Antioxidant and Antitumor Activities of Ethanol Extracts from Unhulled and Hulled Rice Hiami (Oryza sativa L. cv. Hiami) (하이아미의 정조 및 현미 추출물의 in vitro 항산화 및 항암활성)

  • To evaluate the antioxidant and antitumor potential of a newly bred rice, Hiami (Suweon-511), total polyphenol content, $\gamma$-oryzanol content, radical scavenging activities, and antitumor activities were measured (control: Ilpum). Total polyphenol contents were 6.31 and 3.75 mg/g for unhulled (UHR) and hulled rice (HR) of Hiami, which was higher than that of Ilpum (5.66 and 3.47 mg/g). The $\gamma$-oryzanol contents were 33.53 and 39.47 mg/100 g for UHR and HR of Hiami, which was higher than that of Ilpum (24.33 and 28.68 mg/100 g). DPPH radical, hydrogen peroxide, superoxide radical and hydroxyl radical scavenging activities of 70% ethanol extracts of Hiami were higher compared to Ilpum, and UHR was higher than those of HR. Antitumor activities of the 70% ethanol extracts of Hiami and Ilpum were increased with a dose-dependent manner. The extracts of Hiami have higher activities of antitumor activities on gastric and breast cancer cell lines compared to other cancer cell lines, and Ilpum has higher activities of antitumor activities on colon and liver cancer cell lines.

Chemical Components and Enzyme Activity of Hydroponic-cultured Ginseng Roots and Leaves under Different Heating Temperatures (열처리 온도에 따른 수경재배인삼의 화학성분과 효소활성)

  • This study investigated changes in the chemical components and enzyme activities from hydroponic-cultured ginseng roots (HGR) and leaves (HGL) with various heating temperatures (90, 110, 130, and $150^{\circ}C$) for 2 hours. The UV-absorbance and browning intensity of heated ginseng significantly increased with heating temperature. 5-HMF contents also significantly increased with increasing heating temperature. The free sugars (fructose, glucose, and sucrose) were detected and sucrose content decreased, but fructose and glucose content increased with increasing heating temperature. Malic, citric, lactic, and oxalic acid contents were 817.52, 722.25, 122.06, and 18.43 mg%, respectively, in HGR and 682.84, 338.21, 90.37, and 0 mg%, respectively, in HGL at $150^{\circ}C$. Tyrosinase and ACE inhibitory activities significantly increased with heating temperature. These results show that various components and activities of HGT and HGL significantly increase with heating temperature.

Cloning and sequence analysis of Wild Argali short palate, lung and nasal epithelium clone 1 cDNA

  • Objective: Experiments were conducted to clone the sequence of Wild Argali short palate, lung and nasal epithelium clone 1 (SPLUNC1) cDNA, and to lay the foundation for further study the biological function of Wild Argali SPLUNC1. Methods: The complete sequence of Wild Argali SPLUNC1 cDNA was generated by rapid amplification of cDNA ends. The entire coding sequence was inserted into the pPIC9K vector and expressed in Pichia pastoris (P. pastoris) GS115. The recombinant SPLUNC1 protein was detected by Western blot and purified by $Ni^{2+}$ chelate affinity chromatography. The test of effect of the protein on Mycoplasma ovipneumoniae (MO) was performed with real-time polymerase chain reaction. Results: The Wild Argali SPLUNC1 cDNA was 1,076 bp with an open reading frame of 768 bp, which encoded a 26.49 kDa protein composed of 255 amino acids. Its amino acid sequence shared 98.4%, 96.9%, 94.5%, 90.2%, 80.8%, 78.4%, 78.3%, 72.5%, 72.3%, 68.8% identity with those of SPLUNC1 cDNA from Ovis aries (accession no. NP_001288334.1), Capra hircus (accession no. XP_005688516.1), Pantholops hodgsonii (accession no. XP_005979709.1), Bos taurus (accession no. NP_776851.1), Felis catus (accession no. XP_006929910.1), Homo sapiens (accession no. NP_001230122.1), Sus scrofa (accession no. NP_001005727.1), Chinchilla lanigera (accession no. NP_001269294.1), Mus musculus (accession no. NP_035256.2), and Rattus norvegicus (accession no. NP_742028.1), respectively. The recombinant protein corresponded to the expected molecular mass of 25.47 kDa as judged by sodium dodecyl sulfate-polyacrylamide gel electrophoresis, and it was detected in the supernatant of P. pastoris, and it could be purified. The results from the test of inhibition effect of argali recombinant SPLUNC1 protein on MO showed that the product could inhibit MO very well (p<0.01). Conclusion: The amino acid sequence of Wild Argali SPLUNC1 was different from other organisms. The recombinant SPLUNC1 protein has good biological activity.

  • The purpose of this study came to start to be helpful in that the teachers in field produced evaluation subject about the mathematics and applied by analyzing the performance capacity for the students about the subject developed through the application of subject about the teaching-learning model that can utilize the learners' learning experience and the development of items that can evaluate it by utilizing various and concrete operational materials as an object of the first year at the middle schools. The concrete purpose of realizing it is as follows. 1. This study is to develop the teaching-learning model centered on activities adequate to utilize various and concrete operational materials. 2. This study is to develop evaluation data for utilizing the concrete operational materials by making construction through the application of developed teaching-learning model as possible. 3. This study is to present standards initiative for reliable and fair marking about the evaluation data and to analyze the students' practice capacity by using it effectively. For accomplishing the purpose of this research, this study is the work-oriented teaching and learning model using learning data that can be easily found around the surroundings and 8 evaluation programs in order that the experience-oriented learning based on circumstances learning among the learning models of constructivism. Also, it is to examine the result after applying that on a basis of 25 students, consisted of only on class in a year, in the first year at the small size of agricultural middle schools. The result of this study from this research is as follows. First, as a result of frequency survey, of response about the developed evaluation data it showed positive response more than 80% of all the items. The atmosphere of self-directed learning was produced because the developed evaluation data could induce activity-based learning naturally by stimulating the students' curiosity and promoting interest. Second, this study executed t-test for the result of questionnaire about the mathematics propensity, and there were significant differences in 19 items among 25 items. It presented that the application of data about the teaching-learning and evaluation directly using concrete operational materials to the constructional learning by level might have desirable affect on the students' mathematical propensity. It came to be a motive that could increase value recognition and interest on the subject of mathematics by investigating a mathematical principle rule and confirming it through activity learning using the concrete operational materials.

Cassava in Lactating Sow Diets: I. Effects on Milk Composition and Quality

  • The effect on sow milk of variable levels of cassava in lactating sow diets was analyzed in an attempt to explain the beneficial effects reported by producers of including cassava as a basal feed. Twenty crossbred lactating sows were randomly assigned to five dietary treatments. The treatments were: i) broken rice (BR) as the basal feed (BR100), ii) 50% of BR replaced with cassava chip meal (CCM) (CM50), iii) 75% of BR replaced with CCM (CM75), iv) CCM as the basal feed (CM100), and v) dried boiled cassava chips (CCB) as the basal feed (CB100). The hydrocyanide (HCN) content of CCB was reduced to be intermediate between HCN in the no cassava (BR100) and the 50% cassava (CB50) diets. Hydrocyanide was 0.54, 3.24, 4.41, 5.43 and 1.77 ppm in the BR100, CM50, CM75, CM100 and CB100 diets, respectively. Increasing cassava did not affect feed intake (p>0.05), but increased HCN intake (p<0.01). Milk composition was analyzed for protein, fat, lactose, solids not fat (SNF) and total solids (TS). Milk quality was analyzed for total microbes, coliform bacteria, thiocyanate ($SCN^-$), lactoperoxidase (LPO), and glutathione peroxidase (GPx) activity. At farrowing, sow milk composition was not affected by experimental diets (p>0.05), but milk $SCN^-$ increased as the intake of HCN increased in sows diets (p<0.01), $r^2$ = 0.96. At mid-lactation (day 14), milk composition was not affected (p>0.05). The milk quality levels of $SCN^-$ were 9.4, 10.3, 10.5, 11.6 and 9.1 ppm for the BR100, CM50, CM75, CM100 and CB100 diets, respectively (p = 0.01). The LPO contents were 16.41, 42.13, 51.42, 53.94 and 22.81 unit/L, respectively (p = 0.03). There was no GPx activity found in sow milk. When BR was replaced with cassava meal, total microbes and coliforms were reduced 78% and 87%, respectively, by the influences of HCN. The reported beneficial effects of cassava chip meal as a basal feed in lactating sow diets is manifested by improved performance of suckling pigs. This is due to beneficial, non-toxic levels of HCN in the diets. Besides passing HCN to suckling pigs in the form of $SCN^-$, sow milk may also benefit suckling pigs with the observed (day 14) increase in lactoperoxidase content and reduction in coliform bacteria.

Anti-tumor Activity of Fermented Liquid Opuntia humifusa in Cervical Cancer Cells and Its Chemical Composition (손바닥 선인장(Opuntia humifusa) 발효액의 화학적 성분과 자궁경부암 세포주에 대한 항암작용)

  • The purpose of this study is to investigate anti-tumor activities, general composition, elemental composition and mineral contents of fermented liquid stem, root and fruit of Opuntia humifusa. In the general composition, the energy, crude protein, crude lipid and crude carbohydrate contents of fermented liquid stem were 86.21 Kcal, 0.92$\%$, 0.12$\%$, and 20.34$\%$, respectively. Fermented liquid fruit showed 65.32 Kcal, 1.04$\%$, 0.08$\%$, and 15.15$\%$. In mineral analysis, fermented liquid stem and fruit showed 1,800 and 388 mg of calcium per 100g. The ferrous concentrations of fermented liquid stem and fruit were 21 and 10 mg per 100 g, respectively. Methanol, ethanol and water extracts of nonfermented liquid stem and fruit did not inhibit the proliferation in human cervical cancer cells (Caski, SiHa and HaCaT), but the fermented liquid fruit showed the inhibition of Proliferation with dose-response manner in Caski and SiHa cells, but not HaCaT. Therefore, it suggests that fermented cactus may be used as one of potential adjuvant for the treatment of cervical carcinomas.

Effect of Germination Time and Extrusion Temperature on Properties of Germinated Brown Rice (발아시간과 압출성형온도가 발아현미의 성질에 미치는 영향)

  • In order to develop a novel food utilization of germinated brown rice, physicochemical properties of germinated brown rice and its extrudates were investigated. The physical characteristics, the paste viscosity, and the anti-oxidation activity were analyzed. Brown rice was soaked for 10 hr and germinated for 24 and 48 hr at $30^{\circ}C$. Extrusion conditions of brown rice and germinated brown rice were barrel temperature at 100 and $120^{\circ}C$ and moisture content at 20%. The expansion ratio increased with the increase in germination time. It increased at barrel temperature of $100^{\circ}C$, and decreased at $120^{\circ}C$. The bulk density showed negative corporations with the expansion ratio. The paste viscosity of germinated brown rice was decreased with the increase in the germination time. However the paste viscosity of extrudates was lower as barrel temperature increased. Content of polyphenolic compound in extrudates was increased by increasing germination time and barrel temperature.

Nitrite scavenging activity and protective effect of the Puerariae Radix and green tea extract on lead acetate and cadmium-induced liver damage in mice (갈근 및 녹차추출물의 아질산염 소거작용 및 간독성 보호효과)

  • This study was designed to evaluated the nitrite scavenging activity and protective effect of the Puerariae Radix and green tea extract on lead acetate and cadmium-induced liver damage in mice. The quantitative analytical method for major antioxidants, isoflavones, puerarin, catechine and caffeine in galgun (Puerariae Radix) and green tera extract were established by HPLC. Contents of isoflavones, such as daidzin, genistin, daidzein and genistein were 4.23g/100g, 0.13g/100g, 0.07g/100g, and 0.03g/100g, and puerarin contents was 8.99g/100g, respectively. The total catechins and caffeine contents of green tea extract were 49.24g/100g and 6.53g/100g. The nitrite scavenging ability of galgun extract (pH 1.2, 100mg/ml) was 98.07% and it was higher than those of other extracts. It was the highest at the pH 1.2 and more than 64% in 25~100mg green tea extract, and was dependents on pH and concentration of the samples. The hepatoprotective effects of an aqueous extract from the root of gal gun and standard puerarin were evaluated against lead acetate and cadmium-induced liver damage in mice. Galgun extract and standard at a dose of 100mg/kg and 10mg/kg, 50mg/kg were administered orally once daily for successive 5 days and then a lead acetate and cadmium were orally at 3 hrs after the every day administration of galgun. The substantially elevated serum enzymatic activities of alanine and aspartate aminotransferase were due to lead acetate and cadmium treatment was dose dependently restored to the near normal level. In addition, galgun extract also significantly prevented the elevation of hepatic malon-dialdehyde formation in the liver of lead acetate and cadmium intoxicated mice in a dose-dependent manner. The results of this study clearly indicated that green tea and galgun extracts had nitrite scavenging ability and galgun extract had potent hepato-protective effects against lead acetate and cadmium-induced hepatic damage in mice and standard puerarin was also showed similar to the results of the galgun extracts.

Improvement of the Phosphate Solubilization Microorganism by the Introduction of Glucose Dehydrogenase Gene into Aeromonas hydrophila DA33. (Glucose dehydrogenase 유전자의 Aeromonas hydrophila DA33으로의 도입에 따른 인산가용화 균주의 개량)

  • Aeromonas hydrophila DA33 was isolated from cultivated soils as a bacteria having high abilities to solubilize inorganic phosphate. Glucose dehydrogenase gene (gdh) was cloned from Escherichia coli. The recombinant plasmid, pGHS containing glucose dehydrogenase gene was introduced into A. hydrophila DA33 in order to improve the activity of phosphate-solubilizing. The transformant harboring the gdh gene, A. hydrophila pGHS/DA33 increased enzyme activity. The strain also increased the gluconic acid generation that was effective for phosphate solubilization. It was possible that the strain containing pGHS produced higher solubilized phosphate with tri-calcium phosphate as the unique (P) source, in comparison with that of wild type without plasmid. These results suggest that the strain, A. hydrophila pGHS/DA33 is expected as effective biofertilizer for phosphate solubilization.

Cytotoxic Flavonoids from the Whole Plants of Chrysanthemum zawadskii Herbich var. latilobum Kitamura (구절초에서 분리한 Flavonoids의 인체암 세포주에 대한 세포독성 효과)

  • Two known flavonoids, luteolin (1) and acacetin (2) were isolated from a $CHCI_3$ soluble fraction of the whole plants of Chrysanthemum zawadskii Herbich var. latilobum Kitamura, and their structures were determined by NMR analysis. The luteolin (1) was isolated from this plant for the first time. These compounds were examined for their in vitro cytotoxic activities against four human cancer cell lines including HCT116 (colon), UO-31 (renal), PC-3 (prostate) and A549 (lung) by sulforhodamine B(SRB) assay. Acacetin (2) showed significant cytotoxic activity against HCT116 and UO-31 cells with an $IC_{50}$ of 2.44 and $2.89\;{\mu}g/ml$, respectively.
