본 연구는 운동부하와 DNA 및 칡 crude catechin의 급여에 의한 생체내 성분 변화를 알아보기 위하여 흰쥐를 대조군, 운동부하군, 운동부하+DNA 및 운동부하+DNA+crude catechin 첨가군으로 나누어 사육하였다. 각각 0.4% DNA와 0.1% 칡crude catechins을 해당 횐쥐에 급식시켰으며, 운동부하군은 Treadmill에서 20 m/min의 속도로 6주 동안 운동시켰다. 사육실험종료 후 체중 증체량과 체지방무게를 먼저 칭량한 후, 항산화활성과 체지방조성을 kit assay를 이용하여 측정하였다. 6주후 각 군간의 최종 체중에는 유의적인 차이가 없었으나 간장, 부고환주위지방 및 신장주위지방은 운동부하군, 운동부하+DNA 첨가군 및 운동부하+DNA+crude catechin 첨가군이 대조군에 비해 유의하게 낮은 값(p<0.05)을 나타냈다. 또, 혈중 phospholipid, cholesterol 및 triglyceride농도는 운동부하군, 운동부하+DNA 첨가군 및 운동부하+DNA+crude catechin 첨가군이 대조군에 비해 유의하게 낮았으나(p<0.05), HDL-cholesterol은 유의하게 높은 값을 나타냈다. GPT 활성은 각 군간에 유의차가 없었으나, GOT활성은 운동부하를 처리한 군에서 적은 수치를 나타냈으며, TBA가는 각 군간에 유의 차가 없었다. 또, 혈중 NEFA 수준은 운동부하+DNA+crude catechin 첨가군이 대조군에 비해 유의하게 높은 값을 나타냈다.(p<0.05). 이상의 실험에서 운동과 더불어 crude catechin 섭취가 유의한 차이를 나타내지는 않았지만 혈청지질의 성분의 향상에 보조역할을 할 것으로 사료된다.
Crude catechin extracts were prepared using ethyl acetate from Korean Puerariae Radix (PR) and their biological effects on the alcoholic liver damage were investigated in male Sprague-Dawley rats (6 weeks old). Ethanol (5g ethanol/kg body weight/day) administration was performed for 8 weeks and after ethanol consuming rats were treated with one or two% catechin extracts of diet for 8 weeks. At the end of experimental period, lipid hydroperoxides in liver were analyzed using a chemiluminescence-high performance liquid chromatography (CL-HPLC) method. Compared with control animals, ethanol consumed rats showed lighter body weights, lower ratios of liver/body weight, higher activities of GOT and GPT, and increased lipid hydroperoxide amount in liver. With one or two% catechin extracts treatment, GOT and GPT activities returned to normal ranges. Lipid hydroperoxide contents in liver of on or two% PR treated rats lowered to 20% or 25% respectively, compared with the levels of those ethanol consumed animals without catechin extracts treatment. Therefore, we concluded that on or two% PR crude catechins treatment could be effective for alcoholic liver damage caused by lipid peroxidation.
시판 녹차, 오룡차 및 홍차에서 추출된 조카테킨 조성 및 이들 카테킨의 2성분 항산화시스템, 3성분 항산화 시스템의 항산화 효과를 조사하고자 하였다. 녹차, 오공차 및 홍차의 조카테킨의 수율은 각각 5.4%,3.5% 및 2.5%이 었다. EGC, EC, EGCg 및 ECg의 4개의 카테킨류가 확인되었으며, EGCg 성분이 가장 많은 함량을 보였다. 녹차, 오룡차 및 홍차의 총 카테킨의 함량은 각각 926.3ug/g, 713.8ug/g 및 318.0ug/g이었다. 녹차, 오룡차 및 홍차 조카테킨 첨가구의 항산화 효과는 오룡차 > 녹차 > 홍차의 순이었다. 녹차, 오룡차 및 홍차 조카테킨(100ppm)에 $\alpha$-tocopherol(50ppm)을 첨가했을 때의 항산화효과는 녹차 > 오룡차 > 홍차 조카테킨 순으로 녹차에서 가장 강한 상승작용을 나타냈다. 녹차, 오룡차 및 홍차 카테킨(100ppm)에 $\alpha$-tocopherol(50ppm) 및 ascorbyl palmitate(50ppm)를 함께 첨가한 경우, 조카테킨(200 ppm)을 단독으로 첨가한 경우보다 항산화 효과가 강하였다.

Ulmus davidiana var. japonica is a deciduous tree used in traditional medicine. In this study, we examined the antioxidant and anti-inflammatory activity of extracts and compounds isolated from U. davidiana var. japonica stem barks for development of cosmetic phyto-materials. Phytochemical examination of the stem bark led to the isolation and characterization of three known compounds, (+)-catechin (1), (+)-catechin-7-O-${\beta}$-D-apiofuranoside (2), and procyanidin B3 ((+)-catechin-($4{\alpha}{\to}8$)-(+)-catechin) (3). Their bioactivities including antioxidant (DPPH ree radical scavenging assay) activity, anti-inflammatory (nitric oxide nhibition assay) were evaluated. Most of the crude extracts and isolates indicated significant antioxidant potential compared with BHT and ${\alpha}$-tocopherol as controls. Futhermore, all compounds showed higher inhibitory activities for NO production in Raw 264.7 cells than the L-NMMA using the positive control. Eespecially, (+)-catechin (1) and (+)-catechin-7-O-${\beta}$-D-apiofuranoside (2) which could inhibit more than 90% of the NO production at a concentration of 100 ${\mu}g/m{\ell}$, implying excellent anti-inflammatory activity.
The present study was conducted to elucidate the functional property of tea extract obtained from tea extracts, semi-fermented tea(oolong tea) and fermented tea(black tea). Tea extracts exhibited remarkable nitrite-scavenging actions, and the action increased with the increased of the amount of tea extracts. The nitrite-scavenging actionof tea extracts showed pH dependent, highest at pH 1.2 and lowest at pH6.0. The nitrite-scavenging rate in tea extracts/amine/nitrite systems proved to be faster than that in amine/nitrite systems. To screen the nitrite-scavenging factors, tea extracts were fractionated into water-soluble, methanol-soluble, methanol-precipitate and crude catechin fraction. Among these fractions of tea extracts, the crude catechin fraction possessed greater nitrite-scavenging action than the other fractions. The nitrite-scavenging action of tea extracts increased with the contents of total phenols and an absorbance at 280nm, nitrite-scavenging factors were supposed to be and catechins in tea polyphenol compounds.
This study investigated the physiological and antioxidant activities of green, oolong, and black tea extracts. The crude catechin extract yields of green, oolong, and black tea were 4.9%, 3.4%, and 2.5%, respectively. Total phenol contents of green, oolong, and black tea were 40.9%, 43.0%, and 38.5%, respectively. The order of the electron donating abilities of green, oolong and black tea were green tea>oolong tea>black tea extracts. The SOD-like activities of green, oolong and black tea extracts at 5,000 ppm were 21.2%, 17.5% and 13.9%, respectively. The nitrite-scavenging abilities of green, oolong and black tea extracts were higher than that of ascorbic acid (p<0.05). Antioxidant activities in soybean oil substrates at 500 ppm were in the order of green tea>oolong tea>black tea${\geq}$BHT (200 ppm). Therefore these results showed that the physiological and antioxidant activities of green tea were better than those of oolong and black tea.
Antimicrobial effect of tea extracts from green tea(steamed, roasted), oolong tea and black tea was investigated. Minimal inhibitory concentration(MIC) of tea extraxcts against 9 well known strains of foodborne pathogenic bacteria such as the Gram positive and Gram negative bacteria was determined at 37$^{\circ}C$. Significant antimicrobial activity was observed in the steamed green tea and the roasted green tea of the water-soluble fraciton, and the steamed green tea of the methanol-soluble fraction, and the steamed green tea, roasted green tea and the oolong tea of the crude catechin fraction. The MIC of these extracts against B. subtillis were 700$\mu\textrm{g}$/ml, 500$\mu\textrm{g}$/ml and 120 $\mu\textrm{g}$/ml, respectively. The crude catechin fraction possessed greater antimicrobial activity than did the other fractions. Among tea extracts, extracts of steamed green tea, roasted green tea and oolong tea showed higher antimicrobial activity than them of black tea. The MIC of the crude catechin fraction obtained from tea extracts against Gram-positive bacteria such as M. Iuteus, B. subtillis and S. mutans were 30~50$\mu\textrm{g}$/ml, 120~240$\mu\textrm{g}$/ml and 120~180$\mu\textrm{g}$/ml, and against Gram-negative bacteri such as e. aerogenes and V. parahaemolyticus were 50~60$\mu\textrm{g}$/ml and 60~70$\mu\textrm{g}$/ml in the broth medium, respectively. Especially, the MIC to Streptococcus mutans which has known as a causative bacteria of a decayed tooth were 120$\mu\textrm{g}$/ml, 140$\mu\textrm{g}$/ml, 180$\mu\textrm{g}$/ml and above 1,000$\mu\textrm{g}$/ml in steamed green tea, roasted green tea, oolong tea and black tea, respectively. Tea extracts had strong growth inhibition activity against foodborne pathogenic and dental bacteria.
돈지에 대한 녹차 물추출물의 항산화력을 녹차 조카테킨과 대등한 수준으로 향상시키기 위하여 적절한 상승제를 선정하고 그 첨가량을 검토하였다. 녹차 조카테킨의 항산화력은 물추출물보다 약 3.3배 강하였다. 녹차 물추출물의 항산화 상승제로는 ${\delta}-tocopherol$과 레시틴이 적당하였으나 이 두 가지의 상승성은 서로 달랐다. 즉, 녹차 물추출물과 혼합된 ${\delta}-tocopherol$은 첨가량 0.05%까지 현저한 상승효과를 보였으나 그 이후는 변화가 없었으며, ${\delta}-tocopherol$의 상승효과는 녹차 물추출물이 0.1% 이상 첨가되면 오히려 감소하였다. 반면에 녹차 물추출물과 레시틴이 혼합된 시료에서는 각각의 첨가량이 증가할수록 상승효과가 계속 향상되는 경향을 보였다. 한편, ${\delta}-tocopherol$과 레시틴의 복합적인 상승효과를 보기 위하여 이들을 녹차 물추출물과 병용한 결과 대단히 높은 항산화 상승효과가 나타났으며, 돈지에 녹차 물추출물 0.1%, ${\delta}-tocopherol$ 0.05% 및 레시틴 1.0%를 첨가한 시료의 항산화력은 무첨가구의 약 8.1배, 녹차 조카테킨 0.1%와 1%를 첨가한 경우보다 각각 1.6배 및 1.2배 강하게 나타났다.
