A New Soybean Cultivar, "Wonkwang" with Sprout, High Yielding, Disease and Lodging Resistance (내병.내도복 다수성 나물콩 신품종 "원광")

  • "Wonkwang" is a new sprout-soybean cultivar developed from the cross between Danyeobkong and MS91001 at the Honam Agricultural Research Institute (HARI) and National Institute of Crop Science (NICS), RDA, in 2007. The preliminary, advanced and regional yield trials to evaluate the performance of Iksan 56 were carried out from 2003 to 2007. This cultivar has a determinate growth habit, purple flower, grayish brown pubescence, yellow seed coat, Grayish brown hilum, lanceolate leaflet shape and small seed size (10.9 g/100 seeds). The maturity date of "Wonkwang" is three days later than the check variety, "Pungsan". It has a good seed quality for soybean-sprout and resistance to lodging. The soybean-sprouts grown from "Wonkwang" have high isoflavone ($3,481{\mu}g/g$)contents. This cultivar has resistance to soybean mosaic virus (SMV) and necrotic symptom (SMV-N) and other most troublesome soybean diseases which are bacterial pustule and black root rot. The grain yield of "Wonkwang"in the regional yield trials (RYT) for 3 consecutive years was averaged 3.05 ton per hectare, which was 8% higher than that of the check cultivar "Pungsan".

Effect of Various Culture Conditions on the Activity of Extracellular Enzymes from Fomitopsis palustris, Brown Rot Fungi (침엽수 혼합목분에서 배양조건을 달리한 Fomitopsis palustris 의 균체외효소 활성 변화)

  • Extracellular enzyme activities of Fomitopsis palustris were determined by the particle sizes, culture periods and concentrations of wood particle substrate which was mixture of 4 domestic coniferous woods, such as Pinus densiflora, Larix leptolepsis, Pinus koraiensis, and Pinus rigida. The results showed that the culture conditions had an effect on the secretion of most of the extracellular enzymes from Fomitopsis palustris in the mixed wood particle substrate. :The optimal culture conditions for enzyme activities were 80~100 mesh in wood particle size, 7.5% in concentrations of wood substrate, and 4~8 weeks in culture period.

Physiological Responses of Soybean Cultivars to Fusarium solani f. sp. glycines Causing Sudden Death Syndrome

  • Six soybean cultivars having different SDS susceptibility were planted with sorghum seedinoculum infested with F. solani isolate 171 in the greenhouse. First leaf symptoms appeared on unifoliar leaves at 9 days after inoculation and all cultivars showed the typical leaf symptoms at 13 days after inoculation, when trifoliar leaves emerged. Leaf symptoms development in susceptible cultivars was faster than in resistant cultivars. Leaf symptom severities during the period of 25 to 29 days after inoculation showed a significant difference between cultivars which had SDS resistance and sus ceptibility. In this period, area under the diseaseprogress curve (AUDPC) of Hartz 6686 was the highest and that of PI 520733 was the lowest. SDS caused serious damage to the growth of soybean in all cultivars. Average reductions of growth rate of root fresh weight and dry weight were greater than those of plant tops. Duyu-kong showed less severe leaf symptoms than that of SDS suscetible cultivars; however, average growth rate of plants top and roots of this cultivar was less but not significantly different than those of SDS susceptible cultivars. In all cultivars, as severity of leaf symptoms increased, plant top weight decreased. Root rot symptoms were observed in all cultivars before leaf symptoms appeared. Average proportions of tap root reddish-brown discoloration of all cultivars was up to 75 % at 15 days after inoculati on; however there was no significant differenc between cultivars at each rating date. Appearances of leaf symptoms on leaves varied in each cultivar. SDS resistant cultivars had a significantly higher level of crinkling than susceptible cultivars and SDS susceptible cultivars had a significantly higher level of necrosis than resistant cultivars. Further study will be needed to identify the relationships between the physiological growth rate and SDS severities in soybeans.

Sclerotium blight of Phalaenopsis Orchids Caused by Sclerotium rolfsii in Korea (Sclerotium rolfsii에 의한 팔레놉시스 흰비단병)

  • The Sclerotium blight was found on Phalaenopsis spp. at Dong-du-cheon city, and Hwa-seong city, Gyenggido, Korea in September 2009. The symptom included yellowing on lower leaves and wilt of a whole plant. Severely infected plants were blighted and died eventually. White mycelial mats appeared on the surface of basal stem and bulbs and the sclerotia were formed on stems, roots, and sphagnum moss. The sclerotia were spherical, 1-3 mm and white to brown. The optimum temperature for the growth and sclerotia formation was $25-30^{\circ}C$ on PDA. On the pathogenicity test, the first symptom appeared 5 days after inoculation and developed to severe stem rot and blight. On the basis of mycological characteristics and pathogenicity, the causal fungus was identified as Sclerotium rolfsii. This is the first report on the sclerotium blight on Phalaenopsis spp. caused by Sclerotium rolfsii in Korea.

  • The Sclerotium blight was found on Neofinetia falcata at Yong-in city, Gyenggi-do, Korea. The symptom occurred low leaves yellowish and wilt of a whole plant.   • Cellulosic substance which plays an important role in carbon cycle is most abundant in the nature world. Some higher fungi are able to digest cellulose directly to satisfy their carbohydrate requirement. Then, in order to investigate the enzymatic components of Lenzites betulina (Fr.) being wood rot, that fungus was collected in Kwangneung area. The carpophore of the fungus was smashed with cool distilled water, extracted and salted out by ammonium sulfate. And then the precipitate was purified by dialysing with visking tube and dissolved with pH 7.8 ammonia water, and the extract was filtrated. The fraction of filtrate was obtained as light brown powder after lyophilization, and determined cellulolytic activity. Cellulolytic potency of Lenzites betulina (Fr.) was 1. 65 unit/ml. The cellulase of Lenzites betulina (Fr.) was stable at below $45^{\circ}C$ and range of pH $4.5{\sim}6.0$ and is completely inactivated at $60^{\circ}C$ for 15 minutes. The optimum condition for the enzymatic reaction was $40^{\circ}C$ and pH 4.0. The enzyme activity was not influenced by the presence of $Ca^{2+}$ and $Fe^{2+}$.

Effect of Carbon Source on the Hydrolytic Ability of the Enzyme from Fomitopsis pinicola for Lignocellulosic Biomass

  • In this study, effect of carbon source on the hydrolytic ability of the enzyme from Fomitopsis pinicola, a brown rot fungi, for lignocellulosic biomass were examined on two lignocellulosic biomasses (rice straw and wood) without any pretreatment. Cellulase activities of crude enzyme from F. pinicola, which was cultured on softwood mixture as a carbon source, were 19.10 U/$m{\ell}$ for endo-${\beta}$-1,4-gulcanase (EG), 36.1 U/$m{\ell}$ for ${\beta}$-glucosidase (BGL), 7.27 U/$m{\ell}$ for cellobiohydrolase (CBH), and 7.12 U/$m{\ell}$ for ${\beta}$-1,4 xylosidase (BXL). Softwood mixture as a carbon source in F. pinicola comparatively enhanced cellulase activities than rice straw. The optimal pH and temperature of the cellulase was identified to pH 5 and $50^{\circ}C$for the hydrolysis of rice straw. Under these condition rice straw was hydrolyzed to glucose by the cellulase up to $32.0{\pm}3.1%$ based on the glucan amount of the rice straw for 72 h, while the hydrolytic capability of commercial enzyme (Celluclast 1.5${\ell}$) from rice straw to glucose was estimated to $53.7{\pm}4.7%$ at the same experimental condition. In case of addition of Tween 20 (0.1% w/w, substrate) to the cellulase the hydrolysis of rice straw to glucose was enhanced to $38.1{\pm}2.0%$.

Purification and Characterization of Xylanase from Fomitopsis palustris in Rice Straw Culture (볏짚분해과정 중에 생산하는 Fomitopsis palustris 균체 외 Xylanase의 분리정제 및 효소특성)

  • An extracellular xylanase from the brown-rot fungus Fomitopsis palustris grown on rice straw culture was purified to a single protein band. On SDS-PAGE, the molecular mass of purified enzyme was estimated to be about 43 kDa. The amino acid sequence of the proteolytic fragments showed high homology with fungal glycoside hydrolase family 10 xylanases. The $K_m$, $K_{cat}$ and $V_{max}$ for birch xylan were $31mg/m{\ell}$, $2.3{\times}10^4/min$ and 252.3 U/mg, respectively. The optimal activity of the purified xylanase from F palustris was observed at pH 4.0~5.0 and $70^{\circ}C$.

Cladosporium cladosporioides and C. tenuissimum Cause Blossom Blight in Strawberry in Korea

  • Blossom blight in strawberry was first observed in a green house in Nonsan, Damyang, and Geochang areas of Korea, between early January to April of 2012. Disease symptoms started as a grey fungus formed on the stigma, which led to the blossom blight and eventually to black rot and necrosis of the entire flower. We isolated the fungi purely from the infected pistils and maintained them on potato dextrose agar (PDA) slants. To test Koch's postulates, we inoculated the fungi and found that all of the isolates caused disease symptoms in the flower of strawberry cultivars (Seolhyang, Maehyang, and Kumhyang). The isolates on PDA had a velvet-like appearance, and their color ranged between olivaceous-brown and smoky-grey to olive and almost black. The intercalary conidia of the isolates were elliptical to limoniform, with sizes ranging from $5.0{\sim}10.5{\times}2.5{\sim}3.0{\mu}m$ to $4.0{\sim}7.5{\times}2.0{\sim}3.0{\mu}m$, respectively. The secondary ramoconidia of these isolates were 0- or 1-septate, with sizes ranging betweem $10.0{\sim}15.0{\times}2.5{\sim}3.7{\mu}m$ and $8.7{\sim}11.2{\times}2.5{\sim}3.2{\mu}m$, respectively. A combined sequence analysis of the internal transcribed spacer regions, partial actin (ACT), and translation elongation factor 1-alpha (TEF) genes revealed that the strawberry isolates belonged to two groups of authentic strains, Cladosporium cladosporioides and C. tenuissimum. Based on these results, we identified the pathogens causing blossom blight in strawberries in Korea as being C. cladosporioides and C. tenuissimum.

  • Heavy metals were removed from CCA- and CCFZ- treated wood using a brown-rot fungi Tyromyces palustris. The amount of effective elements extracted from treated woods was compared for different treatment methods. The relationship between the amount of heavy metals removed and concentrations of oxalic acid for treated wood was examined. Also, the relationship between mycelia weight and removal rate was examined. The removed quantity of heavy metal from treated wood according to fermentation methods was examined. The extraction amount of chromium and arsenic components increased with increasing oxalic acid concentration, but the extraction amount of copper did not improved much. A 287 mg of mycelia weight can remove chromium and arsenic over 60% in 3 g CCA chips and copper was also removed over 50%. The chromium, copper and arsenic were removed over 60% by shaking fermentation, the removal rate of copper by static cultivation was higher than that of shaking fermentation. The removal rate of chromium, copper and arsenic were 72%, 61% and 59% with air-lift bioreactor, respectively.
