Methods for sampling and analysis of marine microalgae in ship ballast tanks: a case study from Tampa Bay, Florida, USA

  • Ballasting and deballasting of shipping vessels in foreign ports have been reported worldwide as a vector of introduction of non-native aquatic plants and animals. Recently, attention has turned to ballast water as a factor in the global increase of harmful algal blooms (HABs). Many species of microalgae, including harmful dinoflagellate species, can remain viable for months in dormant benthic stages (cysts) in ballast sediments. Over a period of four years, we surveyed ballast water and sediment of ships docked in two ports of Tampa Bay, Florida, USA. Sampling conditions encountered while sampling ballast water and sediments were vastly different between vessels. Since no single sample collection protocol could be applied, existing methods for sampling ballast were modified and new methods created to reduce time and labor necessary for the collection of high-quality, qualitative samples. Five methods were refined or developed, including one that allowed for a directed intake of water and sediments. From 63 samples, 1,633 dinoflagellate cysts and cyst-like cells were recovered. A native, cyst-forming, harmful dinoflagellate, Alexandrium balechii (Steidinger) F. J. R. Taylor, was collected, isolated, and cultured from the same vessel six months apart, indicating that ships exchanging ballast water in Tampa Bay have the potential to transport HAB species to other ports with similar ecologies, exposing them to non-native, potentially toxic blooms.

  • Three strains of pathogenic bacteria isolated from diseased red sea bream at a culture farm in Gyoung sangnam-Do during the midwinter of 1987 were studied taxonomically. Based on their biological and biochemical characteristics, they were identified as Vibrio sp.. They exhibited very similarities to group II (Kusuda et al.,1979) except that the former can grow at lower temperature.

The Residues of Enrofloxacin in Cultured Paralichthys olivaceus (양식산 넙치에서 Enrofloxacin의 잔류)

  • Enrofloxacin is one of the normally used flouroquinolones in mammalian and fish but its withdrawal time and studies were remain obscure. The residual contents of enrofloxacin in fish muscle were analyzed by using HPLC-FLD. More than 0.1 mg/kg of ENR was detected in muscle tissues and the residues were found over 1 year after treatment. The concentration of ENR in Paralichthys olivaceus was not affected by water temperature and lasted for an extended amount of time. The spike recoveries of ENR in the muscle tissue ranged from 78% to 85%. From this results, we need the prescription by veterinarian or aquatic organism disease inspector in ENR usage to assure safety of fish. Future research is required to determine the recommendation dose of ENR for side effects and safety.

  • Formalin, 37% formaldehyde, has been employed as a chemical for controlling ectoparasites and aquatic fungi responsible for infectious fish of diseases in hatcheries and culture facilities (Roberts, 1978; Schnick, 1991; Rach et al., 1997). Regarding the commercial usage of formaldehyde in the aquaculture industry, the U.S. Food and Drug Administration (FDA) approved three commercial products as parasiticides in a species-specific manner: Paracide-F and Formalin-F for bluegill, catfish, largemouth bass, salmon, and trout and Parasite-S for all finfish (FDA, 1998). Withdrawal time for these products was legally zero when used as permitted under the regulations. With the increased production of cultured fish in Korea, such as olive Hounder Paralichthys olivaceus and black rockfish Sebastes schlegeli, application of formalin to diseased fish has become more frequent. Moreover, there is still some concern about environmental exposures caused by effluents from fish culture facilities. The purpose of the present study was to evaluate residues in fish resulting from therapeutic usage of formalin in the aquaculture industry and to document the rate of disappearance of formaldehyde in seawater treated with formalin. Evaluation of Ciliate Euplotes sp. as a Live Food for Marine Fish Larvae (해산 자치어의 먹이생물로써 섬모충 Euplotes sp.의 평가)

  • This study was carried out to evaluate the ciliates Euplotes sp. as a live food for marine fish larvae. The ciliates and the rotifers Brachionus plicatilis, which were cultured with the baker's yeast Saccharomyces cerevisiae and the $\omega$-yeast emulsified with cuttle fish liver oil, were supplied to the larvae of flounder Paralichthys olivaceus and grouper Epinephelus akaara. Considering the size difference between the ciliates 68 $\cdot$ 7 $\mu$n and the rotifers 160 $\cdot$ 20 $\mu$n, the rotifers and ciliates were supplied to the larvae tank with the density of 2 inds./mL and 20 inds./mL, respectively. The survival rate and growth in length of the flounder larvae fed on rotifer were significantly higher than those on Euplotes sp.. In grouper larvae which have a small mouth diameter, even the survival rate of the larvae fed on the ciliates was better than that on the rotifers, it was very low less than $20\%$. Therefore, Euplotes sp. seem to be incongruent as a live food for marine fish larvae.

Establishment and Characterization of Permanent Cell Lines from Oryzias dancena Embryos

  • The development of species-specific fish cell lines has become a valuable tool for biological research. In recent years, marine medaka Oryzias dancena has been recognized as a good experimental model fish but there are no reports of establishment of cell lines from this fish. In this study, two cell lines from O. dancena blastula embryos were established from 41 total trials (4.9%). The two cell lines displayed typical in vitro morphology and have been cultured for >121 passages, which corresponds to 293 days. The doubling times of the cell lines were 29.84 and 28.59 h, respectively, and both possessed the potential to expand in a clonal manner, albeit with significant differences between the two cell lines. The absence of any of the four main medium supplements; i.e., fish serum, fetal bovine serum, basic fibroblast growth factor, and medaka embryo extract, significantly inhibited growth. The proportion of cells possessing normal chromosome number was 45% and 46.7% of the cell lines, respectively. Taken together, two cell lines that proliferate continuously were established from marine medaka and these cell lines may provide a basic tool for characterizing the unique features of this fish species.

Application on Microwave Energy in the Preparation of Fish Samples for Electron Microscopic Observation

  • Chemotherapy can not be applied for the control of fish viral diseases because viruses depend on host machinery for their replication. Although new control strategies including vaccination are under development, avoidance of virus introduction by rapid and correct diagnosis is the best way of fish viral disease control. Although observation of virus particles with an electron microscope is an easy method for virus detection, it take a few days for the sample preparation. In order to shorten the sample preparation time, microwave radiation was applied in the procedure. With this method, 15 seconds was enough for fixation of virus infected fish samples or cultured cells inoculated with infectious hematopoietic necrosis virus, which takes 2-4 hours with routine methods. Also four minutes was enough for polymerization of embedding resin which takes 24-48 hours with routine methods. Samples prepared with microwave were good enough for direct electron microscopic observation and immunogold labeling assay.

Sitological Quality Evaluation of Cultured and Wild River Puffer, Takifugu obscurus (Abe) (양식산과 자연산 황복, Takifugu obscurus (Abe)의 식품학적 품질평가)

  • The present study was performed to evaluate sitological quality of the cultured and wild river puffer, Takifugu obscurus. Proximate composition, the content of extractive nitrogen, the content of nucleotides and their related compounds, total and free amino acid, and fatty acids were analysed and sensory evaluation in the muscle of the river puffer were compared. The cultured river puffer had a higher moisture content compared to the wild fish, while there was no significant difference in crude lipid, crude protein and ash contents. Nucleotides and their related compounds including ATP, ADP, AMP, IMP, HxR and Hx were detected. The result from analyzing ATP-related compound showed difference in total content by wild and cultured river puffer, and IMP content that had largest influence upon the savory taste of sliced raw fish, was higher in the cultured fishes than wild ones. Breaking strength level of the wild river puffer was higher than that of the cultured fish. High levels of C16:0 and C18:0 were shown in all samples and -3 polyunsaturated fatty acid content were not different between the cultured and wild river puffers (P>0.05). Total 17 amino acids were detected in the samples, and most of the samples had high contents of glutamic acid, aspartic acid, lysine and leucine and low contents of cystine, histidine, methionine and tyrosine. The result from surveying free amino acid content of wild and cultured river puffer showed difference in content, but generally taurine and lysine content for the whole free amino acid held the most part. There was no significant difference in texture, flavor and overall acceptance score between the cultured and wild fishes (P>0.05).

Effects of the Culture Broth of Lactic Acid Bacteria Cultured in Herb Extracts on Growth Promotion and Nonspecific Immune Responses of Aquacultured Fish (한약재 추출물에 배양된 유산균 배양액이 양식어류의 사료첨가제로서 성장과 비 특이적 면역에 미치는 영향)

  • We have investigated the effects of the culture broth of lactic acid bacteria (LAB) cultured in herb extract on growth, hematological parameter, nonspecific immune responses and disease resistance of olive flounder (Paralichthys olivacells) and parrot fish (Oplegnathus fasciatus) for 12 weeks. Weight gain of olive flounder fed diet with mixture was not significant among the control group. But In parrot fish, was significantly higher 20g than control group. The feed efficiency of olive flounder were 25% higher in the experimental groups than in the control. There were no significant differences in feed efficiency among each group on parrot fish. Treatment of olive flounder contents of GOT and GPT in serum decreased after 8 weeks. But there were no significant differences in GLU and TP among each group. Also, there was no significant of NBT reduction. The activities of lysozyme were higher in experimental group of olive flounder than in the control after 8 weeks. On the other hand, activities of lysozyme were triple higher in the experimental group of parrot fish than in the control after 12 weeks. In the oliver flounder case, the survival rate (%) after an artificial challenge with $10^7$ CFU/ml of Vibrio anguillarum and Streptococcus iniae per fish, was 18% higher in the experimental groups than the control. The higher survival rate of parrot fish were 17% and 16% in the experimental groups than the control respectively.

Statistical data on fish virus of cultured olive flounder, Paralichthys olivaceus from 2005 to 2007 (2005년부터 2007년 사이 양식 넙치, Paralichthys olivaceus를 대상으로 한 어류바이러스 검출에 대한 통계 자료)

  • The epidemiological study was performed to survey the prevalence of fish pathogens in cultured olive flounder, Paralichthys olivaceus from 2005 to 2007. In this study, the fish pathogens were detected from 1,528 among 2,238 fish samples collected yearly in 5 sites from February, May, August and November. Annual incidences for three years show a yearly increase and there were 60.6% in 2005, 66.7% in 2006 and 72.3% in 2007, respectively. Seasonal prevalence was 63.5% in February, 67.4% in May, 75.1% in August and 64.4% in November for three years. The detection rates of 6 viral pathogens were 35.6% in 2005, 44.6% in 2006 and 24.4% in 2007 and the peak rate was 55.4% at adult size group (above 41cm). Viral nervous necrosis virus (24.7%) has been the most predominant virus in this investigation, while much lower rates were noted in viral haemorrhagic septicemia virus (10.6%) and red sea bream iridovirus (0.9%).
