Isolation and Characterization of 4-hydroxycinnamic Acid with Antimicrobial Activity from Aralia eiata (두릅에서 항미생물활성을 갖는 4-hydroxycinnamic acid의 분리 및 동정)

  • The methanol extracts of AraIia elata showed antimicrobial activity against bacteria. The antimicrobial active principle was successively purified with solvent fractionation, silica gel adsorption column chromatography, Sephadex LH-20 column chromatography, silica gel partition column chromatography and HPLC. The active substance was isolated by HPLC on $C_{18}$ column with an acetic acid-MeOH system. The active substance was identified as trans-4-hydroxycinnamic acid by MS, $^1H-NMR\;and\;^{13}C-NMR$.

  • The Ballast Water Management Convention provides that ballast water treatment systems which make use of active substances shall be approved from IMO according to the procedure developed by the IMO. The Convention described that active substance means a substance or organism, including a virus or a fungus, that has a general or specific action on or against harmful aquatic organisms and pathogens. The Marine Environment Protection Committee of IMO gave basic approval to 13 ballast water management systems and final approval to 4 systems until October 2008. This paper considered the matter of procedure and documents of the basic and final approval based on the "Procedure for approval of ballast water management systems that make use of Active Substances (G9)" and "The Methodology for information gathering and the conduct of work of the GESAMP-BWWG" and summarized the specifications of the treatment systems which was granted the basic or final approval from IMO and raised several points.

  • This research was conducted to analyze the change of surface tension, viscosity, streaming current and conductivity of transformer oil when it were injected with the interface active substances.(anionic:S-111, cationic:S-121, amphoteric:S-131) The changes properties of the surface tension and viscosity of the oil which were injected with the interface active substances were divided into the changes area and the minimum reduction area. The surface tension and viscosity of the oil which were injected with three different kinds of interface active substances showed remarkable change at the point where the concentration of the substance in anionic, in cationic and in amphoteric were 100[ppm], 10[ppm] and 1[ppm] respectively. The streaming current and conductivity of the same sample oil were also changed at the same densities of the surface tension and viscosity. For this factor, it was possibile for us to interpret the mechanism of the streaming current and conductivity. Therefore the interface active substances of the three kinds were injected into the oil within the limit of optimal volume, prevention effects of electrification were showed more excellence than unmixed insulating oil.

Consideration on the Concentration of the Active Substances Produced by the Ballast Water Treatment System (선박평형수 처리장치의 활성물질 농도에 관한 고찰)

  • The International Maritime Organization (IMO) adopted the International Convention for the Control and Management of Ships' Ballast Water and Sediments in 2004 to prevent the transfer of aquatic organisms via ballast water. Thirty-four ballast water treatment systems were granted IMO active substance basic approval, among which twenty systems were granted final approval. This paper is an in-depth consideration of the mechanism principles of the treatment systems that received active substance basic or final approval from IMO, and on the concentration of Total Residual Oxidant (TRO). The TRO maximum allowable discharge concentration was reduced by neutralization equipment, resulting with a concentration lower than 0.2 ppm. However, between various treatment systems TRO maximum allowable dosage showed large differences, ranging from 1 to 15 ppm. The discrepancies of treatment allowable dosage concentration between different treatment systems are largely due to the properties of species and water conditions such as the temperature and turbidity, rather than the characteristics of treatment systems and the type or presence of filters etc.

STUDIES ON INSULIN-LIKE SUBSTANCE IN PANAX GINSENG

  • It was found that water extract of Panax ginseng strongly inhibited adrenaline-induced lipolysis in isolated fat cells of rat epididymal adipose tissue. An antilipolytic action of the water extract was easily inactivated by treatment with pronase, suggesting that the active principle might be a protein or a peptide. Experiments were designed to purify the antilipolytic substance, or insulin-like substance, of the water extract. The water extract was dialyzed against disti'led water. The outer dialysate was subjected to DEAE-cellulose column chromatography, gelfuaration on sephadex G-50 column, avicel cellulose column chromatography and phospho-cellulose column chromatography, successively. The finally purified substance gave one spot on thin layer chromatography. The molecular weight was found to be around 1000. Experiments are now in progress to elucidate the structure of this insulin-like peptide.

Purification and Characterization of an Antibacterial Substance from Aerococcus urinaeequi Strain HS36

  • A bacterial strain inhibiting the growth of Vibrio anguillarum, the causative agent of vibriosis, was isolated from fish intestines. The isolated strain HS36 was identified as Aerococcus urinaeequi based on the characteristics of the genus according to Bergey's Manual of Systematic Bacteriology and by 16S rRNA sequencing. The growth rate and antibacterial activity of strain HS36 in shaking culture were higher than those in static culture, while the optimal pH and temperature for antibacterial activity were 7.0 and 30℃, respectively. The active antibacterial substance was purified from a culture broth of A. urinaeequi HS36 by Sephadex G-75 gel chromatography, Sephadex G-25 gel chromatography, and reverse-phase high-performance liquid chromatography. Its molecular weight, as estimated by Tricine SDS-polyacrylamide gel electrophoresis, was approximately 1,000 Da. The antibacterial substance produced by strain HS36 was stable after incubation for 1 h at 100℃. Although its antibacterial activity was optimal at pH 6-8, activity was retained at a pH range from 2 to 11. The purified antibacterial substance was inactivated by proteinase K, papain, and β-amylase treatment. The newly purified antibacterial substance, classified as a class II bacteriocin, inhibited the growth of Klebsiella pneumoniae, Salmonella enterica, and Vibrio alginolyticus.

Anthelmintic Natural Products against Clonorchis sinensis and the Analogues (간디스토마에 작용하는 천연물 및 그 유사체)

  • Among 230 species of herbal drugs screened, thirty one show the anthelmintic activities in vitro against Clonorchis sinensis, the chinese liver fluke. The active substances have been isolated and their structures identified. Some of the active substances and their derivatives have been synthesized, followed by anthelmintic activity tests. The extracts from the active drugs cause damages in organs of the adult worm in the hepatic duct of rabbit. Some fresh water fish, which function as bad hosts for the fluke, excrete defense substances against the cercaria of the fluke. The defense substances have been isolated from Cyprinus carpio and Cyprinus carpio nudus, followed by structural identification. The results are summarized as follows: l) The bark of Machilus thunbergii as well as the seed of Schizandra chinensis contain meso-dihydro-guaiaretic acid as the anthelmintic component. Among derivatives synthesized, 4-phenyl-1-((3, 4-dihydroxyphenyl)-, 4-phenyl-1-(3-hydroxy-4-methoxy phenyl)-and 4-phenyl-1-(4-hydroxy-3-methoxy phenyl)-2, 3-dimethyl butanes show considerable activities. Administration of the bark extract mainly damages the bladder of the adult worm. 2) The active substance from the roasted fruit of Prunus mume is 2-hydroxymethylfurfural. This substance is produced during the roasting process. Administration of the fruitextract causes a damage of the bladder of the adult worm. 3) The active substance from the root of Scutellaria baicalensis is 5.2'-dihydroxy-6, 7, 8, 6'-tetramethoxyflavone. 4) Beside alantolactone, a very strong anthelmintic component is contained in the root of Inula helenium. Administration of the root extract causes irreversible damage on the worm, affecting mainly the reproductive organs. 5) The cercaricidal substances from the epidermis of C. carpio and C. carpio nudus are ethyl linoleate and linoleic acid, respertively. 6) The cercaricidal substances from various kinds of fresh water fish have different $R_f$ values, implying that the defense substances are species-specific. Unexpectedly, the fish with good host function, for example Pseudorasbora parva, excrete the defense substances, too. The defense substances are possibly organ-specific in individual species; the organs essenstial for the existence of the species excrete the defense substances, allowing other parts to be invaded by the cercaria. 7) The cercaricidal fraction of Carassius carassius is detected only in the fish which have been collected during the summer time from May to September, Its secretion is not dependent on water temperature. Thus, it seems to be possible that the secretion of the defense substance would be stimulated through a contact between the fish and cercaria.

  • Seaweed biotechnology is a multidisciplinary subject to produce food, pharmaceuticals, chemicals, and environmental remediation materials from seaweed resources. It uses various techniques of cell culture, enzyme reaction and genetic manipulation to increase the production efficiency of useful seaweeds or their products. Firstly, an overview of key topics will be introduced in the fields of seaweed tissue culture, strain improvement, genetic analysis briefly as basic techniques. Secondly, some biologically active substances such as anti-inflammatory and antifouling substances that have been screened in my laboratory will be focused.

A Study on the Fabrication of Porous Nickel Substrates Using Graphite Powder (흑연분말을 이용한 다공성 니켈지지체의 제조에 관한 연구)

  • A nickel mesh and an expanded nickel sheet were used as a current collector for supporting active materials of cathode in rechargeable batteries, while a porous nickel substrate was extensively studied because of its 3-dimensional structure which has high capabilities for active materials and current collection. Optimum coating conditions were studied by SEM and two step d. c. constant current electrolysis for the graphite coating and electro-plated nickel on an urethane substance which was highly porous and 3-dimensional structure. The density and the porosity of nickel support obtained by using two step current density and 80 ppi urethane substance were 0.38∼0.40 g /㎤ and 94∼96%, respectively. It was possible to fabricate a highly porous and good packable nickel substrate using two step current density and surfactants at sulfamic acid nickel plating bath.

Screening of Antioxidants to Human Low Density Lipoprotein (LDL) from Marine Resources (수산자원으로부터 Human Low Density Lipoprotein (LDL)에 대한 항산화제의 탐색)

  • This study was undertaken to evaluate antioxidative activities of substances isolated from marine resources against human low density lipoprotein (LDL). Methanol-water extract(80 : 20, v/v) of Sargassum ringgoldianum had the highest antioxidant activity and the active substance was purified by silica gel column chromatography by eluting chlorform : methanol mixture (80 : 20 v/v). The active fraction was seperated to several spots on the TLC in chlorofrom : methanol (10 : 1, v/v) mixture. Antioxidative activity of band 4 of fraction 2 on TLC was highest than that of $\alpha$-tocopherol against human LDL oxidation by the method of thiobarbituric acid reactive substance (TBARS). The band 4 of fraction 2 inhibited the copper mediated oxidation of human LDL with almost completely at 1 or 2 mg/ml.
