Novosphingobium ginsenosidimutans sp. nov., with the Ability to Convert Ginsenoside

  • A Gram-negative, strictly aerobic, non-motile, non-spore-forming, and rod-shaped bacterial strain designated FW-$6^T$ was isolated from a freshwater sample and its taxonomic position was investigated by using a polyphasic approach. Strain FW-$6^T$ grew optimally at $10-42^{\circ}C$ and at pH 7.0 on nutrient and R2A agar. Strain FW-$6^T$ displayed ${\beta}$-glucosidase activity that was responsible for its ability to transform ginsenoside $Rb_1$ (one of the dominant active components of ginseng) to Rd. On the basis of 16S rRNA gene sequence similarity, strain FW-$6^T$ was shown to belong to the family Sphingomonadaceae and was related to Novosphingobium aromaticivorans DSM $12444^T$ (98.1% sequence similarity) and N. subterraneum IFO $16086^T$ (98.0%). The G+C content of the genomic DNA was 64.4%. The major menaquinone was Q-10 and the major fatty acids were summed feature 7 (comprising $C_{18:1}{\omega}9c/{\omega}12t/{\omega}7c$), summed feature 4 (comprising $C_{16:1}{\omega}7c/iso-C_{15:0}2OH$), $C_{16:0}$, and $C_{14:0}$ 2OH. DNA and chemotaxonomic data supported the affiliation of strain FW-$6^T$ to the genus Novosphingobium. Strain FW-$6^T$ could be differentiated genotypically and phenotypically from the recognized species of the genus Novosphingobium. The isolate that has ginsenoside converting ability therefore represents a novel species, for which the name Novosphingobium ginsenosidimutans sp. nov. is proposed, with the type strain FW-$6^T$ (= KACC $16615^T$ = JCM $18202^T$).

A Study on the Application of Manganese Oxidizing Bacteria for Manganese Treatment in Acid Mine Drainage (산성광산배수의 망간처리를 위한 MOB 적용에 관한 연구)

  • Domestic treatment facilities for acid mine drainage (AMD) mostly used a passive treatment process. But some passive treatment facility discharged high manganese concentrations because it is required high pH (>9) for abiotic oxidation of Mn(II) to Mn(IV). This study was focused on the feasibility of biological manganese treatment using the manganese-oxidizing bacteria (Pseudomonas sp. MN5) from AMD and economical application method of it. To investigate the various conditions of water quality the most part of the experiments were based on batch test. And result of it showed that maximum manganese oxidation rate were $10.4mg/L{\cdot}h$ at the pH7. We also performed small column tests in which MOB were attached to the functional polyurethane (FPU) media containing alkaline chemicals. Manganese concentration decreased 42 mg/L to below 6 mg/L. But anaerobic condition formed by excessive bacterial respiration in column resulted in increasing effluent manganese concentration.

Identification and Physiological Characters of Intestinal Bacteria of the Black Soldier Fly, Hermetia illucens (아메리카동애등에 장내세균 동정과 생리적 특징)

  • The black soldier fly, Hermetia illucens, larvae may depend on indigenous bacteria in the intestine to feed and digest diverse food sources. To prove this hypothesis, we isolated and identified the intestinal bacteria of the black soldier fly for their digestive and antimicrobial abilities. The last instar larvae had long digestive tracts, which were about seven times longer than its body length. An individual of H. illucens larvae possessed a total of $5.0{\pm}10^6$ bacteria in the whole intestine, of which more than 98% bacteria were located in the hindgut. Three different bacterial isolates cultured on nutrient agar (NA) medium were detected in the intestine and identified as Morganella morganii, Providencia rettgeri and Bacillus halodurans by Biolog microbial identification system. Analysis of 16S rDNA sequences of the intestinal bacteria detected the additional bacteria of Proteus mirabilis, Providencia alcalifaciens, and Providencia sp. These intestinal bacteria cultured on NA medium exhibited high resistance to 4 antibiotics and inhibited growth of other microbes which are mainly plant pathogens. Also, these bacteria exhibited catalytic activities to degrade cellulose, lipid, proteins, and carbohydrates. These results suggest that H. illucens larvae possess intestinal bacteria that may play crucial roles in their digestive physiology.

A Grub (Protaetia brevitarsis seulensis) Rearing Technique Using Cellulose-digesting Bacteria and Natural Recycling of Rearing Byproduct to an Organic Fertilizer (셀룰로오스 분해균을 이용한 흰점박이꽃무지(Protaetia brevitarsis seulensis) 사육과 부산물 응용 기술)

  • Cellulose-digesting bacteria were isolated from hindgut of Allomyrina dichotoma (Coleoptera: Dynastidae). The bacterial isolates were identified as Yersinia sp. and Bacillus sp. The addition of the identified bacteria to diet increased growth rate of the cetoniid beetle, Protaetia brevitarsis senlensis (Coleoptera: Cetoniidae), probably by digesting cellulose nutrient contained in the oak tree sawdust diet. An additive of wheat flour at more than 10% to the sawdust diet significantly enhanced growth of P. brevitarsis senlensis. Trimmed branches of apple trees have been disposed in the apple farms and could be used for a diet component of the cetoniid beetle when the cellulose-digesting bacteria were mixed with the derived-sawdust. Resulting manure from mass rearing of P. brevitarsis senlensis contained high organic matters and trace amounts of toxic metals. When the manure were splayed on soil, it was effective as a natural compost and significantly stimulated lettuce growth. This research suggests a model technology to use cellulose-digesting bacteria to use for culturing grub, which results in natural recycles of trimmed branches in apple farms as grub diet, and to use grub manure as a natural compost.

Properties of Soil Suppressiveness to Cucumber Wilt, caused by Fusarium oxysporum f. sp. cucumerinum Owen (오이 덩굴쪼김병의 발병(發病)을 억제(抑制)하는 토양(土壤)의 특성(特性)에 관(關)하여)

  • It has been tried to find effective biological control measures involved in nature of soil suppressiveness to fusarium wilt of cncumber caused by Fusarium oxysporum f. sp. cucumerinum Owen. Total 28 soil samples were obtained from Jinju, Haman, Namji, Milyang and Suncheon vinyl house area. The disease response of test soil was quantified in terms of DI50 value which caculated from log-probit transformation of diseases response curves. Soils designated 5 from Jinju, 7 from Suncheon, 22 from Namji were recognized as suppressive to fusarium wilt of cucumber. This suppressiveness was completely nullified after autoclave. The disease suppressiveness of tested soil did not indicate any consistency according to either chemical property or texture of soil. Conidial germination, induction and germination of chlamydospore were markedly inhibited in supprerssive soil compared to those in intermediate or conducive soils, however, mycelial lysis did not appear to have direct relationship with disease suppressiveness of given soil. Population density of fluorescent Pseudomonads and Bacillus spp. in the soil originated from different degree of suppressiveness were not different significantly but the number of lytic bacterial plaques measured by triple layer agar method was remarkably higher in suppressive soil than that in intermediate or conducive soil.

Bacillus ginsengihumi sp. nov., a Novel Species Isolated from Soil of a Ginseng Field in Pocheon Province, South Korea

  • A Gram-positive, aerobic or facultative anaerobic, non motile, endospore-forming bacterial strain, designated Gsoil $114^T$, was isolated from a soil sample of a ginseng field in Pocheon Province (South Korea), and was characterized taxonomically by using a polyphasic approach. It grew well on nutrient agar medium and utilized a limited number of organic substrates as sole carbon sources, including D-xylose and some other carbohydrates, but did not utilize L-amino acids and organic acids. The isolate was positive for oxidase test but negative for catalase, and negative for degradation of macromolecules such as starch, cellulose, xylan, casein, chitin, and DNA. The G+C content of the genomic DNA was 41.8 mol%. The predominant isoprenoid quinone was menaquinone 7 (MK-7). The major fatty acids were $anteiso-C_{15:0}$ (32.1%), $iso-C_{15:0}$ (30.5%), and $anteiso-C_{17:0}$ (30.2%). Comparative 16S rRNA gene sequence analysis showed that strain Gsoil $114^T$ fell within the radiation of the cluster comprising Bacillus species and joined Bacillus shackletonii LMG $18435^T$ with a bootstrap value of 95%. The highest 16S rRNA gene sequence similarities were found with Bacillus shackletonii LMG $18435^T$ (97.6%), Bacillus acidicola DSM $14745^T$ (96.9%), Bacillus sporothermodurans DSM $10599^T$ (96.5%), and Bacillus oleronius DSM $9356^T$ (96.5%). The phylogenetic distance from any other validly described species within the genus Bacillus was less than 96%. DNA-DNA hybridization experiments showed that the DNA-similarities between strain Gsoil $114^T$ and closest phylogenetic neighbors were less than 39%. On the basis of its phenotypic properties and phylogenetic distinctiveness, strain Gsoil $114^T$ (=KCTC $13944^T$=DSMZ $18134^T$) was classified in the genus Bacillus as the type strain of a novel species, for which the name Bacillus ginsengihumi sp. nov. is proposed.

Seasonal Succession of Planktonic Ciliate in Kyungan Stream of Lake Paldang, Korea (팔당호 유입부 경안천의 섬모충 플랑크톤 계절적 분포)

  • Seasonal succession and community composition of planktonic ciliates were studied in Kyungan Stream from December 2000 to December 2001. Oligotrichs accounted for 53% of total ciliates. Total abundance of ciliates peaked in spring (30 March, 6 April) and in summer (8 June, 20 July) reaching values up to $1.9\times10^4$ cells $L^{-1}$. Seasonal succession of dominant species occurred obviously. Large-sized $(>50{\mu}m)$ species (Stylonychia sp1, Phascolodon vorticella and Codonella cratera) dominanted from winter to spring. Small sized $(<30{\mu}m)$ species (Vorticella spp., Rimostrombidium hyalinum and Halteria grandinella) dominanted in summer and autumn. Total abundance of large-sized species coincided with the Chl-${\alpha}$ concencetation during the study (r=0.33, p<0.05, n=39). Among the small-sized species Halteria grandinella was a significant relationship with bacterial abundance (r=0.35, p<0.05, n=39).

Contamination level of commercialized pepper and sterilization effect by intense pulsed light in batch system (시중 판매 후추의 오염도 및 회분식 광펄스 처리에 의한 살균 효과)

  • Twenty-nine pepper products commercially available in the market were collected and investigate for contamination levels. Pepper products purchased from traditional markets had a degree of contamination of $10^6-10^7CFU/g$ aerobic bacteria, $10^4-10^5CFU/g$ Bacillus sp., and less than $10^2CFU/g$ yeast and molds. Organic pepper showed a degree of contamination of $10^4$ aerobic bacteria, $10^2-10^3$ Bacillus sp., and less than $10^1$ yeast and molds. Intense pulsed light (IPL) treatment of 10 min (1,000 V, 5 pps and 4 cm sample-to-lamp distance) showed a bacterial death rate of 1.45-1.55 log for whole peppers, and of 0.8-0.85 log for black and white pepper powder. The sterilization rate using IPL was higher than that using other non-thermal sterilization methods, such as ozone treatment or low-pressure discharge plasma sterilization, indicating that the IPL sterilization method may find potential application in the industry. However, further studies may need to be conducted to enhance the effect of sterilization.

Characterization of Miniimonas sp. S16 isolated from activated sludge (활성슬러지로부터 분리된 Miniimons sp. S16 세균의 특성)

  • Biological factors (e.g. microorganism activity) in wastewater treatment plant (WWTP) play essential roles for degradation and/or removal of organic matters. In this study, to understand the microbial functional roles in WWTP, we tried to isolate and characterize a bacterial strain from activated sludge sample. Strain S16 was isolated from the activated sludge of a municipal WWTP in Daejeon metropolitan city, the Republic of Korea. The cells were a Gram-stain-positive, non-motile, facultative anaerobe, and rod-shaped. Strain S16 grew at a temperature of $15{\sim}40^{\circ}C$ (optimum, $30^{\circ}C$), with 0~9.0% (w/v) NaCl (optimum, 1.0~2.0%), and at pH 5.5~9.0 (optimum, pH 7.0~7.5). Phylogenetic analysis based on 16S rRNA gene sequences indicated that strain S16 was most closely related to the unique species Miniimonas arenae NBRC $106267^T$ (99.79%, 16S rRNA gene sequence similarity) of the genus Miniimonas. The cell wall contained alanine, glutamic acid, serine, and ornithine. Although the isolation source of the type strain NBRC $106267^T$ which considered as a marine microorganism is sea sand, that of strain S16 is terrestrial environment. It might raise an ecological question for habitat transition. Therefore, comparative genome analysis will be valuable investigation for shedding light on their potential metabolic traits and genomic streamlining.

Molecular Mechanisms Involved in Peptidoglycan-induced Expression of Tumor Necrosis Factor-α in Monocytic Cells (펩티도글리칸에 의한 단핵세포의 Tumor necrosis factor-α 발현 기전 연구)

  • Peptidoglycan (PG) is found in atheromatous lesions of arteries, where monocytes/macrophages express inflammatory cytokines, including tumor necrosis factor-alpha ($TNF-{\alpha}$). This study investigated the effects of PG on $TNF-{\alpha}$ expression and examined possible cellular factors involved in $TNF-{\alpha}$ upregulation. The overall aim was to identify the molecular mechanisms underlying inflammatory responses to bacterial pathogen-associated molecular patterns in the artery. Exposure of human THP-1 monocytic cells to PG enhanced the secretion of $TNF-{\alpha}$ and induced its gene transcription. Inhibition of TLR-2/4 with OxPAPC significantly inhibited $TNF-{\alpha}$ gene expression, whereas inhibition of LPS by polymyxin B did not. The PG-induced expression of $TNF-{\alpha}$ was also significantly suppressed by pharmacological inhibitors that modulate activities of cellular signaling molecules; for example, U0126 (an ERK inhibitor), SB202190 (a p38 MAPK inhibitor), and SP6001250 (a JNK inhibitor) significantly attenuated PG-induced transcription of $TNF-{\alpha}$ and secretion of its gene product. $TNF-{\alpha}$ expression was also inhibited by rapamycin (an mTOR inhibitor), LY294002 (a PI3K inhibitor), and Akt inhibitor IV (an Akt inhibitor). ROS-regulating compounds, like NAC and DPI, also significantly attenuated $TNF{\alpha}$ expression induced by PG. These results suggest that PG induces $TNF-{\alpha}$ expression in monocytes/macrophages by multiple molecules, including TLR-2, PI3K, Akt, mTOR, MAPKs, and ROS.
