Social Capital and Parental Stress of Married Mothers with Young Children: Variations by Employment Status (영유아기 기혼 취업모와 비취업모의 사회자본과 양육스트레스)

  • This study attempted to examine whether there is a relationship between parental stress and the accessible and mobilizable social capital of mothers raising young children by the mothers' employment status. The sample included 284 employed and 287 non-employed mothers who have children younger than the age of 5. Three research questions were as follows: 1) do the accessible and mobilizable social capital of mothers and parental stress differ by the mothers' employment status? 2) are the accessible and mobilizable social capital of employed mothers related to the level of parental stress? And finally, 3) are the accessible and mobilizable social capital of unemployed mothers related to the level of parental stress? The results showed that no significant difference was found in accessing social capital by the employment status of mothers. However, employed mothers were capable of mobilizing more social capital for childcare assistance than non-employed mothers. The parental stress of employed mothers was significantly lower than that of non-employed mothers. For employed mothers, their mobilizable social capital contributed to the reduction of parental stress. For non-employed mothers, their capability of accessing social capital significantly predicted lower levels of parental stress. These results indicate that the concept of social capital could be taken into consideration when explaining the parental stress of married mothers raising young children.

A Study on Preference of Wheelchair Users to Toilet Approach Way - Focused on Wheelchair Users' Disability Types and Their Wheelchair Types (휠체어 사용자의 대변기 접근 방식 선호도 분석에 관한 연구 - 휠체어 사용자의 장애 유형 및 휠체어 종류를 중심으로)

  • Purpose: The purpose of this study is to identify the difference in toilet approach way by wheelchair users according to the direction of the door and to suggest the layout of the toilet rooms most accessible to the wheelchair users. Methods: Survey to wheelchair users about approach to toilet have been conducted for the data collection. 90 answers have been analyzed statistically by SPSS 19.0 program. Results: The conclusions of this study are as follows. Firstly, spinal cord disorders prefer diagonal approach and lateral approach to the toilet, but brain lesions prefer diagonal approach and frontal approach to toilet. Secondly, the manual wheelchair users prefer the diagonal approach and the lateral approach to the toilet, and the electric wheelchair users has a different approach way to the toillet depending on the direction of the door. Finally, the layout of accessible toilets have to be designed for the diagonal approach to toilet, which is most preferred by wheelchair users. Implications: The toilet approach varies according to the disability types and the wheelchair types. Therefore, the size of accessible toilets should be larger than now considering the access of variety wheelchair users.

NEUROD1 Intrinsically Initiates Differentiation of Induced Pluripotent Stem Cells into Neural Progenitor Cells

  • Cell type specification is a delicate biological event in which every step is under tight regulation. From a molecular point of view, cell fate commitment begins with chromatin alteration, which kickstarts lineage-determining factors to initiate a series of genes required for cell specification. Several important neuronal differentiation factors have been identified from ectopic over-expression studies. However, there is scarce information on which DNA regions are modified during induced pluripotent stem cell (iPSC) to neuronal progenitor cell (NPC) differentiation, the cis regulatory factors that attach to these accessible regions, or the genes that are initially expressed. In this study, we identified the DNA accessible regions of iPSCs and NPCs via the Assay for Transposase-Accessible Chromatin sequencing (ATAC-seq). We identified which chromatin regions were modified after neuronal differentiation and found that the enhancer regions had more active histone modification changes than the promoters. Through motif enrichment analysis, we found that NEUROD1 controls iPSC differentiation to NPC by binding to the accessible regions of enhancers in cooperation with other factors such as the Hox proteins. Finally, by using Hi-C data, we categorized the genes that directly interacted with the enhancers under the control of NEUROD1 during iPSC to NPC differentiation.

Enhanced Partitioning of Proteins in Metal-Affinity Aqueous Two-Phase Systems (금속 친화성 액 이상분계 시스템에서 단백질의 분배 향상)

  • In metal-affinify aqueous two-phase systems, protein partitioning is affected by a variety of parameters such as pH, the number of surface-accessible histidines, and the amount and partition coefficient of metallated polythylene glyco(PEG) ligand. To enhance partitioning of proteins with surface-accessible histidines, we have synthesized and used a (Cu(II)-ininodiacetic acid)$_2$-PEG20,000($Cu(II)_2IDA_2$-PEG20,000) as well as Cu(II)IDA-PEG5,000 as an affinity ligand. The partition coefficient of $Cu(II)_2-IDA_2$-PEG20,000 in a PEG5,000/dextran two-phase system was 30.1, which corresponded to a 3.8-fold increase over that of Cu(II)IDA-PEG5,000. The partitioning experiments were performed on four proteins, horse cytochrome c, S. cerevisiae cytochrome c, horse myoglobin, and sheep myoglobin. Partitioning of proteins which convey surface-accessible histidines was enhanced dramatically by the addition of $Cu(II)_2IDA_2$-PEG20,000 ligand. These results demonstrate that enhanced partitioning of metal-binding proteins in an aqueous two -phase system can by achieved by using an appropriate metallated PEG ligand.

Ribozyme-Mediated Replacement of p53 RNA by Targeted Trans-Splicing

  • In more than half of human tumors, the p53 tumor suppressor gene is mutated. Thus, restoration of wild-type p53 activity by repair of mutant RNA could be a potentially promissing approach to cancer treatment. To explore the potential use of RNA repair for cancer therapy, trans-splicing group I ribozymes were developed that could replace mutant p53 RNA with RNA sequence attached to the 3'end of ribozymes. By employing a mapping library of ribozymes, we first determined which regions of the p53 RNA are accessible to ribozymes, and found that the leader sequences upstream of the AUG start codon appeared to be particularly accessible. Next, trans-splicing ribozymes were generated that specifically recognized the sequences around these accessible regions. Subsequently, the ribozymes reacted with and altered the p53 transcripts by transferring a 3'exon tag sequence onto the targeted p53 RNA with high fidelity. Thus, these ribozymes could be utilized to repair mutant p53 in tumors, which would revert the neoplastic phenotype.

  • Accidents involving wheelchair accessible vehicles have been frequently occurring since the introduction of these vehicles in the Korean market. However, detailed regulations, which are required to ensure the safety of the wheel-chair occupants, are unavailable. In this study, both domestic and international vehicle safety regulations are analyzed in order to select the regulations that are similar to the transportation environment of Korea. Sled tests with an actual wheel-chair accessible vehicle were carried out based on the analyzed regulation requirements, as well as the values of the HIC, belt loads, dummy movements, and wheelchair movements. The test results showed that the movements of the dummy and the wheelchair did not meet the criteria of the regulation due to the improper positioning of the restraint systems.

  • This study proposes a classification method based on an automated training extraction procedure that may be used with very high resolution (VHR) images of non-accessible areas. The proposed method overcomes the problem of scale difference between VHR images and geographic information system (GIS) data through filtering and use of a Landsat image. In order to automate maximum likelihood classification (MLC), GIS data were used as an input to the MLC of a Landsat image, and a binary edge and a normalized difference vegetation index (NDVI) were used to increase the purity of the training samples. We identified the thresholds of an NDVI and binary edge appropriate to obtain pure samples of each class. The proposed method was then applied to QuickBird and SPOT-5 images. In order to validate the method, visual interpretation and quantitative assessment of the results were compared with products of a manual method. The results showed that the proposed method could classify VHR images and efficiently update GIS data.

  • Recently, there have been active research efforts on devising efficient routing algorithms utilizing location information in ad-hoc mobile networks. In this paper, we propose a new method to improve the performance of the widely recognized Location Aided Routing (LAR) algorithm. To this end, we suggest the concept of accessible zone combined with the basic two schemes associated with LAR algorithm. Through the proposed algorithm, we can confine the flooding area within accessible zone, thereby reducing the routing delay.

  • The advent of the assay for transposase-accessible chromatin using sequencing (ATAC-seq) has shown great potential as a leading method for analyzing the genome-wide profiling of chromatin accessibility. A comprehensive reference to the ATAC-seq dataset for disease progression is important for understanding the regulatory specificity caused by genetic or epigenetic changes. In this study, we present a genome-wide chromatin accessibility profile of 44 liver samples spanning the full histological spectrum of nonalcoholic fatty liver disease (NAFLD). We analyzed the ATAC-seq signal enrichment, fragment size distribution, and correlation coefficients according to the histological severity of NAFLD (healthy control vs steatosis vs fibrotic nonalcoholic steatohepatitis), demonstrating the high quality of the dataset. Consequently, 112,303 merged regions (genomic regions containing one or multiple overlapping peak regions) were identified. Additionally, we found differentially accessible regions (DARs) and performed transcription factor binding motif enrichment analysis and de novo motif analysis to determine new biomarker candidates. These data revealed the gene-regulatory interactions and noncoding factors that can affect NAFLD progression. In summary, our study provides a valuable resource for the human epigenome by applying an advanced approach to facilitate diagnosis and treatment by understanding the non-coding genome of NAFLD.
