Leaf Blight of Perilla Caused by Alternaria alternata (Alternaria alternata에 의한 들깨 잎마름병)

  • A new leaf blight was found on the perilla leaves at the major perilla-cultivating areas such as Kangdong in Busan and Miryang in Kyungnam province. Symptoms of the disease initially appeared on the edge of perilla leaves showing black necrosis and drying, and the infected leaves were finally fell down. The SD1 isolate showing strong pathogenicity and forming abundant conidial spores on the diseased lesions was isolated. Among the tested media, mycelial growth was abundant on PDA (Potato Dextrose Agar) medium at $25^{\circ}C$ under dark condition, but conidial formation was greater on V8A (V-8 juice A8ar) medium than that on PDA medium. Optimal temperatures for mycelial growth and conidial formation on PDA medium were respectively $25^{\circ}C$ and 3$0^{\circ}C$. The rate of conidial germination and the elongation of germ tube were more effective in 10% tomato juice than those in PDB (Potato Dextrose Broth) and sterile water. In 10% tomato juice, the rate of conidial germination and the length of germ tube were 100% after incubation for 24k and 535.2${\mu}{\textrm}{m}$ after incubation for 36hr, respectively. According to the result of pathogenicity, it revealed that conidial suspention with 10% tomato juice was the most effective for pathogenicity test showing as 100% of disease incidence, and the symptoms caused by artificial inoculum were same as those of naturally infected perilla. In this study, the SD1 isolate according to the results of morphological characteristics, the incubation characteristics and pathogenicity was firstly identified A. alternata, and named as leaf blight of perilla.

Responses of Ricania sp. (Hemiptera: Ricaniidae) to light and Attraction to Capturing Device (갈색날개매미충(Ricania sp.) 성충의 광 선호성과 성충포획장치의 유인효과)

  • In order to effectively control the Ricania sp., we investigated the light response to adults and developed an adult capturing device using light with superior attractiveness. The preference among six light sources such as daylight color, green, etc. for the Ricania sp. adult was favored with daylight color 97 > black 79 = red 79 = blue 79 > green 24 > yellow 13. We have developed an adult catching device using the most preferred daylight colors and behavioral habits of the Ricania sp.. The capture device consisted of two daylight compact lamps (30W, 20W), a yellow plate, and a catcher using water, and caught about 700 individuals a day. The capturing device has a large amount of capturing because adult is activity at high nighttime temperatures, but the capturing amount decreased significantly when the temperature dropped below 23℃. More than 85% of the Ricania sp. adults were trapped for 3 hours from 19:00 to 23:00. Therefore, it is considered that the adult capturing device for the control of Ricania sp. is used from mid July to late August when the night temperature is over 23℃, and lights up from 19:00 to 23:00.

Occurrence of Gray Mold on Yacon Caused by Botrytis cinerea (Botrytis cinerea에 의한 야콘 잿빛곰팡이병의 발생)

  • The gray mold disease occurred on tuberous roots of yacon in storage facilities in Gangneung, Korea, in March 2010. Symptoms typically appeared as in the form of dark brown discoloration on the surface of tuberous roots and water-soaked brown lesions in cross sections of the affected portions. A total of five isolates of Botrytis sp. were obtained from the symptomatic portions. All isolates on potato-dextrose agar (PDA) produced abundant conidia which were pale brown, one-celled, mostly ellipsoid or ovoid in shape and $8.2{\sim}14.8{\times}6.5{\sim}9.9\;{\mu}m$ in size. Large numbers of round to irregular, smooth, black, hard sclerotia were produced on PDA over time. The optimal temperature for mycelial growth and sclerotia formation of the fungal isolates was $20^{\circ}C$. On the basis of morphological and cultural characteristics, all the fungal isolates were identified as Botrytis cinerea. Pathogenicity test on host plants showed that the fungus could infect not only tuberous roots but also leaves and petioles of yacon. This is the first report on gray mold of yacon (Smallanthus sonchifolius) caused by Botrytis cinerea in Korea.

Studies on Alternaria and Corynespora Blights of Sesame (참깨 검은무늬병(病) 및 잎마름병(病)에 관한 연구(硏究))

  • Survey of sesame crop in the cultivators' field in the district of Daejeon, Yuseong, Sintanjin, Nonsan and Keumsan revealed that Alternaria and Corynespora blights of sesame were wide spread and caused severe damage to sesame plants. Symptoms of a new disease of sesame in Korea caused by Alternaria sesami were spreading, dark-brown to black, water-soaked lesions which often could be traced the entire length of the stem. In severe infections, several lesions coalesced together involving a major portion of the blade and the infected leaves dried and usually dropped off. Symptoms of Corynespora blight, caused by Corynespora cassiicola, were irregular shaped, concentrically-zoned, light brown to reddish brown lesions which later coalesced and caused defoliation. Stem lesions were long, reddish brown streaks that often coalesced, blighting the plants. The optimum temperatures for mycelial growth of A. sesami and C. cassiicola were about $27^{\circ}C$ and sporulations of these 2 fungi were stimulated under alternating light and darkness. Mycelial growth and sporulation of A. sesami and C. cassiicola were the greatest on sesame oatmeal agar (SOA) and potato dextrose agar (PDA), respectively.

Preparation of Powdered Dried Sea Mussel and Anchovy for Instant Soup (진주담치 및 마른멸치 분말수우프의 제조)

  • As one trials to utilize sea mussel and anchovy effectively, powdered instant soups were prepared and then their quality stability were examined during storage. Powdered instant soup was made by adding $3\%$ sugar, $20\%$ table salt, $5\%$ monosodium glutamate, $0.2\%$ black pepper and garlic powder to the pulverized dried sea mussel or anchovy. Powdered instant soup products, powderd products, and dried round state sea mussel or anchovy were packed with air in laminated film bag (cellophane/polyester/aluminium foil/polyester: $20{\mu}m/15{\mu}m/7{\mu}m/20{\mu}m,\;13{\times}14cm$). The contents of amino-nitrogen and volatile basic nitrogen of these products were showed little significant variations and also water activity and color value (L, a, b) of these products were little changed during storage. Thiobarbituric acid value increased up to 30 days of storage and then decreased slightly. Comparing the quality of powdered-seasoned products with that of dried round state products, there were no significant differences in stability during storage. Judging from the experimental results, the quality of powdered instant soup of sea mussel and anchovy were stable for 100 days at room temperature($25{\pm}3^{\circ}C$).

Firing Conditions and Material Characteristics of Neolithic Potteries from the Goseong Munamri Sites, Korea (고성 문암리 출토 신석기 토기의 재질특성과 소성조건)

  • This study was carried out material characteristics and firing conditions for some potteries in early and middle Neolithic period at the Munamri sites, where the first farmland remails were found in Korea. It is divided into six kinds of surface patterns for potteries such as the raised, bamboo tube and red painted (patternless) from the early Neolithic period (BC 4000 to 6000), and the patterns of horizontal herringbone, short slanted lined or lattice and incised thick line in the middle Neolithic period (BC 3000 to 4000), respectively. Based on the color measures, redness and yellowness of potteries were relatively high as condition of oxidation firings, were also observed black cores on the cross section of potteries. The firing temperature is divided into two groups having under 800℃ and 800to 900℃, the difference in patterns of the potteries are not confirmed. As a microtextures, the bamboo tube pattern potteries show the sericitization biotite, the quartz have developed a suture line textures, and altered alkali feldspars are occurred. X-ray diffraction analysis shows that the main minerals contained in potteries are chlorite and amphibole besides quartz, alkali feldspar and biotite. Considering the geology around the Munamri area is the biotite amphibole granite and soil layers within 10km radius are used as the raw materials for the potteries. The raw materials are presumed that the sources from the metamorphic rocks along the water systems through the mountains around the sites on the basis of well developed suture line textured quartz in potteries. Results on normalized geochemical compositions, the potteries by surface patterns are very similar to sources, and it is judged that was made by using the surrounding soils despite the long time difference from the early to middle in the Neolithic period.

Isolation of a Fibrinolytic Bacterium from Cheongkukjang and Characterization of Its Bioactivity (청국장으로부터 Fibrin 분해 세균의 분리 및 이를 이용한 발효 청국장의 생리활성)

  • In this study, we have isolated and identified a proteolytic bacterium from conventional Cheongkukjang. We also characterized several bioactivity of Cheongkukjang, which was fermented by an isolated strain. One strain out of about $10^4$ strains obtained from Cheongkukjang showed relatively high proteolytic activity was selected and named as a Bacillus subtilis LSH805 strain. White soy-bean Cheongkukjang possessed less odor and more viscous substance than black soy-bean Cheongkukjang. Cheongkukjang showed fibrinolytic activity, and about 1,500 mg fibrin was degraded after 20 h incubation. Although nitric oxide (NO) assays of soy-bean and Cheongkukjang were almost the same, their activities were significantly higher than that of no treatment. Activity of water fraction of Cheongkukjang was somewhat higher than that of soy-bean. Fibrinolytic and NO assays of Cheongkukjang suggest that Cheongkukjang, which was fermented by an isolated strain may be a useful candidate for natural fibrinolytic and macrophage-stimulating agents.

A Revisit to the Recent Human Error Events in Nuclear Power Plants Focused to the Organizational and Safety Culture

  • Objective: This paper presents additional considerations related to organization and safety culture extracted from recent human error incidents in Korea, such as station blackout(i.e., SBO) in Kori#1. Background: Safety culture has been already highlighted as a major cause of human errors after 1986 Chernobyl accident. After Fukushima accident in Japan, the public acceptance for nuclear energy has taken its toll. Organizational characteristics and culture became elucidated as a major contributor again. Therefore many nuclear countries are re-evaluating their safety culture, and discussing any preparedness and its improvement. On top of that, there was an SBO in 2012 in the Kori#1. Korean public feels frustrated due to the similar human errors causing to a catastrophe like Fukushima accident. Method: This paper reassesses Japan's incidents, and revisits Korea's recent incidents. It focuses on the analysis of the hazards rather than the causes of human errors, the derivation of countermeasures, and their implementation. The preceding incidents and conclusions from Japanese experience are also re-analyzed. The Fukushima accident was an SBO due to the natural disaster such as earthquakes and a successive tsunami. Unlike the Fukushima accident, the Kori#1 incident itself was simple and restored without any loss and radioactive release. However, the fact that the incident was deliberately concealed led to massive distrust. Moreover, the continued violation of rules and organized concealment of the accident are serious signs of a new distorted type of human errors, blatantly revealing the cultural and fundamental weakness of the current organization. Result: We should learn from Japanese experiences who had taken pride in its safety technology and fairly high confidence in safety culture. Japan's first criticality accident in JCO facility splashed cold water on that confidence. It has turned out to be a typical case revealing the problems in the organization and safety culture. Since Japan has failed to gain lessons and countermeasure, the issue persists to the Fukushima incident. Conclusion: Safety culture is not a specific independent element, which makes it difficult to either evaluate it properly or establish countermeasures from the lessons. It may continue to expose similar human errors such as concealment of incident and manipulation of bad data. Application: Not only will this work establish the course of research for organization and safety culture, but this work will also contribute to the revitalization of Korea's nuclear industry from the disappointment after the export contract to UAE.

Evaluation of Effect of Low Opening Operation on Increasing Wear of Bearing Bushings of Guide Vanes used in Hydropower Plants (수력발전소 가이드 베인 저개도율 운전에 따른 가이드 베인 베어링 부슁의 마모 가속효과 평가)

  • A guide vane plays a key role in controlling the flow rate of water supplied to the turbine of a hydropower plant. It has been reported that guide vane bearing bushings are subjected to considerable wear, which requires them to be maintained. An ancillary service such as frequency control and black start causes cyclic low opening operation of the guide vanes. It is empirically well known that such operation increases the wear rate of the guide vane bearing bushing. In this study, the effect of low opening operation on the increasing wear of the guide vane bearing bushing is quantitatively assessed via finite element flow analysis, finite element stress analysis, and relative wear evaluation. As a result of the assessment, it is identified that the pressure applied on the guide vane surface increases and the contact length between the outer surface of the guide vane stem and the inner surface of the bearing bushing decreases with a decrease in the opening of the guide vane. In addition, low opening of the guide vanes results in an increase in the relative wear owing to the generation of high contact pressure on the bearing bushing surfaces.

Quality and Physicochemical Characteristics of Rice Cooked along with Various Mixed Grains and by Following Different Cooking Methods (취반방법에 따른 혼합잡곡밥의 품질 및 이화학 특성)

  • This study evaluated the quality characteristics, the presence of polyphenolic compounds, and radical scavenging activity of rice cooked along with various mixed grains (barley, black soybean, adzuki beans, foxtail millet, proso millet, sorghum, glutinous rice) by following two different cooking methods (normal and pressure cooker). The amylogram and water characteristics of mixed grains showed significant differences based on the presence of different types of mixed grains. The chromaticity, palatability characteristics, presence of phenol compounds, and radical scavenging activity of rice cooked along with different mixed grains showed significant differences according to the nature of mixed grains. Total polyphenol contents of before cooking, cooked-rice added to mixed grains cooked in the normal cooker and a pressure cooker were 4.46~5.16, 0.58~0.93 and 0.65~0.96 mg GAE/g, and total flavonoid contents were 250.74~548.89, 129.26~207.04 and $127.41{\sim}218.15{\mu}g\;CE/g$, respectively. The DPPH radical scavenging activity of before cooking, cooked-rice added to mixed grains cooked in the normal cooker and a pressure cooker was 79.25~181.61, 22.07~53.64 and 7.51~39.97 mg TE/100 g, and ABTS radical scavenging activity was 203.25~328.24, 47.28~84.94 and 58.27~99.51 mg TE/100 g, respectively. Accordingly, it is necessary to different combinations of mixed grains according to the cooking method at home and grain industry.
