Chatbot Design Method Using Hybrid Word Vector Expression Model Based on Real Telemarketing Data

  • In the development of commercial promotion, chatbot is known as one of significant skill by application of natural language processing (NLP). Conventional design methods are using bag-of-words model (BOW) alone based on Google database and other online corpus. For one thing, in the bag-of-words model, the vectors are Irrelevant to one another. Even though this method is friendly to discrete features, it is not conducive to the machine to understand continuous statements due to the loss of the connection between words in the encoded word vector. For other thing, existing methods are used to test in state-of-the-art online corpus but it is hard to apply in real applications such as telemarketing data. In this paper, we propose an improved chatbot design way using hybrid bag-of-words model and skip-gram model based on the real telemarketing data. Specifically, we first collect the real data in the telemarketing field and perform data cleaning and data classification on the constructed corpus. Second, the word representation is adopted hybrid bag-of-words model and skip-gram model. The skip-gram model maps synonyms in the vicinity of vector space. The correlation between words is expressed, so the amount of information contained in the word vector is increased, making up for the shortcomings caused by using bag-of-words model alone. Third, we use the term frequency-inverse document frequency (TF-IDF) weighting method to improve the weight of key words, then output the final word expression. At last, the answer is produced using hybrid retrieval model and generate model. The retrieval model can accurately answer questions in the field. The generate model can supplement the question of answering the open domain, in which the answer to the final reply is completed by long-short term memory (LSTM) training and prediction. Experimental results show which the hybrid word vector expression model can improve the accuracy of the response and the whole system can communicate with humans.

The Overexpression of Subtilisin Enzyme Using Mutations on Transition State Regulatory Proteins of AprE Promoter and Development of Bacillus subtilis Host System (AprE Promoter전이상태 조절인자 변이주를 이용한 공업적 효소의 과발현과 고초균 숙주계의 개발)

  • Bacillus subtillis strains with transition state regulator mutations and a spore mutation were developed for the overexpression of apsE and for the enhancement of expression level. Among the many regulator genes, degU and hpr were chosen as a representative positive and negative regulator for the aprE, respectively. Spo II G was used for the construction of asporogeneous strains. All the mutants were constructed from two protease-deleted strain DB104 and the apsE gene was transformed with an integration vector pMK101. DB104(deg$U^h$(32) $his^+$)::pMK101(Cm) and DB104($\Delta$her(Em))::pMKl01(Cm) show 7-fold and about 2-fold increase in aprE expression level, respectively. But the effect of transition state regulator mutation on the aprE expression was diminished when the integrated aprE gene was amplified by the high concentration of chloramphenicol, i. e. 30 $\mu\textrm{g}$/ml. DB104($\Delta$spoIIG(Pm) degUh(32) his+)::pMK101(Cm) and DB104($\Delta$spoIIG(Pm) $\Delta$hpr(Em))::pMK101 double mutant show 10-fold and 3-fold increase in aprE expression level, respectively. The results suggest that sporulation mutation and transition state regulator mutation have independent and additive effect on the aprE expression, and the same gene dosage effect on the transition state regulator mutation was also identified.

  • Precisely reliable and quantitative reporters can provide phenotypes that are consistent with research goals in protein expression. Here, we developed an improved reporter mATglu III 5 by directed evolution using a versatile β-glucosidase ATglu derived from Agrobacterium tumefaciens. When expressed in hosts, a vector containing this mutant distinctly showed a colored or fluorescent phenotype, according to the supplemented substrate, without any inducer. Analysis of mATglu III 5 showed it to be fully functional in fusion state with oligomeric proteins, especially under non-induction conditions, thereby offering an alternative to conventional reporters.

  • As the visual effect frequently used in movies or animations, special effects are well suited for the creation of buildings or materials' destruction and collapse scenes. With the relevant programs developing technologically, the adoption of a real-time physically based-system makes it possible to realistically express dynamic simulations. In the large scale, the visual expression of such effects of destroying is satisfying enough, but most common programs of those effects fail to maximize visual effect generated with the cutting of small materials. Besides, to perform a heavy simulation process needs high-performance hardware and programs, where high costs would become a serious issue. For this reason, this paper suggests a solution optimized for the effect of small materials-cutting. The progress of each step shows technologies which trace movement with the state of the completion of the character's motions and then cut the material in real-time, finally led to the very realistic visual effect. Besides, using vector inner calculation to follow the motions of object and to realize cutting effect, this study provides an experiment that constructs visual effect for visualization from the basis of mathematical algorithm and it would be certainly as an educational material used for further researches.

Enhanced Salt Stress Tolerance in Transgenic Potato Plants Expressing IbMYB1, a Sweet Potato Transcription Factor

  • IbMYB1, a transcription factor (TF) for R2R3-type MYB TFs, is a key regulator of anthocyanin biosynthesis during storage of sweet potatoes. Anthocyanins provide important antioxidants of nutritional value to humans, and also protect plants from oxidative stress. This study aimed to increase transgenic potatoes' (Solanum tuberosum cv. LongShu No.3) tolerance to environmental stress and enhance their nutritional value. Transgenic potato plants expressing IbMYB1 genes under the control of an oxidative stress-inducible peroxidase (SWPA2) promoter (referred to as SM plants) were successfully generated through Agrobacterium-mediated transformation. Two representative transgenic SM5 and SM12 lines were evaluated for enhanced tolerance to salinity, UV-B rays, and drought conditions. Following treatment of 100 mM NaCl, seedlings of SM5 and SM12 lines showed less root damage and more shoot growth than control lines expressing only an empty vector. Transgenic potato plants in pots treated with 400 mM NaCl showed high amounts of secondary metabolites, including phenols, anthocyanins, and flavonoids, compared with control plants. After treatment of 400 mM NaCl, transgenic potato plants also showed high DDPH radical scavenging activity and high PS II photochemical efficiency compared with the control line. Furthermore, following treatment of NaCl, UV-B, and drought stress, the expression levels of IbMYB1 and several structural genes in the flavonoid biosynthesis such as CHS, DFR, and ANS in transgenic plants were found to be correlated with plant phenotype. The results suggest that enhanced IbMYB1 expression affects secondary metabolism, which leads to improved tolerance ability in transgenic potatoes.

  • This paper describe methodology that is distributed on 2-dimensional plane to much high-dimensional facial motion datas using Isomap algorithm, and user interface techniques to control facial expressions by selecting expressions while user navigates this space in real-time. Isomap algorithm is processed of three steps as follow; first define an adjacency expression of each expression data, and second, calculate manifold distance between each expressions and composing expression spaces. These facial spaces are created by calculating of the shortest distance(manifold distance) between two random expressions. We have taken a Floyd algorithm for it. Third, materialize multi-dimensional expression spaces using Multidimensional Scaling, and project two dimensions plane. The smallest adjacency distance to define adjacency expressions uses Pearson Correlation Coefficient. Users can control facial expressions of 3-dimensional avatar by using user interface while they navigates two dimension spaces by real-time.

Construction and Analysis of Food-Grade Lactobacillus kefiranofaciens β-Galactosidase Overexpression System

  • Lactobacillus kefiranofaciens contains two types of β-galactosidase, LacLM and LacZ, belonging to different glycoside hydrolase families. The difference in function between them has been unclear so far for practical application. In this study, LacLM and LacZ from L. kefiranofaciens ATCC51647 were cloned into constitutive lactobacillal expression vector pMG36e, respectively. Furtherly, pMG36n-lacs was constructed from pMG36e-lacs by replacing erythromycin with nisin as selective marker for food-grade expressing systems in Lactobacillus plantarum WCFS1, designated recombinant LacLM and LacZ respectively. The results from hydrolysis of o-nitrophenyl-β-galactopyranoside (ONPG) showed that the β-galactosidases activity of the recombinant LacLM and LacZ was 1460% and 670% higher than that of the original L. kefiranofaciens. Moreover, the lactose hydrolytic activity of recombinant LacLM was higher than that of LacZ in milk. Nevertheless, compare to LacZ, in 25% lactose solution the galacto-oligosaccharides (GOS) production of recombinant LacLM was lower. Therefore, two β-galactopyranosides could play different roles in carbohydrate metabolism of L. kefiranofaciens. In addition, the maximal growth rate of two recombinant strains were evaluated with different temperature level and nisin concentration in fermentation assay for practical purpose. The results displayed that 37℃ and 20-40 U/ml nisin were the optimal fermentation conditions for the growth of recombinant β-galactosidase strains. Altogether the food-grade Expression system of recombinant β-galactosidase was feasible for applications in the food and dairy industry.

Calculation of Magnetic Field for Cylindrical Stator Coils in Permanent Magnet Spherical Motor

  • This paper analyzed the magnetic field produced by the cylindrical stator coils of permanent magnet spherical motor (PMSM). The elliptic equations about the vector magnetic potential were given. Given that the eddy current effects are neglected, the magnet field of the PMSM is regarded as irrotational field, which can be calculated by scalar magnetic potential. The current density of cylindrical stator coil was proposed based on the definition of current density. The expression of current density of stator coil was obtained according to the double Fourier series decomposition and spherical harmonic functions. Then the magnetic flux density for scalar magnetic potential was derived. Further, the influence of different parameters on radial flux density was also analyzed. Finally, the results by the analytical method in this paper were validated by finite element analysis (FEA).

Effects of Heterologous Expression of Thioredoxin Reductase on the Level of Reactive Oxygen Species in COS-7 Cells

  • Thioredoxin reductase (TrxR), a component of the redox control system involving thioredoxin (Trx), is implicated in defense against oxidative stress, control of cell growth and proliferation, and regulation of apoptosis. In the present study a stable transfectant was made by introducing the vector pcDNA3.0 harboring the fission yeast TrxR gene into COS-7 African green monkey kidney fibroblast cells. The exogenous TrxR gene led to an increase in TrxR activity of up to 3.2-fold but did not affect glutathione (GSH) content, or glutaredoxin and caspase-3 activities. Levels of reactive oxygen species (ROS), but not those of nitric oxide (NO), were reduced. Conversely, 1-chloro-2,4-dinitrobezene (CDNB), an irreversible inhibitor of mammalian TrxR, enhanced ROS levels in the COS-7 cells. After treatment with hydrogen peroxide, the level of intracellular ROS was lower in the transfectants than in the vector control cells. These results confirm that TrxR is a crucial determinant of the level of cellular ROS during oxidative stress as well as in the normal state.

  • This paper analyzed orientation simulation of Stewart platform which is a parallel manipulator of 6-DOF. When platform shape had been given, inverse kinematics as the problem about length of actuator could get an answer using a vector function simply, and forward kinematics as the problem solving shape of platform through the length of actuator could get answer using repetitive and manual explaining Newton-Raphson method because it is expressed a high nonlinear polynomial expression. In addition, for control the Stewart platform it could drive simply and it could confirm the state of orientation in real-time.
