한우 비육 전·후기의 등심조직에 있어서 지방합성 유전자 발현 (Lipogenesis Gene Expression Profiling in Longissimus dorsi on the Early and Late Fattening stage of Hanwoo)

  • 지방합성이 왕성하게 진행되는 비육후기의 근육내 지방합성에 있어서, 지방산 및 당의 이용경로 및 이에 따른 근육내 지방합성과정을 규명하기 위해서 지방산 및 당 운반 유전자인 FABP4, GLUT4와 근육내 지방대사 주요유전자인 ACL, ACC, LPL 및 SCD 유전자의 mRNA 발현양상을 분석하였다. 한우 비육전기 및 후기 각 3두를 공시하여 total RNA 추출 및 1st cDNA 합성하여 SYBR green을 이용하여 real-time PCR 분석을 각 유전자별로 3반복씩 수행하였다. FABP4 유전자는 비육전기에 비해 비육후기에 있어서, 3배 이상 유전자 발현이 증가함을 확인할 수 있었고, GLUT4 유전자는 비육전기 및 비육후기에서 큰 차이를 보이지 않았다. 또한 근육내 지방합성 주요유전자인 ACL, ACC, LPL 및 SCD 유전자의 발현량은 비육후기에서 ACL 유전자가 3.8배, ACC 유전자는 2.7배, LPL 유전자는 3.5배, 그리고 SCD 유전자는 7.5배 발현량이 증가하였다. 따라서 본 연구를 통하여 한우의 비육후기에서 근육내 지방합성은 FABP4에 의한 지방산 유입의 증가와 더불어 ACL 유전자에 의해서 지방산 합성의 중간대사물인 acetyl-CoA가 합성되고, 이 중간 대사물을 이용하여 ACC 및 SCD 유전자에 의해서 긴 사슬 지방산 합성이 왕성하게 일어남을 알 수 있었다.

Nutrient requirement for maintenance and nutritional changes of the Hanwoo steers in early-fattening stage under heat stress

  • Four early-fattening Hanwoo steers weighing $247{\pm}13.5kg$ were used within a $4{\times}4$ Latin square design to establish a nutrient requirement for maintenance and to investigate nutritional changes in the steers under heat stress condition. The steers were fed four different energy level diets: 100% (control) and 100%, 115% and 130% of total digestible nutrients (TDN) requirement of the early-fattening Hanwoo steers for maintenance based on the Korean Feeding Standard for Hanwoo. The steers in the control were housed with no stress (temperature $24^{\circ}C$ and humidity 60%), whereas the steers in the other groups were under heat stress (temperature $30^{\circ}C$ and humidity 70%). True digestibilities of dry matter (DM) and other nutrients were not significantly (p > 0.05) affected by heat stress (i.e., control vs T100). This may be the result of a lower DM intake than that of the Korean feeding standard due to the establishment of the nutrients requirement under heat stress. Heat stress and different energy intake levels did not affect the blood metabolite concentrations. Average daily gain (ADG) for T100 (-69.6 g) was lower than that of the control (-44.6 g, numerically), T115 (44.6 g, p < 0.05) and T130 (83.3 g, p < 0.05), respectively. Based on the ADG and TDN intake, the equation (Y = 0.1814X + 111.5) for the TDN requirement of the early fattening Hanwoo steers for maintenance was calculated, indicating that 11.5% of TDN requirement for maintenance under heat stress may be additionally supplied.

한방생균제가 한우의 체중, 혈액성상, 면역성 및 육량·육질에 미치는 영향 (Effect of Herbal Probiotic Supplementation on Weight, Blood Composition, Meat Quality and Immunity in Beef)

  • This study was a report on 60 heads of Hanwoo steers(5 treatments ${\times}$ 4 heads ${\times}$ 3 repeats). Their start weights were $361.2{\pm}14.39kg$ to $380.5{\pm}27.33kg$; 12.7 to 13.6 months of age. The aim was to investigate the effect of herbal probiotic supplementation on the weight, blood composition, meat quality and immunity in beef. In case of body weight gain, con. 1 plot(183.8 kg) in the early fattening stage, T3 plot(115.1 kg) in the middle fattening stage, and T2 plot(163.3 kg) in the late fattening stage significantly increased(p<0.05). There was a statistically significant difference in the blood content of T-Cho and IgG: con. 1 plot and T1 plot in the early fattening stage, T3 plot and T2 plots in the middle fattening stage, and con. 2 plot and T1~T3 plots in the late fattening stage had higher blood content than that of other treatment plots (p<0.05). In addition, the carcass weight was highest in the T1 plot and T2 plot among the total treatment plots(p<0.05). However, no statistical significant difference was observed in the different treatments; all individual treatment plots(T1~T3 plots) were somewhat higher than the control plots(con. 1~con. 2 plots) with respect to the carcass quantity and carcass quality. Combining and analyzing all results revealed that the dietary addition of herbal probiotics for Hanwoo steers had a positive effect on the improvement of weight, amount of meat, meat quality and immunity.

Proteome analysis of the m. longissimus dorsi between fattening stages in Hanwoo steer

  • The objective of this study was to identify proteins in the m. longissimus dorsi between early (12 months of age) and late (27 months of age) fattening stages of Hanwoo (Korean cattle) steers. Using two-dimensional electrophoresis and mass spectrometry, 8 proteins of 11 differentially expressed spots between the 12 and 27 month age groups were identified in the loin muscle. Among those that were differentially expressed, zinc finger 323 and myosin light chain were highly expressed in late-fattening stage, and two catabolic enzymes, triosephosphate isomerase (TPI) and succinate dehydrogenase (SDH) were expressed more in the early versus the late-fattening stage. In particular, the quantification of TPI and SDH by immunoblotting correlated well with fat content. Our data suggested that TPI and SDH are potential candidates as markers and their identification provides new insight into the molecular mechanisms and pathways associated with intramuscular fat contents of bovine skeletal muscle.

점토광물 급여가 한우교잡우의 산육능력 및 육질에 미치는 영향 (Effect of Scoria, Quartz Porphyry and Oak Charcoal Powder Feeding on the Meat Quantity and Quality of Fattening Cross Bred Hanwoo Bulls)

  • This study was conducted to investigate the effects of Scoria, Quartz porphyry and Oak charcoal powder feeding by 1% level of concentrate on the fattening cross bred bulls(5/8 Korean Native Cattle, 2/8 Charolais, 1/8 Brahman germ plasma) Feeding trial was conducted with 4 treatment groups which were T1(Full feeding of concentrate and roughage for 12 months), T2(T1 and Scoria addition), T3(T1 and Quartz porphyry addition) and T4(Oak charcoal addition) for 360 days, consisting of 32 heads whose initially weights were about 299.7kg. The results obtained are summarized as follows; 1. During the over-all period, average daily gains by T3, T4, T2 and T1 were 1.024, 0.987, 0.977kg and 0.964, respectively, without a significant difference by treatments. 2. Concentrate intakes per body weight and TDN intakes required per unit of kilogram gain were lower in addition agent groups than in control, and in all the treatments feed efficiency was higher at early stage for 360 fattening day than at end stage. 3. Carcass weight, dressing percent, back-fat thickness and loin eye muscle area by treatments were 330.9 to 348.4kg(average 340.3kg), 55.2~56.2(average 55.8%), 0.41~0.55(average 0.46cm)and 76.0~80.9$\textrm{cm}^2$(average 77.6$\textrm{cm}^2$), respectively, and were improved in addition agent groups than in control. 4. Meat quality grade is higher in addition agent groups than in control, and heads of 1st grade by treatments was 3, 2, 1 and none in T4, T2, T3 and T1, respectively. 5. Incidence heads of diseases by treatment were not apparently different, but diseases of digestive system was decreased in addition agent groups than in control. 6. The nitrate nitrogen content of fattening bull dung which collected in rectum were 0.082~0.089% (average 0.084%), and the content in addition agent groups was smaller than in controll. 7. Income was higher in order of T4, T3, T2, and T1 and incomes of treatment groups were grater than that of control group by 21.4 to 33.5 percent. According to the above results it may be concluded that fattening bulls may be required to feed no more than 1% of Scoria, Quartz porphyry and Oak charcoal powder based on the concentrate feeding level during the fattening period(12 to 24 month of age) to produce high quality meat and increase income.

Identification of Differentially Expressed Genes Related to Intramuscular Fat Development in the Early and Late Fattening Stages of Hanwoo Steers

  • Marbling of cattle meat is dependent on the coordinated expression of multiple genes. Cattle dramatically increase their intramuscular fat content in the longissimus dorsi muscle between 12 and 27 months of age. We used the annealing control primer (ACP)-differential display RT-PCR method to identify differentially expressed genes (DEGs) that may participate in the development of intramuscular fat between early (12 months old) and late fattening stages (27 months old). Using 20 arbitrary ACP primers, we identified and sequenced 14 DEGs. BLAST searches revealed that expression of the MDH, PI4-K, ferritin, ICER, NID-2, WDNMI, telethonin, filamin, and desmin (DES) genes increased while that of GAPD, COP VII, ACTA1, CamK II, and nebulin decreased during the late fattening stage. The results of functional categorization using the Gene Ontology database for 14 known genes indicated that MDH, GAPD, and COP VII are involved in metabolic pathways such as glycolysis and the TCA cycle, whereas telethonin, filamin, nebulin, desmin, and ACTA1 contribute to the muscle contractile apparatus, and PI4-K, CamK II, and ICER have roles in signal transduction pathways regulated by growth factor or hormones. The final three genes, NID-2, WDNMI, and ferritin, are involved in iron transport and extracellular protein inhibition. The expression patterns were confirmed for seven genes (MDH, PI4-K, ferritin, ICER, nebulin, WDNMI, and telethonin) using real-time PCR. We found that the novel transcription repressor ICER gene was highly expressed in the late fattening stage and during bovine preadipocyte differentiation. This information may be helpful in selecting candidate genes that participate in intramuscular fat development in cattle.

Effects of High Levels of Nutrients on Growth Performance and Carcass Characteristics of Hanwoo Cattle

  • The present study was conducted to evaluate the effects of high levels of nutrients on the growth performance, blood metabolites and carcass characteristics of Hanwoo cattle. Eighteen Hanwoo steers were fed two types of diets: 1) Nine animals were fed the conventional diet including typical levels of crude protein (CP) and total digestive nutrients (TDN), and 2) Nine animals were fed the treatment diet including high levels of CP and TDN. The average body weight (BW) and dry matter intake (DMI) were greater (P < 0.05) in the treatment group than in the conventional group at early and late fattening stages. Also, in the treatment group, the average daily gain (ADG) was greater (P < 0.05) at the late fattening stage. The serum total lipid and cholesterol levels were higher (P < 0.05) in the late fattening stage of treatment group. The carcass weight, total fat weight, longissimus muscle area and the grade of meat quality were also greater (P < 0.05) in the treatment group than the conventional diet group. This study demonstrates that high levels of CP and TDN exhibit a positive effect on the growth performance and carcass characteristics, indicating that high levels of CP and TDN can be used as a cost-effective feeding program for Hanwoo cattle by shortening the feeding period.

Effect of fattening period on growth performance, carcass characteristics, and economic traits of Holstein steers

  • This study was conducted to investigate the effect of different fattening periods on the growth performance, carcass characteristics, and economic traits of Holstein steers. Sixty Holstein steers (8.0 ± 0.28 months old) with an average body weight (BW) of 231.88 ± 2.61 kg, were randomly allocated to five different fattening period treatments: 20, 21, 22, 23, and 24 months (n = 12 in each treatment group). Final BW and average daily gain (ADG) did not differ among the treatment groups during the early fattening period. At the late stage of the fattening period, the final BW of steers in the 24-month treatment group (812.84 kg) was greater (p < 0.05) than that of steers in the 20-month treatment group (750.39 kg). During the same period, steers in the 20- and 21-month treatment groups had a significantly higher (p < 0.05) ADG than those in the 22-month treatment group. The highest ADG (1.36 kg/day) was found in the 20-month treatment group (1.36), followed by the 21- (1.33 kg/day), 22- (1.22 kg/day), 23- (1.21 kg/day), and 24- (1.14 kg/day) month treatment groups. The feed conversion ratio (FCR) increased as the fattening period increased, and the FCR was 12.88% lower in the 20-month treatment group than in the 24-month treatment group. However, no significant differences were detected in back-fat thickness, loin area, marbling score, and chemical characteristics (water, crude protein, and crude fat content) among the treatment groups. The composition of fatty acids including C18:0, C18:1, saturated fatty acids, unsaturated fatty acids, and poly-unsaturated fatty acids did not differ among the experimental groups. As the fattening period increased, production costs increased, resulting in a decrease in gross income. The gross income for steers in the 24-month treatment group was 35.8% and 23.5% lower than that for steers in the 20- and 21-month treatment groups, respectively. Taken together, the best performance, including the ADG, FCR, and gross income, was obtained when the fattening program of the Holstein steers lasted 20 months.

  • This study analyzed the effect on weight and average daily gain, carcass grade, and blood characteristics and immunity in case of dietary addition of pegmatite to Hanwoo steers. Relative to Hanwoo steers, initial body weight was $257.3{\pm}11.74kg$ (8-months old after birth). Forty-eight Hanwoo steers were fed for 24 months using random arrangement (4 treatment plots${\times}$4 heads${\times}$3 repeat). The treatment plot (498.75~500.82 kg) had higher weight during the early fattening stage compared with that of the control plot (474.37 kg) (p<0.05). Regarding total cholesterol of the blood, the value was similar to one during initiation of testing, but the value increased gradually based on the progress of fattening. In addition, the triglyceride was considerably low in T2~T3 plots with high addition of pegmatite during the late fattening stage compared with other treatment plots (p<0.05). This study revealed a tendency in the IgG content that represented immunity as the addition of pegmatite became higher (p>0.05). This study found that the treatment plot (441.55~452.10 kg) had higher carcass weight compared with the control plot (436.30 kg). However, this study revealed that the treatment plot (2.30~2.55 points) had higher carcass quantity grade compared with the control plot (2.11 points). This study found that the control plot (3.55 points) had higher carcass quality grade compared with the treatment plot (2.67~3.09 points) (p>0.05). With studies combined, it was determined that dietary addition of pegmatite to the feed as clay minerals in fattening Hanwoo steers would improve feed intake, body weight gain and carcass quantity grade.

거세한우에 있어서 배합사료의 적정 TDN 수준과 도축 월령 (The Optimal TDN Levels of Concentrates and Slaughter Age in Hanwoo Steers)

  • 본 실험은 성장단계별(육성기-비육전기-비육중기-비육후기) 배합사료의 적정 TDN 농도 구명시험을 위하여 저 TDN (T1; 70-70-71- 72%), 중 TDN(T2; 70-71-72-73%) 및 고 TDN (T3; 70-72-73-74%)의 3처리와 출하월령 차이 시험을 위해 26, 27, 28, 29, 30, 31개월령의 6처리를 두고 수행하였다. 공시축은 6개월령 수송아지 90두를 입식하여 외과적 수술로 거세를 실시한 후, 18개 우방에 5두씩 배치하였다. 육성기, 비육전기, 비육중기의 배합사료 급여량은 일당 증체 0.7-0.9kg가 되도록 제한급여 하였고, 비육후기는 무제한 급여하였다. 조사료는 육성기에 오차드그라스 건초(Dactylis glomerata L.)만을 급여하였고, 비육전기동안에는 건초를 점차적으로 볏짚으로 대체하였고, 비육중기 이후에는 건초만을 급여하였다. 월령별 TDN 수준에 따른 체중과 사료섭취량의 차이는 없었다. 일당증체량은 19-21개월령 경에 1일 0.9kg 이상의 증체성적을 보였다가 다시 점차 감소하였으나, 최저 0.7kg 이상은 유지하였다. 모든 공시축의 배합사료 섭취량 평균을 체중비로 계산한 결과를 보면, 육성기 1.0%-1.3% 범위에서 비육전기에 1.5%까지 증가하고, 비육중기 이후부터 다시 감소하여 비육후기로 전환되는 시점에서 1.4% 수준까지 감소하였다. 그 후에도 계속 감소하여 30개월령에는 1.0% 수준이었다. 출하월령이 늦어짐에 따라서 등심단면적, 등지방 두께 그리고 근내지방도가 꾸준히 증가하였고, 특히 29개월령을 기점으로 유의성있는 증가(P<0.05)를 보였다. 월령별 1등급이상 출현율은 27, 28개월령이 86, 80%로 가장 낮았고, 26개월령이 93%, 그리고 29, 30, 31개월령은 100%의 출현율을 보였다. 도체율은 출하 월령이 증가함에 따라 증가하였으나(P<0.05), 거래정육율은 도체율 증가에 따라 감소하였다(P<0.05). 1일 두당 순이익은 26- 28개월령에 출하할 경우에는 차이가 없었으나, 29개월령 이후에는 약 32-46%가 향상되었다. 본 실험과 같은 사양관리 조건 하에서는 TDN 수준간에 증체, 사료섭취량, 도체등급 등에서 차이가 없었고, 육질 1등급이상 출현율이 100%로 나타나기 시작한 29개월령이 적정 출하월령으로 판단된다.
