Real-Time Voltammetric Assay of Lead Ion in Biological Cell Systems

  • Trace lead detection for cyclic voltammetry (CV) and square-wave (SW) stripping voltammetry was performed using mercury immobilized onto a carbon nanotube electrode (HNPE). Using the characteristics of mercury and the catalytic carbon nanotube structure, a modified technique, the $0.45{\mu}g/l$ detection limit of lead ion was attained. The developed method can be applied to pond water, fish tissue, plant tissue, and in vivo direct assay.

남해안 양식어류의 fluoroquinolone계 항균제 잔류량 (Residual Concentrations of Fluoroquinolones in Farmed Fish in the Southern Coast of Korea)

  • Fluoroquinoles have a wide range of antimicrobial properties and are effective in the treatment of bacterial diseases in fish. The use of fluoroquinoles continues to grow steadily. Fluoroquinolone antibiotics are probably the most important class used among synthetic antibiotics in human and veterinary medicines because of their broad activity spectrum and good oral absorption. This study was conducted to estimate the residue of antibiotics in four species of farmed fishes, including olive flounder (Paralichthys olivaceus), black rock fish (Sebastes schlegeli), red sea bream (Pagrus major), and sea bass (Lateolabrax japonicus), collected from fish farms located in the southern coastal area of Korea. The residues of fluoroquinolones were determined using high performance liquid chromatography (HPLC) with a fluorescence detector. Residuals of five fluoroquinolones in muscle tissue of farmed fish were analyzed. We found that enrofloxacin was the most common agent in fish muscle, and that ciprofloxacin was the next most common. The range of detected concentrations of fluoroquinolones in olive flounder muscle was 0-0.859 mg/kg in 32.6% of all samples. Enrofloxacin was commonly detected in sea bass muscle at a range of 0-0.143 mg/kg in 38.9% of all samples. Fluoroquinolones were detected in 6.9% of black rock fish muscle and in 16.6% of sea bream, although the detected concentration was below 0.01 mg/kg. The maximum detection value of enrofloxacin and ofloxacin in olive flounder at the time of shipping was 0.102 mg/kg and 0.09 mg/kg, respectively; no other antimicrobials were detected. We detected no antimicrobial substances in red sea bream.

장흥댐에 설치되어 있는 어도와 담수어류의 이용 분석 (Freshwater Fish Utilization of Fishway Installed in the Jangheung Dam)

  • 댐이나 보와 같은 하천내 구조물의 건설시 어도의 설치는 어류의 이동에 있어서 중요한 역할을 한다. 탐진강에 건설되어 있는 장흥 다목적댐에는 어류의 상류로 이동을 원활히 하기 위해 엘리베이터식 어도에 포함되는 트럭식 댐체어도가 방류구 옆쪽에 위치하고 있다. 이러한 종류의 어도는 효율적으로 어류를 포집하기 위한 트랩이 함께 설치된다. 본 연구에서는 장흥댐에 설치되어 있는 어도 트랩의 어류 이용정도를 파악하기 위하여 PIT telemetry 방식을 적용, 총 15종 254개체의 어류에 tag을 삽입하여 모니터링을 하였다. 붕어, 떡붕어, 돌고기, 갈겨니, 피라미, 눈동자개 6종 36개체가 감지되어 14.2%의 감지율을 나타냈다. 붕어가 43개체 중 19개체가 감지되어 44.2%로 가장 높은 감지 비율을 보였고 돌고기는 14.3%의 비율을 나타냈으며, 갈겨니와 피라미는 각각 5%와 7.7%의 감지 비율을 나타냈다. 일부 개체들은 한번에 어도내로 이동하지 않고 장기간에 걸쳐 꾸준히 신호가 감지되었다. 이동한 개체들과 이동하지 않은 개체들 사이에 크기 (전장, 체장, 체중)는 차이가 없었으며(Mann-Whitney U test, p>0.05), 주로 댐에서 방류가 이루어지는 시간에 트랩으로 이동하는 것으로 파악되었다 (Mann-Whitney U test, p<0.001). 시간대별 분석에서는 주로 야간시간대보다 주간시간대에서 더 높은 감지빈도를 보였다(Mann-Whitney U test, p<0.001). 본 연구의 결과에 의하면, 어류가 트랩으로 이동하는 데는 방류시간과 같은 외적인 요소와 각 종별 생태적 특성(산란기, 주행성, 야행성)이 중요한 역할을 하고 있었다. 따라서 어류의 어도 이용율을 높이기 위해서는 어류의 생태적 특성을 고려하여 방류량과 더불어 방류시간, 방류시간대를 적절하게 변화시키는 전략적 방류가 필요할 것으로 판단된다.

Screening for down syndrome using trophoblast retrieval and isolation of the cervix: preliminary study

  • Purpose: Trisomy 21, the cause of Down syndrome (DS) with various medical problems, is the most common aneuploidy during the fetal period. For diagnosis, a non-invasive screening test using maternal blood, which cannot be confirmed and invasive confirmation test with a risk of miscarriage, may be performed. The trophoblast retrieval and isolation of the cervix (TRIC) have been proposed by some researchers as an alternative to overcome the limitations of current tests. We experimented using TRIC to identify the possibility of trisomy 21 for the first time in Asia. Materials and Methods: Three cases of DS were analyzed confirmed by invasive tests (chorionic villus sampling, amniocentesis). All samples of trophoblasts immediately were immersed in phosphate-buffered saline and processed with formalin for fixation. The trophoblasts were isolated using an anti-human leukocyte antigen-G antibody coupled to magnetic nanoparticles. β-human chorionic gonadotropin (hCG)-expressing cells were considered as trophoblast cells, and the detection rate calculated. DS was confirmed by fluorescence in situ hybridization (FISH). Results: The mean trophoblast detection rate using β-hCG was 78.1%, and the detection rate using FISH was 22.2%. In all cases, the trisomy of chromosome 21 was identified. Conclusion: Trophoblast can be obtained from the five weeks of gestation and has a high detection rate, so it is noted that it can replace the current prenatal genetic test. To realize the clinical application as a prenatal genetic test, we will need additional efforts to identify trisomy 21 as well as other chromosomal abnormalities in future large-scale studies.

어안 이미지의 배경 제거 기법을 이용한 실시간 전방향 장애물 감지 (Real time Omni-directional Object Detection Using Background Subtraction of Fisheye Image)

  • This paper proposes an object detection method based on motion estimation using background subtraction in the fisheye images obtained through omni-directional camera mounted on the vehicle. Recently, most of the vehicles installed with rear camera as a standard option, as well as various camera systems for safety. However, differently from the conventional object detection using the image obtained from the camera, the embedded system installed in the vehicle is difficult to apply a complicated algorithm because of its inherent low processing performance. In general, the embedded system needs system-dependent algorithm because it has lower processing performance than the computer. In this paper, the location of object is estimated from the information of object's motion obtained by applying a background subtraction method which compares the previous frames with the current ones. The real-time detection performance of the proposed method for object detection is verified experimentally on embedded board by comparing the proposed algorithm with the object detection based on LKOF (Lucas-Kanade optical flow).

사람 Y 염색체 특이 DNA Probe의 개발과 이를 이용한 FISH 기술의 임상적 적용 (Identification of a Human Y Chromosome Specific DNA Probe and Their Clinical Application by Fluorescence in situ Hybridization Techniques)

  • Objective: The present study was undertaken to synthesize a human Y chromosome specific probe and to confirm the usefulness of the probe for fluorescence in situ hybridization (FISH) in various types of human cells. Methods: An approximately 400 bp DNA fragment of the DYZ1 sequences was synthesized by PCR using digoxigenin labeled dUTP (dig-PCR). The fidelity of probe was tested by FISH for cultured and uncultured human lymphocytes, amniocytes, chorionic villus cells, embryos, sperms, and germ cells of seminiferous tubule. Results: The human Y chromosome specific probe hybridized specifically to Y chromosome of the cells that had been tested. This probe assigned to the Yq12 region where the DYZ1 repetitive sequence is concentrated. Conclusion: We have identified a human Y chromosome specific probe that hybridized specifically to the Y chromosome by FISH for various types of uncultured as well as cultured cells. Therefore FISH technique using human Y chromosome specific probe should be useful for clinical application as a diagnostic tool for the detection of human Y chromosome.

Dynamics of Hexavalent Chromium in Four Types of Aquaculture Ponds and Its Effects on the Morphology and Behavior of Cultured Clarias gariepinus (Burchell 1822)

  • Hexavalent chromium is a bio accumulative toxic metal in water and fish. It enters aquaculture ponds mainly through anthropogenic sources. Hexavalent chromium concentrations and its effects on the morphology and behavior of Clarias gariepinus were investigated from four aquaculture ponds for 12 weeks. Chromium was measured using diphenyl carbohdrazide method; alkalinity and hardness were measured using colometric method and analyzed with Bench Photometer. Temperature and pH were measured using pH/EC/TDS/Temp combined tester. Temporal and spatial replications of samples were done with triplicates morphological and behavioural effects of the metal on fish were observed visually. Chromium ranged from no detection to 0.05 mg/L, alkalinity 105 to 245 mg/L, hardness 80 to 165 mg/L, pH 6.35 to 8.03 and temperature 29.1 to $35.9^{\circ}C$. Trend in the chromium concentrations in the ponds is natural > earthen > concrete > collapsible. There was a significant difference (P < 0.05) in chromium, alkalinity, water hardness, pH and temperature among the four ponds. Significant positive correlation also existed between alkalinity, water hardness, pH, with chromium. Morphological and behavioural changes observed in the fish include irregular swimming, frequent coming to the surface, dark body colouration, mucous secretion on the body, erosion of gill epithelium, fin disintegration, abdominal distension and lethargy. High chromium concentration in natural pond was due to anthropogenic run-off of materials in to the pond. Acidic pH, low alkalinity, low water hardness also contributed to the high chromium concentration. Morphological and behavioural changes observed were attributed to the high concentrations, toxicity and bio accumulative effect of the metal. Toxicity of chromium to fish in aquaculture could threaten food security. Watershed best management practices and remediation could be adopted to reduce the effects of toxicity of chromium on pond water quality, fish flesh quality and fish welfare.

  • 양식어류의 세균성 질병의 신속한 진단을 위하여 틸라피아(O. niloticus)로부터 분리된 $\beta$용혈성 연쇄구균인 Streptococcus sp. KST-2균주를 사용하여 면역효소법으로 검출실험을 하였으며, 면역효소법의 현장사용의 용이함과 검출의 민감도를 알아보기 위하여 counterimmunoelectrophoresis(CIE), 및 이중면역확산법등과 비교하여 보았다. 다른 어류 병원균과의 교차반응 결과 교차반응이 전혀 일어나지 않는 것으로 보아 이 방법은 $\beta$용혈성 연쇄구균(KST-2균주)에 매우 특이적임을 알 수 있었다. 연쇄구균의 검출실험한 결과 면역효소법은 $1{\times}10^3CFU/ml$의 농도까지 검출가능하였으며, 검출의 민감성이 CIE에 비해 $10^2$배, 이중면역확산법에 비해 $10^4$배나 높은 것으로 나타났다. 면역효소법은 이번 연구에서 사용된 진단법 중에서 현장사용이 가장 용이하며, 민감성이 가장 뛰어난 것으로 나타났다.

생선가시의 전산화 단층촬영 자료에 삼차원 재구성법을 적용한 연구 (Three Dimensional Reconstruction on Computed Tomograms of Fish Bones)

  • Background and Objectives This study was conducted to gather three dimensional images from computed tomogram in detecting and gaining information about fish bones. Materials and Methods Thirty-nine fish bones and 2 chicken leg bones were used. Bones sandwiched between the cotton sheets were placed into the 3 plastic boxes. Computed tomogram of the bones in the boxes were reconstructed three-dimensionally by Xelis (Infinitt, Korea) to make images of MPR, MIP and VR images. The images were compared with real chicken bones and fish bones as to possibility of detection, finding of location and orientation, and evaluation of shape and calculation of size. Results All 41 bones were detected in all reconstructed images. Distance to the bones from reference point can be measured and orientation can be checked. Shapes were similar between matched images of all real bones. Conclusion The results indicate the usefulness of 3D reconstruction technique in getting informations about fish bones.

SinGAN 딥러닝 모델을 이용한 넙치 질병 이미지 증강 (Image Augmentation of Paralichthys Olivaceus Disease Using SinGAN Deep Learning Model)

  • 수산 양식장에서 어류 질병을 초기에 발견하지 못하는 경우 밀폐된 공간 안에서 확산하기 때문에 집단 폐사로 이어질 확률이 매우 높다. 이런 이유로 질병의 조기 발견은 양식업에서 매우 중요하다. 양식장에서 질병의 확산을 막기 위해서는 초기에 병이 든 어류를 자동식별이 가능한 방법이 필요하다. 최근 딥러닝 기반의 어류 질병 자동식별 방법이 많이 사용되고 있는데, 어류의 질병 이미지가 충분하지 않아 객체 식별에 많은 어려움이 있다. 본 논문은 질병 자동식별 예측을 위한 질병 이미지의 부족 문제를 해결하기 위해서 SinGAN 딥러닝 모델을 이용하여 정상 이미지와 질병 이미지를 합성해 다양한 어류 질병 이미지를 자동 생성하는 방법을 제안한다. 넙치에서 가장 빈번히 발생하는 3가지 질병 스쿠티카병, 비브리오증, 림포시스티스에 대해서 SinGAN 기반으로 질병 이미지를 증강한다. 본 연구에서는 넙치 정상 이미지 11장에 각 질병 패턴 10가지를 합성하여서 스쿠티카병 110장, 비브리오증 110장, 림포시스티스 110장으로 총 330장을 만들었고 이를 통해 생성된 이미지는 4배수 하여 1,320장의 이미지를 생성할 수 있었다.
