As basic study to evaluate the treatability of oil-contaminated environment with bacteria, isolation and characterization of crude oil-degrading bacterium were carried out. A bacterial strain SH-14 capable of degrading crude oil was isolated from contaminated soils by enrichment culture technique and identified as Acinetobacter sp. by morphological, cultural and biochemical characteristics, and so named Acinetobacter sp. SH-14. The optimal medium composition and cultural conditions for the growth and emulsification of crude oil by Acinetobacter sp. SH-14 used were crude oil of 2.0%, $KNO_3$ of 0.2%, $K_2HPO_4$ of 0.05%, and $MgSO_4\;{\cdot}\;7H_2O$ of 1.0%, along with initial pH 7.0 at $30^{\circ}C$. Acinetobacter sp. SH-14 showed to be resistant to chloramphenicol and utilized various hydrocarbons such as dodecane, hexadecane, isooctane, cyclo-hexane etc., as a sole carbon source. Acinetobacter sp. SH-14 harbored a single plasmid. By agarose gel electrophoresis and curing experiment it was found that the genes for crude oil components degradation were encoded on the plasmid.
Scientific investigation of cultural heritage can provide important information to understand the context of the object. To know the characteristics of the material is also an essential part of objects management and conservation. However, the identification and characterization of organic dyes used in archaeological and historical textiles are often limited by the restrictions in sampling. To cope with the difficulties, applications of high-performance techniques of surface analysis, such as Time of Flight-Secondary Ion Mass Spectra (TOF-SIMS) could be considered as a non or micro-destructive option. This study aims to examine the applicability of TOF-SIMS analysis to the detection of organic dyes from historical textiles. A group of silk fabrics dyed with vegetable dyes were artificially photo-aged to different degrees and analyzed with TOF-SIMS. Molecular and fragment ions from indigo were successfully detected from the aged samples; however, only some fragment ions were observed from gardenia and safflower dyed fabrics. Further studies with actual historical samples with extended examination scope would be necessary to assess the validity of this technique.
This study investigates the parameters that affect the choice of Korean medical services, as well as the relationship between the cultural characteristics and the behavioral intention to use Korean medical services. The cultural characteristics of Chinese and Russian individuals have an impact on the national image and image of Korea's medical services, and that this affects their behavioral intention to use Korean medical services. Nevertheless, there were differences among the cultural characteristics, in terms of specific elements. Among Chinese individuals, the national image is affected by 'power distance', 'individualism-collectivism', and 'long-term orientation'; for Russians, it is affected by 'individualism-collectivism', 'power distance', 'uncertainty avoidance', and 'long-term orientation'. On the other hand, for Chinese, the image of Korean medical services is affected by 'power distance' and 'long-term orientation', while for Russians it is affected by 'power distance' and 'uncertainty avoidance'. These in turn influence their behavioral intention to use Korean medical services. As such, by recognizing these cultural properties, and by designing and offering suitable services with these in mind, Korean medical institutions can nurture among foreign visitors greater satisfaction and a desire to revisit.
This study was carried out to investigate occurence of microbiales density and characteristics of xylanase produced by those in Janggyeong Panjeon. Cladosporium cladosporioides H1, Penicillium citreonigrum H3, Penicillilum toxicarium H4, Aspergillus versicolor H6, Acremonium alternarium H7 isolated from Janggyeong Panjeon produced xylanase, which had different production rates and specialized activities in an acidic condition. Cladosporium cladosporioides H1, Aspergillus versicolor H6, and Acremonium alternatum H7 produced xylanase at a faster rate than other fungi. A xylanase of Cladosporium cladosporioides H1 and Penicillilum toxicarium H4 showed a high thermostability in an acidic condition. As results, this study may lead to the development of a strategy for preservation of organic cultural heritages.
We examined characterizations of Paraloid B72 films when it dissolved in four different solvents-acetone, MEK, toluene and xylene. The surface before and after coating were observed by optical microscope and AFM. Thickness and contact angle of films was measured and surface energy was calculated for grasping properties of films. We also tested adhesive strength of films by scratch tester and durability of films by performing yellowing test. The result was that xylene has better coating property than other solvents. Because 10% Paraloid B72 in xylene film is even, non-mottled, thin, stabilized in yellowish and has proper hydrophobic, we confirmed xylene is good solvent for bronze artifact conservation. However, other tests will need for finding appropriate concentration of Paraloid B72.

The materials of sculptured stone property around the Hyeonleung Royal Tomb in Guri consist mainly of high weathered of granitic rocks with magnetite-series. Deterioration characteristics occurred highly with microorganism, soil inflow and black contaminants at the burial mound zone. As a result of deterioration evaluation, stone surface around the burial mound zone show serious comprehensive damage of soil inflow (50.5%) and lichen coverage (47.6%) which are the major damaging factors, and there are about 8.6% of biological growth noticeably in the north side. Surface contaminants and the discoloration had the complex causes. Gypsum occurred between joints of stones and the major contaminant type, darkening which was analyzed organic bodies caused discoloration. From result of ultrasonic velocity measurements, there are mean value of 2,195 m/s with highly weathered (HW) grade. Most of the stone properties showed 4 to 5 weathered grade. Therefore, it turned out that sculpture stone properties require conservation treatments. To remove soil accelerating damage factors and lichen occupying high percents, the cleaning process is necessary and consolidation, rejoining and filling are needed as well. Also, consideration on removing conservation materials containing Ca and e fflorescence is required and retreatments need to be considered.

The purpose of the present study is to examine the objects excavated from the archaeological site of the Nogye Village in terms of their relationships with the iron production facilities such as 1ho smithing hearth and 2ho smelting furnace. 32 samples including slags, iron ores, and wall were analyzed to identify the mineralogical and chemical characterization. In addition, in the case of the 1ho smithing hearth slag, differing points of the cross-section were analyzed to examine its formation in depth. The analysis results suggest that the slags from the each site adjacent to the 1ho smithing hearth and 2ho furnace are related to smithing and smelting process respectively. Furthermore, it is possible to draw some conclusions that the aspects of the increased contents of nonferrous elements such as CaO in the slags in comparison with those of the iron ores are due to various factors such as charcoal ash and analytical reasons rather than an addition of CaO as flux.
In this study, materials and color formation techniques were assessed for black potteries excavated from the Janghyeon-dong, Jungsan-dong and Gyodong-ri sites during the Proto-Three Kingdoms period in Ulsan, Korea. Although the black potteries were black superficially, the inner cores were either black or reddish yellow. Microscopy analysis identified that body clay was used for reddish iron oxide rich soils with quartz, alkali feldspar and mica, along with grains of myrmechite texture. Additionally, as marginal differences exist in the contents of SiO2, Fe2O3 and CaO, the composition of the host rock and clay distributed around the sites was affected. Thus, we can deduce that pottery was made by soiling at a short distance. Raman spectroscopy results revealed that the black layer of the black pottery was used as amorphous combustion carbon. In addition, as a transparent layer of brown lacquer was observed on the substrate that was in contact with the surface layer, the black layer of the pottery induced black color development by a combination of combustion carbon and lacquer. Based on the mineral composition and microtexture of the body clay, the firing temperature of the potteries seemed to range from 750 to 850℃, whereas the lacquer layer was pyrolyzed at 468℃ by thermal analysis. Therefore, a combined layer of combustion carbon and lacquer, which formed the black color, was painted after the body clay was fired.
Optimal media and cultural conditions for the production of red pigment were established using Monasurs anka KFCC 4478. The optimal temperature and initial pH for the production of red pigment were 30$^{\circ}C$ and 7.0, respectively. Glucose turned out to be most suitable carbon source for red pigment production. Optimal glucose concentration was 3.0%. Addition combined of nitrogen sources of peptone and NaNo3 induced good red pigment production. Thiamine-HCI and nicotinic acid were increased the production of red pigment. Under optimal conditions, maximum red pigment production and cell growth were observed after 5 days of incubation.
Several bacterial strains utilizing crude oil as their sole carbon and energy sources were isolated from marine. One of the strains named KCL-1 showed the highest degradative activity for crude oil and the best growth rate. This strain was identified as a Klebsiella sp. based on the morphological, biochemical, and physiological characteristics. The optimum cultural conditions were as follows; $32^{\circ}C$~$37^{\circ}C$ for temperature and 7.0 for initial pH. Additionally, the optimal concentration of sodium chloride was 3.0%, indicating that this strain was derived from seawater. KCL-1 could use several kinds of n-alkane hydrocarbons from octadecane to hexacosane as a sole carbon source. The degradation of crude oil by KCL-1 was stimulated by addition of octadecane in the culture. The emulsifying activity by KCL-1 was highest after 3 days of cultivation under the condition of 3.0% sodium chloride, pH 7.0 and $37^{\circ}C$.
