Comparison of Physicochemical and Organoleptic Characteristics of Omija Wines Made by Different Methods (제조방법에 따른 오미자 발효주의 이화학적 및 관능적 특성의 비교)

  • Physicochemical and organoleptic characteristics of omija wines made by traditional method, adding grape juice and sugar solution periodically, and with dry omija were compared. The pH values of all omija wines were ranged $2{\sim}3$ during fermentation. The acidity value of omija wine made by traditional method was 2.5%, that of omija wine made by adding grape juice and sugar solution periodically decreased from 3.3% to 0.8%, and that of omija wine made with dry omija increased from 0.2% to 3.9%. Sucrose and alcohol contents were $6.5{\sim}34.5^{\circ}Brix$ and 12% at the end of fermentation, respectively. The viable cell numbers of yeast decreased from $5.7{\sim}6.9\;\log\;CFU/mL$ to $4.3{\sim}4.6\;\log\;CFU/mL$. Omija wine made by adding grape juice and sugar solution periodically had the highest sensory scores for color, taste, flavor, swallowing, and overall acceptability, and was significantly different from the both omija wines made by traditional method and with dry omija. Because omija is rarely fermented due to the little fermentative sugar content, omija wine made by adding grape juice and sugar solution periodically was shown to be the most appropriate.

  • Commercial greenhouse plant factories are highly efficient for controlling external factors such as floods, drought, insects, air pollution etc. However, they require substantial startup & maintenance investments and experimental research to optimize production. These facilities are especially useful for urban farming where high efficiency in small spaces is required. In this study, we investigated whether light emitting diode (LED) lights with mixed dominant wavelengths (650 nm : 550 nm : 445 nm=8:1:1, 650 nm : 445 nm=6:4) can increase the growth rate and bio-active compound content of carrots in comparison to that of fluorescent light (FL). LED with mixed wavelength (650 nm : 550 nm : 445 nm=8:1:1) increased the total weight and root circumference of carrots compared to FL. However, ${\beta}$-carotene contents were not significant in LED (650 nm : 550 nm : 445 nm=8:1:1). However, LED (650 nm : 445 nm=6:4) increased the ${\beta}$-carotene (FL: 7.27, LED: 10.48 mg/g ${\beta}$-carotene dried weight). These results suggested that using LED light at the ideal wavelength, at the antithesis color of the plant, might enhance plant growth and bio-active compound contents.

Biological Activity of Ethanol Extracts from Amelanchier asiatica Fruits (채진목(Amelanchier asiatica) 열매 에탄올 추출물의 생리활성)

  • Amelanchier asiatica fruits have been used as a traditional medical food. This research was investigated to assess angiotensin converting enzyme, xanthine oxidase (XOase) and elastase inhibitory activity and antioxidant activities. The content of total phenolic compounds in A. asiatica fruits extracts was 17.6mg/mL. In extracts, the electron donating ability by 1,1-diphenyl-2-picrylhydrazyl free radical scavenging test of A. asiatica fruits extracts was 90.18% at $200{\mu}g/mL$. The 2,2'-azinobis(3-ethylbenzothiazoline-6-sulfonic acid radical decolorization of A. asiatica fruits extracts was 98.81% at $200{\mu}g/mL$. The inhibition rate of the antioxidant protection factor was 1.03, and thiobarbituric acid reactive substance was 73.27% at $200{\mu}g/mL$. The XOase inhibition activity of A. asiatica fruits extracts of showed to be 13.19% at $200{\mu}g/mL$. The angiotensin converting enzyme activity was significantly inhibited by A. asiatica fruits extracts as 82.52% inhibitory rate at $200{\mu}g/mL$. Elastase inhibitory activity in the A. asiatica fruits extracts (41.48% at $200{\mu}g/mL$) was higher than vitamin C (12.8% at $200{\mu}g/mL$). These results suggests that A. asiatica fruits extracts have the greatest property as a functional food and functional cosmetic source.

  • Animal cell culture industry has a large market and an exponential growth rate among biological industry field. Chines hamster ovary(CHO) cells are the most widely used cell lines for recombinant protein production. They can avoid infection from polio, herpes, hepatitis B, HIV, measles, adenovirus and etc. Moreover it is easy to transfection recombinant genes and possible to suspension culture. Serum free media is one of the most important factor of protein production. Because serum has problems. Serum is not defined the contents until now, it has a number of proteins, lipids, carbohydrates and unknown molecules that cause of risk involve in infection and high cost of product purification. CHO cell line cultured using serum free media were the basis of a very successful method to produce(glyco-)protein in mammalian cells, which are then used as pharmaceutical products. Also, the low protein content of the developed medium facilitates downstream processing and product purification. But non-adapted CHO cells have a limit of proliferation cultured using serum free media and it takes very long time to adapt non-adapted cells to serum free media. There are a number of causes of a limit of proliferation using serum free media. Absence of growth factors and growth stimulating molecules is a major factor of the reasons. It makes growth signals and moves cell cycle. And increase of cellular stress is another reason. It induces increase of intraceullar ROS concentration. The purpose of this study is about improvement of proliferation capacity of non-adapted CHO cells cultured using serum free media without adaptation process.

Comparative Study on Protein and Amino Acid Composition of Wild and Cultured Eel (천연 및 양식산 뱀장어의 단백질 및 아미노산 조성비교)

  • The muscles of wild and cultured eel, Anguilla japonica, were analyzed for the protein composition and amino acid profile. The differences of the subunit distribution for the sarcoplasmic and myofibrillar proteins were discussed with sodium dodecylsulfate(SDS) polyacryamide gel electrophoresis. The muscle protein in wild eel was composed of $30.78\%$ of sarcoplasmic, $59.02\%$ of myofibrillar, $9.73\%$ of residual intracellular and $2.47\%$ of stroma fraction. That in cultured eel was composed of $31.81\%,\;58.37\%,\;8.16\%\;and\;1.80%$, respectively, The sarcoplasmic and myofibrillar proteins were composed of 16 and 14 subunits in wild eel, and 22 and 15 subunts in cured eel. The sarcoplasmic protein between wild and cultured muscles showed a similar trend in the subunits, except a few subunits such as 36,500, 46,000, 58,500, 75,000, 170,000 and 235,000 daltons in cultured eel. Only the existence of 45,000 dalton subunit was the difference between wild and cultured eel in myofibrillar protein. The distribution patterns of total amino acid in muscles of wild and cultured eel were found to be very similar trend, although glycine content in wild eel was slightly higher than that in cultured one.

  • RFID that provide automatic identification by reading a tag attached to material through radio frequency without direct touch has some specification, such as rapid identification, long distance identification and penetration, so it is being used for distribution, transportation and safety by using the frequency of 125KHz, 134KHz, 13.56MHz, 433.92MHz, 900MHz, and 2.45GHz. Also it is one of main part of Ubiquitous that means connecting to net-work any time and any place they want. RFID is expected to be new growth industry worldwide, so Korean government think it as prospective field and promote research project and exhibition business program to linked with industry effectively. RFID could be used for access control of person and vehicle according to section and for personal certify with password. RFID can provide more confident security than magnetic card, so it could be used to prevent forgery of register card, passport and the others. Active RFID could be used for protecting operation service using it's long distance date transmission by application with positioning system. And RFID's identification and tracking function can provide effective visitor management through visitor's register, personal identification, position check and can control visitor's movement in the secure area without their approval. Also RFID can make possible of the efficient management and prevention of loss of carrying equipments and others. RFID could be applied to copying machine to manager and control it's user, copying quantity and It could provide some function such as observation of copy content, access control of user. RFID tag adhered to small storage device prevent carrying out of item using the position tracking function and control carrying-in and carrying-out of material efficiently. magnetic card and smart card have been doing good job in identification and control of person, but RFID can do above functions. RFID is very useful device but we should consider the prevention of privacy during its application.

Effect of pH Control on Physico-chemical Characteristics of Chicken Breast Surimi (닭가슴살 Surimi의 이화학적 특성에 미치는 pH 조절의 영향)

  • This study was conducted to determine the effect of pH adjustment on physico-chemical characteristics of chicken breast surimi. The chicken breast meat was ground with distilled water, of which pH was then adjusted to 2.5 (T1), 3.0 (T2), 10.5 (T3) and 11.0 (T4) for surimi manufacture, respectively. Water content was higher in order of T4>T1>T3>T2 (p<0.05). Crude protein and crude fat were higher in T3 and T4 compared with T1 and T2 (p<0.05). $L^*$ values, myofibrillar protein and water holding capacity of T2 and T4 were higher than those of T1 and T3 (p<0.05). T4 had the lowest yield among the treatments (p<0.05). T1 was higher in yield and pH, whereas breaking force and deformation were higher in T1 (p<0.05). $a^*$ was higher in order of T3>T2>T4>T1 and $b^*$ was lower in T1 compared with other treatments (p<0.05). In textural properties, the chewiness values of T2 and T3 were higher than those of T1 and T4, the hardness was higher in order of T2>T3>T4>T1 (p<0.05). Cohesiveness and gumminess of T1 showed higher values than those of other treatments (p<0.05). In sensory evaluation, the note for appearance was higher in T2 than other treatments (p<0.05), however other traits were not significantly different (p>0.05). Therefore, the alkaline processing (T4, pH 11.0) would be recommended.

Anti-thrombotic Activities of Hot-water Extracts Prepared from Various Parts of Nelumbo nucifera Gaertner (다양한 부위의 연(Lotus) 열수 추출물의 항혈전 활성)

  • To investigate the anti-thrombotic activities of the lotus (Nelumbo nucifera Gaertner), various hot-water extracts were prepared from the leaf, pod of seed (PS), seed, embryo of seed (ES), root, and the node of root (NR) of the lotus. The highest extraction ratio was found in the NR (20.3%), followed by the seed, root, leaf, ES, and PS. These extracts had pH and acidity levels ranging from 5.6~6.5 and 0.06~0.20%, respectively. The seed extract showed 70% brix, whereas the leaf and PS extracts showed less than 0.1% brix. The highest contents of total polyphenol (179.7 mg/g), total flavonoids (161.4 mg/g), and reducing sugar (161.4 mg/g) were observed in the leaf extract, and the highest total sugar content (873.0 mg/g) in the seed extract. Anti-coagulation assay of the extracts of NR, leaf, and PS showed strong activities. In particular, at a concentration of 5 mg/ml, the PS extract had 15-fold extended thrombin, prothrombin, and activated partial thromboplastin times. However, only the ES extract showed activities inhibitory to platelet aggregation, with treatment with 0.25 mg/ml of ES extract decreasing platelet aggregation to 25.1%, a reduction comparable to that effected by aspirin. The extracts other than the seed extract showed no hemolysis activities against human RBC at treatments of up to 1 mg/ml. These results suggest that the NR, PS, seed, and ES, all byproducts of the lotus agriculture industry, have high potential as novel sources of anti-thrombotic agent.

Changes of Chlorophyll Contents in Spinach by Growth Periods and Storage (성숙시기 및 저장에 따른 시금치의 클로로필 함량 변화)

  • This study was carried out to determine the changes of chlorophyll contents in spinach during growth stages and storage with two packaging materials. The chlorophyll contents increased to a maximum level in the spinach leaves after 35 days of planting, while the level in the roots extremely decreased and that in the stems did not change during the growth period. The total chlorophyll content was higher in the leaves than that in the stems and roots. The ratios of chlorophyll a/b in the leaves and stems were almost similar to those reported for spinach elsewhere. When stored for 1, 3, and 5 days without packaging, the remaining percentage of chlorophyll was $96.29\%,\;90.79\%\;and\;85.74\%$, respectively, at $5^{\circ}C,\;and\;82.75,\;44.63,\;33.07\%$, respectively, at $20^{\circ}C$. The chlorophyll contents of the spinach stored in polyethylene bags were significantly higher than of that those without packaging and of that packaged with newspaper during storage. From the results, it was found that spinach packaged in polyethylene bags suppressed the degradation of chlorophyll and maintained the freshness during low temperature storage and that the chlorophyll contents increased during the growth period.

Comparison of Food Component between Conger eel (Conger myriaster) and Sea eel (Muraenesox cinereus) as a Sliced Raw Fish Meat (생선회로서 붕장어 (Conger myriaster) 및 갯장어 (Muraenesox cinereus)의 식품성분 비교)

  • In Korea, the conger eel, Conger myriaster and sea eel, Muraenesox cinereus have been eaten as a sliced raw ash meat for a long time. This study was carried out to compare the food component and sensory differences of the conger eel and sea eel as a sliced raw fish meat. The yield and ash content of conger eel were slightly higher than those of sea eel, and no significant differences in moisture, crude protein and crude lipid. The major fatty acids of conger eel were 16:0 ($19.9\%$), 18 : 1n-9 ($36.1\%$) and 22: 6n-3 ($8.3\%$), while those of sea eel were 16:0 ($24.0\%$), 16: 1n-7 ($12.1\%$) and 18:1n-9 ($21.9\%$), The contents of (n-3)-fatty acids were low in all of the two samples. The contents of total amino acid in the conger eel and sea eel were 19,147.2 mg/100 g and 18,527.1 mg/100 g, respectively. The major amino acids of two samples were aspartic acid, glutamic acid and lysine. The chemical score in protein of conger eel was higher about $6\%$ than that of sea eel, and the Ca and P contents of conger eel were also higher than those of sea eel. Although the nutritional value of conger eel as a sliced raw fish meat was generally higher than that of sea eel, the sensory scores in taste and texture of conger eel were lower than those of sea eel.
