Rich Phase Separation Behavior of Biomolecules

  • Phase separation is a thermodynamic process leading to the formation of compositionally distinct phases. For the past few years, numerous works have shown that biomolecular phase separation serves as biogenesis mechanisms of diverse intracellular condensates, and aberrant phase transitions are associated with disease states such as neurodegenerative diseases and cancers. Condensates exhibit rich phase behaviors including multiphase internal structuring, noise buffering, and compositional tunability. Recent studies have begun to uncover how a network of intermolecular interactions can give rise to various biophysical features of condensates. Here, we review phase behaviors of biomolecules, particularly with regard to regular solution models of binary and ternary mixtures. We discuss how these theoretical frameworks explain many aspects of the assembly, composition, and miscibility of diverse biomolecular phases, and highlight how a model-based approach can help elucidate the detailed thermodynamic principle for multicomponent intracellular phase separation.

Transparent cryogenic thermosiphon using $N_2\;and\;CF_4$ mixture as the working fluid

  • A mixed working fluid has a potential to widen the operation temperature range of the thermosiphon. In this study, the thermosiphon using $N_2\;and\;CF_4$ mixture as the working fluid is fabricated and tested to verify its transient thermo hydraulic characteristic. A transparent pyrex glass tube was used for the thermosiphon itself and the vacuum chamber was also fabricated by glass to visualize the internal state of thermo siphon. Onset of condensation temperature was related to the partial pressure of $CF_4$. Two solidifications were observed and condensate temperature range of mixed working fluid was from 160K to 70.7K with $N_2$ 25% composition.

  • We investigate the effect of photon loss on pair production in a hybrid atom-light system. The loss of light field not only affects the generation of photons, but also prevents the generation of atomic collective excitation, although the atoms are not influenced directly. We propose an unbalanced homodyne detection of the number of atomic collective excitation that overcomes the challenge caused by counting uncertainty in practical measurement. In discussion, we show that the intermode correlations and the number correlation is closely related to the initial input state, while the quadrature correlations are independent of the initial state and always exhibit opposite intermode correlations even in the presence of loss.

Action Mechanism of Enhancers for Activating Gene Transcription

  • Enhancers are cis-elements to regulate transcription of cell/tissue-specific genes in multicellular organisms. These elements locate in upstream or downstream regions of target genes and are found in a long distance up to 100 Kb in some cases. Transcription factors and coactivators bind to enhancers in a chromatin environment. Enhancers appear to facilitate the transcription of target genes by communicating with promoters and activating them. As transcription activation mechanism of enhancers, chromatin looping between enhancers and promoters, tracking of enhancer activity to promoters along the intervening regions, and movement of enhancers and promoters into transcription condensates have been suggested based on various molecular and cellular biology studies. These mechanisms are likely to act together rather than exclusive each other for gene transcription. Understanding of enhancer action mechanism may provide a way to regulate the transcription of cell/tissue-specific genes relating with aging or various diseases.

Inhibitory Effect of Radish Juice on the Mutagenicity and Its Characteristics (무즙의 돌연변이 억제 효과 및 그 특성)

  • The inhibitory effects of radish juice on the mutagenicities of cigarette smoke condensate (CSC), methanol extract of charred part of fried or broiled saury pike (MECS) and 2-aminofluorene (2-AF) were examined by the use of Ames assay toward Salmonella typhimurium TA 98 strain. Radish juice exhibited inhibition percentage of about 100, 100 and 87 on the mutagenicities of CSC, two kinds of MECS and 2-AF, respectively. Except for juices of cabbage and leek, radish juice has inhibited more effectively the mutagenicity of CSC than other fruit or vegetable juices studied. Inhibitory effect of radish juice might be originated from the components with molecular weight above 50,000 and decreased sharply in 5 min by heat treatment at $100^{\circ}C$, but hardly changed at low and moderate storage temperatures such as $4^{\circ}C,\;10^{\circ}C,\;25^{\circ}C\;and\;35^{\circ}C$ for about 2 weeks. Precipitate obtained from ammonium sulfate saturation from 30 to 80% had inhibitory effect on the mutagenicity of CSC. Extracts from 3 bands of non-denaturing gel of $30{\sim}80%$ amnonium sulfate precipitate have exhibited the inhibitory effects on the mutagenicity of CSC.

Field Case Study for the Productivity Analysis of B2 Reservoir, Donghae-1 Gas Field (동해-1 가스전 B2 저류층의 생산성 분석에 관한 Field Case Study)

  • This paper presents the user-interactive productivity analysis model based on material balance as well as deliverability equations equipped with EOS model to perform a productivity analysis for Gorae V structure, Donghae-1 gas field. This model is designed to be able to analyse the productivity in the case of reservoir contacting with the aquifer. Also, in order to investigate the effect of condensation on productions, condensation phenomenon is considered as an apparent skin effect in the computation of bottomhole pressure from average reservoir pressure. By utilizing the developed model, we investigate the productivity analysis for B2 layer of Garae V structure with the various production cases in volumetric and non­volumetric reservoirs that contact with aquifer. From the results in the case of 5500 MMSCF/year of production and reservoir-aquifer contacting angle 270$^{\circ}C$ with aquifer size of 10 times greater than reservoir, B2 layer could maintain peak production rate even after 8.5 years of production by considering the bottomhole pressure which is estimated above the operating pressure of 1298 psia. It is also found that condensate will be formed after 1100 days of production and existed throughout the reservoir at 1270 days. Note that the computed reservoir pressure of B2 layer is maintained sufficiently high enough for production due to the water influx into the reservoir, and skin effect caused by condensation is not significant.

Extraction of organic carbon from the condensate of food waste dry feed process (음식물류폐기물의 건식사료화 공정에서 발생되는 응축수로부터 유기탄소 추출)

  • In this study, organic carbon was extracted from the condensed water of food waste drying process to estimate the applicability as external organic carbon sources. The COD, TN, TP and TS of condensed water were 21,374 mg/L, 148 mg/L, 4.19 mg/L, and 455.7 mg/L, respectively. In addition, the content of biodegradable organics in condensed water was 47%. The fractional distillation and the vacuum evaporation were employed for extracting organic carbon. There were 8 extraction conditions, but 4 conditions were available for extraction. They were 1) 0mmHg, 110℃ 2) -600mmHg, 70℃ 3) -500mmHg, 80℃ 4) -600mmHg, 80℃. All 4 conditions showed the highest organic concentration and the highest quantity of organics when extracted 10% of initial volume. It was estimated that optimum conditions were 80℃, -600mmHg and 10% extraction. Then, extraction concentration, extraction quantity, extraction efficiency, extraction time, BOD/TCOD ratio, TVFAs/TCOD ratio and NH3-N were 174,200 mg/L, 8,710 mg, 46%, 10 min, 0.97, 0.74 and 75.5 mg/L respectively. Therefore, the extracted organic carbon can be utilized as external organic carbon sources.

The Effects of Reductants on the Behaviors of Fe Selective Chlorination using an Ilmenite Ore (일메나이트 광의 Fe 선택염화 거동에 미치는 환원제의 영향에 관한 연구)

  • In this study, the behaviors of Fe selective chlorination in ilmenite ore by using PVC or $CO-Cl_2$ gas mixture as reducing agents under the condition of 1173 K, for 60 minutes were investigated. The weight loss ratio was 28% when PVC was applied as the reducing agent. The condensate formed at the outlet of reaction tube was identified as $FeCl_2$ by X-ray diffraction analysis. From these results, it was observed that iron in ilmenite ore reacted with HCl gas and Fe was selectively removed in the form $FeCl_2$. However, when $CO-Cl_2$ gas mixture was used as a reducing agent, the weight reduction ratio was 54%, and the condensate formed at the outlet of reaction tube after the experiment was estimated to be $FeCl_3$. It was observed that the ilmenite ore reacted with the $CO-Cl_2$ gas mixture and was simultaneously removed in the form of $FeCl_3$ and $TiCl_4$. However, the results of X-ray diffraction of ilmenite ore after the reaction showed that Fe was almost removed.

Development of the Pre-treatment Technology for LNG-FPSO (LNG-FPSO용 천연가스 전처리 기술 개발)

  • Submarine gas fields have focused because of the increasing fuel cost, the environmental regulations, and the safety & NIMBY problems. LNG-FPSO which is available for acid gas removal, recovery of the condensate & LPG and Liquefaction in topside process is one of high technology offshore structures. On the other hands, it is necessary to verify the pre-treatment efficiency by the ship motion and to apply to the design for LNG-FPSO. This study is to develop the pre-treatment technology for LNG-FPSO as taking account to the process efficiency by ship motion effects and the area optimization. Based on the simulation results, it founds that hybrid process shows the low circulate rate, the low heat duty and the small size of column dimensions compared to typical amine process. It will be verified the process efficiency in the various conditions by sea states as performing the 6-DOF motion test and CFD simulation.

Analysis of Geometrical Effects on Heat Transfer Characteristics in a Modular Flat Tube-Bundle Heat Exchanger (모듈형 편평원관군 열교환기의 열전달 특성 해석)

  • Flow channels with non-circular cross-sections are encountered in a wide variety of heat exchangers. Accurate friction factor and Colburn j factor data are essential for the design and viable applications of such heat exchangers. In this study, an analysis is con ducted on heat transfer and pressure drop characteristics for tube-bundle heat exchanger with various arrangements of tubes, of which their geometry could easily be modified from a circular one in a harsh environment. The parameters investigated are aspect ratio, pitch, and inclined angle of tubes. The results obtained are: (1) Aspect ratio has larger influence on the j and f factor than pitch; (2) As aspect ratio increases, both j and f factors decrease; (3) The high performance is achieved when the pitch and aspect ratio are in the range of 1.5${\~}$2.5 and 1.25${\~}$2.0, respectively; and (4) the inclined arrangements of tubes show unfavorable results for both heat transfer and pressure drop characteristics in spite of the positive possibility of condensate removals in a latent heat recovery system.
