The Analysis of the Genetic Variance and Combining Ability in some Quantitative Characters by Daillel Crosses of the Silkworm, Bombyx mori L. (이면교잡에 의한 가잠의 몇가지 실용형질의 유전변이와 조합능력 분석)

  • The genetic variances and combinding ability of some useful characters were analyzd on sixty four combinations in the 8$\times$8 diallel cross using the four Japanese races(Jam 107, Jam 113, Jam 117 and Jam 119) and the four Chinese races(Jam 108, Jam 114, Jam 118 and Jam 120). The eight quantitative characters were the total larval stage period(TP), the fifth larval instar period(FP), the female cocoon total weight(FW), the male cocoon total weight(MW), the female cocoon layer weight(FL), the male cocoon layer weight(ML), the female cocoon layer ratio(FR), and the male cocoon layer ratio(MR). The results were as follows: The analysis of the genetic variance and the combining ability in the TP and the FP. In TP and FP, h2N was less than h2B. The GCA, SCA and RCA were at a high significant level. Hl/D and (Hl/D)1/2 ere large. The heterosis were small minus. E and D were large. The r was in the positive direction, because the recessive genes were mainly expressed as a short rearing periods. The regressions of the characters were passed below 0 point, because the characters in the TP and the FP were appeared overdominant. The order of the dominance in the TP of the parents were in the order of Jam 119>Jam 113>Jam 117>Jam 108>Jam 120>Jam 114>Jam 107>Jam 118, and that in the FP of the parents were followed in the orders of Jam 117>Jam 113>Jam 108>Jam 114>Jam 119>Jam 107>Jam 120>Jam 118. The analysis of the genetic variance and the combining ability of the FW and the MW. In the FW and the Mw, h2N was less the h2B. The GCA and SCA were large but RCA was little. Hl/D and (Hl/D)1/2 in the parents were large. Heterosis was large. E was appeared large in the FW, and small in the MW. D was small. The r was of the minus direction, because the dominance genes were less expressed. The regression of the these characters were padded below 0 point, because the characters in FW and MW were appeared overdominant. The orders of the dominance in the FW of the parents were as the order of Jam 107>Jam 108>Jam 119>Jam 113>Jam 114>Jam 120>Jam 117>Jam 118, and in the MW of them in the order of Jam 114>Jam 120>Jam 108>Jam 113>Jam 107>Jam 119>Jam 117>Jam 118. Heterosis was large. The r was in the negative direction, because the dominance genes were less expressed. The regression of the characters of FL and ML were appeared overdominant. The dominance in the FL of parents ere in the order of Jam 120>Jam 114>Jam 119>Jam 119>Jam 118>Jam 107>Jam 117>Jam 113, and the ML of them in the order of Jam 114>Jam 108>Jam 120>Jam 117>Jam 118>Jam 107>Jam 119>Jam 113. The analysis of the genetic variance and combining ability of the FR and the MR. In the FR and the Mr, h2N was less than h2B. GCA was large. The SCA and RCA were little. In the FW, Hl/D was large but (Hl/D)1/2 was a little. In MR, Hl/D and (Hl/D)1/2 both were a littel. Heterosis was a little. E in the FR was in the negative direction, because the dominance genes were less expressed but that in the MR was the positive direction because the recessive genes were mainly expressed. The order of the dominance in the FR of the parents were in the order of Jam 117>Jam 114>Jam 108>Jam 120>Jam 118>Jam 119>Jam 107>Jam 113 and that in the MR these were in the order of Jam 114>Jam 117>Jam 108>Jam 118>Jam 107>Jam 119>Jam 120.

Studies on the Comparative Performance of Victory-1, S-36 and Kanva-2 Mulberry Genotypes and their Impact on Silkworm Rearing under Telangana Conditions of Andhra Pradesh.

  • A new high yielding mulberry genotype, Victory-1 (V-1) was recommended to the field for irrigated areas by Central Sericultural Research and Training Institute (CSRTI), Mysore during 1996-1997. As K-2 and S-36 mulberry genotypes are already in the field, a study on the comparative performance of V-1, 5-36 and K-2 varieties, in terms of leaf yield, yield attributes, leaf moisture and moisture retaining capacity and their impact on silkworm rearing was carried out under Telangana conditions of Andhra Pradesh during 1999-2001. Among the three genotypes the leaf yield was found to be significantly (P < 0.01) higher in V-1 (64,130 kg/ha/yr) followed by S-36 (44,064 kg/ha/yr) and K-2 (23,990 kg/ha/yr). The maximum leaf moisture was also observed in V-1 (71.84%) compared to S-36 (66.86%) and K-2 (64%). The moisture retaining ability was more in V-1 (70.17%) followed by S-36 (66.20%) and K-2 (60.08%). The yield attributes were found to be significantly (P < 0.01) enhanced in V-1 over S-36 and K-2. Silkworm rearing results (bioassay) indicated a significant (P < 0.01) reduction in larval duration and improvement in larval weight, effective rate of rearing by number and weight, single cocoon and shell weights and shell ratio in the silkworm lots fed on V-1 variety followed by S-36 and K-2. Hence, in this study, V-1 was found to be superior among the three varieties studied.

Light and Electron Microscopic Study on the Development of Nosema Bombyics Naegeli in the Midgut of Silkworm Bombyx Mori L.

  • Infection effect of $Nosema$ $bombyics$ on the midgut of silkworm $Bombyx$ $mori$ and subsequent appearance of spores and the performance of larvae was studied. Autopsy of larvae showed white pustules on the surface of midgut at 5 days of post infection (pi). At later stage, important organs like midgut, silk gland and gonads reduced in size and all these organs showed white pustules. Light microscope observation of pustules revealed enormous spores. Spore multiplication was at a faster rate in young larvae. Infection of the adult larvae resulted in pebrinized pupa and moths. Larval weight, cocoon weight and cocoon shell ratio reduced as the post infection period increased. Transverse sections of midgut showed $N.$ $bombycis$ infection limited to a few columnar cells at 3-5 days of pi. At 7 days pi, cell volume increased, cells were swollen and elongated. Heavily infected cells looked like sacks filled with parasite and the apical region of certain cells were bulging into the gut lumen. Later at 8-9 days of pi, spores or its developing stages leaked into the lumen either freely or enclosed within the globules of host cytoplasm. Besides columnar cells, development of $N.$ $bombycis$ was observed in the regenerative cells and rarely in goblet cells. Development of $N.$ $bombycis$ was also observed in both longitudinal and circular muscles at the late pi period. The histopathological changes, deformities and spore production time in the host were all influenced by the spore dosage and age of the host.

Effects of Number of Days After Urea Foliar Spray on Mulberry Leaf Components and Silkworms (요소 엽면시비후 경과일수가 뽕잎 화학성분과 누에에 미치는 영향)

  • The effect of short term duration(1, 2 and 3 days) after urea foliar spray on leaf quality was investigated by chemical analysis of leaves and silkworm rearing with leaves treated with urea in autumn. Foliar spray increased water content by 0.4~0.7% and total nitrogen by 0.19~0.51% in leaves compared to the conventional treatment. Foliar spray increased NO3-N concentration in leaf 503~1107 ppm more than the conventional treatment. NO3-N concentration in leaf decreased with days after foliar spray. Whole cocoon weight and cocoon shell weight increased by 0.02~0.14g and 0.5~3.1 cg, respectively in foliar spray than in the conventional treatment. Though foliar spray raised NO3-N concentration in leaf, leaves harvested one day after foliar spray were no harmful to silkworm.

The Developmental Characterization of the Sericin jam(Bombyx mori), a Mutant in the Synthesis and Secretion of Fibroin (세리신잠(Nd-s. $Nd^H$의 발육 특성)

  • This experiment was done to know developmental characteristics of Sericin jam. Incubation periods were 10 day 2 hr, and 10 day 1 hr. 11 day 1 hr for Nd-s jam, N $d^{H}$ jam, and Baegok jam, respectively. Hatching rates were 83.9, 83.3 and 96.0% for Nd-s jam, N $d^{H}$ jam, and Baegok jam. Larval periods were, 20 days 1 hr for Nd-s jam, 20 days 5 hrs for N $d^{H}$ jam, and 22 days 12 hrs for Baegok jam. Death rate of larvae was highest in N $d^{H}$ iam, followed by Nd-s jam and Baegok jam. Pupation rate was highest in Baegok Jam followed by Nd-s jam and that of N $d^{H}$ jam was the lowest among the three. Cocoon weight was 1.39, 1.08, and 2.01 g for Nd-s jam, N $d^{H}$ jam, and Baegok jam, respectively. Shell weight were 13, 3, and 48 cg for Nd-s jam, N $d^{H}$ jam, and Baegok jam. Cocoon shell ratios were 9.0% for Nd-s jam, 2.8% for N $d^{H}$ jam and 23.9% for Baegok jam. Cocoon sizes were 30.6${\times}$15.8 mm for Nd-s jam, 24.7${\times}$14.9 mm for N $d^{H}$ jam and 35.8 ${\times}$ 20.5 mm(1${\times}$w) for Baegok jam.w) for Baegok jam.

Studies on Silkworm Selection by Use of Anesthetic(1) (The Effect of Silkworm Analysis through Anesthesia) (마취제처리에 의한 잠아선발 연구(I))

  • The author is the first man who tried to use an anesthetic on insect specially for silkworm in orde to evaluate the silkworm health and silk yielding ability and the obtained results are as followings. 1. The necessary ether vapor induction for narcosis on silkworms is varied by the glowth of the silkworm which the larger worm is, the longer induction is required. For instance, it was 2∼3 minutes for the worms of third day fifth instar silkworm in case use of ether anesthetic. 2. The longer anesthetic induction for silkworms, the longer recovery needs from anesthesia. In case five minutes ether vapor induction, silkworms recovered narcosis during in 5∼130 minutes which had varied very much by the health variation. 3. The ether induction caused silkworm to vomit digestive juice from a few per cent of the worms, but the chloroform induction showed majority of the worms to vomit the digestive juice out of mouth. So, the ether was found as better anesthetic for silkworms. 4. When ether induction last more than 20∼30 minutes, the recovered silkworms can eat mulberry, but when it gets more than three hours they can not eat mulberry. And when it last more than ten minutes, the silkworm may eat mulberry leaf, but they can not spinn cocoon properly. 5. In case five minutes ether induction for silkworms on third day fifth instar, the stronger variety is, the rallier recovered from narcosis. 6. The recovering duration from narcosis varies regarding with each worm health which shows Poisson′s distribution even in a same variety silkworm. 7. The female worms recover from narcosis earlier than male worms which means the female worm is stronger than male one. 8. The later recovered silkworm from narcosis spinned more rich cocoon silk and ended with smaller pupae weight. Such a tendency showed until at some recovery duration, then the silk yield droped down on the worms recovered in more longer duration. The author (Choe) had named such a relation curve as "Silk Yield Curve against Silkworm Health." 9. The silk yield or cocoon layer ratio had varied from 13 to 27% even in a same worm varity cocoon which showed serious variation and call attention carefulness for the duplication work of a variety silkworm eggs. 10. Not always the rich silk yielding worm is the best worm during the silkworm selection and it should be considered with the silkworm health evaluation. 11. At present situation, only specific breeding expert is allowed to join in the selection service because of need many years experience by use of visual observation, but the ether anesthesia method may help such an evaluation with more accuracy and easy way even for the people in fresh on the field. 12. The effect of the narcosis on the silkworm for the next generation or hybrid worm will be reported in next publication.

The Combining Ability Analysis and Heterosis for some Quanitatives Traits in the Silkworm (Bombyx mori L.)

  • Recently two breeding programs were carried out for isolation of new parental inbred lines in Iran. This study was undertaken in order to estimate the combining ability effects and heterosis of these lines. For this purpose an $8\times8$ diallel cross analysis including eight inbred lines of silkworm with four lines from each program were studied for their five quantitative traits. The results indicated that reciprocal effects appeared insignificant for most of traits. Japanese lines of 111 and 113 were best combiners for shell weight, cocoon weight and shell percentage traits. With respect to pupation rate, Chinese line 110 was the best and according to general combining ability (GCA) and specific combining ability (SCA) variances, it is superior in specific combinations. Hybrid $109\times110$ showed better characters for both productive and viability traits. Hybrids with high heterosis had high SCA too.

Effect of Different Periods of Cold Storing of Bivoltine Eggs on Subsequent Generation Rearing Performance

  • Different methods of cold storing of bivoltine eggs are in practice to postpone hatching. Bivoltine eggs undergo hibernation if they are not acid treated within 20 - 24 hrs of oviposition, which depends on race, ambient temperature and humidity. The schedules adopted for cold storing include hibernation schedule (Hib), ordinary chilling (OC), short term chilling (STC) and acid treated layings (AT). Peanut cocooning race ${NB_4}{D_2}$ has been subjected for the present assessment. Cocoons harvested from the crop pertaining to all the four methods of cold storing have been used for producing different combinations and acid treated followed by rearing. The performance in respect of chawki loss, maximum larval weight 4({5^th} age)$, yield/ 10,000 larvae (no), cocoon and shell weight showed maximum values for hibernation ${\times}$ hibernation combination followed hibernation with OC and hibernation with AT. Lowest performance was recorded when STC batch source females were used.

  • In this study, hair care effects of the hair cosmetics including low molecular weight silk peptide, hydrolysate which is produced from cocoon were investigated. After producing the hair cosmetics including silk peptide which has 300-500 molecular weight, we measured its hair care effects through the various tests; change of the hair weight, hair thickness, absorbance rate and hair moisture value and micro structure analysis. As a result, S.P.T. (Silk peptide Treatment) was effectively penetrated into the hair which has been damaged by chemical treatments, increased the hair weight, thickness and hair moisture value and also recovered the cuticle of the hair. Thus, the treatment of hair with silk peptide hydrolysate would be effective to recover the damaged hair into the normal conditions.

Breeding of New Silkworm Variety Golden silk, a Yellow Cocoon Color for Spring Rearing Season (황색실크 생산용 특수누에 품종 '골든실크' 육성)

  • A new silkworm variety Golden silk for spring rearing season is $F_1$ hybrid between Jam311, a Japanese race bred from introduction breeding and Jam312, a Chinese race from introduction breeding. In the local adaptability test performed at 8 local areas in spring of 2006, "Golden silk" was showed 7 percent higher in hatchability than authorized silk-worm races laval. "Golden silk" was receded higher 1.7 percent, 2.2 percent in pupation rate, best cocoon rate respectively than Chunsujam. The larval period shortened for 1.12 day even though low record to the several items including productivity than Chunsujam. "Golden silk" was showed 1,031 m, 2.97 denier in length of cocoon filament, weight of cocoon filament respectively. The concentration of Deoxynojirimycin(DNJ) was measured 4.34 mg in "Golden silk", high then to "Chunsujam"(3.81 mg). In the Paecilomyces tenuipes production ability test, the pupal weight of "Golden silk" was 1.20g.
