Regulation of Development in Aspergillus nidulans and Aspergillus fumigatus

  • Members of the genus Aspergillus are the most common fungi and all reproduce asexually by forming long chains of conidiospores (or conidia). The impact of various Aspergillus species on humans ranges from beneficial to harmful. For example, several species including Aspergillus oryzae and Aspergillus niger are used in industry for enzyme production and food processing. In contrast, Aspergillus flavus produce the most potent naturally present carcinogen aflatoxins, which contaminate various plant- and animal-based foods. Importantly, the opportunistic human pathogen Aspergillus fumigatus has become the most prevalent airborne fungal pathogen in developed countries, causing invasive aspergillosis in immunocompromised patients with a high mortality rate. A. fumigatus produces a massive number of small hydrophobic conidia as the primarymeans of dispersal, survival, genome-protection, and infecting hosts. Large-scale genome-wide expression studies can now be conducted due to completion of A. fumigatus genome sequencing. However, genomics becomes more powerful and informative when combined with genetics. We have been investigating the mechanisms underlying the regulation of asexual development (conidiation) and gliotoxin biosynthesis in A. fumigatus, primarily focusing on a characterization of key developmental regulators identified in the model fungus Aspergillus nidulans. In this review, I will summarize our current understanding of how conidiation in two aspergilli is regulated.

  • This study was performed to compare the efficacy of carboxymethylchitosan fabric (CMCF) with that of the combination of CMCF and low molecular weight heparin (LMWH) for the prevention of postoperative uterine adhesion in rats. Adhesions were induced by suturing both the uterine serosa and peritoneum abrased until petechial bleeding occurred. Fourteen days later, adhesions were evaluated clinically and histopathologically. The mean tensile strength was significantly decreased in the CMCF and CMCF+LMWH groups compared to that of control group, and the CMCF+LMWH group had the lowest tensile strength. The distance of adhesion site was highest in the CMCF group and slightly decreased in the CMCF+LMWH group comparing to that of control group. The inflammatory cell infiltration and neovascularization of the CMCF group were significantly lower than those of the control group. It was observed that the damage at intestinal serosa was significantly decreased in the CMCF+LMWH group comparing to that of control group. Therefore, it was concluded that CMCF and LMWH may be useful to prevent postoperative uterine adhesion in rats.

Effectiveness of Home Health Care Service for Elders after Spinal Surgery

  • Purpose: This study was done to evaluate effectiveness of home healthcare services (HHCS) specialized for elders who received spinal surgeries. Methods: A non-equivalent control group pre-post test quasi-experimental study was performed. HHCS was developed based on the Rice model of dynamic self-determination for self-care. For data collection, a control group (n=23) and an experimental group (n=23) were selected by matching age, BMI, pain, general characteristics and type of spine surgery. Measurement tools to evaluate uncertainty and knowledge were developed by the authors. The Numeric Rating Scale (NRS) and Japanese Orthopedic Association Back Pain Evaluation Questionnaire (JOABPEQ) were used to evaluate pain levels. Muscular strength in the legs was measured using a digital muscle tester and tape ruler. Questionnaires were used to evaluate disability in performing ADL and psychological distress levels. Results: The experimental group showed significant decrease in uncertainty (p=.028), increased knowledge (p=.038), and partially decreased pain (p=.003-.331). Partial muscle strength increased significantly (p=.021-.644). Disability in performing ADL and psychological distress in the experimental group decreased significantly compared to control group (p=.002, p=.004). Conclusion: Results indicate HHCS is an efficient home care nursing program for these elders. Further experimental studies with larger samples are required to confirm effects of HHCS.

니스타틴의 응집 특성 및 용혈 활성 (The Aggregation State and Hemolytic Activity of Nystatin)

  • The aggregation behavior of nystatin (NYS) in the presence of pluronic F127, triblock copolymer of poly (ethylene oxide) (PEO) and poly (propylene oxide) (PPO), was measured and correlated with hemolytic activity. Antifungal activity was also studied using Saccharomyces cerevisiae as a model strain. The critical aggregation concentrations (CAC) of the drug were 50.1, 108.0, 134.2, 154.3, and $217.9\;{\mu}M$ at 0.1%, 0.5%, 1.0%, 1.5%, and 2.0% pluronic F127 solution, respectively. The levels of NYS required to start lysis of erythrocytes were about 80, 100, 125, 150, and $200\;{\mu}M$ at 0.1%, 0.5%, 1.0%, 1.5%, and 2.0% pluronic F127 solution, respectively. It was $50\;{\mu}M$ in the absence of the polymer. Minimal inhibitory concentration (MIC) and minimal fungicidal concentration (MFC) of NYS-pluronic F127 lyophilizate were same at $3\;{\mu}g/ml$, while MIC and MFC of pure NYS are $3\;{\mu}g/ml$ and $12\;{\mu}g/ml$, respectively. By modulating the aggregation behavior of NYS, pluronic F127 was able to reduce the toxicity of the drug without compromising the MIC and MFC.

Performance-based Evaluation for Efficiency of Landfill Liner Systems

  • Efficiency of landfill liners system is usually evaluated based on leakage rate and mass flux. In this study, composite liner systems including the GCL(geosynthetic clay liner) composite liner, the Subtitle D liner, the Wisconsin NR500 liner, and the recently utilized double composite liner, which is a combination of the GCL composite liner and Subtitle D-type liner, have been examined. The leakage rate through circular and long defects in the geomembrane (GM) of the liner system was analyzed with the aids of analytical and numerical methods. For the mass flux criterion, contaminant transport through defects in the GM of landfill liners can be evaluated based on the calculated leakage rates. The diffusion rate of volatile organic compounds through intact landfill liners was evaluated by performing a one-dimensional numerical model. Cadmium and toluene were adoptted in the analyses as typical inorganic and organic substances, respectively, which will be chemical species encountered during landfill operation. The performance-based evaluation indicates that the double composite liner systems are superior to the other types of liner.

Which Individual Characteristics Influence Mothers' Health Information-seeking Behavior?

  • Historically, mothers have been noted as active health information seekers, reflecting their roles as health mangers and caregivers for their family members. However, previous studies have focused on health-related information behavior among mothers in native populations or mothers of children with specific diagnoses. To fill this research gap, this study focused on health information behavior among mothers of healthy infants and toddlers. Using Wilson's (1997) information-seeking model, this study aimed to uncover the relationships between mothers' demographic characteristics and their health information source use. Online surveys were completed by 851 mothers: 255 U.S.-born mothers, 296 Korean-born mothers, and 300 Korean immigrant mothers living in the United States. Results indicated that there were statistically significant relationships between mothers' nine demographic characteristics (mother's age, education level, household income, employment status, the number of children, years living in the U.S. (or Korea), fluency in speaking English, size of household, housing status) and their health information source use. Based on the results, the implications for information professionals at diverse organizations are discussed when they provide health information services to this specific population.

수분함량에 따른 율무가루의 점도변화 및 동적물성 특성에 관한 연구 (Viscosity and Dynamic Rheological Properties of Job's-tears as a Function of Moisture Content)

  • Changes in viscosity and dynamic theological properties of Job's-tears were measured by Bohlin dynamic tester as a function of moisture, and measurement was performed within a linear viscoelastic range. The result of the shear stress vs shear rate of Job's-tears at different moisture contents $(50{\sim}75%)$ was applied to mathematical models and Herschel-Bulkley model showed the highest correlation coefficient. Lower moisture content (55%) produced higher yield stress and consistency index, but lower flow behavior index, whereas higher moisture content showed reverse effects. Job's-tears with $50{\sim}70%$ moisture contents showed a higher storage modulus (G') than loss modulus (G') at all frequencies, showing a higher concentrated polymer characteristics. However, higher moisture content (>75%) showed crossover point between G' and G', and frequency dependency. As the moisture content was increased, the amount of viscoelastic properties such as G', G', complex viscosity decreased during heating, and initial temperature and miximum value of viscoelastic properties shifted to higher temperatures, representing the moisture-dependence of Job's-tears upon theological properties.

반도체 및 LCD 제조 공정의 AGV Controller 개발 (Development of an AGV Controller in Semiconductor and LCD Production Systems)

  • In this paper, LAC(Look-ahead AGV Controller) has been developed for efficient routing of parts in semiconductor and LCD production systems. Several procedures have been developed as sub-modules. LACP(Look-ahead AGV Control Procedure) which controls AGVs using the information on the current and future status of the systems is the main element of the LAC. To support LACP, DSP(Destination Selection Procedure) which determines a destination of a part and AGV call time, SSP(Source Selection Procedure)which selects a part coming next to a buffer when the buffer becomes available. and RTM(Response Time Model) which estimates empty travel time of AGVs and waiting time for an available AGV have been developed. A simulation experiment shows that LAC reduces part's flow time, AGV utilization, average and maximum inventory level of a central buffer, empty travel time of an AGV, and waiting time for an available AGV.

A two-stage damage detection approach based on subset selection and genetic algorithms

  • A two-stage damage detection method is proposed and demonstrated for structural health monitoring. In the first stage, the subset selection method is applied for the identification of the multiple damage locations. In the second stage, the damage severities of the identified damaged elements are determined applying SSGA to solve the optimization problem. In this method, the sensitivities of residual force vectors with respect to damage parameters are employed for the subset selection process. This approach is particularly efficient in detecting multiple damage locations. The SEREP is applied as needed to expand the identified mode shapes while using a limited number of sensors. Uncertainties in the stiffness of the elements are also considered as a source of modeling errors to investigate their effects on the performance of the proposed method in detecting damage in real-life structures. Through a series of illustrative examples, the proposed two-stage damage detection method is demonstrated to be a reliable tool for identifying and quantifying multiple damage locations within diverse structural systems.

Influencing Knowledge Sharing on Social Media: A Gender Perspective

  • Online Word-of-Mouth communication, or eWOM, has dramatically changed the way people network, interact, and share knowledge. Studies have examined why consumers choose to share knowledge online, especially online product reviews, as well as the motivations of individuals to share product ideas online. However, the role of gender in shaping the motivation and types of knowledge shared online has been given little consideration. Using concepts from Social Exchange Theory and the Theory of Reasoned Action, we address this research gap by developing and testing a model of gender's influence on knowledge sharing in a social media context. A PLS analysis of survey data from 257 students indicates that reputation, altruism, and subjective norms are key motivators for knowledge sharing intention in social media. More importantly, that gender plays a moderating role within the motivation-knowledge sharing relationship. We also find that subjective norms have a greater impact on knowledge sharing with women than with men. Collectively, our research results highlight individualized factors for improving customer participation in external facing social media for marketing and product innovation.
