Induction of Phase I, II and III Drug Metabolism/Transport by Xenobiotics

  • Drug metabolizing enzymes (DMEs) play central roles in the metabolism, elimination and detoxification of xenobiotics and drugs introduced into the human body. Most of the tissues and organs in our body are well equipped with diverse and various DMEs including phase I, phase II metabolizing enzymes and phase III transporters, which are present in abundance either at the basal unstimulated level, and/or are inducible at elevated level after exposure to xenobiotics. Recently, many important advances have been made in the mechanisms that regulate the expression of these drug metabolism genes. Various nuclear receptors including the aryl hydrocarbon receptor (AhR), orphan nuclear receptors, and nuclear factor-erythoroid 2 p45-related factor 2 (Nrf2) have been shown to be the key mediators of drug-induced changes in phase I, phase II metabolizing enzymes as well as phase III transporters involved in efflux mechanisms. For instance, the expression of CYP1 genes can be induced by AhR, which dimerizes with the AhR nuclear translocator (Arnt) , in response to many polycyclic aromatic hydrocarbon (PAHs). Similarly, the steroid family of orphan nuclear receptors, the constitutive androstane receptor (CAR) and pregnane X receptor (PXR), both heterodimerize with the ret-inoid X receptor (RXR), are shown to transcriptionally activate the promoters of CYP2B and CYP3A gene expression by xenobiotics such as phenobarbital-like compounds (CAR) and dexamethasone and rifampin-type of agents (PXR). The peroxisome proliferator activated receptor (PPAR), which is one of the first characterized members of the nuclear hormone receptor, also dimerizes with RXR and has been shown to be activated by lipid lowering agent fib rate-type of compounds leading to transcriptional activation of the promoters on CYP4A gene. CYP7A was recognized as the first target gene of the liver X receptor (LXR), in which the elimination of cholesterol depends on CYP7A. Farnesoid X receptor (FXR) was identified as a bile acid receptor, and its activation results in the inhibition of hepatic acid biosynthesis and increased transport of bile acids from intestinal lumen to the liver, and CYP7A is one of its target genes. The transcriptional activation by these receptors upon binding to the promoters located at the 5-flanking region of these GYP genes generally leads to the induction of their mRNA gene expression. The physiological and the pharmacological implications of common partner of RXR for CAR, PXR, PPAR, LXR and FXR receptors largely remain unknown and are under intense investigations. For the phase II DMEs, phase II gene inducers such as the phenolic compounds butylated hydroxyanisol (BHA), tert-butylhydroquinone (tBHQ), green tea polyphenol (GTP), (-)-epigallocatechin-3-gallate (EGCG) and the isothiocyanates (PEITC, sul­foraphane) generally appear to be electrophiles. They generally possess electrophilic-medi­ated stress response, resulting in the activation of bZIP transcription factors Nrf2 which dimerizes with Mafs and binds to the antioxidant/electrophile response element (ARE/EpRE) promoter, which is located in many phase II DMEs as well as many cellular defensive enzymes such as heme oxygenase-1 (HO-1), with the subsequent induction of the expression of these genes. Phase III transporters, for example, P-glycoprotein (P-gp), multidrug resistance-associated proteins (MRPs), and organic anion transporting polypeptide 2 (OATP2) are expressed in many tissues such as the liver, intestine, kidney, and brain, and play crucial roles in drug absorption, distribution, and excretion. The orphan nuclear receptors PXR and GAR have been shown to be involved in the regulation of these transporters. Along with phase I and phase II enzyme induction, pretreatment with several kinds of inducers has been shown to alter the expression of phase III transporters, and alter the excretion of xenobiotics, which implies that phase III transporters may also be similarly regulated in a coordinated fashion, and provides an important mean to protect the body from xenobiotics insults. It appears that in general, exposure to phase I, phase II and phase III gene inducers may trigger cellular 'stress' response leading to the increase in their gene expression, which ultimately enhance the elimination and clearance of these xenobiotics and/or other 'cellular stresses' including harmful reactive intermediates such as reactive oxygen species (ROS), so that the body will remove the 'stress' expeditiously. Consequently, this homeostatic response of the body plays a central role in the protection of the body against 'environmental' insults such as those elicited by exposure to xenobiotics.

초현실주의 표현기법을 활용한 코트 디자인 개발 (A Design Development for Coat Using Expression Technique of Surrealism)

  • The purpose of this study is to examine the concept of surrealism and its expression technique and to develop creative and artistic women's coat design based on the analysis of various expression techniques of surrealistic artists and designers. To this end, literature review was done and design and actual works of women's coats were made. Surrealism was applied to fashion by Schiaparelli in the 1930s and it has constantly been applied by many fashion designers. The examples used in fashion were examined by shape of body parts or natural objects, metaphor and transformation of objects, position change and optical illusion. Based on this examination, motifs were drawn from paintings of Rene Magritte, Salvador Dali, Vladimir Kush, and Rafal Olbinski, who most inspired contemporary fashion designers with their works. The creative and artistic sense of five women's coats for 20's and 30's were designed using surrealistic paintings and fashion design expression techniques in a view of social problems. This study has a significance in that it expanded the expression techniques of fashion design and its field with original design which is inspired by surrealistic formativeness and expression technique with social criticism. It is also possible for this study to contribute to the development of fashion design if various designs are presented through grafting various artistic expression techniques onto fashion.

A Screen for Genetic Loci on the X Chromosome Required for Body-Wall Muscle Development during Embryogenesis in Caenorhabditis elegans

  • We have screened available chromosomal deficiencies on the X chromosome for genetic loci whose zygotic expression is required for body-wall muscle development during embryogenesis in Caenorhabditis elegans. Previously, it had been reported that no sign of muscle development was detected in nullo-X embryos arrested at an early stage of embryogenesis. Based on this observation, it has been suggested that genetic loci exist on the X chromosome whose zygotic expression is essential for body-wall muscle formation. In order to identify such myogenic loci, 9 chromosomal deficiencies covering approximately 45% of the X chromosome have been tested. Homozygous embryos from these deficiency strains were collected and terminal phenotypes of arrested embryos were observed by Nomarski microscopy. As a secondary assay, monoclonal antibodies against two myosin heavy chain (MHC) isoforms, the products of the myo-3 and unc-54 genes, were used to detect body-wall muscle differentiation. All the homozygous deficiency embryos were positively stained with both MHC antibodies and muscle twitching movement was observed in most cases. Combined with previously analyzed deficiencies, our deficiency screen has covered approximately 70% of the X chromosome. We conclude that the regions covered by the available deficiencies on the X chromosome do not include any myogenic locus required for body-wall muscle formation. Alternatively, the possibility that nullo-X embryo may not form body-wall muscle due to a general failure to differentiate during embryogenesis remains to be tested.

성인의 분노표현 방식과 뇌졸중 위험 요인 (Anger Expression Style and Risk Factors for Stroke)

  • Purpose: As anger may be strongly related to stroke, this study examined the relationship between anger expression style (anger-in, anger-out, anger-control) and risk factors for stroke to investigate the possible attribution of anger expression style to stroke incidence. Methods: The cross sectional study design was employed. The 291 participants signed consent forms and completed questionnaires including the anger expression style scales and risk factors for stroke. Data were analyzed using descriptive statistics, t-test and One-way ANOVA. Results: The highest score among anger expression styles of the participants was from anger-control. Participants with higher mean value of anger-out were current non-smokers, doing regular exercise, having scores above 23 in Body Mass Index, having scores above 90 cm (male) or 80 cm (female) in abdominal circumference, and having scores below 85 mmHg in diastolic blood pressure. Participants with more than 5 risk factors to stroke had the highest score of anger-in. Conclusion: The results of this study implied anger expression style was psychological risk factor for stroke. Further studies to differentiate the anger expression style contributing to the risk of stroke are needed with a longitudinal study design.

  • This study is to compare and analyze two types of 'transparency' concept expressed in architecture and garment which has not only been an important theme in the post-modern expression of architeture but been shown in various types of concept. This study examined the characteristics of transparency and through theoretical studies divided them into four types; frankness(purity ambiguity comparison exposure and eroticism) Purity building would use the matrial which is transparent glasses for the expprssion of its purity and clothing would use the transparent matrial for the expression of the natural beauty of human body and the purity of human of itself. Ambiguity is expressed in the outer side of architecture by duplicating transparent materials and by phenomenon of optical illusion and inflection. On the contrary in garment it is shown in the outward formation of underclothing. Comparison in architecture is expressed by the use of void but opaque materials as well as solid but transparent materials. Extinction is expressed with the visual effect of context of the surrounding by the use of opaque glass and reflexive glass. In garment the effect of comparison is expressed by the use of mixing transparent materials and opaque materials. Building would express the inside of itself by the way of its exposure and clothing would also express its frankness through the human body is partly opened or clossed at the same thime that arises from eroticism of the way of the expression of human feelings.

생쥐 고환 및 부고환에서 galectin-3의 발현 (Expression of galectin-3 in the testis and epididymis of mouse)

  • The testis and epididymis are important organs of the male reproductive system; the functionis to produce, mature, transport, and store sperm. It is important to understand the localization and expressionof specific proteins based for the studies of its physiological processes. In this study, we investigated theexpression and distribution of galectin-3, one of beta-galactoside-binding proteins, in the testis andepididymis of mouse using western blot and imunohistochemistry. Western blot analysis revealed that theexpression of galectin-3, 29 kDa protein, was low in the testis. In the epididymis, high expression wasdetected in the body and tail part, but moderate expression in the head part. By immunohistochemicalanalysis, we found that positive localization of galectin-3 was detected in some myoid cells and Leydigin the epithelium of epididymis, especially in the epithelium of both body and tail of epididymis. Collectively,these results suggest that galectin-3 is constitutively expressed in the testis and epididymis of mouse withvarying intensity, and the role of galectin-3 in the male reproductive organ may be involved in the specificfunction of its structures.

패션 디자인에 나타난 가상공간의 페르소나 표현 (Virtual Space Persona Expressed in Fashion Illustration)

  • Persona means the mask of personality where the internal ego exposes itself to the outside. The complicated structure and diversity of contemporary society has provided men with a more variegated and diversified persona; in addition, advancements in internet and information communications creates the possibility for the emergence of more variegated and new persona. This study probes the persona of virtual space expressed in fashion designs; subsequently, this study reviews the concept and various characteristics of persona and examines the type of persona in the virtual space from the perspective of the manifestation of identity. The type of persona in the virtual space canbe categorized into flexible identity and falsified identity; subsequently, the characteristics and meanings of virtual space persona in fashion designs are analyzed. Flexible identity-oriented personas expressed in fashion designs are revealed through the images of animals or dolls. This is a childlike persona expression that expresses ego through other substances in an effort to express infantile substances such as dreams and fantasy cherished in childhood. Falsified identity-oriented personas are expanded as a realm of expression of the body and the realities are camouflaged as transformed persona. Negating existence itself by covering the eyes or face as well as the concealment of the body with masks or veils is an expression of a self-denying persona.

맥시멀리즘 특징을 이용한 패션디자인의 표현성 연구 (The Expression of Fashion Design Using a Maximalism Character)

  • Maximalism is the code which has a power to interpret a fashion phenomenon expressed complexly and variously in the multicultural society. In the 21st century, maximalism which reappeared is not the expression by a method only. It is presenting the experimental design for the new pursuit through the mixture such as expansion, splendor, variety, decoration, distortion of configuration or transformation, and composition. Therefore, this study aims at analyzing expression of fashion design by four types(expansion, decoration, mixture, non-structure) materializing a feature of maximalism expressed in the domestic and foreign collection works from 2005 to 2010. The research results are as follows. 1. Expansion: It was showed artificial, formative, unique, and odd teatures through the expansion of the upper half of the body, the lower half of the body, and both of them. 2. Decoration: It was showed the actual decoration considering functionality and practicality and the decoration emphasizing and unique character and featuring domination and emphasis. 3. Mixture: It was showed through the mixture of the contrary textiles or the different sex image. 4. Non-structure: It showed the non-structure of the avant-garde trend and non-structure through decoration, detail, adjustment direction of a dress.

총재정부 시기 앵크루아야블(Incroyables) 복식의 낭만주의 특성 (Romanticism Characteristics of the Incroyables Fashion during the Directoire)

  • Romanticists realized the ideals of a more fantastic and better society for the present day in which they lived; in addition, dress was an important medium of expression of this period. The Incroyables were a group that expressed eccentric forms and abuses of luxury through their clothing, centering around the second generation of financial elite groups during the French Directoire. Incroyables created their own fashion that expresses their new image in dress, expressing the intense personal innerity of life, which was influenced by the romanticism of pursuing an internal expression of the subjective and emotional individual. This study used a literature review to analyze the characteristics of the Romanticism expressed in the Incroyables fashion. The research results were as follows. First, it was an expression of an emotional desire for and ancient regime. Incroyables fashion were based on bourgeois nostalgia for the days of the ancient regime that resulted in an emphasis on individual and original human views. Second, it was also a hybrid of Romantic classicism. It was a form emphasizing body form where body beauty expressed a classical form through a dress under the influence of neo-classicalism that desires to return to nature.

In Vitro에서 Retrovirus Vector System을 이용한 인간 성장 호르몬 유전자의 발현 (Expression of Human Growth Hormone Gene using Retrovirus Vector System In Vitro)

  • Human growth hormone (hGH), one of the most important hormones in medicine, is secreted from anterior pituitary gland. Its broad physiological function includes body growth, cell regeneration, increasement of muscle volume, bone density, body fat reduction, and so on. Due to the wide range of therapeutic effects, the hGH produced from E. coli has been commercialized already. In this study, we asked whether it is possible to produce recombinant hGH efficiently from various cultured mammalia cells. To meet this purpose, we chose a retrovirus vector system for transfer and expression of the hGH gene in various mammalian cells. Analyses of RT-PCR, ELISA, and Western blot to determine expression of the hGH gene showed the highest production of the hGH was determined from chicken embronic fibroblast (CEF) cells with the concentration of 8.58 ${\mu}g$/ml. The biological activity of the hGH was similar to the commercially available counterpart. These results suggest that mass production of hGH is possible not only in the E. coli but also in the various mammalian cells.
