containing 5 and 10% Undaria powder, 2% wasabi leaf, 2% wasabi stem, and 0.5% herb (Obosan) for 8 weeks. Survival was not affected by the different dietary additives (P>0.05). The highest weight gain and feed efficiency offish fed the diet containing 0.5% herb were significantly higher than those of fish fed the diets containing 10% Undaria powder (P<0.05). No significant differences were found in contents of moisture, crude protein, lipid and ash of whole body (P>0.05). Fish fed the diet containing 10% Undaria powder showed the highest moisture and the lowest crude lipid contents in the liver. Although hematological parameters (red blood cell, hematocrit and hemoglobin) and serum constituents (glucose, total cholesterol and glutamic-oxaloacetic transaminase) contents of fish varied between treatments, no specific trend was observed throughout feeding periods. Lysozyme activity in the serum and nitroblue tetrazolium reduction of macrophage in the head kidney from fish fed the diets containing herb was significantly higher than those of fish fed the control diet. The results of this study suggest that herb as an additive in this formulated diet may improve growth and non-specific immune response of juvenile flounder.

치어기 넙치의 면역 증강에 대한 셀레늄의 적정 첨가량 평가 (Evaluation of the Optimum Dietary Selenium (Se) Level to Improve Immune Responses in Juvenile Olive Flounder, Paralichthys olivaceus)

  • This experiment was conducted to examine the utilization of added dietary selenium (Se) as an immune stimulant in juvenile olive flounder, Paralichthys olivaceus. Fish averaging $4.0{\pm}0.1\;g$ ($mean{\pm}SD$) were fed one of seven semi-purified diets containing 0.56, 1.07, 2.86, 4.56, 43.15, 90.71, or 161.74 mg of Se/kg ($Se_{0.56}$, $Se_{1.07}$, $Se_{2.86}$, $Se_{4.56}$, $Se_{43.2}$, $Se_{90.7}$ and $Se_{161.7}$, respectively) for 12 weeks, respectively. At the end of the feeding trial, the fish fed diets containing more than 43.2 mg of Se/kg showed above 90% mortality. There were no significant differences in weight gain, feed efficiency, specific growth rate, protein efficiency ratio, or hematological characteristics among the fish fed the $Se_{0.56}$, $Se_{l.07}$, $Se_{2.86}$, and $Se_{4.56}$ diets. Se concentrations of the gill, kidney, muscle and liver tissues occurred in dose-dependent manners. Alternative complement pathway activation and the chemiluminescene responses of the fish fed the $Se_{1.07}$ diet were significantly higher than those of the fish fed the other diets (P<0.05). These results indicate that the optimum dietary supplementation level of Selenium as selenoyeast could be 1.07 mg of Se/kg based on the non-specific immune responses of juvenile oilve flounder, Paralichthys olivaceus.

부산 일대에서 시판되는 붕장어의 Anisakis 유충 감염상 (Infection status of larval anisakids in Astroconger muyias.for collected from the Southern Sea near Pusan)

  • 인체 소차관의 급성 질환 띤 만성으로 호산구성 육아종증을 일으키는 것으로 알려져 있는 Anisakis류에 있어서 중간 숙주가 되는 우리나라 남해안산 허산 어류 중 부산 각 지역별 광안리, 자갈치, 송도와 다대포에서 붕장어를 구입하여 유충류를 채집한 후 헙대학적인 조사를 시도하였던 바 Anisakis type과 Contracaecum type의 두가지 유충을 검출하였다. 붕장어 382마리 중 259마리가 양성으로 67.8%의 감염률이었으며 붕장어 마리당 감염를은 전체 충체 1,768마리로써 4.6마리였다. 넉장어의 크기에 따른 선충류의 감염률은 꽐수록 증가되었다. 통장어의 각 장기별 선충류의 감염률은 망 1,430(80.8%), 소장 166(9.38%), 위장 107(6.0%), 퍼부 32(1.8%), 근육 18(1.0%), 간장 13(0.7%) 그리고 두부 2(0.1%)의 순위이었다. 이상의 결과에 의하면 붕장어회의 생식으로 Anisakis류의 인체 감염의 가능성이 충분하다고 판단된다.

Suppression of Fatty Acid Synthase by Dietary Polyunsaturated Fatty Acids is Mediated by Fat itself, not by Peroxidative Mechanism

  • This study examined the effect of dietary polyunsaturated fatty acids (PUFA) that were supplemented with vitamin E on lipid peroxidation, glutathione-dependent detoxifying enzyme system activity, and lipogenic fatty acid synthase (FAS) expression in rat liver. Male Sprague-Dawley rats were fed semipurified diets containing either 1% (w/w) corn oil or 10% each of beef tallow, corn oil, perilla oil, and fish oil for 4 wk. Alpha-tocopherol was supplemented in perilla oil (0.015%) and fish oil (0.019%). Hepatic thiobarbituric acid reactive substances, an estimate of lipid peroxidation, were not significantly different among the dietary groups. The glutathione peroxidase, glutathione reductase, and glutathione S-transferase activities were all elevated by the polyunsaturated fats, especially fish oil. The activity of FAS was reduced in the polyunsaturated fat-fed groups in the order of fish oil, perilla oil, and corn oil. The mRNA contents decreased in rats that were fed the 10% fat diets, particularly polyunsaturated fats, compared with the rats that were fed the 1% corn oil diet. Similarly, the inhibitory effect was the greatest in fish oil. These results suggest that lipid peroxidation can be minimized by vitamin E; PUFA in itself has a suppressive effect on lipogenic enzyme.

Norcardia 감염증에 의한 양식 가물치의 대량 폐사 (Mass Mortality Caused by Nocardial Infection in Cultured Snakehead, Channa arga in Korea)

  • 2005년 여름에 국내에서 사육되는 가물치에서 대량폐사를 수반하는 새로운 질병이 발견되었다. 감염 어류는 복부팽만과 항문 주위 출혈 이외의 특이한 외부 증상을 나타내지 않았으나 복부를 절개하면 많은 수의 백색 결절이 간, 비장 및 신장을 포함한 내부 장기에서 관찰되었다. 특히 내부 장기의 조직병리학적 관찰에서 granuloma가 관찰되었다. 이들 감염 어류에서 Gram 양성의 사상균이 분리되었다. 분리균은 Nocardia spp.와 non-Nocardia 균의 16S rRNA 염기 분석에 기초한 Nocardia 특이 primer를 사용한 PCR 방법으로 확인하였다. 이것은 어류에서 발생한 Nocardia 감염증의 국내 첫 사례이다.

저수온기 참돔 치어의 성장 및 체조성에 미치는 사료내 지질의 영향 (Effects of Dietary Lipid Level on Growth and Body Composition of Juvenile Red Sea Bream (Pagurus major) during Winter)

  • This study was conducted to investigate effects of dietary lipid level on growth and body composition of juvenile red sea bream in low temperatures. Duplicate groups of fish (initial body weight of 79 g) were fed one of three isonitronic diets (47% crude protein) containing different lipid levels (10%, 17% and 22%) for 25 weeks during the winter season. Weight gain and survival of fish fed 17% lipid diet were significantly (P<0.05) higher than those fish fed the 10% or 22% lipid diet. Protein efficiency ratio, daily feed intake, condition factor, hepatosomatic index and viscerasomatic index were not affected by dietary lipid level, but feed efficiency of fish fed 10% lipid diet was significantly (P<0.05) lower than those fish fed the 17% or 22% lipid diet. Proximate composition of the whole body, liver, viscera and dorsal muscle were not significantly different among all groups except for crude protein content of dorsal muscle. The contents of 16:0, 18:0, 20:4n-3 and 20:5n-3 of the whole body were significantly (P<0.05) affected by dietary lipid level. The results of this study suggest that an increased dietary lipid level from 10% to 17% can improve growth of juvenile red sea bream in low temperature periods.

포르말린과 중성포르말린 약욕한 어류의 Formaldehyde 잔존량 측정과 병리학적 관찰 (Determination of formaldehyde residue and histopathological observation in formalin and neutral-formalin treated Korean rockfish(Sebastes schlegeli))

  • 이 실험에서는 조피볼락을 대상으로 중성 formalin과 formalin으로 약욕하여 잔존량과 잔존기간 및 병리 조직학적 변화를 관찰하고 온도가 미치는 영향에 대해서 알아보았다. formalin 과 중성 formalin의 잔존량은 $15^{\circ}C$일 때 약욕 직후 약간의 차이를 제외하고는 별다른 차이를 보이지 않았다. 잔존기간은 수온 $15^{\circ}C$$25^{\circ}C$은 조건에서 각각 72 hr과 24 hr으로 온도의 영향을 많이 받는 것으로 나타났다. 어체에 미치는 독성은 중성 formalin보다 formalin이 강하게 나타나고 수온과 농도가 증가 할수록 조직에 미치는 영향이 심해졌다. formalin 약욕으로 인하여 아가미 조직에서는 부종과 상피세포의 박리, 뒤틀림, 괴사증상이 그리고 간에서는 부종과 핵농축, 공포화 현상이 나타나고 신장에서는 수증성 퇴행적 병변과 세뇨관 상피세포의 과립화 중상, 괴사가 일어나고 피부에서는 점액 세포의 증가와 표피의 괴사 및 피하조직과 진피층의 균열이 나타났다.

Dietary Optimum Phosphorus Level of Juvenile Korean Rockfish (Sebastes schlegeli)

  • A 10-week feeding experiment was conducted to determine the phosphorus requirement of juvenile Korean rockfish (Sebastes schlegeli). Three replicate groups of fish initially averaging 4.2g were fed the semipurified experimental diets containing graded levels of $NaH_2PO_4\;\cdot\;2H_2O$ to provide from $0.1\%$ to $1.32\%$ total phosphorus level in a flow-through seawater system. Korean rockfish muscle and casein were used as the protein sources of the basal diet. Weight gain, feed efficiency and protein retention of fish fed the $0.35\%$ phosphorus were higher than those of fish fed the $0.1\%$ phosphorus, although no significant improvements $(P>0.01)$ were observed above the level. Determined phosphorus requirement using the broken line model was found to be $0.3\%$ for weight gain. Moisture, protein and lipid contents of whole body and muscle were not affected by dietary phosphorus levels $(P>0.01)$. Lipid contents of liver in fish fed the $0.1\%$ phosphorus were lower than those in fish fed the $0.35\%$ and $1.32\%$ phosphorus $(P<0.01)$. Dietary phosphorus increased ash and phosphorus contents of the whole body, while those of bone were not affected $(P>0.01)$. The data obtained in this study indicate that a $0.3\%$ dietaryphosphorus level could be recommended for the optimum growth and efficient nutrient utilization of juvenile Korean rockfish.

Growth and Fatty Acid Composition of Juvenile Olive Flounder Paralichthys olivaceus Fed Diets Containing Different Levels and Ratios of Eicosapentaenoic Acid and Docosahexaenoic Acid

  • This study was carried out to investigate the influences of dietary levels, ratios and sources of eicosapentaenoic acid (EPA) and docosahexaenoic acid (DHA) on the growth and fatty acid compositions of juvenile olive flounder Paralichthys olivaceus. Sixteen diets containing five levels of EPA (0.5%, 1.0%, 1.5%, 2.0%, and 4.0%), five levels of DHA (0.5%, 1.0%, 1.5%, 2.0% and 4.0%), three ratios of EPA/DHA (75/25, 50/50 and 25/75), two levels of squid liver oil (5% and 10%) and a control diet containing 5% soybean oil were hand-fed to triplicate groups of fish (average weight, $9.7{\pm}0.3g$) for 8 weeks. Survival, specific growth rate, feed efficiency and protein efficiency ratio of fish were not affected by dietary EPA and DHA levels or ratios. Also, the dietary treatment had no significant effect on the lipid and protein contents of muscle and whole body of fish. A corresponding increase in the EPA and DHA contents of fish occurred with increasing EPA and DHA levels in their diets. Our results suggest that juvenile olive flounder require a dietary EPA level of approximately 0.32% in the presence of 0.74% DHA for suitable survival and growth, and that EPA and DHA levels in fish muscle can increase to as much as 32% and 53%, respectively, of the total fatty acid content.
