Evaluation of the Degradation of Carbohydrate-based Material During Anaerobic Digestion for High-efficiency Biogas Production

  • Purpose: In this study, the potential for biogas production, degradation rates, and lag-phase of diauxic growth of carbohydrate-based material, which is one of the proximate compositions, were investigated. Methods: This study was conducted using starch as a carbohydrate-based material. In experimental condition 1, the biogas potential of carbohydrate-based material was measured. In experimental condition 2, the effect of feed to microorganism ratio (F/M ratio) on lag-phase of diauxic growth from carbohydrate-based material was tested. Biochemical methane potential tests were performed at five different feed to microorganism ratios (0.2, 0.4, 0.6, 0.8, and 1.0) under mesophilic conditions. The biogas production patterns, lag-phase, total volatile fatty acids to total alkalinity ratio (TVFA/TA ratio), and time required for 90 percent biogas production were used to evaluate biogas production based on the biochemical methane potential tests. Results: In experimental condition 1, unlike previous studies, biogas was produced in the TVFA/TA ratio ranging from 1.131 to 2.029 (approximately 13-19 days). The methane content in the biogas produced from the digesters was 7% on day 9 and increased rapidly until approximately day 27 (approximately 72%). In experimental condition 2, biogas yield was improved when the feed to microorganism ratio exceeded 0.6, with an initial lag-phase. Conclusions: Even if the TVFA/TA ratio was greater than 1.0, the biogas production was processed continuously, and the $CO_2$ content of the biogas production was as high as 60%. The biogas yield was improved when the F/M ratio was increased more than 0.6, but the lag-phase of carbohydrate-based material digestion became longer starting with high organic loading rate. To clarify the problem of the initial lag-phase, our future study will examine the microbial mechanisms during anaerobic digestion.

  • Fat replacers are divided into three categories. These include carbohydrate-based, protein-based and fat-based replacers. Carbohydrate-based replacers occupy half of the fat replacers market. The main ingredients of carbohydrate-based are gums, starch, modified starch, cellulose and fiber. The functional properties of fat replacers are to retain moisture, to retard staling, to provide mouthfeel and texture, to emulsify, to stabilize emulsion, and to reduce fat. Using these functionalities, fat replacers are used in various foods such as baked goods, salad dressing, sauces, meat products, dairy products, frying foods, bakery, and confectionery. Success factors of fat replacers in the market are sensory equivalent, texture and safety as food ingredients.

  • During last few decades dietary guidelines for the weight management mainly have focused on a low-fat, high carbohydrate diet. Carbohydrate was supposed to be low-dense, highly satiating as well as it affects little on the lipogenesis. Although low-fat diet has significant weight-reducing effect, the loss was modest and usually regained after cessation of the diet. Furthermore, low-fat, modest-carbohydrate diet did not impact on the ever increasing rates of overweight and obesity. Alternative approaches include low-carbohydrate diet, high-carbohydrate diet and low-glycemic index diet. Although none of above mentioned diet have sufficient evidence for standard weight management diet, short-term efficacy and safety are being approved continuously. Low-carbohydrate diet contains less than 45% of carbohydrate in daily energy consumption, it is claimed to have more satiating effect and to improve metabolism. However, low compliance due to the limitation of food choice should be considered on prescribing the diet. High-carbohydrate which contains 90% of carbohydrate in total daily energy consumption, is effective in providing satiety and lowering total calorie intake and cholesterol. On the other hand, nutritional unbalance should be took into account. Low-glycemic index diet is based on the theory that contemporary diet contains significantly less fiber and unrefined carbohydrate, therefore insulin secretion is disturbed. Because low glycemic index food slowly increase blood glucose and insulin level, it induces much satiating effect and may decrease calorie ultimate intake. However, poor standardization of glycemic index is one of the main obstacle for the diet to be applied in the clinic. Meanwhile, high fructose food and beverage should be discouraged because it has little satiating effect and may cause insulin resistance. High fiber food is another recommendation for healthy, lean diet.

Investigation of dry dog food market trends and valuation of carbohydrate sources for dog diets

  • Plant-based, grain-free, and gluten-free diets have been the focus of recent research, as part of the premiumization trend in dog food. However, the lack of comprehensive information regarding carbohydrates in dog food makes it difficult to produce nutritionally balanced meals. Therefore, we aimed to evaluate the nutritional value of carbohydrate ingredients commonly used in commercial dog foods. First, using data from Euromonitor, we investigated the trends in the dry dog food industry and analyzed the annual volume of dry dog food sales, as well as the dog population from 2010 to 2021 in Korea. The growth of the market was greatest in 2021, when the total value of sales of premium dry dog food was 254.3 billion KRW (Korean won) more than in 2010. In addition, sales of dry dog food by age category were marked by a significant increase (404.8%) in the market for dry food for senior dogs. Second, we examined the frequency of carbohydrate ingredients used in dry dog food and conducted a proximate analysis of the top 10 most frequently used ingredients. Subsequently, we analyzed the crude protein, crude fat, and nitrogen-free extract in the carbohydrate ingredients based on dry matter (DM) and calculated the ratio of metabolic energy content supplied from each nutrient in the carbohydrate ingredients. The ratio of metabolic energy content in the carbohydrate ingredients indicated that mung beans had the highest crude protein content (31.3%), whereas oats exhibited the highest crude fat content (23.3%). Sweet potatoes had the highest nitrogen-free extract content, measuring at 93.6%. The results of this study are expected to provide a valuable foundation for the growth and development of the domestic dog food industry, while also contributing to a deeper understanding of the specific role of each ingredient as a carbohydrate source.

A Review on Use of Carbohydrate-based Fillers and Pigments in Packaging Paper

  • As one of traditional packaging materials, paper and paperboard are being more popular and beneficial thanks to their environmental sustainability and have been widely used in packaging applications, from light weight infusible tissue for tea/coffee bags to heavy duty boards for the distribution. Papermakers have to design the products having a desired customized function with their paper machine. Globally, the use of filler and pigment in papermaking is now a very common practice to meet the needs of customers. Many benefits can be achieved as a result of filler addition, which mainly includes cost and energy savings. The replacement of traditional mineral fillers and pigments with biodegradable and renewable carbohydrate polymers is a very interesting and promising research topic due to the concern of environmental impact. In this review paper, the use of traditional and novel carbohydrate fillers and pigments in cellulosic paper is highlighted. It is noteworthy that there are still some challenges and technical barriers associated with the use of these organic materials in point of structural stabilities and manufacturing costs, although most of them are available in market as the commercialized products. With the emerging nanotechnologies, it is believed that the use of carbohydrate-based filler and pigment for papermaking will increase and bring technical advantages to industry.

Dietary patterns based on carbohydrate nutrition are associated with the risk for diabetes and dyslipidemia

  • Several studies have been conducted on dietary patterns based on carbohydrate nutrition in Asian populations. We examined the cross-sectional associations in dietary patterns based on carbohydrate nutrition, including the glycemic index (GI) with dyslipidemia and diabetes among the Korean adult population. We analyzed 9,725 subjects (3,795 men and 5,930 women, ${\geq}$ 20 years) from the Fourth Korea National Health and Nutrition Examination Survey. Dietary information was collected using single 24-hour recall. Reduced rank regression was used to derive dietary patterns from 22 food groups as predictor variables and four dietary factors related to the quantity and quality of carbohydrates as response variables. Two dietary patterns were identified: 1) the balanced pattern was characterized by high intake of various kinds of foods including white rice, and 2) the rice-oriented pattern was characterized by a high intake of white rice but low intake of vegetables, fruits, meat, and dairy products. Both patterns had considerable amounts of total carbohydrate, but GI values differed. The rice-oriented pattern was positively associated with hypertriglyceridemia in men and low high density lipoprotein-cholesterol in both men and women. The balanced pattern had no overall significant association with the prevalence of dyslipidemia or diabetes, however, men with energy intake above the median showed a reduced prevalence of diabetes across quintiles of balanced pattern scores. The results show that dietary patterns based on carbohydrate nutrition are associated with prevalence of dyslipidemia and diabetes in the Korean adult population.

Effects of a Very Low Carbohydrate (Pork Rind-Based) Diet on Weight Gain, Serum Levels of Cholesterol, Triacylglycerol, Glucose, Ketone Bodies and Insulin and Body Composition in Adult Rats

  • A study was carried out to determine the effect of a very low-carbohydrate diet on weight gain, body composition, and serum levels of cholesterol, triacylglycerol, glucose, ketone bodies and insulin. Twenty rats (mean initial weight, 212 g) were divided into two groups and each group was assigned a conventional high-carbohydrate diet (control) or a very low-carbohydrate diet containing 59.8% ground pork rind snack and fed the diet for four weeks. Average daily body weight gain was not different between the two groups during the first two weeks, but was significantly lower in rats fed the very low-carbohydrate diet than in those in the control group during third (p<0.05) and fourth weeks (p<0.01). Feedintake as well as energy intake was lower in rats fed the very low-carbohydrate diet than in those in the control group. The very low-carbohydrate diet reduced (p<0.01) serum triacylglycerol (34$\pm$83 vs 82$\pm$8 mg/l00 mL) and insulin (3.90$\pm$0.53 vs 7.60$\pm$0.61 $\mu$IU/mL) levels, while increasing (p<0.01) ketone body level (368$\pm$25 vs 236$\pm$24 $\mu$mol/L), compared with the control. Serum glucose and total cholesterol levels were not different (p>0.05) between the two dietary treatments. Proximate analysis of carcasses showed that the very low-carbohydrate diet decreased (p<0.01) body fat (26.1$\pm$1.04 vs 30.5$\pm$0.86%), while increasing (p<0.01) body protein (63.1$\pm$0.94 vs 59.4$\pm$0.70%) contents. Results indicate that short-term feeding of a very low-carbohydrate diet is beneficial for alleviating risk factors known to involve cardiovascular diseases or artherosclerosis. However, more studies with model animals as well as humans are recommended to examine the long-term health benefits of low-carbohydrate diets.

Relationship between the Methane Production and the CNCPS Carbohydrate Fractions of Rations with Various Concentrate/roughage Ratios Evaluated Using In vitro Incubation Technique

  • The objective of the trial was to study the relationship between the methane ($CH_4$) production and the Cornell Net Carbohydrate and Protein System (CNCPS) carbohydrate fractions of feeds for cattle and the suitability of CNCPS carbohydrate fractions as the dietary variables in modeling the $CH_4$ production in rumen fermentation. Forty-five rations for cattle with the concentrate/roughage ratios of 10:90, 20:80, 30:70, 40:60, and 50:50 were formulated as feed samples. The Menke and Steingass's gas test was used for the measurement of $CH_4$ production. The feed samples were incubated for 48 h and the $CH_4$ production was analyzed using gas chromatography. Statistical analysis indicated that the $CH_4$ production (mL) was closely correlated with the CNCPS carbohydrate fractions (g), i.e. CA (sugars); $CB_1$ (starch and pectin); $CB_2$ (available cell wall) in a multiple linear pattern: $CH_4=(89.16{\pm}14.93)$ $CA+(124.10{\pm}13.90)$ $CB_1+(30.58{\pm}11.72)$ $CB_2+(3.28{\pm}7.19)$, $R^2=0.81$, p<0.0001, n = 45. Validation of the model using 10 rations indicated that the $CH_4$ production of the rations for cattle could accurately be predicted based on the CNCPS carbohydrate fractions. The trial indicated that the CNCPS carbohydrate fractions CA, $CB_1$ and $CB_2$ were suitable dietary variables for predicting the $CH_4$ production in rumen fermentation in vitro.

Combinatorial Methylerythritol Phosphate Pathway Engineering and Process Optimization for Increased Menaquinone-7 Synthesis in Bacillus subtilis

  • Vitamin K2 (menaquinone) is an essential vitamin existing in the daily diet, and menaquinone-7 (MK-7) is an important form of it. In a recent work, we engineered the synthesis modules of MK-7 in Bacillus subtilis, and the strain BS20 could produce 360 mg/l MK-7 in shake flasks, while the methylerythritol phosphate (MEP) pathway, which provides the precursor isopentenyl diphosphate for MK-7 synthesis, was not engineered. In this study, we overexpressed five genes of the MEP pathway in BS20 and finally obtained a strain (BS20DFHG) with MK-7 titer of 415 mg/l in shake flasks. Next, we optimized the fermentation process parameters (initial pH, temperature and aeration) in an 8-unit parallel bioreactor system consisting of 300-ml glass vessels. Based on this, we scaled up the MK-7 production by the strain BS20DFHG in a 50-l bioreactor, and the highest MK-7 titer reached 242 mg/l. Here, we show that the engineered strain BS20DFHG may be used for the industrial production of MK-7 in the future.

한국 성인 비만 집단의 탄수화물 급원 식품 섭취 분포 및 노출 위험도: 대규모 한국인유전체 역학 자료 활용 (Distribution and Exposure Prevalence of Carbohydrate-based Food Intake among Obese Korean Adults Based on the Health Examinees (HEXA) Study)

  • Objectives: The purpose of this study was to estimate the distribution and exposure prevalence of total carbohydrate intake and the carbohydrate-based foods such as rice, noodles, sweet potatoes, sweet drinks, milk and fruits and to characterize intake patterns among obese Korean adults. Methods: Subjects included 137,363 adults aged 40 years or older who participated in a Health Examinees (HEXA) Study. Multiple regression analysis of data from Semi-Quantitative Food Frequency Questionnaire (SQFFQ) identified food sources of carbohydrates. Weight, height and waist circumstance (WC) were measured, and the body mass index (BMI) was calculated. Obesity was defined as $BMI{\geq}25kg/m^2$ and abdominal obesity as $WC{\geq}90cm$ and ${\geq}85cm$ for males and females, respectively. Results: Obese adults appeared to have a higher total carbohydrate intake in the univariate analysis but had eaten less after adjustment for general and lifestyle factors, compared to normal weight adults (OR 0.78, 95% CI 0.73-0.82 for general obesity; OR 0.79, 95% CI 0.74-0.85, for abdominal obesity; P trend < 0.0001, respectively). Based on advance analysis for the food sources of carbohydrates, obese subjects had lower intake of rice (OR 0.86, 95% CI 0.68 -1.09 for general obesity; OR 0.87, 95% CI 0.67-1.13, for abdominal obesity; P trend < 0.0001, respectively) and higher intake of noodles (OR 1.21, 95% CI 1.16-1.27 for general obesity; OR 1.25, 95% CI 1.19-1.32, for abdominal obesity; P trend < 0.0001 respectively). With regard to other food sources of carbohydrates such as milk and fruits, intake was lower among obese compared to normal weight subjects. Conclusions: In the Korean middle-aged and older obesity groups, the intake of carbohydrates and the related foods was lower than in normal weight subjects, except for noodles. We conclude that a higher intake of noodles may enhance weight-gain. Therefore, this study suggested that the guidelines should consider the types of carbohydrate sources and the amount consumed from foods in order to provide proper guidance with regard to control and prevent obesity among Korean adults.
