Correlation among Conformation Parameters in 'Fuji' Apple Fruit ('후지' 사과 과실의 형태 결정 요인 간 상관)

  • Factors affecting fruit conformation of 'Fuji' apples (Malus domestica Borkh.) were investigated through correlation study of fruit weight, seed numbers, and seed weight in relation to fruit conformation indices. Apples were harvested from two orchards in 'Andong' and 'Youngyang' areas at commercial maturity and grouped by fruit weight. Average number of seeds and seed weight were 8.9 and 0.64 g in 'Andong' orchard, while 6.3 and 0.47 g in 'Youngyang' orchard, respectively. When pooled data from the both orchards were analyzed, correlation of fruit weight with seed weight was significantly positive, whereas that with seed number was not significant. Fruit shape index determined by length/diameter ratio was positively correlated with fruit weight, while relationship of the shape index with seed number or with seed weight was not significant. Asymmetry index tended to be low in apples from the 'Andong' orchard in which higher seed numbers and seed weight were observed. The asymmetry index showed highly negative correlations with seed numbers and with seed weight.

Simultaneous Biocatalytic Synthesis of Panose During Lactate Fermentation in Kimchi

  • As a functional additive for intestinal microflora, panose ($6^2-{\alpha}$-D-glucopyranosylmaltose) was synthesized during kimchi fermentation using the glucose transferring reaction of glucansucrase from Leuconostoc mesenteroides. For the glucose transferring reaction, sucrose and maltose were added ($2\%$ each, w/v) to dongchimi-kimchi as the glucosyl donor and acceptor molecule, respectively. After five days of incubation at $10^{\circ}C$, referring to the initial phase for the production of lactic acid in kimchi, over $60\%$ (w/v) of the total sugars were converted into panose and other branched oligosaccharides. Thereafter, the kimchi was stored at $4^{\circ}C$ and the amount of panose remained at a constant level for three weeks, thereby indicating the stability of panose to microbial degradation during the period of kimchi consumption. The use of maltose as the acceptor molecule in the kimchi also facilitated a lower viscosity in the kimchi-juice by preventing the synthesis of a dextran-like polymer which caused an unfavorable taste. Accordingly, the application of this new method of simultaneous biocatalytic synthesis of oligosaccharides during lactate fermentation should facilitate the extensive development of new function-added lactate foods.

Operational Management System and Characteristics Analysis on the Rural Experience Programs: the Case of Comprehensive Rural Village Development Projects (농촌체험프로그램 운영 유형 및 실태분석 : 농촌마을종합개발사업을 중심으로)

  • The comprehensive rural village development projects (CRVDP) have been carried out as the core one of the rural development schemes in Korea since 2004. CRVDP included the various rural experience programs to increase rural income and in order to promote rural community development in the project area. This study analyzed the operating management conditions, types and characteristics of the rural experience programs targeting the 168 CRVDPs have been completed so that the recommendations and lessons which were found the usefulness, challenges and improvements to the CRVDP can be provided to be better the same kinds of rural development projects. We identified the relationships between performances such as increasing village income and utilization of rural amenity resources to the CRVDP and operational management types of the rural experience programs as well. Employing principle component analysis and cluster analysis technique, this study found 5 clusters of rural experience programs among 168 CRVDPs. The results of analysis of variance indicated that there were significant the mean differences between clusters such as the utilization of rural amenity resources(0.01), income of rural experience programs(0.1). According to the result of the Chi-squire test, there was very significant differences between internet homepage operation and clusters(0.01). Finally, the analysis of covariance about the income of rural experience programs showed that there were significant the mean differences between clusters(0.05).

Palynotaxonomic Study on the Korean Pyrolaceae (한국산 노루발과(Pyrolaceae)의 화분분류학적 연구)

  • Pollen grains of seven taxa on the family Pyrolaceae, known to be distributed in Korea (Pyrola dahurica(Andreses) Kom., P. incarnata (DC.) Fisch. ex Kom., P. japonica Klenze ex Alefeld, P. minor L., P. renifolia Max., Orthilia secunda (L.) House, Chimaphila japonica Miq.), were examined by the light and the scanning electron microscope in order to evaluate their taxonomic significances. The pollen units of genus Pyrola, Orthilia, Chimaphila were tetrad, monad, polyad respectively. This characteristics was good character for delimiting the genus, and Orthilia secunda (L.) House was more proper scientific name than Pyrola secunda L. based upon this character. Pollen size, tricolporate length, surface sculpturing pattern were different slightly among the treated taxa, and surface sculpturing patterns of Pyrola dahurica, P. incarnate, P. renifolia, Chimaphiza japonica were different from previous reports. Evolutionary trends of pollen grains in Pyrolaceae were inferred from the pollen units and aperture shapes.

Screening for Antioxidative Activities of Extracts from Aerial and Underground Parts of Some Edible and Medicinal Ferns (식용 및 악용 양치식물에서 부위별 추출물의 항산화활성 탐색)

  • The different parts of 8 edible and medicinal ferns were extracted with 80% ethanol and the bioactive substances and antioxidant activities were investigated. Total polyphenol content was highest in Cyrtomium fortunei rhizome ($57.32mg{\cdot}g^{-1}$) and followed by C. fortunei root, Pyrrosia lingua aerial part and Onoclea sensibilis var. interrupta aerial part. Total flavonoid contents ranged from 1.95 (Onoclea sensibilis var. interrupta rhizome) to $27.51mg{\cdot}g^{-1}$ (O. sensibilis var. interrupta aerial part). Among the samples tested, C. fortunei root and rhizome were found to be most effective in both DPPH and ABTS radical scavenging assay. Except in Selaginella tamariscina underground part and Osmunda japonica aerial part, most of the tested samples showed strong potential (above 87%) in inhibition rate on linoleic acid oxidation.

Morphological and Immunological Characterization of Hemocytes in Larvae of Pentodon quadridens bidentulus (Famaire, 1887) (둥글장수풍뎅이 혈구 종류와 식균작용)

  • The purpose of this study was to analyze the characteristics of hemocytes in the hemolymph of the larvae of Pentodon quadridens bidentulus and the characteristics of the hemocytes responsible for cellular immunity during pathogen infection. Granulocytes, plasmatocytes, oenocytoids, spherulocytes, prohemocytes and adipohemocytes were found in the circulating hemocytes. Among them, granulocyte were observed as cells responsible for immunological phagocytosis during entry of foreign substances. In particular, it was observed that the most active phagocytic action occurred within 12 hours in vivo, and that after 24 hours, the immune activation was reduced and converted to a normal state. Plasmatocytes were occasionally observed as immunological response, but the remaining hemocytes were not related to immunological activation.

Dynamics of Early Fermentation of Italian Ryegrass (Lolium multiflorum Lam.)Silage

  • The dynamics of fermentation were studied with Italian ryegrass ensiled in the laboratory silos. The silos were kept in the room set at 25$^{\circ}C$, and then were opened on 0.5, 1, 2, 3, 5, 7 and 14 days after ensiling, respectively. The samples were taken from three silos at each sampling time for chemical analyses. Mono-and disaccharides composition was determined for glucose, fructose and sucrose by high performance liquid chromatography. The Italian ryegrass silage succeeded to achieve lactate type fermentation; high values of lactic acid (85.83 g/kg) and lactic acid/acetic acid at the end of ensiling (14 day), low values of pH (3.74), acetic acid (5.38 g/kg), ethanol (19.20 g/kg) and $NH_3-N/Total\;N$ (75.84 g/kg), no or only small amounts of butyric acid, valeric acid and propionic acid. The fermentation dynamics showed a fast and large pH decrease caused by a fast and large production of lactic acid during the first 5 days. Mono-and disaccharides composition largely decreased within initial 0.5 day (12 h) of ensiling. Sucrose disappeared rapidly within initial 0.5 day of ensiling, and fructose and glucose contents showed an initial rise during ensiling, and then decreased gradually. These indicated that the enzymes of plant tissue were active within 2 days of ensiling, which caused the initial rise in fructose and glucose from the hydrolysis of sucrose and fructans. After 5 days of ensilage, glucose was consumed completely, suggesting that glucose was the first fermentation substrate. After 2 days of ensiling, sum amounts of lactic acid and remaining mono-and disaccharides proved to be larger than the quantity of mono-and disaccharides in the initial grass. From the facts mentioned above, it was suggested that considerable amounts of lactic acid were produced from some other substrate such as fructans than initial mono-and disaccharides.

A Molecular Marker Discriminating the Soybean Podworm, Matsumuraeses phaseoli and the Podborer, M. falcana (Lepidoptera: Tortricidae) (팥나방(Matsumuraeses phaseoli)과 어리팥나방(M. falcana)의 판별 분자마커)

  • Two closely related species, the soybean podworm, Matsumuraeses phaseoli, and the podborer, M. falcana, gives differential economic damages on crops. It is difficult to discriminate these potential sympatric species by morphological characters. The goal of this study was to develop a discriminating molecular marker based on polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP). A partial genomic fragment (500 bp) of mitochondrial cytochrome oxidase I (mtCOI) was sequenced in both species, in which restriction site by Rsa I was selected as a dichotomous marker. PCR-RFLP in the mtCOI region clearly discriminated both species.

Identification of Hepatitis C Virus Core Domain Inducing Suppression of Allostimulatory Capacity of Dendritic Cells

  • Hepatitis C virus (HCV) is remarkably efficient at establishing chronic infection. One of the reasons for this appears to be the suppression of the accessory cell function of professional antigen presenting cells. In the present study, the immunosuppressive activity of HCV protein was examined on dendritic cells (DCs) generated from mouse bone marrow progenitor cells in vitro. We found that the DCs forced to express HCV protein have defective allostimulatory ability. DCs expressing HCV protein were phenotypically indistinguishable from normal DCs. However, they were unable to produce IL-12 effectively when stimulated with lipopolysaccharide. The functional domain of the HCV protein essential for immunosuppression was determined using a series of ${NH_2}-and$ C-terminal deletion mutants of HCV core protein. We found that amino acid residues residing between the 21 st and the 40th residues from the ${NH_2}-terminus$ of HCV core protein are required for immunosuppression. These findings suggest that HCV core protein suppresses the elicitation of protective Th1 responses by the inhibition of IL-12 production by DCs.

Optimal Culture Conditions for Production of Yellow Pigments from Monascus purpureus in Liquid Culture (액체 배양에 의한 Monascus purpureus가 생산하는 황색색소의 최적 생산조건)

  • The optimum cultural conditions for production of yellow pigment by Monascus purpureus MMK2 were investigated in liquid culture. Monascus purpureus MMK2 was shown to give the maximum production of yellow pigment in the medium containing of 3.0% wheat flour, 0.15% $NaNO_3$, 0.25% $Na_2HPO_4\;12H_2O\;and\;0.15%\;MgSO_4\;7H_2O$. The optimal culture conditions for temperature and initial pH were $30^{\circ}C$ and 6.5, respectively. The yellow pigment production reached a maximum level at the 7th day of cultivation.
