Screening for Antitumor Efficacy from the medical plants in Korea and Japan (韓國과 日本産 抗腫瘍性 資源의 Screening에 대하여)

  • 21plants, which collected from Korea and Japan, were applied to antitumor and cytotoxic screening tests against sarcoma 180 a ascitec in mice. The results are summariged as follows : 1) The total packed cell volume method has been used for the antimeoplastic screening for from natural higher plants in Korea. By this method, we have found out that Selaginella involves, Patrinia hispida, Archyranthes japonica and Solanum nigrum having significant activity and also Cydonia sinensis and Rubia akane showed slight activity to antitumor 2) The total packed cell volune method has been used for the antineoplastic screening for from natural higher plants in Japan. Among the 21 tested plants, lsodon japonicus having strong antitumor activity and also Torilis japonica, Aralia elata, Leonurus sibiricus and Rubia cordifolia showed significant activity to anticancer tumor while Forsythia spp and Solanum nigrum showed slight activity to antitumor. 3) Among the 21 tested Korea plants, lsodon excisus and Forsythia Koreana showed strong antitumor activity by the V79 cytotoxic cell screening test.

Pristimerin Inhibits Breast Cancer Cell Migration by Up-regulating Regulator of G Protein Signaling 4 Expression

  • Background/Aim: Pristimerin isolated from Celastrus and Maytenus spp can inhibit proteasome activity. However, whether pristimerin can modulate cancer metastasis is unknown. Methods: The impacts of pristimerin on the purified and intracellular chymotrypsin proteasomal activity, the levels of regulator of G protein signaling 4 (RGS 4) expression and breast cancer cell lamellipodia formation, and the migration and invasion were determined by enzymatic, Western blot, immunofluorescent, and transwell assays, respectively. Results: We found that pristimerin inhibited human chymotrypsin proteasomal activity in MDA-MB-231 cells in a dose-dependent manner. Pristimerin also inhibited breast cancer cell lamellipodia formation, migration, and invasion in vitro by up-regulating RGS4 expression. Thus, knockdown of RGS4 attenuated pristimerin-mediated inhibition of breast cancer cell migration and invasion. Furthermore, pristimerin inhibited growth and invasion of implanted breast tumors in mice. Conclusion: Pristmerin inhibits proteasomal activity and increases the levels of RGS4, inhibiting the migration and invasion of breast cancer cells.

  • There are many cyclic peptides with diverse biological activities, such as antibacterial activity, immunosuppressive activity, and anti-tumor activity, and so on. Encouraged by natural cyclic peptides with biological activity, efforts have been made to develop cyclic peptides with both genetic and synthetic methods. The genetic methods include phage display, intein-based cyclic peptides, and mRNA display. The synthetic methods involve individual synthesis, parallel synthesis, as well as split-and-pool synthesis. Recent development of cyclic peptide library based on split-and-pool synthesis allows on-bead screening, in-solution screening, and microarray screening of cyclic peptides for biological activity. Cyclic peptides will be useful as receptor agonist/antagonist, RNA binding molecule, enzyme inhibitor and so on, and more cyclic peptides will emerge as therapeutic agents and biochemical tools.

Screening of New Antibiotics Inhibiting Bacterial Methionyl-tRNA Synthetase (세균의 Methionyl-tRNA Synthetase를 저해하는 새로운 항생물질의 스크리닝)

  • Aminoacyl tRNA synthetases of bacteria are known as potential targets for new anti-microbial agents. To isolate new inhibitors of bacterial methionyl-tRNA synthetases from natural sources, a new target-oriented screening system using whole cells which are over-expressing a target enzyme was developed. Approximately 8,000 culture broths of microorganisms from soils were tested by this screening system. Among them, ten culture broths was found to contain inhibitory activity against methionyl -tRNA synthetases of Escherichia coli. For the validation of the screening system, this new method was compared with in vitro enzymatic method. Seven out of 10 culture broths showed inhibitory activity against methionyl-tRNA synthetases of E. coli. This result showed that the new screening system was comparable to the enzyme assay. Thus we believe that our screening system as a new method can be applied for the screening of new antibiotics inhibiting bacterial methionyl-tRNA synthetases from natural products.

Factors Affecting Early Cancer Screening for Lung Cancer: Focusing on Lung Cancer Screening Subjects (폐암의 조기 암검진 여부에 미치는 요인: 폐암 검진 사업대상자를 중심으로)

  • Objectives: The purpose of this study is to examine the current status of cancer screening among subjects in the lung cancer screening cycle and to analyze the factors affecting the cancer screening of subjects in the lung cancer screening cycle. Methods: This study used the 'National Health and Nutrition Survey 7th Year (2017)' surveyed nationwide as the main data. The subjects are lung cancer screening projects, the dependent variable is early cancer screening, the independent variables are gender, age, marital status, household income level, education level, national health insurance type, private health insurance, The number of chronic diseases, general health examination, smoking status, drinking status, moderate intensity physical activity, stress perception rate, and weight control efforts were determined. Results: The results of this study showed that factors affecting early cancer screening of lung cancer screening subjects were gender, age, marital status, education level, national health insurance, smoking status, drinking status, moderate physical activity, and weight. Irrespective of the control effort, it was found that the private medical insurance, the number of chronic diseases, the medical examination, and the stress perception rate were affected. Conclusion: If the lung cancer screening subjects recognize the importance of early cancer screening themselves and create a social environment to increase their participation rate, lung cancer screening patients and their families will help them to live a healthy life.

Screening Method for Antihepatotoxic Activity Using $CCl_4-induced$ Cytotoxicity in Primary Cultured Rat Hepatocytes (일차 배양 흰쥐 간세포에서 $CCl_4$ 유발 세포독성을 이용한 간보호 효과 검색방법)

  • To devise an in vitro screening method for antihepatotoxic activity, $CCl_4-induced$ cytotoxicities in primary cultures rat hepatocytes were examined. When rat hepatocytes were intoxicated with 0.5, 1.0 or 1.5 mM $CCl_4$ for 1.5, 3 or 19hr, in order of LDH>GOT>GPT release form hepatocytes was increased in a dose-dependent manner. Treatment with 1.5 mM $CCl_4$ for 1.5 hr showed maximum increase in activity of LDH, GOT or GPT released in the medium compared with the control. At this experimental condition, well known antihepatotoxic substances, glycyrrhizin and silybin markedly inhibited $CCl_4-induced$ cytotoxicities. These results demonstrated that the screening method using $CCl_4-induced$ injury in primary cultured rat hepatocytes might be suitable in vitro assay for antihepatotoxic activity.

Effect of Prochloraz on Electrolytic Leakage and Spore Germination of Puccinia recondita Causing Wheat Leaf Rust

  • The effects of prochloraz on membrane permeability and germination of uredospores of Puccinia recondita were investigated to determine its potential mode of action on wheat leaf rust control activity. Disease control activity of ergosterol biosynthesis inhibitors (EBIs) and their activities on uredospore membrane permeability and germination were examined with wheat leaf rust pathogen, both in vitro and in vivo. While wheat leaf rust was not controlled by prochloraz, electrolytic leakage and spore germination of P. recondita uredospore was the highest with the use of prochloraz among the eight fungicides tested. Prochloraz stimulated uredospore of P. recondita to germinate at a higher ratio. Although certain EBIs, such as hexaconazole, showed excellent control activity, their effects on uredospore membrane permeability and germination was much inferior to prochloraz. Therefore, results of this study suggest that effects of EBIs on membrane permeability and germination of uredospore are not always correlated with their disease control activity.

  • The Antioxidant activity screening identification of five kind compounds in Artemisiae annuae herba with the on-line screening high performance liquid chromatography (HPLC) $ABTS^+$ assay. The various experimental variables such as the extraction time (h) and extraction solvent composition (%) of dipping method were investigated efficiently extraction at the room temperature $25^{\circ}C$. The results, the highest yield of total extract amount (0.458 g, 15.250%) was obtained by dipping method with 100% water and extraction time to 3 h. And the on-line screening HPLC-$ABTS^+$ assay method was rapid and efficient to search for bioactivity from natural products.

  • Purpose: The purpose of this study was to examine the factors affecting of health screening among persons with disabilities. Method: The study examines the factors affecting of health screening using over 20 years of age who participated in the 2017 National survey of the disabled data. Subjects were 6,332 person with disabilities over 20 years of age. Data were analyzed using descriptive statistics, X2-test, multiple logistic regression with the SPSS win 21.0 program. Results: Health screening among persons with disabilities is differentiated with education level, marital status, Subjective House economic status, Health insurance, Economic activity, Type of disability, Grade of disability, Stress recognition, Depressive symptom, Suicidal thinking, Suicidal attempt, and grade of disability. The significant predictors of the health screening were age, Subjective House economic status, Economic activity, and Grade of disability. Conclusion: We should consider age, disability level, and economic level when developing a screening program for people with disabilities. Especially, the development and promotion of the health screening program for persons with disabilities and related education are required.

  • For screening of the antiviral agent from soil, about 520 strains of microorganisms were evaluated for their antiviral activity, About 6.9% of strains showed more than 95% antiviral activity against Herpes Simples Virus (HSV)-1. Two strains among 30 strains active against HSV-1 virus showed a quite strong activity against human immunodeficiency virus.
