Matrix Development in Cast Iron by Addition of Mischmetal Hydrides (주철(鑄鐵)의 기지조직(基地組織)에 미치는 Mischmetal Hydride의 첨가효과(添加效果))

  • In this study we have investigated the effects of Mm, MmH and $MmH_2$ on the matrix development in cast iron, The conclusive summary is as follows: The spheroidal graphite was observed when 0.5wt.% or more of mischmetal was added and the matrix was of ledeburite structure, but bull's eye structure was not observed. On the other hand, the bull's eye structure was observed when 0.25wt.% of MmH, or 0.25wt.% to 0.5wt.% of $MmH_2$ was added. Above limit of the additives, the matrix changed into ledeburite structure. As the hydrogen content of mischmetal compound increased from MmH, the range of additives to obtain bull's eye structure expanded. This reveals the significant effect of mischmetal hydride on matrix development in cast iron and the possibility of practical use of the additives.

  • A 2-month-old, intact female white Bull Terrier presented because of suspected deafness. The coat color was predominantly white and the iris color, of both eyes, was brown. The dog did not respond to the owner's voice when the sound stimuli were presented outside of the visual field; however, the dog responded to visual gestures. The other physical, neurological, otoscopic, radiographic, and blood examinations were unremarkable. To assess the apparent deafness, brainstem auditory evoked responses (BAER) were recorded and analyzed in the dog with suspected deafness as well as a normal littermate. The response in the normal littermate consisted of a series of five wave peaks (I-V) with decreased amplitude and prolonged latency as the stimulus intensity decreased. The BAER from the dog suspected of deafness appeared as a flat line and did not reveal identifiable peaks that corresponded to those found in the normal littermate. Thus, congenital, sensorineural and bilateral deafness was confirmed by the BAER.

Change of Sperm Viability and Acrosome Integrity of Post-thawed Korean Jeju Black Bull Spermatozoa according to Glycerol Concentration (제주 흑우 동결 정액 제조에 있어 Glycerol의 농도에 따른 생존율 및 정자 첨체 양상의 변화)

  • This study was carried out to establish most suitable freezing condition, to evaluate the different glycerol concentration of freezing and thawing rates on motility, viability, membrane integrity and acrosome intecrity of frozen Korean Jeju Black Bull spermatozoa, Semen was collected from a Korean Jeju Black Bull using an artificial vagina and transported to the laboratory. The semen was extended gradually 1:5 then cooled slowly for 2 hrs to 4$^{\circ}C$. The semen was diluted 1:1 with cryoprotectant extenders (3%, 5% and 7% glycerol) and equilibrated for 2 hrs at cold chamber and packed to 0.5 ml straws. The semen straws were located above 3 cm of liquid nitrogen for 5 minutes, above 5 cm for 10 min and above 8 cm for 10 min. And then the frozen straw was plunged into LN$_2$. The presented straws were examined the viability and motility after thawed at 37$^{\circ}C$ water bath. The viability and membrane integrity immediately post-thawing were significantly higher in samples frozen in 7% glycerol than 3% and 5% glycerol (p<0.05). After CTC staining to assess acrosome integrity, F pattern was significantly increased, but B pattern was significantly decreased in 7% glycerol (p<0.05). Freezing distance of 5 cm from liquid nitrogen and pre-cooling for 10 min yield better survival and membrane integrity, but not significant difference. However, AR pattern according to CTC staining was significantly decreased in 3 cm for 5 min.

  • This study was performed, by Finite Element Method, to evaluate the stress distribution on the periodontal tissue according to activation of the various closing loops and to predict the pattern of movement of maxillary incisors. At the same time, bull loop, key-hole loop, T-loop, combination loop and asymmetrical T-loop which were used for retraction of maxillary incisors was analysed by Finite Element Method. The following results were obtained 1. Horizontal force was the greatest in bull loop, the followed by key-hole loop, combination loop, T-loop and initial tooth movement exhibited uncontrolled tipping. 2. Horizontal force in asymmetrical T-loop compared to other closing loops was remarkably decreased, and the intrusive force on the incisors occurred. 3. As torque was increased, the moment was increased as a linear increment. 4. As moment was increased, initial movement of tooth changed to root movement from uncontrolled tipping.

FACTORS INFLUENCING FEED EFFICIENCY AND BACKFAT THICKNESS IN STATION TESTED BEEF BULLS

  • Records taken on 372 young beef bulls tested at the Ellerslie Bull Test Station, Alberta, Canada from November 1981 to April 1987 were analyzed to quantify the effects of age of dam, on-test age, on-test liveweight and herd of origin of bull on feed efficiency (feed/gain, kg/kg) in the test period (n = 231) and ultrasonic measurement of bakcfat thickness (mm) at the end of the test (n = 372). The reduction in $R^2$ due to each influencing factor (i.e. the variation accounted for by the factor) was used to indicate the importance of the influencing factor. Age of dam and on-test age of bull were not important factors on feed/gain and ultrasonic backfat thickness, as they accounted for less than 0.5% of the variation in feed/gain and ultrasonic backfat thickness, respectively (p > 0.1). On-test liveweight had some influence on feed/gain and ultrasonic backfat thickness, accounting for 3.5% (p < 0.01) and 0.4% (p < 0.05) of the total variation, respectively. The regression coefficients of feed/gain and ultrasonic backfat thickness on on-test liveweight were 0.016 (kg/kg)/kg and .013 mm/kg, respectively, both being significant (p < 0.05), indicating that lighter bulls entering the test were generally more efficient in feed utilization in the test period and had less backfat at the end of the test than heavier entering bulls. Herd of origin of bull accounted for a substantial amount of the total variation (> 16%) in feed/gain and ultrasonic backfat thickness (p = 0.08), indicating that a prolonged aqjustment period was needed to reduce the influence of herd of origin when assessing aggregate genetic merit of beef bulls for growth rate, feed efficiency and lean meat production using a central station performance testing program.

Effect of Ethylene Glycol and Antioxidant Combination on Function of Frozen-thawed Spermatozoa in Korean Jeju Black Bull (Ethylene glycol과 항산화제가 제주흑우 동결정액에 미치는 영향)

  • We investigated the effect of ethylene glycol and antioxidants such as taurine, hypotaurine and trehalose with extenders during cryopreservation of Korean Jeju Black Bull spermatozoa. The cryopreservation of freshly collected spermatozoa was conducted with four different conditions. As a control, spermatozoa were cryopreserved with Tris egg-yolk extenders added 5% ethylene glycol (EG). Taurine (20 mM), hypotaurine (20 mM) and trehalose (20 mM) were individually added into tris egg-yolk extenders with 5% EG. After thawing of frozen spermatozoa with four different conditions, sperm viability, motility, acrosomal integrity, and membrane integrity were investigated. The significant (p < 0.05) improvement of sperm viability showed in all antioxidant treated thawed spermatozoa (taurine; $68.1%{\pm}4.4$, hypotaurine; $69.2%{\pm}6.7$ and trehalose; $68.0%{\pm}4.4$) when compared to control ($63.4%{\pm}5.6$). Neither positive nor detrimental effects of three antioxidants were shown sperm motility after thawing. The results of hypo-osmotic swelling test showed that the membrane integrity of taurine, hypotaurine or trehalose treated thawed spermatozoa ($64.1%{\pm}5.4$, $61.5%{\pm}3.7$ and $59.0%{\pm}4.0$, respectively) had significantly (p < 0.05) higher rate of the swollen sperm compared to control ($53.7%{\pm}9.7$). Hypotaurine treated frozen-thawed spermatozoa had siginificantly higher (p < 0.05) F pattern ratio than taurine, trehalose and control treated frozen-thawed spermatozoa. Trehalose added frozen-thawed spermatozoa had significantly higher (p < 0.05) acrosome reaction pattern ratio than taurine and hypotaurine added frozen-thawd spermatozoa. In this study, we found that antioxidants such taurine, hypotaurine and trehalose treatments during cryopreservation process could reduce damage of spermatozoa of Korean Jeju Black Bull and improved sperm capability of fertilization.

Effect of Low Density Lipoprotein (LDL) on Motility, Viability, Membrane Integrity and Acrosome Integrity of Frozen-thawed Sperm in Korean Jeju Black Bull (제주흑우 동결정액 제조 시 Low Density Lipoprotein (LDL)의 첨가가 동결 융해 후 정자의 성상에 미치는 영향)

  • This study was designed to determine whether low-density lipoporoteins (LDL) extracted from egg yolk in extender improve the function of Korean Jeju Black Bull semen. The semen was cryopreserved with 5% ethylene glycol (EG) or 7% glycerol (G) extenders containing 10% egg yolk (EY), 4% LDL and 5% EY or 8% LDL. Frozen-thawed sperm were evaluated sperm motility, viability, membrane integrity and acrosome integrity. Post-thawed sperm motility has been significantly higher (p<0.05) in 4% LDL + 5% EY ($69.00%{\pm}4.18$; EG and $63.00%{\pm}9.75$; 7% G) than 8% LDL ($57.00%{\pm}5.70$; EG and $52.00%{\pm}4.47$;G). Treatment of 4% LDL + 5% EY-EG ($66.85%{\pm}5.06$) has been significantly improved sperm viability compared to other treatments except 10% EY - EG. Moreover, in membrane integrity, swollen sperm ratio has been only significantly increased (p<0.05) in 4% LDL + 5% EY - EG ($64.65%{\pm}6.10$) among all treatments. In assess to detect acrosome integrity, especially, AR pattern ratio has been significantly decreased (p<0.05) in 4% LDL + 5% EY - EG among all treatments. In sperm viability as time passes, between 4% LDL + 5% EY and 10% EY, there was no significant difference, but 8% LDL was significantly decreased sperm viability in EG (1 and 2 hrs) and G (30 min, 1, 2, 5 and 12 hrs) extender. However, there were no significant differences among all treatments except 8% LDL-G in sperm membrane integrity. 8% LDL-G has been significantly decreased swollen sperm ratio at 5 hrs after thawed. It is concluded from these results that 4% LDL + 5% EY to the freezing extender showed more positive effect on the frozen-thawed spermatozoa in Korean Jeju Black bull.

The Effects of Licorice Fraction and Glycyrrhizin on Prostaglandin Synthetase Activity of Bull Seminal Vesicle (감초의 분획과 Glycyrrhizin이 황소정랑의 Prostaglandin Synthetase활성에 미치는 효과)

  • The investigation aimed to study the effects of methanol fraction of licorice (FM 100) and glycyrrhizin on prostaglandin synthetase activity, in relation to their analgesic effects. Effects of FM 100 and glycyrrhizin on the activity of prostaglandin synthetase extracted from bull seminal vesicles were examined by the modified method of Takeguchi et al. The analgesic effect of FM 100 was tested in mice by the acetic acid writhing method. FM 100 was administered orally to mice. BSV prostaglandin synthetase activity was inhibited significantly by FM 100 in a dose-dependent manner, whereas the activity was slightly inhibited by glycyrrhizin. Statistically significant analgesic effects were also observed with FM 100. The results suggest that analgesic effect of licorice may be due to the inhibition of prostaglandin synthesis.

Genomic selection through single-step genomic best linear unbiased prediction improves the accuracy of evaluation in Hanwoo cattle

  • Objective: Genomic selection (GS) is becoming popular in animals' genetic development. We, therefore, investigated the single-step genomic best linear unbiased prediction (ssGBLUP) as tool for GS, and compared its efficacy with the traditional pedigree BLUP (pedBLUP) method. Methods: A total of 9,952 males born between 1997 and 2018 under Hanwoo proven-bull selection program was studied. We analyzed body weight at 12 months and carcass weight (kg), backfat thickness, eye muscle area, and marbling score traits. About 7,387 bulls were genotyped using Illumina 50K BeadChip Arrays. Multiple-trait animal model analyses were performed using BLUPF90 software programs. Breeding value accuracy was calculated using two methods: i) Pearson's correlation of genomic estimated breeding value (GEBV) with EBV of all animals (rM1) and ii) correlation using inverse of coefficient matrix from the mixed-model equations (rM2). Then, we compared these accuracies by overall population, info-type (PHEN, phenotyped-only; GEN, genotyped-only; and PH+GEN, phenotyped and genotyped), and bull-types (YBULL, young male calves; CBULL, young candidate bulls; and PBULL, proven bulls). Results: The rM1 estimates in the study were between 0.90 and 0.96 among five traits. The rM1 estimates varied slightly by population and info-type, but noticeably by bull-type for traits. Generally average rM2 estimates were much smaller than rM1 (pedBLUP, 0.40 to0.44; ssGBLUP, 0.41 to 0.45) at population level. However, rM2 from both BLUP models varied noticeably across info-types and bull-types. The ssGBLUP estimates of rM2 in PHEN, GEN, and PH+ GEN ranged between 0.51 and 0.63, 0.66 and 0.70, and 0.68 and 0.73, respectively. In YBULL, CBULL, and PBULL, the rM2 estimates ranged between 0.54 and 0.57, 0.55 and 0.62, and 0.70 and 0.74, respectively. The pedBLUP based rM2 estimates were also relatively lower than ssGBLUP estimates. At the population level, we found an increase in accuracy by 2.0% to 4.5% among traits. Traits in PHEN were least influenced by ssGBLUP (0% to 2.0%), whereas the highest positive changes were in GEN (8.1% to 10.7%). PH+GEN also showed 6.5% to 8.5% increase in accuracy by ssGBLUP. However, the highest improvements were found in bull-types (YBULL, 21% to 35.7%; CBULL, 3.3% to 9.3%; PBULL, 2.8% to 6.1%). Conclusion: A noticeable improvement by ssGBLUP was observed in this study. Findings of differential responses to ssGBLUP by various bulls could assist in better selection decision making as well. We, therefore, suggest that ssGBLUP could be used for GS in Hanwoo proven-bull evaluation program.

Analysis of Estimation of Relative Conception Rate on Korean Proven Bull Number Semen using Non-return Rate (비발정 재귀율을 이용한 한우 보증씨수소 수정능력의 상대적 추정치 분석)

  • The decreased fertility is frequently thought to be problem of cattle production. However, studies figure out that number of these problems is related to bull factors especially in artificial insemination setting. Therefore, this study was designed to investigate the fertility status of bull by their estimated relative conception rate of cows that were inseminated by frozen semen from Korean proven bulls. Here we use the non-return rate (NRR) to access the bull fertility whereas, the NRR was define as the proportion of bulls that semen were used to inseminate cows and the number of cows that did not return for another service within 60 days. The data from 54,388 artificial inseminations (AI) were analyzed from 88 KPN semen. The NRRs of highest and lowest fertile bull were 83.81 and 51.33%, respectively. And mean NRR was 68.27%. In comparison to previously reported study, our data shows 17.38% higher NRR and the absolute value of difference in 50%>NRR and 50%
