Antimicrobial and Antioxidant Activity of Camellia japonica Extracts for Cosmetic Applications (동백나무 추출물을 향장소재로 사용하기 위한 항균, 항산화 활성 탐색)

  • This study was carried out to investigate the antimicrobial and antioxidative activities of Camellia japonica extracts for cosmetic applications. Antimicrobial effects of C. japonica were determined against Bacillus cereus by methanol extract of new leaf, stem and stem-leaf; Malassezia pachydermatis, by methanol extract of new leaf and stem-leaf. A methanol extract of new leaf of C. japonica showed strong antimicrobial effect using paper disc method against most species especially in Staphylococcus aureus. Antioxidative activities of C. japonica seed oils were determined by DPPH and ABTS radical scavenging activities. The value of $EC_{50}$ of DPPH scavenging activity was 500 mg/mL and that of ABTS scavenging activity was 96.10 mg/mL. C. japonica oil extracts showed lower antioxidative activities than those of gallic acid and ${\alpha}$-tocopherol. Electron microscopic observation of damaged virgin hairs of different ages gave a stabilizing effects after C. japonica seed oil treatment. These results indicated that the extracts of stem, leaf and seed of C. japonica could be used as cosmetic ingredient combined with appropriate formula.

General Characteristics, Antherpometric Values and Health-Related Factors according to Sasang Constitution among Elderly People in Rural Areas (농촌지역 50세 이상 성인 및 노인의 사상체질에 따른 일반적 특성, 신체계측 및 건강관련인자에 관한 연구)

  • The purpose of this study was to elucidate the general characteristics, antheropometric values and health-related characteristics of the rural elderly population according to their Sasang constitutions. The participants in this study were 31 in Taeeumin group, 29 in Soyangin group, and 15 in Soeumin group. The present study was performed using self-administration questionnaires, antheropometric and biochemical analysis. There were no significant differences in height, weight, BMI, diastolic blood pressure and alcohol habits between the Sasang constitutions. In the analysis of physical activity, Soyangin had a higher value than the other groups. Compared with the other groups, Soeumin also had considerable resistance to stress($101.1{\pm}26.2$ point). Furthermore, in the analysis of fatigability caused by stress, Soeumin was mostly shown to feel less fatigue than the other groups. On the other hand, Taeeumin appeared to feel more fatigue. In the biochemical analysis of blood, there was no significant difference between Sasang constitutions.

  • Three different strains of carotenoid accumulating XantlwphyUomyces dendrorhous mutants, JH1, JH2, and JH3, were isolated by NTG (N-methyl-N'-nitro-N-nitrosoguanidine) mutagenesis, which might potentially be useful for animal feed as well as for studies on the regulation and biosynthesis of astaxanthin. Mutants were selected based on the capability of growth and carotenoid production on the YM agar plate containing chemical inhibitor, $\beta$-ionone. Astaxanthin-overproducing mutant JH1 produced 4.032 mg astaxanthinlg dry cell weight, and this value was about 15-folds higher than that of wild-type. $\beta$-Carotene-overproducing mutant JH2 produced 0.273 mg $\beta$-carotene/g dry cell weight, and this was 4-folds increase from that of wild-type. In contrast, JH3 was a white-colored mutant that was unable to produce carotenoid pigment.

  • The Biochemical Oxygen Demand(BOD) indicates that microbes are proliferating or that oxygen is being spent by breathing action when examining water under the same aerobic condition. In this research of the mesurement of BOD are the poisonous elements of heavy metal ions such as Cu-ion, Cr-ion, Pb-ion and Zn-ion. They exert an unfavorable influence in the analysis of BOD and research was performed to provide certain data of minimum negative influence by the poisonous matters. The results of the research confirm that heavy metal ion(Cu, Cr, Pb, Zn) do direct an influence upon the normal growth of aerobic microbes in actual tests of chemical analysis of portable water or sewage. The most critical concentration for a negative effect on lowering oxygen quantity and disturbing the aerobic mocrobes normal growth was found to be 0.01 mg/l. Therefore, test results are not valid if the heavy metal concentration is to or greater than 0.0mg/l, To improve comprehension through out the research the author uses the following abbreviations: 1. The Cu-ion is to be excluded before experimental analysis if it is over 0.01mg/l inorder to obtain a real value for the BOD. 2. The Cr-ion is to be excluded before experimental analysis if it is over 0.01mg/l in order to obtain a real value for the BOD. 3. The Pb-ion is to be excluded before experimental analysis if it is over 0.01mg/l in order to obtain a real value for the BOD. 4. The Zn-ion is to be excluded before experimental analysis if it is over 0.01mg/l in order to obtain a real value for the BOD.

  • Administrating PalMoolGoonJaTang and SipIMiGwanJungTang to 40-weeks-(old Wistar rat for 10 weeks so, I researched into weight change, weight change of internal organ, and hemtological and serological changews. Then I got these conclusions. 1. Observed gain in weight than control group form of SipIMiGwanJungTang to aged Wistar rat. 2. Is thought to promote activation of living body action gaining intestine weight along with gain in weight. 3. Displayed decrease of MDA's content of serum than control group form of SipIMiGwan JungTang to aged Wistar rat. 4. Change that is Wistar rat's hematological value by aging according to 10, 30, 50 week WBC, RBC, Hgb, monocytes, eosinophil etc. increase, and HCT, PLT etc. showed tendency that decrease according to old-week, and observed improvement that is hematological value than control group form of SipIMiGwanJungTang 5. Functional Characterization of Drosophila melanogaster CYP6A8 Fatty Acid Hydroxylase

  • Genomic analysis indicated that the genome of Drosophila melanogaster contains more than 80 cytochrome P450 genes. To date, the enzymatic activity of these P450s has not been extensively studied. Here, the biochemical properties of CYP6A8 were characterized. CYP6A8 was cloned into the pCW vector, and its recombinant enzyme was expressed in Escherichia coli and purified using Ni2+-nitrilotriacetate affinity chromatography. Its expression level was approximately 130 nmol per liter of culture. Purified CYP6A8 exhibited a low-spin state in the absolute spectra of the ferric forms. Binding titration analysis indicated that lauric acid and capric acid produced type I spectral changes, with Kd values 28 ± 4 and 144 ± 20 µM, respectively. Ultra-performance liquid chromatography-mass spectrometry analysis showed that the oxidation reaction of lauric acid produced (ω-1)-hydroxylated lauric acid as a major product and ω-hydroxy-lauric acid as a minor product. Steady-state kinetic analysis of lauric acid hydroxylation yielded a kcat value of 0.038 ± 0.002 min-1 and a Km value of 10 ± 2 µM. In addition, capric acid hydroxylation of CYP6A8 yielded kinetic parameters with a kcat value of 0.135 ± 0.007 min-1 and a Km value of 21 ± 4 µM. Because of the importance of various lipids as carbon sources, the metabolic analysis of fatty acids using CYP6A8 in this study can provide an understanding of the biochemical roles of P450 enzymes in many insects, including Drosophila melanogaster.

  • Polyethylene terephthalate (PET) is a plastic material commonly applied to beverage packaging used in everyday life. Owing to PET's versatility and ease of use, its consumption has continuously increased, resulting in considerable waste generation. Several physical and chemical recycling processes have been developed to address this problem. Recently, biological upcycling is being actively studied and has come to be regarded as a powerful technology for overcoming the economic issues associated with conventional recycling methods. For upcycling, PET should be degraded into small molecules, such as terephthalic acid and ethylene glycol, which are utilized as substrates for bioconversion, through various degradation processes, including gasification, pyrolysis, and chemical/biological depolymerization. Furthermore, biological upcycling methods have been applied to biosynthesize value-added chemicals, such as adipic acid, muconic acid, catechol, vanillin, and glycolic acid. In this review, we introduce and discuss various degradation methods that yield substrates for bioconversion and biological upcycling processes to produce value-added biochemicals. These technologies encourage a circular economy, which reduces the amount of waste released into the environment.

Efficacy of I-123/I-131 Metaiodobenzylguanidine Scan as A Single Initial Diagnostic Modality in Pheochromocytoma: Comparison with Biochemical Test and Anatomic Imaging (갈색세포종의 초기 진단에서 I-123/I-131 Metaiodobenzylguanidine 스캔의 단일 검사로써의 진단 성능: 생화학적 검사, 해부학적 영상과 비교)

  • Purpose: We underwent this study to evaluate the diagnostic potential of I-123/I-131 metaiodobenzylguanidine (MIBG) scintigraphy alone in the initial diagnosis of pheochromocytoma, compared with biochemical test and anatomic imaging. Materials & Methods: Twenty two patients (M:F=13:9, Age: $44.3{\pm}\;19.3$ years) having the clinical evaluation due to suspicious pheochromocytoma received the biochemical test, anatomic imaging modality (CT and/or MRI) and I-123/I-131 MIBG scan for diagnosis of pheochromocytoma, prior to histopathological confirmation. MIBG scans were independently reviewed by 2 nuclear medicine physicians. Results: All patients were confirmed histopathologically by operation or biopsy (incisional or excisonal). In comparison of final diagnosis and findings of each diagnostic modality, the sensitivities of the biochemical test, anatomic imaging, and MIBG scan were 88.9%, 55.6%, and 88.9%, respectively. And the specificities of the biochemical test, anatomic imaging, and MIBG scan also were 69.2%, 69.2%, and 92.3%, respectively. MIBG scan showed one false positive (neuroblastoma) and one false negative finding. There was one patient with positive MIBG scan and negative findings of the biochemical test, anatomic imaging. Conclusion: Our data suggest that I-123/I-131 MIBG scan has higher sensitivity, specificity, positive predictive value, negative predictive value and accuracy than those of biochemical test and anatomic imaging. Thus, we expect that MIBG scan is e tectively used for initial diagnosis of pheochromocytoma alone as well as biochemical test and anatomic imaging.
