A study on the inflammatory response induced by LPS of the Arthrospira platensis ethanol extract

  • Arthrospira platensis has been reported to contain a variety of substances such as phycocyanin, ${\beta}$-carotene, vitamin E and other carotenoids. In this study, zebrafish were treated with indoor cultivation spirulina ethanol extracts(ICAE) to determine toxicity(coagulation rate, hatching rate, heart rate). We used the DCFH-DA staining method to detect the effect of reactive oxygen species(ROS) generation on lipopolysaccharide(LPS)-induced zebrafish embryos ROS various concentrations(0.01, 0.05, 0.1, 0.5mg/ml) of ICAE. Cell toxicity was measured by WST-1 assay on RAW 264.7 macrophage cell line. Also, measured the inhibitory effect of nitric oxide(NO) and prostaglandin $E_2$ ($PGE_2$) production in RAW264.7 macrophages induced by LPS at various concentrations of ICAE. The results of embryo coagulation rate, hatching rate, heart rate of zebrafish at various(0.01, 0.05, 0.1mg/ml) of ICAE was no toxicity. The ICAE treated group had an inhibitory effect on NO and $PGE_2$ production compared and decreased with concentration. The results of this study ethanol extract of Arthrospira platensis has an anti-inflammatory effect and suggest that is worthy of use cosmetics for skin protection.

Characterization of Partially Purified Lipoxygenase of Mackerel Gill (고등어 아가미에서 추출한 Lipoxygenase의 특성)

  • Lipoxygenase of mackerel gill exhibited the highest reactivity toward eicosapentaenoic acid (EPA) followed by arachidonic acid, linoleic acid. The optimum pH were pH 4.5, 5.0 and 4.8 for EPA, arachidonic acid and linoleic acid, respectively. The enzyme was the most stable at pH 5.5. Optimum temperature was $25^{\circ}C$ for all substrate fatty acids. For linoleic and arachidonic acids the highest thermal stability was observed at $8^{\circ}C$ whereas, for (EPA) at $20^{\circ}C$. Optimum ionic strength was 0.22M, $Sn^{2+}$, vitamin E and catechin completely inhibited the enzyme at the concentration of 1.0mM. Molecular weight of the enzyme was 42,000 dalton.

Studies on the Blood Chemical Values in Thoroughbred Foals (Thoroughbred종 망아지의 혈액화학식에 관한 연구)

  • The considerable researches have been done on the change,1 associated with age in the blood picture of domestic animals, but a survey has not been of the blood picture in Thoroughbred fears in Korea. Therefore, the purpose of this study is to outline the range of values in clinically not\\\\`- mal Thoroughbred foals. Some blood chemical parameters from 3 days to 8 weeks of age were exam-ined to obtain normal values in Thoroughbred foal7. Blood samples were obtained from the jugular vein in 11 Thoroughbred foals and collected at 3 days, and at 1,2,4 and 8 weeks of age, respectively. The results obtained ill this study were summarized as follows: The packed cell volume (PCV) at 4 weeks of age was significantly lower than that at 3 days of age (P<0.01) and that al 2 and 8 weeks of age were straightly lower than that at 3 days of age (P<0.05). The total protein (TP) at 8 weeks of age was higher than that at 3 days, 1 and 2 weeks of age (P<0.05). The activity of aspartate ami-notransferase (AST) at 2,4 and 8 weeks of age were significantly higher than that at 3 days of age (P<0.01). The total bilirnbin showed a tendency to decrease gradually according to the weeks and that at 2,4 and 8 weeks of age were significantly lower than that at 3 days of age (P<0.01). The BUN at 1 and 8 weeks of ago were lower than that at 3 days of age (P<0.05). The concentration of Ca at 1 week of alee was higher than that at 3 days of age (P<0.05). The vitamin A and vitamin E showed a tendency to decrease gradually from 3 days to 4 weeks of age and increased slightly at 8 weeks of age.

Physico-chemical Composition and Anti-Allegic Effects of Walnut Oil (호두 기름의 성분조성과 항알레르기 효과)

  • This study was conducted to investigate physicochemical properties and anti-allergic properties of walnut oil. pH, acid value and iodine value of walnut oil were respectively 4.9, 0.8 and 117. Most of general composition of walnut oil was crude fat(99.9%) Vitamin A and E were 0.06 and 10.25mg/100g and the major fatty acids of walnut oil was linoleic acid(62.8%). Total phenolics and antiallergic effects of walnut oil were estimated 27mg% and 62.82% at the concentration of 0.5% ethanol walnut oil. These results suggest that the walnut oil can provide one of the valuable resource for the functional foods.

  • The methanol extract of Taraxacum coreanum Nakai (Compositae) was examined on the in vitro oxidation of human low density lipoprotein (LDL). It is well known that LDL oxidation induced artherosclerosis, if we can protect LDL oxidation process, excess plasma lipoprotein accumulation into the arterial lesion prone areas can be blocked. The methanol extract was treated with oxidized LDL which was incubated with $16\;{\mu}M$ $Cu^{2+}$ for metal catalyzed oxidation and TBA value, mobility on agarose gel and formation of conjugated diene and change of vitamin E were determined for the evaluation. The extract showed antioxidative effect at concentration $200\;{\mu}g/ml$ on LDL oxidation.

  • Mulberry heart disease was associated with vitamin E and selenium deficiency of pigs. Anatomical findings of this disease were hydropericadium, extensive patchy hemorrhage of epicardium, endocardium, and discoloration of dark red color of myocardium. In histological findings were characterized by acute myocardial degeneration, extensive hemorrhage, fibrinoid degeneration of arterioles, PAS positive material deposition of arterioles and capillary thrombosis. In affected herds, feed and serum tocopherol and selenium concentration were less than normal values.

  • Biological fixation of carbon dioxide using microalgae is known as an effective CO$_2$reduction technology. However, many environmental factors influence microalgal productivity. Optimal cultivation factors were determined for the green alga, Euglena gracilis Z, which offers high protein and vitamin E content for animal fodder. In batch culture in a photovioreactor, it was found that theinitial pH, temperature, CO$_2$concentration in air, and light intensity during the optimal cultivating conditions were 3.5, 27$^{\circ}C$, 5-10% and 520 ${\mu}$mol/㎡/s, respectively. When tap water and freshwater were used as cultivating media unsterilized tap water was found to be effective. A kinetic model was considered to determine the relationship between the specific growth rate and the light intensity. The half-velocity coefficient (K(sub)I) in the Monod model under photoautotrophic conditions was 978.9 ${\mu}$mol/㎡/s.

PREVENTION OF HYDROXYL RADICAL-INDUCED ERYTHROCYTE HEMOLYSIS BY PROTEIN THIOLS

  • A system for studying oxidative hemolysis has been used by controling UV-irradiation and concentration of a novel molecular probe, N,N'-bis(2-hydroperoxy-2-methoxyethyl)-1,4,5,8-naphthalenetetra-carboxylic diimide (NP-III), which generates hydroxyl radical upon longer wavelength photoirradiation (> 350 nm). NP-III induces 25~30% of hemolysis at low concentration (50 $\mu$M) for 3h-irradiation of UVA. The simultaneous treatment of N-ethylmaleimide (NEM) with NP-IH completely hemotyzed erythrocytes under the same conditions as NP-III alone by both decreasing thiol group and increasing lipid peroxidation in erythrocyte membrane. However. thiol-reducing agents prevented the protein-crosslinking and lipid peroxidation on the NEM-synergistic hemolysis by partially scavenging hydroxyl radical and maintaining the thiol group of erythrocyte membrane in the reduced state. In addition, erythrocytes pretreated with 2,2,5,7,8-pentamethyl-6-hydroxychromane (PMC), vitamin E homologue was able to delay and decrease the lipid peroxidation when compared to cells pretreated with both NEM and PMC. We suggest that the presence of reduced thiols in inner membrane protein by GSH can prevent the protein-crosslinking and the lipid peroxidation, and eventually prevent the oxidative hemolysis of erythrocyte.

Carnitine and/or GABA Supplementation Increases Immune Function and Changes Lipid Profiles and Some Lipid Soluble Vitamins in Mice Chronically Administered Alcohol

  • This study evaluated the effects of carnitine and/or GABA supplementation on immune function, lipid profiles and some vitamins in mice chronically administered alcohol. BALB/c mice were fed with either AIN-76 diet (N), control diet plus alcohol (4 g/kg bw, E), E plus 0.5 g/kg bw carnitine (EC), E plus 0.5 g/kg bw GABA (EG), or E plus 0.5 g/kg bw carnitine plus 0.5 g/kg bw GABA (ECG) for 6 weeks. Administrations of the carnitine and/or GABA prevented alcohol-induced increases in triglyceride concentrations in serum and liver. However, there was no difference among the supplemented groups. Serum vitamin E concentration was higher in mice supplemented with EC and EG, but not in mice given ECG. Phagocytic activity of peritoneal macrophages was increased in EG group compared with E group. The subpopulations of murine splenocyte's TH cells were increased significantly in EC and ECG groups. These data suggest that immune function, lipid profiles and some immune-related lipid soluble vitamins were positively changed by supplementation of carnitine or GABA, but do not show any synergistic effect of mixed supplementation.

Effect of Water and Ethanol Extracts of Persimmon Leaf and Green Tea Different Conditions on Lipid Metabolism and Antioxidative Capacity in 12-month-old Rats (추출 조건을 달리한 감잎과 녹차의 물 및 에탄올 추출물이 노령쥐의 지방대사와 항산화능에 미치는 영향)

  • This study was performed to investigate effects of dried leaf powders, water, 75% and 95% ethanol extracts of persimmon leaf and green tea on lipid metabolism, lipid peroxidation and antioxidative enzyme activity in 12-month-old rats. Fifty-four male Sprague-Dawley rats weighing 542$\pm$4.5g were blocked into groups according to their body weight and were raised for four weeks with the diets containing 5%(w/w) dried leaf powders of persimmon(Diospyros kaki Thunb) and green tea(Camellia Sinensis O. Ktze), water or 75% and 95% ethanol extracts from same amount of each dried tea powder. Food intake was not significantly different among all groups, but weight gain of green tea powder group was significantly lower than that of control group. Plasma and liver lipid levels of all the tea diet groups were lower than those of control group. Especially, 75% ethanol extract of persimmon leaf decreased total lipid and triglyceride concentrations in plasma and 95% ethanol extract of persimmon leaf decreased liver total lipid level. However, there was no difference between 75% ethanol extracts groups and 95% ethanol extracts groups in lipid metabolism. Superoxide dismutase(SOD) and catalase activities in erythrocyte were remarkably increased by all the green tea diets. SOD, catalase and glutathione peroxidase activities in liver were increased by the feeding of ethanol extracts from green tea and persimmon leaf powder. Liver xanthine oxidase activity was not different among all groups. Plasma Thiobarbirutic acid reactive substance(TBARS) concentrations of all the green tea diet groups were significantly low. It was thought that high flavonoids in green tea inhibited plasma lipid peroxidation by promoting SOD, catalase activities in erythrocyte. 95% ethanol extract of persimmon leaf also inhibited plasma lipid peroxidation by high vitamin E and beta-carotene. Persimmon leaf powder decreased liver TBARS concentration by vitamin E, betacarotene and vitamin C and by increasing activities of antioxidative enzymes with flavonoids. In conclusion, dried leaf powders, water, 75% and 95% ethanol extracts of persimmon leaf and green tea were effective in lowering lipid levels and inhibiting lipid peroxidation in 12-month-old rats. Above all, ethanol extracts of persimmon leaf decreased plasma and liver lipid levels and persimmon leaf powder effectively inhibited liver lipid peroxidation. Extracts of green tea leaf inhibited plasma lipid peroxidation. In lowering lipid levels and inhibiting lipid peroxidation, ethanol extracts were more effective than water extracts, but there was no difference between 75% ethanol extracts and 95% ethanol extracts in lipid metabolism.
