A Comparison of Nicotine Diffusive Sampler and XAD-4 Tube for Determination of Nicotine in ETS (공기 중 니코틴 포집에 있어 국산 확산포집기와 능동포집기의 비교 평가)

  • It is widely known that Environmental Tobacco Smoke(ETS) is not good for health. ETS is composed of a lot of chemicals. So indicators are needed to evaluate the risk of ETS in air. One of the indicators is Nicotine. Active sampler has been used to measure nicotine concentration in air. The experiments were conducted to compare the active sampler method with diffusive sampler in exposure chamber and smoking areas, respectively. Sampling rate was 40.5 ml/min in exposure chamber. Experimental sampling rate (40.5 ml/min) was more than theoretical sampling rate (33.52 ml/min). And the higher was the concentration in air, the higher was experimental sampling rate. The average desorption, rate was 113.6%. The overall precision was 7.31 %. The overall accuracy was 18.96%, which were under NIOSH criteria. The average(GM) concentrations of nicotine by two sampling methods were $8.29{\mu}g/m^{3}$ (active sampler), $7.54{\mu}/m^{3}$ (diffusive sampler) in smoking area and smoking room. There was no regression between active sampler and diffusive sampler ($R^{2}=0.2397$). But slope, coefficient of determination was 1.017, 0.9292, respectively after removing outliers. And the slope (1.017) was close to the theoretical slope (1). In conclusion, this study indicated that diffusive sampler can be used to evaluate concentration of nicotine in air instead of active sampler.

  • Three different commercial particle counters were used to measure the PM2.5 particles in this study. An Aerosol Spectrometer (AS) 1.109 model of Grimm and a Particle Sensor (PS) 3030 model of HCT were compared with an Aerodynamic Particle Sizer (APS) 3321 model of TSI. The responses of these instruments were compared for four sizes ($1.0{\mu}m$, $1.5{\mu}m$, $2.0{\mu}m$ and $2.5{\mu}m$) of polystyrene latex (PSL) particles and indoor air particles of the office room. The mode diameter, particle size distribution and total particle number concentration of PSL particles were measured by each instrument. In the office room, the total particle number concentration was measured for 25 minutes. In results of particle size distribution and mode diameter, the APS 3321 (52 size-channels) was more accurate than the AS 1.109 (31 size-channels) and PS-3030 (10-szie channels) since the APS has more number of size-channels than the other instruments. However, AS 1.109 and PS-3030 provided similar results of total particle number concentration to those from the APS 3321. In results of office room test, there were no significant difference from each instrument similar to results of PSL test.

  • We developed a method to rapidly evaluate the freshness of fish using a sensor gas chromatography (SGC) system. Mackerel Scomber japonicus was stored at $4^{\circ}C$ for 12 days to create an environment similar to the natural decomposition process. Trimethylamine (TMA) content in mackerel muscle was measured at 3-day intervals using a spectrophotometer. The gas-phase concentration of TMA in whole mackerel was also determined using an SGC system. The muscle TMA content increased over time during storage, as did the gas-phase concentration of TMA. Therefore, this study demonstrated that an SGC system can be used to rapidly measure the gas-phase concentration of TMA in fish during processing.

Effect of corn grain particle size on ruminal fermentation and blood metabolites of Holstein steers fed total mixed ration

  • Objective: This study was conducted to investigate the effect of corn grain particle size on ruminant fermentation and blood metabolites in Holstein steers fed total mixed ration (TMR) as a basal diet to explain fundamental data of corn grain for cattle in Korea. Methods: Four ruminally cannulated Holstein steers (body weight $592{\pm}29.9kg$) fed TMR as a basal diet were housed individually in an auto temperature and humidity modulated chamber ($24^{\circ}C$ and 60% for 22 h/d). Treatments in a $4{\times}4$ Latin square design were TMR only (control), TMR with whole corn grain (WC), coarsely ground corn grain (CC), and finely ground corn grain (FC), respectively. The corn feeds substituted for 20% energy intake of TMR intake. To measure the ruminal pH, ammonia N, and volatile fatty acids (VFA), ruminal digesta was sampled through ruminal cannula at 1 h intervals after the morning feeding to determine ruminal fermentation characteristics. Blood was sampled via the jugular vein after the ruminal digesta sampling. Results: There was no difference in dry matter (DM) intake between different corn particle size because the DM intake was restricted to 1.66% of body weight. Different corn particle size did not change mean ammonia N and total VFA concentrations whereas lower (p<0.05) ruminal pH and a ratio of acetate to propionate, and higher (p<0.05) propionate concentration were noted when the steers consumed CC compared with WC and FC. Concentration of blood metabolites were not affected by different particle size of corn grain except for blood triglyceride concentration, which was significantly (p<0.05) increased by FC. Conclusion: Results indicate that feeding CC may increase feed digestion in the rumen, whereas the FC group seemed to obtain inadequate corn retention time for microbial degradation in the rumen.

Catechins Content and Color Values of Silk Fabrics Dyed with Korean Green Tea Extracts (녹차 염색 견포의 카테킨 함량 및 색상변화)

  • Despite several recent attempts to measure the concentration of individual catechins by HPLC, it has not been so easy to separate catechins accurately. The aim of the present work is to provide a proper condition for separating each component of catechins by HPLC analysis, and also to evaluate the experimental variables including color differences, and metal ion contents after dyeing and mordanting. Four kinds of Catechins, (-)-epicatechin(EC), (-)-epicatechin gallate(ECG), (-)-epigallocatechin(EgC), (-)-epigallocatechin gllate(EgCG) were isolated from the residues after dyeing. Catechins in Korean green tea leaves are richer when e tea leaves are younger. Higher concentration of catechins owes it to e way it is processed. The contents of catechins adsorbed in silk fabrics after dyeing were in order of EGCG>ECG>EGC>EC. We have found $68\%$ uptake of EGCG, and 116.8mg of EGCG in the silk fabrics after it was dyed with $1\%$ Korean green tea extracts. The absorbance intensity and K/S values of dyed silk fabrics were increased with dyeing temperature and time. Only the surface color of the silk fabric dyed with green tea extracts was yellowish red, but it changed from yellowish red to red with an increase in the mordant concentration. Post-mordanted silk fabrics with ferrous sulfate changed from yellowish red to red and purple color shade. In a practical evaluation, there is no significant change in color after twenty times of the continuous dry cleaning process. However, post-mordanted fabrics with ferrous sulfate faded the brightness of color after dry cleaning. Dyeing silk fabrics with a Korean tea extract reduced the metal ion contents in the silk fabrics when compared to the untreated silk fabrics. Metal contents in silk fabrics dyed and post-mordanted with $1\%$ each metal solution were 0.194 ppm for Aluminum, 1.601ppm for Copper, and 0.334 ppm for Iron. After the post-mordanting process, the heavy metal ion absorption increased, which was mainly attributed to the catechins and polyphenols in dyed silk fabrics.

Analysis and Assessment by Thermal Desorption Method of Mixed Organic Solvents Collected on Activated Carbon(AC) and Activated Carbon Fiber(ACF) (AC 및 ACF에 포집된 혼합 유기용제의 열탈착 방법에 따른 분석 및 평가)

  • This study was conducted to evaluate desorption efficiencies accuracy and precision by $CS_2$ and thermal desorption method for polar and non-polar organic solvents collected on activated carbon(AC), activated carbon fiber(ACF), carbosieve SIII, materials tested were Methyl alcohol, n-Hexane, Benzene, Trichloroethylene, Methyl isobutyl ketone and methyl cellosolve acetate and six different concentration levels of samples were made. The results were as follows ; 1. Accuracy on kind adsorbent and desorption method was low. In case of $CS_2$ desorption solvent, Overall B and Overall CV on AC and ACF were 43% and 6.63%, respectively. In case of thermal desorption method, accuracy of thermal desorption method appeared higher than solvent desorption method by AC 18.0%, 3.54%, ACF 2.6%, 2.57%, Carbosieve SIII 13.7% and 1.97%, respectively. 2. In the concentration level III, accuracy of thermal desorption method on adsorbent was in order as follow ; ACF > Carbosieve SIII > AC in the methyl alcohol and Carbosieve SIII > ACF > AC in the rest of them all subject material and Concentration levels showed good precision at EPA recommend standard (${\leq}{\;}30%$) 3. DEs by type of organic solvent adsorbent and desorption method are as follows ; In the case that desorption solvent is $CS_2$, DE of Methyl alcohol is AC 47.5%, DE of all materials is ACF about 50%. In the case of thermal desorption method, DE of Methyl alcohol is AC 82.0%, ACF 97.4%, Carbosieve SIII 86.3%. DE of the later case is prominently improved more than one of former. In particular, Except that DE of EGMEA is ACF 88.5%, DE of the rest of it is more than 95% which is recommend standard MDHS 72. With the result of this study, in order to measure various organic solvent occurring from the working environment, in the case of thermal desorption method, we can get the accurate exposure assessment, reduce the cost, and use ACF as thermal desorption sorbent which available with easy.

An Experimental Study on Thermal Conductivity Change of Water-Al2O3 Nanofluid with the Elapse of Time, Stirring, and Adding Dispersing Agent (시간경과, 교반 및 분산제 첨가에 따른 물-Al2O3 나노유체 열전도도 변화에 관한 실험적 연구)

  • A water-$Al_2O_3$ nanofluid was manufactured, and its thermal conductivity was measured in this study. The measurement was performed at volumetric concentrations of 0.5%, 1%, 2%, and 3%, and the nanoparticle sizes were 20 nm and 70 nm. Experimental test equipment, using the transient hot wire method, was installed to measure the thermal conductivity of the nanofluid, and the measured results were confirmed by measuring pure water with a measurement error of 0.92% at $20^{\circ}C$. The thermal conductivity enhancement ranged from 4.8% to 13.6% for the 20 nm particle size, and from 3.1% to 8.8% for the 70 nm particle size at a concentration range of 0.5% to 3%. The enhancement increased with a decrease in particle size and an increase in concentration. With the elapse of time after manufacturing the nanofluid, the thermal conductivity enhancement decreased significantly from 5 to 9 h, and this trend was measured under all of the measurement conditions. After 24 h, the enhancement ranged from 1.2% to 3.5% for the 20 nm particles, and from 0.6% to 2.3% for the 70 nm particles. The enhancement trends with the elapse of time were almost identical with and without stirring the nanofluid. SDBS (Sodium Dodecyl Benzene Sulfonate) was added as a dispersing agent, and the decrease in the thermal conductivity enhancement was delayed.

Distribution on the sorbic acid in cooked meat produced from meat processing plants of Gyeongbuk province

  • Sorbic acid and potassium sorbate are widely used food additives with high efficiancy and they are approved and recommended by FAO and WHO. Sorbic acid is one of preservatives that is mostly used in general food. Sorbic acid and potassium sorbate are used to prevent food, such as cheese, bread, beverage, and so on, from staling and molding. The world market for sorbic acid and potassium sorbate is growing and the market in Asian region is also expected to grow rapidly. This study was carried out to measure the amount of the sorbic acid in cooked meat produced from meat processing plants in Gyeongbuk province during 2000∼2003. One thousand one hundred and thirty-five samples of cooked meat(430 spices added meats, 486 grinding cooked meats, 3 bacons, 23 jerked meats, 68 sausages, 125 hams) were collected from meat processing plants and analyzed for the concentration of preservative sorbic acid by using of high performance liquid chromatography(HPLC). Sorbic acid was not detected in spices added meats, grinding cooked meats and bacons, except on jerked meats, ham and sausage. But the concentration range of sorbic acid in jerked meat was 0.00∼1.5g/kg, average 0.37g/kg, and in sausage was 0.00∼1.31g/kg, average 0.53g/kg, and in ham was 0.00∼ 1.22g/kg, average 0.56g/kg. There was no sample that sorbate concentration exceeded the legal permitted level 2.0g/kg in cooked meat.

Measurement of Fluorescence Correlation Function by Using Size and Concentration of Fluorescence Particles (형광입자들의 크기와 농도에 따른 형광 상관 분광함수 측정)

  • The concentration and hydrodynamic radius of nano-sized fluorescence particles diffusing in solution were compared by using fluorescence correlation spectroscopy (FCS), which can measure the variation of the correlation function of a fluorescence signal by size and number of particles. The used nano-sized fluorescence particles are Alex Fluor 647, quantum dots, and fluorescence beads, and three kinds of sample solutions with different concentrations were prepared by dilution to 1/10 and 1/100 with distilled water for each kind of particles. The effective focal volumes were calculated by using the known diffusion coefficient of Alexa Fluor 647 particles, and the diffusion time, number of particles in focal volume, and variation of concentration according to the dilution could be measured by the FCS system. Through this study, we determined that the concentrations of arbitrarily diluted sample solutions can be measured by a home-built FCS setup in the range of 0.1 nM ~ 10 nM and that the diffusion coefficient of the quantum dot was $27{\pm}1{\mu}m^2/s$.

A Study on the Fire Risk Assessment and Prevention in the Recycling Process of Used Refrigerators (냉장고 파쇄 공정에서의 화재 위험성 및 예방에 관한 연구)

  • In the recycling procedure of the refrigerator, the fire frequently breaks out. In this study, to clarify the exact cause of the fire, the components and concentration of the materials produced in the process are analysed as well as the problems in the process system, and the protective measure to prevent the fire and the explosion fundamentally is proposed. In this procedure, the preventive measures of fire by removing the combustible materials such as polyurethane and inflammable gases, by removing the ignition sources and by reducing the oxygen concentration to the minimum are proposed along with the protective measures to reduce the damage in the fire. In the crushing procedure where the fire or explosion can break out in diverse ways, the forced ventilation or exhaust system applied to the small partial ventilation facility are installed to reduce the concentration of inflammable gas mixture to lower than the inflammable limit by injecting and exhausting the air forcibly.
