The Effect of Ethanol Extracts of Herba Prunellae, Flos Lonicerae and Radix Scutellaria on 6-OHDA Induced Oxidative Damage in the SH-SY5Y Cell Line (SH-SY5Y 세포주에서 하고초, 금은화, 황금 에탄올 추출물의 6-OHDA로 유도된 산화적 손상에 미치는 영향)

  • Objectives : Categorized as 'cheongyeol' herbs, Herba Prunellae, Flos Lonicerae and Radix Scutellaria have been proven to have effect on degenerative brain disease, cerebrovascular disease and brain tumor because of their anti inflammation, antioxidant, or anticancer effects. In this study, we studied activity against reactive oxygen species and anti inflammation effect of these three 'Cheongyeol' herbs. Methods : We measured each herb's yield of ethanol extracts, phenolic contents and activities against DPPH, hydroxyl radical and superoxide anion. Also through 6-hydroxydopamine (6-OHDA) induced oxidative damage in SH-SY5Y human neuroblastoma cell line, we measured antioxidant effect and NO activity of the three herbs. From the three herbs, we chose Prunella Herba, which showed the highest antioxidant effect, and studied its cell survival rate and anti inflammation effect through COX-2 and iNOS. Results : All three herbs showed significant results, and especially Prunella Herba showed significant effect on phenol contents, antioxidant effect on various active oxygen and antioxidant, and anti inflammation effect through cell line. Conclusions : Further study of the origin concept of 'cheongyeol' and research into specific mechanisms and role in treatment of cranial nerve disease, seems warranted.

A Study on the Effect of Electroacupuncture at Hyunjong (GB39) on Anti-Oxidation (현종에 시술한 저주파 전침자극의 항산화 효과에 대한 실험적 연구)

  • Objectives & Methods : The purpose of this study is to investigate the anti-oxidative effects of electroacupuncture at Hyunjong (GB39) in rats. The author performed several experimental items including change of body weight, relative liver weight, level of albumin, total bilirubin, LDL-Cholesterol, LDH, GOT and GPT in blood serum, level of SOD, glutathione, catalase, NO and MDA in liver, histological change of liver. Results : 1. In the GB39-EA group, body weights were decreased in the early stages, but increased later on gradually. 2. In the GB39-EA group, the level of LDL-cholesterol was significantly decreased in comparison with that of the holder group, sham-EA group and GB39-NR group. 3. In the GB39-EAgroup, liver SOD, glutathione, catalase activity were increased, and NO density was decreased in comparison with those of the holder group, sham-EA group and GB39-NR group. MDA activity was significantly decreased in comparison with that of the holder group. 4. In the GB39-EA group, the density of liver tissue was more similarly maintained to the normal group in comparison with that of the holder group and sham-EA group. 5. The results of the GB39-EA group showed similar tendency with those of the GB39-EA group, but the effects were lower than those of the GB39-EA group. Conclusions : These results suppose that electroacupuncture at GB39 has an anti oxidative effect in human.

Studies on the Anti-apoptotic Effect of the Mudanpi (목단피가 세포고사의 억제에 미치는 영향에 관한 연구)

  • Mudanpi (Cortex Moutan Radicis; the root cortex of Paeonia suffruticosa Andrews) is an important Chinese crude drug used in many oriental prescriptions. 1,2,3,4,6-Penta-O-galloyl-beta-D-glucose (PGG), a major component of this crude drug, has been shown to possess potent antioxidant, anti-mutagenic and anti-proliferative effects. In this study, I examined whether PGG could protect Neuro 2A cells, a kind of neuronal cell lines, from oxidative damage through the induction of HO-1 expression and HO activity. Exposure of Neuro 2A cells to PGG (10-50μM) resulted in a concentration- and time-dependent induction of HO-1 mRNA, and protein expressions and heme oxygenase activity. PGG protected the cells from hydrogen peroxide-induced cell death. The protective effect of PGG on hydrogen peroxide-induced cell death was abrogated by zinc protoporphyrin IX (ZnPP IX), a HO inhibitor. These results indicate that PGG is a potent inducer of HO-1 and HO-1 induction is responsible for the PGG-mediated cytoprotection against oxidative damage.

YJI-7 Suppresses ROS Production and Expression of Inflammatory Mediators via Modulation of p38MAPK and JNK Signaling in RAW 264.7 Macrophages

  • Chalcone, (2E)-1,3-Diphenylprop-2-en-1-one, and its synthetic derivatives are known to possess anti-oxidative and anti-inflammatory properties. In the present study, we prepared a novel synthetic chalcone compound, (E)-1-(4-hydroxyphenyl)-3-(2-(trifluoromethoxy)phenyl)prop-2-en-1-one name (YJI-7), and investigated its inhibitory effects on endotoxin-stimulated production of reactive oxygen species (ROS) and expression of inflammatory mediators in macrophages. We demonstrated that treatment of RAW 264.7 macrophages with YJI-7 significantly suppressed lipopolysaccharide (LPS)-stimulated ROS production. We also found that YJI-7 substantially decreased NADPH oxidase activity stimulated by LPS, indicating that YJI-7 regulates ROS production via modulation of NADPH oxidase in macrophages. Furthermore, YJI-7 strongly inhibited the expression of a number of inflammatory mediators in a gene-selective manner, suggesting that YJI-7 possesses potent anti-inflammatory properties, as well as anti-oxidative activity. In continuing experiments to investigate the mechanisms that could underlie such biological effects, we revealed that YJI-7 suppressed phosphorylation of p38MAPK and JNK stimulated by LPS, whereas no significant effect on ERK was observed. Furthermore, LPS-stimulated production of ROS, activation of NADPH oxidase and expression of inflammatory mediators were markedly suppressed by treatment with selective inhibitor of p38MAPK (SB203580) and JNK (SP600125). Taken together, these results demonstrated that YJI-7, a novel synthetic chalcone derivative, suppressed LPS-stimulated ROS production via modulation of NADPH oxidase and diminished expression of inflammatory mediators, at least in part, via down-regulation of p38MAPK and JNK signaling in macrophages.

Antioxidant and Anti-Melanogenic Activities of Hyssopus officinalis Extracts (히솝 추출물의 항산화 효과 및 멜라닌 생성 저해효과)

  • Hyssopus officinalis is a herbaceous plant of the genus Hyssopus. Due to its properties as an antiseptic, cough reliever and expectorant, it is commonly used as an aromatic herb and medicinal plant. This study was performed to investigate the anti-oxidative and anti-melanogenic properties of Hyssopus officinalis extracts (HE) using in vitro assays and cell culture systems. As a result, HE showed higher DPPH and ABTS radicals scavenging activity in a dose-dependent manner. Also, HE inhibited the prodution of intracellular ROS and melanin contents in B16F10 melanoma cell as well as tyrosinase activity. We also found that HE inhibit mRNA expression of MITF, tyrosinase and TRP-2 gene. These findings suggest that HE may be beneficial for preventing oxidative damage and melanogenesis of skin.

Anti-Oxidative Effect of Seungyangikki-tang Decoction in Spleen, Pancreas and Stomach Cells of SD Rats (승양익기탕(升陽益氣湯) 전탕액이 노화쥐의 비장, 췌장, 위장 세포의 항산화능에 미치는 영향)

Objectives: The purpose of this study is to investigate the anti-aging and anti-oxidative effects of Seungyangikki-tang decoction(SY) in spleen, pancreas and stomach cells of Sprague-Dawley(SD) rats. 2. Methods: This experiment was used by the tissue of spleen, pancreas and stomach cells of 6, 52 and 68 weeks old SD rats. We devided each group by three. One group as normal group was non-treated cells, another group as control group was saline-treated cells, and the other group as experimental group was SY treated cells. After culture for 48 hours, each groups measured the level of SOD, GSH, MDA and NO in the tissue of kidney, bladder and spleen cells. 3. Results and Conclusions: SOD activity was significantly increased in spleen cell of 6, 68w-SY group, pancreas cell of 52,68w-SY group and in stomach cell of 52w-SY group compared with those of the control groups. GSH concentration was significantly increased in spleen cell of 6,68w-SY group and in pancreas cell of 6w-SY group compared with those of the control groups. MDA concentration was significantly decreased in spleen cell of 68w-SY group and in stomach cell of 68w-SY group compared with those of the control groups. NO concentration was significantly decreased in spleen cell of 68w-SY group, stomach cell of 68w-SY group compared with those of the control groups.

Anti-apoptotic Effect of Steam Exploded Quercus variabilis

  • We hypothesized that the extract from steam exploded Q. variabilis might be cytoprotective for tenofibroblasts cells during oxidative stress. In the present study, the extracts obtained from steam exploded (severity log Ro 4.68) Q. variabilis contained high quantities of phenolics and flavonoids contents. Also, the extracts appear to have, on these tenofibroblasts, a protective effect against oxidative stress. Tenofibroblasts cells incubated with the extracts and stressed with $H_2O_2$ showed an increase in cell viability by MTT assay. The extracts is found to inhibit $H_2O_2$-induced apoptosis in tenofibroblasts cells, as shown by Annexin V/PI double staining analysis. Western blot data showed that in the extracts/$H_2O_2$-treated cells, the extracts inhibited the $H_2O_2$-dependent phosphorylation of ERK and p38. From these results, it is suggested that the extracts showed the protective effect on $H_2O_2$-mediated oxidative stress. The main chemical compounds of the extract was identified as 1,8-cineole by GC-MS analysis. The anti-apoptosis activity is accordingly believed to be attributable to the 1,8-cineole.

Antioxidant and anti-inflammatory effects of silymarin (실리마린의 항산화 및 항염증 효과)

  • We investigated the antioxidant and anti-inflammatory effects of silymarin. The antioxidant activity was evaluated using 1,1-diphenyl-1-picrylhydrazyl radical (DPPH) and 2,2'-azino-bis (3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) radicals scavenging assays. Silymarin scavenged 71% of DPPH radicals and 78% of ABTS radicals at a concentration of 1 mg/mL, respectively. Silymarin effectively inhibited the oxidative damage of DNA, and the oxidative modifications of human serum proteins and Cu,Zn-SOD. Also silymarin effectively inhibited H2O2- and LPS-induced cell death as well as the generation of reactive oxygen species and DNA fragmentation by H2O2 and LPS. The results suggested that silymarin might be an effective natural antioxidant and anti-inflammatory material.

Effect of SHT on the anti-oxidant activity and atopic dermatitis related inflammatory cytokines (석자해기탕(石紫解肌湯)이 아토피피부염 유관 인자인 산화적 손상과 염증 인자에 미치는 영향)

  • In order to investigate the possibility of SHT as therapeutic for the treatment of atopic dermatitis (AD), cytotoxicity, anti-oxidant activity, modulatory and suppression activities of SHT were tested. 90% or higher cell viability was observed in all tested groups from 25 to 200 ug/ml using Raw 264.7 cells. SHT showed dose-dependent DPPH scavenging activity, with more than 80% scavenging activities at 400 and 800 ug/ml concentrations. SHT showed dose-dependent suppression activity of ROS production, especially at 200 ug/ml of 57.4%. SHT decreased NO production activity dose dependently, expecially at 200 ug/ml of 28.8%. IL-$1{\beta}$, IL-6, MCP-1, TNF-${\alpha}$ production rate were decreased by 45.7%, 15.5%, 8.9%, 16.5% respectively when Raw 264.7 cells were treated with LPS and with SHT of 200 ug/ml. However, only IL-6 and TNF-${\alpha}$ showed significant changes. The results above indicate that SHT significantly reduces the effect of oxidative and inflammatory cytokines. The use of SHT in dermatitis can be widely suggested.

Resistance Activity of Kyung-Ok-Ko on Thermal Stress in C. elegans (경옥고(瓊玉膏)의 열 스트레스에 의한 피부노화 억제 활성)

  • Objectives : This study was conducted to reveal the scientific mechanism of the anti-skin aging activity of Kyung-Ok-Ko(KOK), which is highly useful as a Korean traditional medicine and functional food. Methods : The skin wrinkle and aging inhibitory activity of KOK was confirmed through in vitro experiments of human dermal fibroblast neonatal cell(HDFn) and in vivo of C. elegans, and hairless mouse(SKH-1). Results : The amount of the C-terminus of the collagen precursor in the HDFn cell culture medium treated with KOK using an enzymes-linked immunoassay kit. The group treated with KOK 200㎍/㎖ was a 28.3% increase of collagen precursor compared to the control group. KOK showed inhibitory activity of MMP-1 compared to the control group at a concentration of 200㎍/㎖. In addition, KOK 200㎍/㎖ showed significant inhibitory activity of thermal stress and an oxidative stress compared to the control group in C. elegans. Furthermore, KOK showed a concentration-dependent(100mg/kg and 500mg/kg) anti-wrinkle formation effect in UV-irradiated hairless mouse(SKH-1). Additionally, when KOK was administered to UV-irradiated hairless mice, an increase in procollagen -1 and -3 genes expression was observed, and mmp-1 and mmp-9 genes, which increase collagen decomposition, decreased with the administration of KOK. Conclusions : The skin aging inhibition mechanism of Kyung-Ok-Ko(KOK) is presumed to be achieved through suppressing thermal stress and oxidative stress, suppressing mmp-1 and mmp-9 genes, and increasing procollagen-1 and procollagen-3.
