고품질 무핵 포도 품종 '흑이슬' 육성 (Breeding of a Seedless Table Grape Cultivar 'Heukisul' (Vitis sp.) with High Quality)

  • 무핵 포도 품종을 육성하고자 2000년에 'Kyoho'와 'Thompson seedless' 교잡으로부터 '흑이슬'이 육성되었으며, 2004년도에 1차 선발되었다. 2004년부터 2005년까지 두 곳에서 KTS014의 이름으로 지역적응시험을 실시하여 2006년에 최종 선발하였다. '흑이슬'은 'King Dela'에 비하여 화방의 과실 착과가 우수하여 화진현상이 낮았다. 발아기는 4월 23일경, 만개기는 6월 9일경이며 수확기는 8월 29일경으로 중생종 품종으로 생각된다. 평균 과립무게는 4.0g이고 당도는 $21.9^{\circ}Brix$이다. 과피색은 과분이 많은 자흑색이고 과육은 단단하다. 착립밀도가 높아 알솎기가 필요 없다. 또한 열과율이 대단히 낮은 특성을 가지고 있다.

에탄올 농도에 따른 머루(Wild Grape, Vitis coignetiae) 추출물의 폴리페놀 함량 및 항산화 활성 (Total Polyphenol Content and Antioxidative Activity of Wild Grape (Vitis coignetiae) Extracts Depending on Ethanol Concentrations)

  • 추출용매를 달리하여 마이크로웨이브로 추출한 머루의 생리활성을 측정하였다. 전자공여능, 아질산염 소거능은 모든 추출구에서 농도 의존적으로 활성이 증가하였고 폴리페놀도 농도 증가에 따라 함량이 높게 나타났다. 세 가지 추출용매 중 50% 에탄올 추출 시 항산화 활성이 높았는데 이는 총 폴리페놀 함량 측정 결과와도 일치하여 머루의 항산화 활성이 폴리페놀과 밀접한 관련이 있다고 사료된다. 전반적인 활성은 전자공여능이 약 $13{\sim}101%$로 특히 1.6 g/dL 농도의 50% 에탄올 추출물의 활성의 경우 1% L-ascorbic acid보다 우수했다. 아질산염 소거능은 pH가 낮을수록 활성이 높은 경향으로 pH 1.2에서 활성이 $34{\sim}99%$로 가장 높게 측정되었다. 총 폴리페놀 함량은 농도에 따른 폴리페놀 함량의 차이가 컸으며 50% 에탄올 추출, 1.6 g/dL 농도에서 약 38.76 mM gallic acid equivalent로 가장 높았다. SOD 유사활성은 $30{\sim}50%$로 전자공여능이나 아질산염 소거능만큼 우수하진 않았지만 0.01% L-ascorbic acid보다 50%, 100% 에탄올 추출구에서 높은 활성을 보였다. 머루의 항산화 활성과 폴리페놀 함량은 밀접한 관련이 있는 것으로 보이며 폴리페놀 함량이 풍부한 머루를 이용한 항산화 건강기능성 식품 개발의 기초 자료로 제공하고자 하였다.

Comparison of the Antioxidative Effects and Content of Anthocyanin and Phenolic Compounds in Different Varieties of Vitis vinifera Ethanol Extract

  • This study was a quantitative HPLC analysis of four anthocyanins and five phenolic compounds contained in the skins, vines and seeds of the Campbell Early, Muscat Bailey A and Neo Muscat grape varieties. In the phenolic analysis, the seeds of the Campbell Early were found to contain 1.9, 1.8 and 1.6 times higher quantities of gallic acid, catechin and epicatechin relative to other grape seeds. Three anthocyanins, cyanidin, peonidin and pelargonidin, were also found to be higher in the skins of the Campbell Early relative to other grape skins. Therefore, the Campbell Early is the most useful grape variety with regard to the extraction of these six compounds from these grape seeds and skins. The free radical scavenging effects of grape seeds were also compared, and the results indicated that the Campbell Early seeds were most effective among them.

Polyphenolic Contents and Antioxidant Activities of Underutilized Grape (Vitis vinifera L.) Pomace Extracts

  • Grape pomace is an abundant source of underutilized winery by-products. Polyphenols were extracted from grape pomace using cellulase and gluco-amylase enzymes. 2,2-diphenyl-1-picrylhydrazyl (DPPH) and Folin-Ciocalteu's assays were used to measure antioxidant activity and total polyphenolic contents. Both cellulase, and gluco-amylase digested grape pomace showed efficient radical scavenging activity. In addition, the total polyphenolic contents of cellulase digested grape pomace showed lower concentrations were effective compared to higher concentrations, whereas glucoamylase enzyme did not show remarkable variations. The DPPH radical scavenging activity and total polyphenolic contents were significantly higher in the cellulase digested grape pomace compared to the gluco-amylase digested and the not digested grape pomace. It is notable that enzymatic digestions were efficient for extracting polyphenols from grape pomace. The underutilized grape pomace polyphenols can be further used for food safety as a natural antioxidant.

Distribution and Features of the Six Classes of Peroxiredoxins

  • Peroxiredoxins are cysteine-dependent peroxide reductases that group into 6 different, structurally discernable classes. In 2011, our research team reported the application of a bioinformatic approach called active site profiling to extract active site-proximal sequence segments from the 29 distinct, structurally-characterized peroxiredoxins available at the time. These extracted sequences were then used to create unique profiles for the six groups which were subsequently used to search GenBank(nr), allowing identification of ~3500 peroxiredoxin sequences and their respective subgroups. Summarized in this minireview are the features and phylogenetic distributions of each of these peroxiredoxin subgroups; an example is also provided illustrating the use of the web accessible, searchable database known as PREX to identify subfamily-specific peroxiredoxin sequences for the organism Vitis vinifera (grape).

천연식품의 색과 향기를 첨가한 기능성 칼라 묵의 제조 (Manufacturing of Functionalized ColorMook by Addition of the Color and Flavor from Natural Foods)

  • This study was carried out to make the new products of color moot as a functional food by variation of the color and flavor through addition of gamgyul, Daccus Carota Var. Sativa, Vitis vinifera, soy sauce, and Pimpinella brachycarpa N., respectively. For all the products, the color was observed by Colorimeter and the sensory evaluation was performed to evaluate their effectiveness and usefulness. The A$_w$ (water activity) test of color mooks has performed to examine their water activity individually. The A$_w$ of color moot was higher than that of the white one. The characters of texture of color mook as a function of the amount of the color and the flavor added were measured by a texture analyzer (XT-RA, Texturometer). By the TPA test, the rainbow and orange color mook examined were high in the springiness.

가온 재배 시 '피오네' 포도(Vitis vinifera × V. labrusca)의 엽과비에 따른 과실 특성 (Fruit Characteristics Based on Leaf to Fruit Ratio in 'Pione' Grapevine (Vitis vinifera × V. labrusca) during Cultivation with Heating)

  • BACKGROUND: Defoliation in grapevine cultivation is practically used to improve light environment within the canopy and thereby fruit quality. Effects of defoliation in five-year-old 'Pione' grapevine during cultivation with heating were investigated to find out optimum ratio of leaf area to fruit cluster weight (L/F). METHODS AND RESULTS: The grapevines were defoliated with berry-thinning 20 days after full bloom to provide various levels of L/F. At harvest, total leaf area values of fruit bearing branches were between 0.23 and $0.60m^2$. With increasing L/F, soluble solids and anthocyanin contents curvilinearly increased ($R^2=0.76^{**}$). At L/F over $0.6m^2/kg$, soluble solids content (SSC) leveled off. With increasing L/F, titratable acidity (TA) linearly decreased ($R^2=0.87^{**}$), but the ratio of SSC to TA linearly increased ($R^2=0.86^{**}$). Anthocyanin content was significantly correlated with SSC and the ratio of SSC to TA ($R^2=0.80^{**}$ and $0.82^{**}$, respectively). When total leaf area per fruit bearing branch was maintained $0.40m^2$, soluble solids and anthocyanin contents linearly decreased ($R^2=0.79^{**}$ and $0.85^{**}$, respectively), but TA linearly increased with increasing fruit cluster weight ($R^2=0.70^{**}$). Fruit was low in quality when the L/F was below $0.6m^2/kg$. CONCLUSION: L/F is recommended to be maintained at least $0.6m^2/kg$ in 'Pione' grapevine during cultivation with heating to produce higher-quality fruits.

토양 및 기후정보 통합 최대저해인자법에 의한 복숭아와 포도의 적지 평가 (The maximum limiting characteristic method-based land suitability assessment for peaches (Prunus persica) and grapes (Vitis vinifera L.) using rasterized data of soil and climate on agricultural land in South Korea)

  • 본 논문에서는 남한지역을 대상으로 토양학적 그리고 기후학적 적지 기준을 통합하여 최대저해인자 방법으로 과수 2종(복숭아와 포도)에 대해서 재배적지를 구분하였다. 복숭아는 최적지 2.21%, 적지 19.21%, 가능지(저위생산지 포함) 12.07%, 부적지 66.52%로 구분되었고, 포도는 최적지 3.65%, 적지 17.98%, 가능지(저위생산지 포함) 11.85%, 부적지 66.52%로 구분되었다. 토양과 기후 조건의 통합에 의해 구분한 복숭아와 포도의 적지는 토양 조건만으로 구분한 적지와 유사한 것으로 분석되었고, 토양 조건에 의한 구분한 적지(최적지 포함) 면적이 기후 조건에 의해 구분한 면적보다 적은 것으로 분석되어서 토양 조건이 적지구분의 저해인자로 확인되었다. 어떤 행정구역(도)에서 적지(최적지 포함)로 구분된 면적이 많으면 많을수록 더많은 과수가 재배될 것이라는 가정 하에, 행정구역별 복숭아와 포도의 실재 재배면적 비율과 토양 및 기후 조건 통합의 최대저해인자법에 의해 구분한 적지면적과 비교하였을 때, 실재 과수 재배면적의 비율이 적은 행정구역에서는 해당 과수의 재배면적 확대를 고려할 필요가 있을 것이다. 다만, 과수 적지구분의 정확도 향상을 위해서는 분석적 계층화법(AHP)과 같은 개선된 방법과 농장단위의 과수 생산량과 수익자료를 추가하여 비교 분석할 필요가 있다.

포도와 미국자리공의 세포현탁배양계에 있어서 배지내 무기염 농도가 색소축적에 미치는 영향 (Effects of Salt Concentrations on Accumulation of Pigments in Cell Suspension Cultures of Vitis vinifera and Phytolacca americana L.)

  • 포도 및 미국자리공의 세포현탁배양계에서 세포생장 및 색소축적에 미치는 배지내 무기 염농도의 효과를 알아 보았다. 포도의 세포 증식은 control구에서만 정상적인 대수증식기를 보였고, 희석배지에서는 4일째에서 6일째까지 완만한 대수증식기를 나타냈다. Anthocyanin은 모든 처리구에서 배양후 12일째에 최대로 축적이 되었다. 미국자리공에 있어서는 모든 처리구에서 세포증식과 동조적으로 betacyanin이 축적되어, 배양 8일 이후에 최대축척을 나타냈다. 포도의 세포배양에서는 sucrose 87.6mM까지는 세포증식이 증가하였으나 그 이상의 농도에서는 증식이 급격히 떨어졌다. Anthocyanin은 146mM에서 최대로 축적되었으며 그 이상의 농도에서도 다량의 anthocyanin이 축적되었다. 미국자리공에서는 sucrose농도가 높아질수록 세포증식 및 색소축적이 증가하는 경향을 보여, 포도의 배양세포와 대조적 인 결과를 보였다. 이상의 결과로써 포도의 anthocyanin과 미국자리공의 betacyanin이 같은 적색색소이지만, 배양세포내에서의 생성기작 및 그 축적과정이 서로 상이하다는 것을 보여주었다.

Effect of Herbal Extracts Mixtures on Antioxidant System in Chronic Enthanol-treated Rats

  • Disturbance of antioxidant system is very common in chronic alcoholics and herbal or natural products with antioxidant activity have been used for its treatment. This study was to investigate the effect of Vitis vinifera extract(V), Schisandra chinensis extract(S), Taraxacum officinale extract(T), Gardenia jasminoides extract(G), Angelica acutiloba extract(A) and Paeonia japonica extract(P), and their combinations on the antioxidant and ethanol oxidation system. Male Sprague-Dawley rats were subjected to Lieber-DeCarli ethanol liquid diet(ED) and were then given different herbal extract mixtures for 6 weeks including VST(V 100+S 150+T 150mg/kg/day), VSG(V 100+S 150+G 150mg/kg/day), VTG(V 100+T 150+G 150mg/kg/day), and VAP(V 100+A 150+P 150mg/kg/day). When the activity of alcohol dehydrogenase(ADH) and acetaldehyde dehydrogenase(ALDH) were compared between ED only group and herbal extracts treatment group, the differences were statistically significant. Phase I and II(glutathione-S-transferase, phenol sulfatransferase) enzyme activities were found to be significantly higher in the VAT treatment group compared to the ED group. Herbal extracts not only repressed the ethanol-induced elevation of malondialdehyde level, but also protected against ethanol-induced decrease in glutathione content, glutathione reductase, glutathione peroxidase, catalase and superoxide dismutase activities. The administration of the herbal extracts was found to be effective in eliminating lipid-peroxides induced by long-term consumption of alcohol by activating various enzyme systems and physiological active compound formation system. After a chronic consumption of alcohol, Angelica Radix protected the liver via activating the ethanol-metabolism enzyme system, and Paeoniae Radix via activating the ethanol-metabolism enzyme and the phase I, II-metabolism enzyme system. Taraxaci Herba was also effective in liver protection via activating the ethanol-metabolism enzyme system and the phase I, II-metabolism enzyme system, Gardeniae Fructus via activating the phase II-metabolism enzyme system and the anti-oxidation system enzyme, and Schisandra Fructus and a grapestone via activating the anti-oxidation system. Our data suggest that these herbal extracts may be useful as a health functional food or new drug candidate for fatty liver and hepatotoxicity induced by chronic alcohol consumption.
