  • The aims of this study was to elucidate the correlation between cotyledon numbers and seedling growth and some other traits of Pinus koraiensis. The results are summurized as follows; 1. A highly significant correlations between numbers of coty ledons and juvenile needles was found but no significances between cotyledon numbers and juvenile needle length, width and stomata row numbers. 2. The accessory resin canals did not begin to appear before June but began to appear only small numbers of seedling after July. It was observed that the number of accessory resin canals of juvenile leaves at various growing stages were not related with the number of cotyledons. 3. In the case of adult leaves, there was a significant difference in the number of leaves and the number of cotyledons, but no significant difference between the number of cotyledons and needle length, needle width and stomata row. 4. There were significant differences in the number of cotyledons and top height growth, and root caliper growth among individuals and due to ages. 5. Seedlings with more than 12 cotyledons showed better growth than those with 9 to 11 cotyledons.

Measurement of and Changes in L-carnitine Levels in Developing Cucumber Cotyledon (오이 떡잎의 발달 과정에서 carnitine의 검출과 변화)

  • Mobilization of storage lipids is critical for the germination of oil seeds, as they supply carbon and energy until photosynthesis commences in cotyledons. In this study, we determined the levels of plant carnitine and associated changes in these levels from seed germination to cotyledon senescence. We also examined changes in the content of unsaturated fatty acids throughout seedling development. Carnitine levels peaked on day 3 at 14.5 nM in cotyledons and decreased sharply to 7.2 nM on day 4. On development day 3 carnitine levels were maintained at around 3 nM until day 7. The unsaturated fatty acid content dropped by half at the same time as carnitine peaked (day-3), and storage lipids were almost depleted by day 5. Thereafter, carnitine was hardly detected until the second stage of cotyledon senescence, at which stage the carnitine content was 6.8 nM, similar to that on day 4 at the time of fatty acid depletion in the cotyledons. Unsaturated fatty acids levels remained constant in green cotyledons but slightly increased in the senescing cotyledons. The latter can be explained by intracellular breakdown of membrane lipids. This is the first such discovery in developing cotyledons and may offer clues regarding other roles of the acetyl unit transport system in plants. The expression of BOU was closely associated with carnitine metabolism during seed germination and cotyledon development. The results provide support for the possibility of carbon re-routing during the glyoxylate cycle in the supply of energy for early germination and development.

  • In order to study the rate control step in the protein metabolic course of the chlorophyll formation, the transaminase activities which are obtained freely in the extracts of cotyledons of germinating peas at the light and the dark places, are measured in Beckman mopel D.U, spectrophoto meter at 490 mu. In this case, of two enzymatic reaction products; oxalacetic acid and pyruvic acid, the former is converted to pyruvic acid by aniline citrate and after each pyruvate phenyl hydrazones are extracted by toluenes: when this is treated with strong alcoholic alkali, a colored hydrazone is formed and it is measured by above apparatus. The estimated G.O.T. and G.P.T. in the germinated cotyledons at dark and light places considerably differ in their activities; G.O.T. and G.P.T. activities which are formed at the light are more increased than at the dark and also they differ in their rates through germination, though G.O.T. activity increment is smoothly but that of G.P.T. is more sharply, and they are considered to be directly affected to the chlorophyll formation and indirectly to the growth. G.O.T. and G.P.T. in each fractions of cell in the cotyledons should be formed by dissociation of zymogens in the microsomal fractions and it seems to promoted by light. In the formation of the chlorophyll, the protein metabolism occurred mainly in the microsomal fractions and the rate determining step is found at the point where the zymogene that is able to produce G.P.T. is activated, and this activation is promoted by light as noted above.

  • The Merit variety of soybean (Glycine max L.), harvested in 1971 was germinated in the dark at $21{\sim}25^{\circ}C$ for days. The soybean sprouts were divided into cotyledons and seedling axis (=hypocotlyplus root) and subjected to the determination of dry weight, crude fat content and lipid components (esterified sterols, triglycerides, free fatty acids, free sterols and phospholipids) at two-day intervals during the germination periods. The results are summarized as follows: 1) During the germination period, the dry weight and crude fat content in cotyledons decreased continuously, but the dry weight seedling axis increased continuously and crude fat content remained almost constent. 2) The triglyceride content in crude fat from cotyledons decreased and free fatty acid content increased continuously, but triglyceride content in crude fat from seedling axis showed no change until 6th day and increased slightly after 8th day, and free fatty acid content showed increase after 4th day and decrease after 6th day. Phospholipids, free and esterified sterols content in cotyledons increased continuously, but their content in seedling axis remained unchanged.

  • Developmental changes of chlorophyll-protein complexes (CPs) and plastid membrane proteins in greening mung bean cotyledons and the effect of spermine therein were examined by SDS-polyacrylamide gel electrophoresis. The changes in the amounts of CPs became larger with the progress of greening and light-harvesting chlorophyll a/b protein (LHCP) was the main CP in the early greening stage up to f h. As the greening proceeded, chlorophyll-protein of the photosystem I (CPI) accumulated. Application of spermine were effective in accumulating CPs of the thylakoid membrane in the early phase of greening. In the profiles of the plastid membrane proteins, quantitative and qualitative changes were observed with the onset of greening up to 72 h. 56 kD protein of major intensity was observed in all greened chloroplasts and 24 kD protein increased remarkablly in both control and spermine-treated cotyledons. The thylakoids from spermine-treated cotyledons showed hither amounts of thylakoid proteins as compared to the controls. The results suggest that spermine may play a role in the regulation of plastid development and stabilizes the membrane function during greening.

  • This experiments were carried out to determine the optimum concentrations of plant growth regulators for the direct embryogenesis from the cotyledon culture of Paeonia lactiflora Pall. Zygotic embryos rescued from true seeds were developed abnormally in the medium containing ABA. But somatic embryogenesis from cotyledons of abnormal seedlings was induced more effectively. Also the somatic embryogenesis from cotyledons was promoted in the medium containing ABA. The frequency of embryogenesis was maximum(59.9%) from the cotyledons cultured on MS medium containing 0.5 mg/L ABA on which the frequency of somatic embryos with two cotyledons was 22.6%.

Effect of carbon sources on somatic embryogenesis and cotyledon number variations in carrot (Daucus carota L.)

  • In order to investigate the effect of carbon sources on somatic embryogenesis and cotyledon number variations in carrot, embryogenic callus were cultured in the medium supplemented with various concentrations of sucroseor glucose, and with combination of 2% sucrose and various concentrations of mannitol or sorbitol. The maximum number of somatic embryos formed per flask (1,555.70) was obtained in the medium supplemented with 2% sucrose rather than glucose alone or a combination of mannitol or sorbitol and 2% sucrose, and the number of somatic embryos was decreased with the increasing of mannitol or sorbitol concentration. The frequencies of somatic embryos with two cotyledons were 35.14% for sucrose, 19.88% for glucose, 32.55% for mannitol + 2% sucrose, and 38.59% for sorbitol + 2% sucrose, respectively, and the frequencies of abnormal somatic embryos having 3 or more cotyledons were 64.86% for sucrose, 80.12% for glucose, 67.44% for mannitol + 2% sucrose, and 61.41% for sorbitol + 2% sucrose, respectively. Particularly, the frequency of somatic embryos with two cotyledons (59.16%) was the highest in the 2% sucrose medium compared to the frequency of abnormal somatic embryogenesis with three or more cotyledons, and the frequency gradually decreased with increasing concentration of glucose, mannitol or sorbitol. According to these results, it was found that the ratio of abnormal somatic embryo was higher than the normal somatic embryo in carrot, and was shown that somatic embryogenesis and the cotyledon number was affected by the concentrations of sucrose, glucose as carbon source, and mannitol and sorbitol as osmotic agents in culture medium.

  • The Merit variety of soybean (Glycine max L.), harvested in 1971 was germinated in the dark at $21{\sim}25^{\circ}C$ for 10 days. The soybean sprout were divided into cotyledons and seedling axis and subjected to the determination of lipoxygenase activity and fatty acid composition of triglycerides, free fatty acids, phosholipids and crude fat fractions at two-day intervals during the germination periods. The results are summarized as follows 1) The lipoxygenase activity in cotyledons declined sharply after second day, but the activity in seedling axis inclined slightly after second day. However, the decrease of lipoxygenase activity in cotyledons coincided with decrease of linoleic and linolenic acids in cotyledons and increase of lipoxygenase activity in seedling axis coincided with increase of those acids in seedling axis. 2) The iodine value of neutral fat in cotyledons decreased continuously, but the iodine value of the neutral fat in seedling axis remained almost constant. iodine value in cotyeldons was greater than in seedling axis. 3) In the fatty acid composition of triglycerides in cotyledons, palmitic acid did not changes significantly, stearic acid increased continuously, oleic acid changed irregularly, linoleic and linolenic acids continuously decreased significantly. But in the fatty acid composition of triglycerides in seedling axis, palmitic acid remained unchanged, linoleic and linolenic acids slightly increased continuously, stearic and oleic acids changed irregularly. 4) Composition of free fatty acids in cotyledons and seedling axis changed irregularly, suggesting that all fatty acids produced by hydrolysis of triglycerides by lipase are used(or either biosynthetic Purpose or energy Production at random. 5) Fatty acids with odd-numbered carbon chain were not detected in the triglycerides and free fatty acid fractions during the germination periods, suggesting that all fatty acids are utilized as $C_2$-unit in degradation and biosynthesis. 6) The changes of fatty acids composition of Phospholipid in cotyledons and seedling axis during the germination were similar to these of triglyceride fraction.

Early Selection of Double Flowers Based on Cotyledon Shape in Cut Stock (Matthiola incana L.) Flowers

  • Selection of double-flowered plants at the seedling stage is one of the main purposes of stock breeding programs. Eight stock cultivars of Matthiola incana L. named 'Nobel', 'Cinderella', 'Pacific', 'Avalanche', 'Midblue', 'Lavender', 'Goddess' and 'Esfahan', with different percentage of double-flowered plants were used for examining the relationship with three morphological types of cotyledons. The results of a chi-square test indicated that in heart-shaped (HC) and cup-shaped cotyledon (CC) populations, the number of plants with double flowers was much more than that of single flowers and CC seedlings rarely produced single flowers. Therefore, increasing the number of CC seedlings can improve the percentage of double flowers. The highest and lowest numbers of CC seedling were observed in high double and single flower cultivars, respectively. Single flower cultivars showed the maximum count of dumbbell-shaped cotyledons. Chromosome pairing of these cultivars was evaluated using the squash technique. Aneuploid cells were found in 'Nobel' and 'Goddess' cultivars, which showed the highest percentage of CC seedling. Based on morphological measurements, the highest value of inflorescence size was observed in the seedlings with cup-shaped cotyledons.
