This study was carried out to investigate on the survival and in vitro developmental rates of bisected bovine embryos by microblade, micropipette and pronase methods. Bisected embryos cultured for 1∼7 days in TCM-199 media with 10 FCS+hormones. Survival and in vitro developmental rates was defined on in vitro culture or FDA-test. The results are summarized as follows ; 1. The survival and in vitro developmental rates of bisected bovine embryos by microblade, micropipette and pronase methods were 22.2, 16.7, 15.0% and 22.2, 23.3, 18.8%, respectively. In vitro developmental rate of bisected bovine embryos was significantly lower than that of non-bisection embryos(27.8% and 25.0%). 2. In vitro developmental rates of bovine embryos bisected for 1, 2, 4, 8, 16 cells stages during in vitro culture in 10% FCS+TCM-199 media were 25.0, 20.0, 20.0, 15.0 and 6.7%, respectively. 3. In vitro developmental rates of intact and free-zona pellucida of bisected demi-embryos during in vitro culture in 10% FCS+TCM-199 media were 25.6, 16.7%, respectively. 4. In vitro developmental rates of biopsied embryos and biopsied blastomeres during in vitro culture in 10% FCS+TCM-199 media were 20.0, 11.1%, respectively.
Many in vitro developmental toxicity assays have been proposed over several decades. Since the late 1980s, we have made intermittent attempts to introduce in vitro assays as screening tests for developmental toxicity of inhouse candidate products. Two cell-based assays which were developed two decades apart were intensively studied. One was an assay of inhibitory effects on mouse ascites tumor cell attachment to a concanavalin A-coated plastic sheet surface (MOT assay), which we studied in the early days of assay development. The other was an assay of inhibitory effects on the differentiation of mouse embryonic stem cell to beating heart cells (EST assay), which we assessed more recently. We evaluated the suitability of the assays for screening in-house candidates. The concordance rates with in vivo developmental toxicity were at the 60% level. The EST assay classified chemicals that inhibited cell proliferation as embryo-toxic. Both assays had a significant false positive rate. The assays were generally considered unsuitable for screening the developmental toxicity of our candidate compounds. Recent test systems adopt advanced technologies. Despite such evolution of materials and methods, the concordance rates of the EST and MOT systems were similar. This may suggest that the fundamental predictivity of in vitro developmental toxicity assays has remained basically unchanged for decades. To improve their predictivity, in vitro developmental toxicity assays should be strictly based on elucidated pathogenetic mechanisms of developmental toxicity.
Purpose: This study examined the perceived quality of life of families of individuals with developmental disabilities who regularly participate in physical activities. It also explored differences in the quality of family life based on variables such as family relationships, family income, gender, disability type, and age. Methods: A survey was administered to 69 family members of individuals with developmental disabilities who regularly participate in physical activities. The collected data were analyzed using descriptive statistics, independent samples t-test, and one-way analysis of variance. Results: The average overall quality of life for families of individuals with developmental disabilities who participate in continuous physical activities was 3.49. After analyzing each subtype, psychological health was found to be 3.91, the burden for people with developmental disabilities was 2.75, community participation was 3.76, cohesion was 3.83, and openness was 3.49. No statistically significant differences were found in family relationships or family classes, disability type, age, or gender of the individuals with developmental disabilities across the variables. Conclusion: When planning service development and support to improve the quality of life for families of individuals with developmental disabilities, it is necessary to develop systematic programs at the family level rather than providing support based on various variables.
Purpose: This study used a descriptive investigative design to identify educational needs for developmental supportive care for the purpose of establishing a developmental supportive care education program for nurses in neonatal intensive care units. Methods: A survey was conducted on 93 nurses working in neonatal intensive care units located in a metropolitan area, and the collected data were analyzed using descriptive statistics and a paired t-test with the SPSS 25.0 program. For the purpose of analyzing educational needs, we further analyzed data through an importance-performance analysis (IPA) and the Borich needs assessment and the locus for focus models. Results: The average importance of the developmental supportive care of nurses in neonatal intensive care units was 3.74±0.78 out of 5, and the average performance was 3.46±0.81 out of 5. A t-test on the difference between the IPA and Borich needs assessment showed the highest educational need in the categories of 'individualized care', and 'I serve on the Developmental Care Committee at my institution'. In addition, according to the results of deriving the priorities of educational needs using the Borich needs assessment and the locus for focus model, the highest priority was 'I provide appropriate pain management when noxious procedures are necessary'. Conclusion: These findings can be used as basic data to design a developmental supportive care program suitable for nurses in neonatal intensive care units to meet the educational needs for developmental supportive care.
The problems of growth & development due to maladjustment are gradually increasing while need for the treatment of children's diseases is decreasing. The level of developmental deficiency or delay correlates with neonatal birth weight and also with gestational age, i.e. degrees of prematurity. There-fore, developmental defects and potential risk factors' are more Common in premature infants than in full term infants. The purpose of this study is to define the difference in the growth at developmental status between premature and full term infants, and to define the relation between the developmental status and the physical growth during the first 3 years' Data were collected from January 10, 1985 to April 6, 1985 at 3 hospitals including St. Mary's Hospital, and through home visiting. The subjects of this study consisted of 79 Premature infants (G.A. <37wks. & B.W. <2.5kg) and 94 full term infants (G.A.≥37 wks. & B.W.≥2.5kg). The study method used was a questionnaire, anthropometric assessment and DDST for normative data of growth & development. The collected data were analyzed using descriptive statistics, chi-square test and t-test. The results of the study were as follows: Hypothesis: 1 : That the prematures will differ from the full term infants in the physical growth status during the first 3 years was partially supported (p<0.02) : The prematures reached up the full term infants in the physical growth status in the first 6 months. And, the first hypothesis was supported (P<0.01) : There are more cases which is below‘the Korean children's physical. growth standards’in prematures than in full term infants. Hypothesis 2 : That the prematures will differ from the full term infants in the developmental status during the first 3 years was supported (P< 0.001);‘Normal’developmental status due to DDST was less in prematures than in full term infants. And, the second hypothesis was Partially supported (P<0.02) : The developmental status of the pre-matures was different from that of the full term infants within the first 3 months by analysis of passed items in DDST, Hypothesis 3 : That the prematures' developmental status will relate to their physical growth during the first 3 years was supported (P<0.001) : If the prematures' developmental status is in delayed status, then, their physical growth status is also in delayed status. This study shows that the prematures differed significantly from the full term infants in the growth at developmental status during their infancy. This means that the nurse can foster the growth & development of the prematures by supportive care during their infancy. Further longitudinal study is needed to verify these findings for the environmental factors.
Purpose. The purpose of this study is to investigate effects of group play therapy program on improvement of social skills of children with developmental disabilities. Methods. With 4-year children with developmental disabilities with developmental problems, group play therapy program with 16 sections, 40minutes for each section for 16 weeks was conducted, and by using Korean infant social skills test (K-SSRSP), social skills index (SSQ) between before and after applying group play therapy program on children with developmental disabilities were compared. Results. When group play therapy are applied on children with developmental disabilities, it was found that there was improvement of social skills including problem solving, emotional expression, sense of order and self-confidence. Conclusions. According to the result of this study, if more specific studies regarding group play therapy program are conducted and they can reflect on other integrated programs, then it is expected that there will be greater synergy effects on improving not only children with developmental disabilities but also social skills of them.
Embryonic stem (ES) cells have potential for use in evaluation of developmental toxicity because they are generated in large numbers and differentiate into three germ layers following formation of embryoid bodies (EBs). In earlier study, embryonic stem cell test (EST) was established for assessment of the embryotoxic potential of compounds. Using EBs indicating the onset of differentiation of mouse ES cells, many toxicologists have refined the developmental toxicity of a variety of compounds. However, due to some limitation of the EST method resulting from species-specific differences between humans and mouse, it is an incomplete approach. In this regard, we examined the effects of several developmental toxic chemicals on formation of EBs using human ES cells. Although human ES cells are fastidious in culture and differentiation, we concluded that the relevancy of our experimental method is more accurate than that of EST using mouse ES cells. These types of studies could extend our understanding of how human ES cells could be used for monitoring developmental toxicity and its relevance in relation to its differentiation progress. In addition, this concept will be used as a model system for screening for developmental toxicity of various chemicals. This article might update new information about the usage of embryonic stem cells in the context of their possible ability in the toxicological fields.

In vitro developmental rates of bisected bovine embryos co-cultured in 10% FCS+TCM-199 media containing PMSG+hCG, PMSG+$\beta$-estradiol, hCG+$\beta$-estradiol, PMSG, hCG 0 to 3 days and 4 to 7 days were 16.7~30.0% and 11.1~25.0%, respectively. In vitro developmental rates of bisected embryos co-cultured in 10% FCS+TCM-199 media containing hormones significantly higher than that of non co-culture. 2. In vitro developmental rates of bisected bovine embryos co-cultured 10% FCS+TCM-199 media containing oviductal epithelial cells 0 to 3 days and 4 to 7 days were 25.0% and 22.2%, respectively.
Purpose: This study was conducted to estimate the prevalence of questionable development according to developmental screening testing for children, and to explore biological and environmental risk factors of developmental delays. Method: The study participants were 153 children under two years of age. A questionnaire and Home Observation for measurement of the Environment (HOME) inventory were used to collect data, and Korean Denver II was administered. Results: The overall prevalence of questionable development was 11.1%. Significant differences in biological factors between the normal development group and questionable development group were the rate of prematurity, twins, the educational level of mother and father's and the presence of an illness in the mother (p<.10). Maternal acceptance in HOME was significantly different between the two groups. The factors related to questionable development that were significant in the logistic regression analysis were prematurity (OR=3.56, p=.026), and maternal acceptance in HOME score (OR=.629, p=.028). Conclusion: Early developmental screening tests seem necessary for all children, especially for prematurely born children. Also, child rearing environments were identified as significant factors in the development of all children. These findings suggest that the HOME scores might be useful in identifying children at risk for developmental delays, and that interventions for these children will probably be more effective if their mothers are helped to provide a more appropriate social environment.
Purpose: To develop an Infant Developmental Assesment Scale appropriate for Korean infants that includes cultural context. Method: The total sample included 990 infants aged birth to 24months. The developmental tests were conducted at hospital well baby clinic, daycare centers, public health centers and homes located in city, urban and rural areas. Item analysis was done to examine content validity of the scale. Test responses were analyzed by SPSS to examine the inter-rater reliability, and construct validity. Result: Based on the naturalistic observation and analysis of other developmenta l instruments, 260 Korean infant developmental items were created. The mean score of the developmental quotient of 966 infants was 101.63±10.89. Correlation coefficient of inter-rater reliability was .99. In addition, 84.6% of total items showed Kappa .70 and above. The development quotient was significantly different depending upon the residential area and father's occupation, therefore, construct validity using known group technique was supported. Conclusion: The developed instrument can be used to assess the development of handicapped infants, as well as normal infant's general abilities and to study individual differences within the normal range for Korean.
