하수오 물 추출물이 마우스 골다공증에 미치는 영향 (Antiosteoporotic Effects of Polygoni Multiflori Radix (PMR) in Ovariectomized (OVX)-Induced Osteoporosis ddY Mice)

  • Polygoni Multiflori Radix (PMR), a dried root tuber of Polygonum multiflorum Thunberg with bioactivities in bone metabolism is one of the most famous tonic traditional medicines. To observe in vivo anti-osteoporotic efficacy of PMR extracts, we orally administered once a day for 28 days (Qd ${\times}$ 28) to bilateral ovariectomized (OVX)-induced osteoporosis ddY mice after 1 week of recovery periods at 125, 250 and 500 mg/kg (of body weight). A positive control drug, Alendronate (FOSA) 10 mg/kg-dosing group was added. As results of OVX-induced osteoporotic process, estrogen-deficient osteoporotic changes were also dramatically decreased in all PMR extracts-dosing groups. Especially middle dosage of PMR extracts, 250 mg/kg constantly and significantly (p < 0.01 or p < 0.05) inhibited the loss of bone strength and bone quality. Based on the results, it was concluded that PMR extracts (125, 250 or 500 mg/kg; orally dosing) has relatively good favorable effect to prevention and/or treatment of OVX-induced osteoporosis. Therefore, although the efficacy was slighter than that of Alendronate on the inhibition of bone loss, it is expected that PMR extracts will be promising as a new anti-osteoporotic agents for prevent the fracture induced in osteoporotic patients because natural herbal medicine origin PMR extracts will be dose not show serious side effects especially the problem in upper alimentary irritation by bisphosphonate and hypercalcaemia of parathyroid hormone analogs.

Female SD Rat mice underwent bilateral ovariectomy for prevention study that test article was administered from 2 days after ovariectomy for 6 weeks, for therapeutic study it was conducted from 6 weeks after ovariectomy for three months.   • Among aging diseases, the most basic problem is a decrease in bone mineral density. Patients with osteoporosis are steadily increasing in the world. This study was investigated to show the relationship between osteoporotic effects and oxidative damage. Water extracts of 15 medicinal plants and ethanol extracts of 14 medicinal plants with known anti-osteoporotic effects, were tested for their radical scavenging activity using DPPH, ABTS, SRSA and FRAP assay. Water extract of Cornus officinalis, Rubus coreanus and ethanol extract of Rubus coreanus, Viscium album var. coloratum, Cimicifuga heracleifolia showed about 15-20 mg/g of total phenolic contents. Water extract of Cornus officinalis, Rubus coreanus and Epimedium koreanum showed high radical scavenging activity. Ethanol extract of Drynaria fortunei, Cornus officinalis, Rubus coreanus, Gentiana scabra and Astragalus membranaceus showed high radical scavenging activity. Water extract of Drynaria fortunei, Cornus officinalis, Nelumbo nucifera, Epimedium koreanum, and Gentiana scabra showed very strong protective effect against oxidative damage to DNA. These results suggest the correlation between anti-osteoporotic effects and antioxidative effects.

  • Polygoni Multiflori Radix (PMR) a dried root tuber of Polygonum multiflorum Thunberg with bioactivities in bone metabolism is one of the most famous tonic traditional medicines in China, Japan and Korea.To observe the anti-osteoporotic effect of PMR, we observed the effects on proliferation and alkaline phosphatase (ALP) activity of primary osteoblasts, bone nodule formation, pit formation of osteoclasts and osteoclastogenesis in vitro. As the result of that, although PMR extracts have no meaningful changes on the the proliferation of primary osteoblasts, significant (p < 0.01) increases of ALP acitivity of osteoblasts were dose-dependently observed in all PMR extracts treated groups tested compared to that of vehicle control. It means that it enhanced the differentiation (ALP activity) and bone nodule formation of osteoblast. In addition, PMR extracts dose-dependently inhibits the pit formation and the number of multinucleated osteoclast-like cells (OCLs), osteoclastogenesis in vitro. Therefore these results show the possibility of PMR extracts as another anti-osteoporotic agents.

MC3T3-E1 조골세포주와 RAW 264.7 파골세포주에서 길경을 함유한 한약재 추출물의 항골다공증 효과 (Anti-osteoporotic Activity of Mixed Herbal Extract Involving Platycodon Grandiflorum Root in Osteoblastic MC3T3-E1 and Osteoclastic RAW 264.7 Cells)

  • 목 적: 본 연구는 길경(Platycodon grandiflorum root)를 함유한 복합 한약재 추출물(ExMH-PGR)의 골다공증 예방 및 치료효과를 알아보기 위해 MC3T3-E1 조골세포주와 RAW 264.7 파골세포주를 이용하여 in vitro 수준에서 검증하였다. 방 법: 배양액에 다양한 농도의 ExMH-PGR를 첨가하여 MC3T3-E1 세포와 RAW 264.7 세포의 세포증식을 비교하였다. MC3T3-E1 세포에서 Alkaline phosphatase (ALP) 활성, 콜라겐 합성, 오스테올칼신 생성, 무기질 축적을 분석하였다. RAW 264.7 세포에서 Tartrate-resistant acid phosphatase (TRAP) 활성과 actin ring 형성 정도를 분석하였다. 결 과: ExMH-PGR는 $25{\mu}g/mL$ 농도까지 유의한 수준으로 ALP 활성, 콜라겐 합성, 그리고 오스테올칼신 형성을 증가시켰다. $50{\sim}200{\mu}g/mL$의 ExMH-PGR은 TRAP 활성과 actin ring 형성을 유의하게 억제했다. 결 론: ExMH-PGR은 조골세포의 활성을 촉진하고 파골세포의 활성을 억제하는 효과가 있어 골다공증 치료에 유용할 가능성이 있다.

The Inhibitory Effects of Glycyrrhiza uralensis on human Platelet Aggregation and Thrombus Formation

  • Platelets are activated at the sites of vascular injury by a number of molecules, including adenosine diphosphate, collagen and thrombin. The full platelet aggregation is absolutely essential for the normal hemostasis. Glycyrrhiza glabra is a well-known medicinal herb that grows in various parts of the world and is known to have various effects such as antioxidant, anti-inflammatory, anti-atherogenic, anti-osteoporotic and skin-whitening. However, the platelet inhibitory effect of Glycyrrhiza glabra extract (GGE) has not been identified. In this study, we investigated if GGE inhibited platelet aggregation. We observed that GGE inhibited collagen-induced platelet aggregation, Ca2+ mobilization, and thromboxane A2 generation. In addition, GGE suppressed phosphorylation of phosphatidylinositol-3 kinase (PI3K), Akt and elevated phosphorylation of inositol 1,4,5-trisphosphate receptor (IP3R), vasodilator stimulated phosphoprotein (VASP). Taken together, GGE showed strong antiplatelet effects and may be used to block platelet-mediated cardiovascular diseases.

  • Panax ginseng C.A. Meyer (P. ginseng) is known to exert a wide range of pharmacological effects both in vitro and in vivo. Although studies on ginsenoside, antioxidant activity, and anticancer effect of wild simulated ginseng (WSG) have been conducted, there is little research on the effect of WSG on bone metabolism. In this study, we investigated the potential anti-osteoporotic properties of WSG on the growth and differentiation of MC3T3-E1 cells. WSG significantly increased the viability and proliferation of MC3T3-E1 cells. WSG activated intracellular alkaline phosphatase (ALP) activity in MC3T3-E1 cells. In addition, WSG increased the mineralized nodules in MC3T3-E1 cells. Furthermore, WSG increased the expression of genes such as Runx2, ALP, OPN and OCN associated with osteoblast growth and differentiation in a dose-dependent manner.

Acer tegmentosum Maxim Prevents Bone Loss by Inhibiting Osteoclastogenesis and Promoting Osteoblast Mineralization in Ovariectomized Mice

  • Osteoporosis is a worldwide disease leading to significant economic and societal burdens globally. Osteoporosis is caused by unbalanced bone remodeling between the rate of osteoclast bone resorption and osteoblast bone formation. Acer tegmentosum Maxim (AT) is a traditional herbal medicine containing multiple biological activities such as anti-oxidant and anti-inflammatory purposes. However, its role in osteoporosis has not been fully studied. Therefore, we investigated whether AT has a potent inhibitory effect on osteoporosis and its mechanism through a systemic evaluation in ovariectomized (OVX) mice. OVX mice were orally administrated with the AT at doses of 50, 100, and 200 mg/kg for 10 weeks. Histological images and histomorphometry analyses were performed by H&E and Toluidine blue satin, and the expression levels of receptor activator for nuclear factor-kB ligand (RANKL), nuclear factor of activated T cells cytoplasm 1 (NFATc1), c-Fos, and matrix metalloproteinase 9 (MMP9) related to the osteoclast differentiation were investigated using immunohistochemical analysis. Administration of AT prevented bone loss and the alternations of osteoporotic bone parameters at the distinct regions of the distal femur and spongiosa region in OVX mice. Further, administration of AT increased periosteal bone formation in a dose-dependent manner. Meanwhile, AT inhibited not only the expression of NFATc1 and c-Fos, which are two major regulators of osteoclastogenesis but also reduced bone resorbed encoding expression of MMP9 and RANKL. Our results indicated that administration of AT prevented bone loss and the alternations of osteoporotic bone parameters at the distinct regions of the distal femur and spongiosa region in OVX mice. Also AT has the bone protective effect through the suppression of osteoclast and promotion of osteoblast, suggesting that it could be a preventive and therapeutic candidate for anti-osteoporosis.

새로운 11β-HSD1 저해제인 KR-67500의 약물동태 (Pharmacokinetic Characterization of KR-67500, a Novel 11β-HSD1 Inhibitor)

  • KR-67500, trans-4-(2-(4-methyl-1,1-dioxido-6-(2,4,6-trichlorophenyl)-1,2,6-thiadiazinan-2-yl)acetamido)adamantane-1-carboxamide, is a novel $11{\beta}$-HSD1 inhibitor with its therapeutic effects of its anti-diabetic, anti-adipogenic and anti-osteoporotic activity. This study was performed to evaluate in vitro and in vivo pharmacokinetic properties of KR-67500 as a new drug candidate. KR-67500 was stable and highly bound to proteins in rat plasma. The microsomal stabilities of KR-67500 in human and rat liver were high. The inhibitory effect of KR-67500 for five cytochrome P450 enzymes was low. Preclinical pharmacokinetic studies have been carried out with intravenous or oral administrations of KR-67500 (10 mg/kg) to male rats and monkey. KR-67500 showed low clearance (0.68 l/h/kg) and high oral bioavailability (102%) in male rats. These results suggest that KR-67500 has good drug-like pharmacokinetic properties with a low first-pass effect and high bioavailability for an oral therapeutic agent of diabetes and osteoporosis.
