Sustained Production of Amino Acids by Immobilized Analogue- resistant Mutants of a Cyanobacterium Anacystis nidulans BD-1

  • Batch cultures of Anacystis nidulans BD-1 resistant to azaleucine and fluorotyrosine produced and liberated a wide range of amino acids, notably glutamic acid, alanine, phenylalanine, leucine, isoleucine, cysteine and methionine. Sustained liberation for prolonged periods was achieved after immobilization on calcium alginate and the net concentration in the medium was 0.18-0.2 g $I^{-1}$. While acetohydroxy acid synthase in azaleucine-resistant mutant lost leucine- and isoleucine-sensitivity, fluorotyrosine-resistant strain turned phenylalanine activating. The activities of nitrate assimilating enzymes were also higher in the mutants and were relaxed from ammonium-repression. The metabolic adjustments involved in amino acid overproduction are discussed.

  • The response properties of quartz crystal coated with stearic acid LB films to organic gases were investigated by measuring the shift of resonant frequency. Stearic acid was used as a sensing material and deposited on the surface of quartz crystal using the Langmuir-Blodgett(LB) method. The effect of temperature on the quartz crystal coated with stearic acid LB films was also investigated by Scanning Maxwell-stress Microscopy(SMM). As a result, the sensitivity of the quartz crystal coated with LB films to organic gases is dependent on temperature, thickness of LB film and molecular weight of organic gas.

  • In order to improve the sensitivity of the current assay methods of selenium in dried-yeast preparations, atomic absorption spectrophotometry (AAS), high performance liquid chromatography (HPLC) and UV-Vis spectrophotometry were employed. The sample was prepared with the digestion by acid mixture of hydrochloric acid, nitric acid and perchloric acid after elimination of ether-soluble substances. The range of quantitation of selenium was $1.0{\sim}6.0\;{\mu}g/ml$ by UV-Vis spectrophotometry, $5.0{\sim}20.0\;{\mu}g/ml$ by HPLC and $0.03{\sim}0.10\;{\mu}g/ml$ by AAS.

  • Glycans released from ovalbumin by PNGase F were analyzed by matrix-assisted laser desorption/ionization time-of-flight (MALDI-TOF) mass spectrometry using three different dihydroxybenzoic acid (DHB) matrix systems: 2,5-DHB, 2,6-DHB, and a 2,5-DHB/2,6-DHB binary matrix. Relative to the results obtained with the single-component matrices (2,5-DHB or 2,6-DHB), the 2,5-DHB/2,6-DHB binary matrix boasted lower background noise and higher sensitivity. A total of 16 glycan peaks were observed using the 2,5-DHB/2,6-DHB binary matrix, while only 10 and 9 glycan peaks were observed using the 2,5-DHB and 2,6-DHB matrices, respectively.

  • Five acid hydrazides as precolumn fluorescence derivatization reagents for carboxylic acids in HPLC, which have the benzofuran or benzothiazole moiety conjugated to a furan, thiophene or oxazoline ring, were synthesized and examined in view of reactivity, separability and sensitivity. Of these hydrazides, 2-(5-hydrazinocarbonyl-2-oxazolyl)-5,6-dimethoxybenzothiazole (BTOH) was most favorable. The detection limit of lauric acid as a model acid was 0.1 pmol per $10-{\mu}l$ injection volume at S/N=3, which was roughly equal to that of an analogous compound, 2-(5-hydrazinocarbonyl-2-furyl)-5,6-dimethoxybenzothiazole. The reagent allowed rapid assays of carboxylic acids ($C_{12:0}-C_{20:4}$) within 20 min with satisfactory scparability. The method was applied to the determination of fatty acids in human sera from healthy volunteers as well as from patients with diabetes or thyroid dysfunction.

  • The membrane fatty acid composition of tumor cell can be modified either in cell by altering the lipid composition of the medium of during growth in animals by changing the dietaty fat composition. These modifications are associated with changes in membrane physical properties and certain cellular functions, including carrier-mediated transport and enzyme contained within the membrane. Such effects influence the transport of nutrients and chemotherapeutic agents in cancer cells .Fatty acid modification also can enhance the sensitivity of the neoplastic cell to chemotherapy. The alteration in plasma membrane composition will be affected through dietary supplementations and the potential value to cancer patients could be a better understanding of the effects of diet on responsiveness of neoplasms to chemotherapy, i.e. cancer patients' chances for a "cure" can be improved by diet changes prior to treatment.

  • High performance amino acid analyzing method has been developed for the routine analysis of taurine in preparations. Ion-exchange resin #2619 Hitachi Custom Ion-Exchange Resin, $2.6(I.D.){\times}150$(length)mm was used as column, buffer I, pH 3.3 as mobile phase. The retention time of taurine was 7 minutes. Calibration curve by peak height for standard taurine was linear from 2.5ppm to 25ppm. The reproducibility showed relative standard deviation $\pm$1.9% when analyzed 10 times for standard solution. The samples could be continuously analyzed without regenerating the resin between samples. Five samples were applied to column every 12 min. and then the resin was regenerated for 30 min. during one analyzing cycle time, 90 min. The automatic amino acid analyzer has made it possible to assay multiple samples in a relatively short period of time using the analytical magnetic program card. The high sensitivity and specificity of the analytical column of the automatic amino acid analyzer permits the routine analysis of taurine in preparations.

  • This technology is a water leak repair technology using composite materials of concrete structures that block leakage of structures by injecting rapid microcement into the face of underground concrete to block water and injecting flexible glycidylacrylate. Rapid micro cement system repair materials are mixed with fine fibers to improve the flexural sensitivity of the material and to form a layer that blocks stabilized water at the back of the structure by allowing rapid and tight spatial filling during injection with high cohesion The glycidylacrylate repair material can control the expansion rate, and the external stress also has the characteristic that the form of the material is not destroyed or separated, which can also be applied to vibrating induced structures that produce repetitive fatigue loads, and has an effective durability in saline, alkali, acid (chloric acid, sulfuric acid, nitric acid).

  • To synthesize a new positive photosensitive polyimide precursor, parts of carboxylic acid groups in poly (amic acid) were esterified with 4,5-dimethoxy-2-nitrobenzyl bromide in the presence of K$_2$CO$_3$/HMPA followed by the chemical imidization of residual carboxylic acid units. The chemical structure of resulting polymer was characterized by $^1$H-NMR, UV/vis and FT-IR spectroscopic methods, and its thermal properties were examined by DSC and TGA. Upon UV irradiation, 4,5-dimethoxy-2-nitrobenzyl moiety underwent the photodegradation. As a result, the polymer became soluble in alkaline developer due to the formation of carboxylic acid moiety, which was used to make a micron-sized positive pattern. Sensitivity curves were obtained from the gel fraction experiments with respect to the various 4,5-dimethoxy-2-nitrobenzyl ester contents. From those curves, the sensitivity was ranged iron 4000 to 6000 mJ/㎠, and the contrast was measured to be from 3.1 to 4.9.

The Effect of Using Image Media for Environmental Education on Students' Environmental Sensitivity (환경교육 영상매체 활용 수업이 환경 감수성에 미치는 영향)

  • Today, environmental pollution and destruction due to the industrialization of our modem society have been the global issues. Although now we can lead affluent lives through science technologies and economic development, these environmental problems have resulted in striking the harmonious balance between human beings and nature and threatened human lives with abnormal weather caused by the global warming, destruction of the ozone layer, and El Nino phenomenon, acid rain, decrease in species diversity, movement of hazardous materials, and harmful waste increase. We are aware of the importance of environmental education, but in reality, it seems impossible to implement appropriate environmental education on account of our educational climate which exclusively focuses on the entrance examination. However, environmental education is the most ultimate solution for those problems in that only when students understand our environment fully and grow habits to protect it through environmental education, the present environmental problems can be solved and more serious problems that can be resulted in the future can possibly be prevented. Thus, this study has examined the effect of using image media in the environment subject on students' environmental sensitivity. According to the results, it was shown that it had positive effects on 'sensitivity of environment', 'attitudes towards environment', and 'environmental affinitive behaviors'.
