In vitro Multiplication through Single-Node Culture of Sea-Milkwort (Glaux maritima L.) (갯봄맞이(Glaux maritima L.) 실생의 단마디배양을 통한 기내증식)

  • This study was conducted to establish an in vitro propagation system for sea-milkwort (Glaux maritima L.), which is an endangered coastal plant species with high horticultural value. Two phenotypes, 'Red type (RT)' and 'Pistachio type (PT)' based on the colors of stem and flower, were obtained from a personal horticulturist in 2009 and used for this study as plant materials. The stock plants showed typical morphologies in flower, capsule, and seed appearances as previously reported. Low temperature treatment at $4^{\circ}C$ for four or more weeks after in vitro sowing maximized seed germination percentage, indicating that imbibition of seed and subsequent low temperature treatment are crucial for its germination. The in vitro seedlings had phenotypic variation, falling into 'RT' and 'PT' classes like the stock plants. Although slight differences depending on genotype and medium were recognized, the fourth or fifth nodes detached from the in vitro seedlings revealed the best multiplication efficacy when estimated on the basis of total number of nodes of newly developed axillary shoots. In addition, the nodes from 'RT' and 'PT' regenerated the most shoots on medium supplemented with $0.5mg{\cdot}L^{-1}$ BA alone and $0.5mg{\cdot}L^{-1}$ BA plus $0.5mg{\cdot}L^{-1}$ IAA, respectively. The node culture-derived plantlets were well acclimatized in a culture room ex vitro and completed the pseudo-annual life cycle coincident with that in the natural salt march habitat with the current cultivation method of applying fresh water-irrigation under an inland environment. This work represents the first report of in vitro propagation of sea-milkwort. Thus, our study will contribute to exo-habitat conservation and natural habitat restoration of this endangered species in addition to development of a horticultural product.

Association between physical activity measured using an accelerometer and arterial stiffness based on pulse wave velocity and ankle-brachial index in healthy adults (건강한 성인에서 가속도계로 측정한 신체활동과 맥파전달속도 및 상완-발목 간 혈압비에 기반한 동맥경화지표와의 관계)

  • Purpose: Physical activity (PA) has a beneficial effect on the prevention of arteriosclerosis in healthy adults. The purpose of this study was to analyze the relationship between PA measured using an accelerometer and arterial stiffness in healthy Korean adults. Methods: This study involved 87 subjects (36.8% women) aged 20-64 years. PA was evaluated using an accelerometer (wGT3X-BT, ActiGraph, Florida, USA) for 7 days. Based on the results of the accelerometer measurement, subjects were classified into active and inactive groups according to the World Health Organization (WHO) PA guidelines. The brachial-ankle pulse wave velocity (baPWV) and ankle-brachial index (ABI) to assess arterial stiffness were measured by a non-invasive vascular screening device (VP-1000 Plus, Omron). Results: The average age of the study subjects was 47.7 ± 11.3 years and the WHO PA guideline achievement rate was 29.9%. There was no significant difference in arterial stiffness (baPWV and ABI) between the active and inactive groups. In females, the time spent in light PA were positively correlated with ABI (r = 0.396; p < 0.05) and the number of sedentary bouts over 50 minutes was inversely correlated with ABI (r = -0.402; p < 0.05). However, there was no significant correlation between PA and arterial stiffness in males. Conclusions: The results of this study suggest that light PA and sedentary behavior have a positive correlation with arterial stiffness in females.

Effect of tissue proliferation and somatic embryo induction in Larix kaempferi following treatment with organic nitrogen sources and plant growth regulators (일본잎갈나무(Larix kaempferi) 유기질소원 및 식물생장조절물질 처리에 따른 조직증식 및 체세포배 유도 효과)

  • This study was conducted to evaluate the effects of different types and concentrations of organic nitrogen sources (${\small{L}}$-Glutamine and casein hydrolysate, CH) and plant growth regulators (auxins and cytokinins) on embryogenic tissue proliferation and somatic embryo production in L. kaempferi. Overall, the highest tissue fresh weight was obtained at either 2 or 4 weeks in culture when 1,000 mg/L ${\small{L}}$-Glutamine was added to the culture medium, which showed similar results with other treatments. In experiments with different types and concentrations of plant growth regulators on somatic embryo production, the highest production (426.3/90 mg tissue) was found when 0.2 mg/L IBA was added; however, no somatic embryos were induced following treatment with 0.2 mg/L BA or Kinetin. The effect of various concentrations of IBA on somatic embryo production was also tested. The best result (303/90 mg tissue) was obtained when plants were treated with 0.2 mg/L IBA; 1.0 mg/L IBA was also effective (281/90 mg tissue). The lowest result (109.3/90 mg tissue) was obtained with 5.0 mg/L IBA.

Is Heart Rate Measured by Smartwatch during Exercise Reliable? Analysis of Correlation and Agreement Between Heart Rates of Polar and Smartwatch (운동 중 스마트워치 심박수 믿을 만한가? 폴라와 스마트워치 심박수 간 상관과 일치도 분석)

  • The purpose of this study is to investigate the correlation and agreement between heart rates of Polar heart rate monitor and a smartwatch in order to confirm the accuracy of heart rate measured by the smartwatch. Heart rates of fifty college students were measured for a total of 12 minutes under four conditions: rest, walk, Zumba, and cycle. As a result of correlation and agreement analysis between heart rates of the two devices, correlation coefficient (r) was 0.995 at rest, 0.991 at walk, 0.923 at Zumba, 0.932 at cycle, and Bland-Altman ratio (BA ratio) was 0.02 at rest, 0.03 at walk, 0.06 at Zumba, 0.04 at cycle. Heart rate from smartwatch showed high correlation and agreement with heart rate from Polar in all conditions, representing that smartwatch can be considered an reliable apparatus to measure hear rate.

  • Efficient plant regeneration system of birdsfoot trefoil (Lotus corniculatus L.) was development. The factors affecting the somatic embryo formation, its proliferation and regeneration capacity of leaf and stem explants of Empire cultivar was investigated. The highest number of somatic embryos was obtained on B5 medium supplemented with 1 mg/L NAA and 1 mg/L BA. Depending on different explants, highest frequency of embryogenic callus and regeneration were observed in Empire with leaf explants. The response from stem explants was slower and callus induction was less than that from leaf explants. Regenerated shoots formed complete plantlets in on 1/2 MS medium supplemented with 1 mg/L IBA. Regenerated plants were morphologically uniform with normal shape and growth pattern.

  • Objective: The aim of this study is to find the most helpful midsagittal reference plane for diagnosis in PA cephalometry compared with 3D CT. Methods: The subjects consisted of 25 adults who showed no facial asymmetry by gross inspection. 3D CT and posteroanterior cephalogram of the subjects were taken. To find the most helpful midsagittal reference plane in PA cephalometry, we considered five kinds of midsagittal planes from which the distances to five landmarks were measured and compared the result with that of 3D CT. The midsagittal plane for 3D CT was determined by the landmarks Nasion, Sella and Basion. Results: PA measurements using the midsagittal reference plane on a perpendicular plane lying through the midpoint of the right and left latero-orbitales was closest to those of 3D CT. Conclusions: It was considered that latero-orbitale perpendicular could be used as the helpful midsagittal reference plane to assess facial asymmetry in PA cephalometry.

Study on Low-Temperature Solid Oxide Fuel Cells Using Y-Doped BaZrO3 (Y-doped BaZrO3을 이용한 저온형 박막 연료전지 연구)

  • In this study, we fabricate and investigate low-temperature solid oxide fuel cells with a ceramic substrate/porous metal/ceramic/porous metal structure. To realize low-temperature operation in solid oxide fuel cells, the membrane should be fabricated to have a thickness of the order of a few hundreds nanometers to minimize IR loss. Yttrium-doped barium zirconate (BYZ), a proton conductor, was used as the electrolyte. We deposited a 350-nm-thick Pt (anode) layer on a porous substrate by sputter deposition. We also deposited a 1-${\mu}m$-thick BYZ layer on the Pt anode using pulsed laser deposition (PLD). Finally, we deposited a 200-nm-thick Pt (cathode) layer on the BYZ electrolyte by sputter deposition. The open circuit voltage (OCV) is 0.806 V, and the maximum power density is 11.9 mW/$cm^2$ at $350^{\circ}C$. Even though a fully dense electrolyte is deposited via PLD, a cross-sectional transmission electron microscopy (TEM) image reveals many voids and defects.

Plant Regeneration Via Direct Adventitious Roots from Free Root Segments of Ulmus davidiana Planch (당느릅나무(Ulmus davidiana Planch) 부정근 절편으로 부터 부정아 유도를 통한 식물체 생산)

  • Micropropagation of Ulmus davidiana Planch was established via adventitious shoot formation from the segments of adventitious roots. Adventitious roots were produced directly from root segments of seedlings on a 1/2 SH medium plus various concentrations of IBA. The maximum growth of adventitious roots was observed in the presence of 2.0 mg/L IBA. After the segments of adventitious roots were cultured on various cytokinins (zeatin, 2-iP, BA, kinetin) and cytokinins plus auxin (IBA), formation of adventitious shoot was investigated. Among cytokinin treated, kinetin was the most effective on both adventitious shoot induction and number of shoots. Especially, 2.0 mg/L kinetin was the best to increase adventitious shoot induction (95.8%) and a number of shoots (8.4). Adventitious shoots were rooted on 1/2 WPM medium and the plantlets were acclimated 100% on composed soil (peatmoss : vermiculite 1 : 1).

High Resolution Elements Analysis in N-E Pacific Sediments using XRF Core Scanner (XRF 코어스캐너를 이용한 북동태평양 퇴적물 내 원소의 고해상분석)

  • The XRF core scanner was used, to analyze high resolution chemical elements in deep sea sediment cores from Clarion-Clipperton fracture zone of the northeastern Pacific. Comparison of data estimated by the XRF core scanner with ICP-AES showed relatively weak correlation coefficients between elements (especially Ba, Pb, Sr, Zr) except for Mn contents ($r^2$ > 0.89). However down-core variations of most elements seemed to be well matched each other and furthermore, XRF core scanner data reflected changes of sedimentary facies characterized by sediment colors. Mn/Al ratio dramatically changed at boundaries of facies in BC08-02-05 and BC08-02-13 but progressive changes occured in BC08-02-02, BC08-02-09 and BC08-02-10 where the sediments have been affected by bioturbations. The difference of Mn/Al ratio in each facies (Facies I, Facies II, Facies III) has been caused by redox condition of depositional environment. Vertical change of Mn/Al ratio were divided into two types probably affected by activities of benthic organisms in the study area.

Multiple Shoot Induction from Radicle-derived Callus and in Vitro Propagation of Silene Acaulis Subsp. Arctics (극지식물 Silene acaulis subsp. arctica의 유근 유래 캘러스로부터 다신초 유도와 기내 증식)

  • We describe here an efficient in vitro propagation method of Silene acaulis subsp. arctica (Caryophyllaceae), one of the higher arctic angiosperms, through the multiple shoot regeneration after callus induction from the radicle. The seeds of S. acaulis subsp. arctica collected from Svalbard, the Norwegian Arctic, were germinated and calli were induced from the radicle on solid MS media supplemented with 0.25mg/L 2,4-D and 1mg/L $GA_3$ at both $10{\pm}1^{\circ}C\;and\;23{\pm}1^{\circ}C$ Two weeks after callus induction, the multiple shoots were efficiently regenerated on the MS media supplemented with 0.25 g/L BA and 0.05mg/L HPh. The total biomass increment of regenerated shoots increased most efficiently of S. acaulis subsp. afctica was showed the maximum efficiency in at $23{\pm}1^{\circ}C$ on 1/2 MS salt strength. The multiple regenerated plantlets of S. acaulis subsp. arctics were grown to normal plants on soil.
