Wound Healing Effect of Low Molecular PDRN on Experimental Surgical Excision Rat Model (저분자화된 Polydeoxynucleotide (PDRN)의 흰쥐에 대한 외과적 창상 치유 효과)

  • This study was performed to investigate the wound healing effect of skin regeneration cosmetics utilizing low molecular weight Polydeoxynucleotide (PDRN). High purity PDRN was prepared from salmon testes poly-deoxy-ribonucleotide through protein and toxin removal process and molecular weight reduction. In order to evaluate the wound healing effect of PDRN in SD rats, 4 sites of dorsal skin of each animal were excised by using biopsy punch and $500{\mu}L$ of test solution was topically applied once daily for 4 weeks. The tissue changes were observed for every week during the application periods. After applying the PDRN to the wound, the skin was cut flower and contraction of the wounds more quickly, and the coating of PDRN in the wound area was reduced significantly as compared to the positive control group $Fucidin^{(R)}$ applied. The microscopic observation of stained tissue showed that a positive control was most rapid in re-epithelialization ability followed by the PH group, PDRN group, HA group. In addition, transforming growth factor ($TGF-{\beta}$) and vascular endothelial growth factor (VEGF), such as in the growth factor was similar to the results of staining of tissue lesions. In conclusion, it is determined that the low molecular weight PDRN has the therapeutic effect to the wound, and could be used as a functional material of cosmetics and medical industries.

  • Adequate acidic environment in wound healing prevents the inflammation of virus, increases the cell activity, promotes cell proliferation and regular rearrangement of fibroblast, and results in matured epithelialization. In this study, we prepared dressing materials consisting of pectin and carboxymethylcellulose (CMC) with varied ratios. These dressing materials showed different pH values according to the composition ratio. The effect of acidity of pectin/CMC dressing materials on wound healing rates, degree of epithelialization, collagen deposition, and so on, in 3 types of wound models (fresh surgical wounds, $3^{rd}$ degree burn wounds, and infection wounds) were investigated by animal tests. From the results of wound contraction, wound healing, and epithelialization, it can be deduced that dressing material having pectin/CMC ratio of 16/19 (pH 4.67) is most effective among the 3 types of wound models.

  • Sex hormones have long been considered to play an important role in bone turnover rate, periodontal diseases, and wound healing. We have studied the effect of porcine testis steroid extract (PTSE), an extract of porcine testes, which holds a good ratio of 19-nortestosterone (nandrolone), testosterone, androstenedione, $17{\beta}$-estradiol, and estrone, on the healing rate of a standardized full-thickness linear wound on the back of the rat. Skin punch or carbon dioxide ($CO_2$) laser methods were used to create the deep skin injury in two groups of animals. The animals were treated with the PTSE cream, control cream and Vaseline (control) to find out the effect in re-epithelialization, contraction, and formation of granulation and scar tissues. Histological examination after 21 days showed 100, 87.4, and 80.5% recovery of epidermis, dermis, and hypodermis, respectively in the PTSE-treated animals. Similarly, on the 15th day of treatment, complete healing of intact skin was observed in the PTSE cream-treated animals among the laser radiation group. Even though the beginning of re-epithelialization phase and completion of serum crust formation was also observed in the base cream- and Vaseline-treated animals respectively, the complete healing cycle was observed only in the PTSE-treated group. The white blood cell count in the PTSE-treated group showed that PTSE cream is nontoxic to animals.

Use of the Full-thickness Free Skin Mesh Graft for Reconstruction of Wound in a Dog (개의 피부손상에 대한 전층망상식피술 증례)

  • A 6-year-old male dog was referred to Animal Medical Center, Chonbuk National University with large distal extremity abrasion wound to the hind limb by automobile accident. The wound was reconstructed with an autogenous, full-thickness skin mesh graft. The graft was harvested from the ventrolateral abdominal wall and transplanted to the wound. The skin mesh graft survived successfully without any noticeable complications. Successful grafting requires asepsis, an adequately prepared recipient bed through the surgical debridement and lavage, proper harvesting and preparation of the graft. Full-thickness skin mesh graft has lesser skin contraction which prevents formation of scar. Meshing the graft provides more graft flexibility over uneven, convex and concave surfaces and allows adequate drainage. The full-thickness skin mesh graft can be successfully used for the treatment of large distal skin wounds in dogs.

  • In this study, we investigated the effects of total ginseng saponin (TGS) on the cutaneous wound healing process using histological analysis. A total of 24 ICR mice, 5-weeks-old, were used for all in vivo experiments. Mice were divided into control and TGS-treated groups and four equidistant 1-cm full-thickness dorsal incisional wounds were created. The wounds were extracted at days 1, 3, 5, and 7 post-injury for histomorphometrical analysis including wound area and contracture measurements, keratinocyte migration rate, and calculation of infiltrating inflammatory cells. The results showed that the wound area was smaller and keratinocyte migration rate was higher in the TGS-treated group than that of the control group from days 3 to 7. Inflammatory cells in the TGS-treated group at days 1 and 3 were reduced compared to the control group. Wound contraction in the TGS-treated group was greater than in the control group on days 3 to 5, and collagen deposition in the TGS-treated group was higher than in the control group during wound healing. The results indicate a beneficial effect of TGS when used to treat skin wounds.

  • This study was performed to investigate the effect of silk protein (BM-1) treatment on epidermal growth factor (EGF) expression in the wound healing process by excision on the hairless mouse. Significant wound healing activity was observed BM-1 treated group. In BM-1 treated groups ($100{\sim}116 ug/day$), epithelialization of the incision wound was laster with a high rate of wound contraction. In expression of EGF and EGF mRNA, the lesion of BM-1 treated group made EGF to more induce significantly than control lesion. These data suggest that silk protein (BM-1) treatment have wound healing effect by excision on the hairless mouse.

  • Sutures are used not only for wound closure but also for oriental medicine field, beauty and even for plastic surgery. Especially, it is popular in the field of cosmetic surgery. In this paper, we produced sutures using PCL and PLLA with better strength than PDO sutures, which was widely used in the past. To learn about the mechanical characteristics of the PCL and PLLA sutures, the contraction change, tensile strength, and elongation were measured. And SEM was also analyzed for diameter and surface. The contraction change Ratio of sutures are stabilized after a certain period of time regardless of temperature. Also, it can be seen that the higher the temperature, the higher the contraction increase rate. And the rate change of mechanical properties is different according to the temperature before and after the glass temperature. Also the higher the temperature, the faster the molecular motion and the lower the tensile strength. The diameter of the PLLA and PCL sutures is opposite to the contraction change ratio. And it is considered that the sterilization temperature of PLLA sutures is best to set at $45^{\circ}C$ and the sterilization temperature of PCL sutures is best to set at $35^{\circ}C$.

  • This is a report of cases that post-operative orocutaneous through & through open wounds are healed & closed by wound contraction if soft tissue drainage was well established, which seldom is interfered with by local or systemic abnormalities. Authors also found that the localization & seguestration of infected mandible are far better performed by natural mechanisms than by cutting across involved bone.

  • Laryngotracheal stenosis is a congenital or acquired narrowing of the airway, representing a continuum of disease that may affect the glottis, subglottis, and/or trachea. The larynx and trachea are semirigid tubular structures in which concentric scar contraction - a normal wound healing process - tends to narrow the lumen. The diversity of causes, severity, location of stenosis, and association with swallowing and phonation make this disease difficult to compare across patient populations and treating otolaryngologists. The wide array of surgical techniques for any given stenosis attests to the complexity and less than perfect results of the treatment. This review will address the etiology, diagnosis, and management of laryngotracheal stenosis.

An immunohistochemical study on the effects of low-level laser irradiation on expression of actin filaments of human gingival fibroblasts in vitro (저출력레이저조사가 배양치은섬유아 세포의 actin filaments발현에 미치는 영향에 관한 면역조직화학적 연구)

  • The induction of a phenotype with preoperties may have clinical significance in the acceleration of the wound-healing process. Wound contraction involves a specialized cell known as the myofibroblast. The myofibroblasts can be identified by their intense staining of actin bundles with anti-actin antibody. Tissue-specific actin distribution is correlated with the contractile activity of the myofibroblasts and smooth muscle etc. This study was performed to determine the expression of actin filaments in the cytoplasm of cultured human gingival fibroblsts after GaAs laser(BIOSAER, Korea) irradiation. Human gingival fibroblasts were cultured from explants of normal interdental gingival tissue. The third-generation fibroblasts were used for immunohistochemical study. The cultured fibroblasts were exposed $0.53joule/cm^2$(lmW, 7 mimutes) of energy density, and then observed by immunohistochemical method using, rabbit anti0gelsolin, hen smooth muscle polyclonal antibody(Chemicon international inc.), and biotinylated goat anti-rabbit IgG(Vectastain) 24-, 36-, 48-hour after laser irradiation Following results were obtained ; 1. In nonirradiated cultures, round shaped active fibroblasts with abundant cytoplasm and prominet nucleoli were observed. 2. In 24- and 36-hour cultures after laser irradiation, spindle shaped cells with long process were observed. The intensity of stain was seen in cytoplasm of these modified fibroblasts. 3. In 48-hoour cultures after laser irradiation, stained spindle shape cell were not observed. The results suggest that the effect of the galium-arsenide laser treatment on cultured gingival fibroblasts is the rapid development of cytoplasmic actin filaments.
