생굴로부터 bacteriophage의 분리 및 부분특성 (Isolation and Partial Characterization of Bacteriophage from Oyster)

  • 시판중인 생굴을 대상으로 Vibrio 속을 숙주로 하는 bacteriophage의 분리를 시도하였다. 4 종의 Vibrio 속과 5 혈청형의 Vibrio parahaemolytius를 실험대상으로 한 결과 배brio parahaemolyticus 2 혈청형에서만 bacteriophage가 분리되었다. 분리된 phage의 plaque 크기는 0.4mm였으며 전자현미경적 형태는 미부가 뚜렷하지 않은 육각형의 두부가 관찰되었고 크기는 67nm$\times$83nm 였으며, PFV/ml은 1.25$\times$$10^{11}$이었다. 분리된 phage는 chloro-form에 감수성을 나타내었다. 분리된 phage의 genomic 특성을 규명하기 위하여 핵산을 분리한 결과 DNA로 판명되어졌으며 두 혈청형 모두 제한효소 처리한 결과 Eco R I으로 4부위의 절단양상과 Hind III로부터 14부위 절단양상이 관찰되었다.

Vibrio anguillarum O1이 생산하는 Outer Membrane Vesicle (OMV)의 분리 및 OMV 내의 단백질 특성 (Isolation and characterization of the outer membrane vesicle (OMV) protein from Vibrio anguillarum O1)

  • Vibrio anguillarum is a gram-negative bacterium that causes vibriosis in approximately 80 different fish species. V. anguillarum produces several exotoxins are correlated with the pathogenesis of vibriosis. This study is focused on the composition of the outer membrane vesicle. Most of gram-negative bacteria produce outer membrane vesicle (OMV) during cell growth. OMV was formed from the outer membrane surface of cell and than released to extracellular environment. OMV consists of outer membrane lipids, outer membrane protein (OMP), LPS, and soluble periplasmic components. Also, they contain toxins, adhesions, and immunomodulatory. Many gram-negative bacteria were studied out forming OMV. In Vibrio sp., formation of OMV by electron microscopy has been reported from V. cholerae and V. parahaemolyticus. In present study, we isolated OMV from V. anguillarum and OMV protein was separated by SDS-PAGE. Magor band was sliced and analyzed by MALDI-TOF. The major protein band of 38kDa was identified as OmpU by MALDI-TOF MS analysis.

A Novel Marker for the Species-Specific Detection and Quantitation of Vibrio cholerae by Targeting an Outer Membrane Lipoprotein lolB Gene

  • Vibrio cholerae O1 and O139 are the major serotypes associated with illness, and some V. cholera non-O1 and non-O139 isolates produce cholera toxin. The present study describes a quantitative polymerase chain reaction (qPCR) assay for the species-specific detection and quantitation of V. cholera using a primer pair based on an outer membrane lipoprotein lolB gene for the amplification of a 195 bp DNA fragment. The qPCR primer set for the accurate diagnosis of V. cholera was developed from publically available genome sequences. This quantitative PCR-based method will potentially simplify and facilitate the diagnosis of this pathogen and guide disease management.

Vibrio parahaemolyticus에 대한 Immunoglobulin yolk (IgY)의 생산과 특이성 (Production and Specificity of Imunoglobulin yolk (IgY) on Vibrio parahaemolyticus)

  • 본 연구는, Vibrio parahaemolyticus에 대한 면역 글로블린 Y(IgY)를 생산하기 위하여 시행되었다. Vibrio parahaemolyticus는 중요한 병원성 균이며, 위장염을 유발시키는 원인 균이다. 따라서, 난황에서 Vibrio parahaemolyticus에 대한 항체를 생산하기 위하여 항원 LPS(lipopolysaccharide)를 암탉에 주사하였다. 면역은 2주 간격으로 4차까지 실시하였으며, 추가면역으로 2주 간격으로 3차까지 실시한 후 ELISA로 그 항체 역가를 측정하였다. 난황에서의 항체생성은 1차 면역 후 1주일부터 나타나기 시작하였으며, 7주에 최고 역가에 도달하였고, 이러한 역가는 13주 이후까지 지속되는 것으로 나타났다. 한편 혈청에서의 항체 역가도 난황에서의 경향과 유사하였다. 정제시킨 IgY를 본 연구의 water-dilution방법과 상업용 IgY분리 kit로 그 정제 정도를 비교해본 격과 큰 차이를 나타내지 않았으며, Vibrio parahaemolyticus외의 다른 Vibrio 속 및 세균들과의 교차반응성 조사에서 특별한 교차반응이 없었다. 본 연구에 나타난 결과로, 아주 적은 양의 LPS 항원으로 역가가 높은 항체의 생산이 가능하였으며, 이러한 결과들로 보아 난황은 유행성 위장염을 유발하는 병원성 균의 특이 항체 개발을 위한 좋은 공급원이 될 것으로 생각된다.

2018년 경남 양식어류에서 검출된 병원체 모니터링 (Monitoring of Pathogens Detected in Cultured Fishes of Gyeongnam in 2018)

  • The major cultured marine fishes in sea off the coast Gyeongsangnam-do Province, South Korea, were assessed and included 9.3% rockfish Sebastes schlegelii, 7.8% red seabream Pagrus major, and 2.1% rock bream Oplegnathus fasciatus. The number of insurance payments related to disease mortality in cultured fish in 2017 was fourfold that in 2016. Economic loss in aquaculture due to disease in cultured fish is high and represents an important inhibitory factor affecting marine fishery productivity. In 2018, diseases led to severe production losses in several aquaculture species: 40.0% in rockfish, 11.4% in olive flounder Paralichthys olivaceus, 10.0% in filefish Thamnaconus modestus, and 9.3% in red seabream. Fish-parasitic pathogens such as Microcotyle sebastis, Alella spp., and Dactylogyrus spp. enter mainly via the gills and skin surface. Among bacterial pathogens, Vibrio species were most common, with Vibrio harveyi being the dominant species causing infections in these fishes. The bacterium Lactococcus garvieae is thought to exhibit host specificity in fish. The fish species in the present study exhibited a higher tendency for infection by heterologous pathogens than by a single pathogen; therefore, it is necessary to devise new strategies for treating diseases in cultured fish.

2016년도 국내 해양환경내 병원성 비브리오균의 분포 및 해양환경인자간의 상관성 분석 (Distribution of Pathogenic Vibrios in the Aquatic Environment Adjacent to Coastal Areas of South Korea and Analysis of the Environmental Factors Affecting Their Occurrence)

  • Objectives: The pathogenic Vibrios genus denotes halophilic bacteria that are distributed in aquatic environments, including both sea and freshwater. Vibrio cholerae, Vibrio vulnificus, and Vibrio parahaemolyticus are the most important species since they can be potent human pathogens and leading causes of septicemia, wound infections, and seafood borne gastroenteritis. The recent emergence of a potential pandemic clone, V. cholera serotype O1 and the cholera outbreak in South Korea in 2016 indicates the importance of consistent surveillance of pathogenic Vibrio genus within coastal areas. Methods: The present study was undertaken to determine where and how vibrios live in the aquatic environment adjacent to coastal areas of South Korea. For this survey, a total of 838 samples were obtained at 35 different sites in South Korean coastal areas during the period from January 2016 to December 2016. Pathogenic vibrios was determined using the real-time PCR method, and its clones were isolated using three selective plating media. We also monitored changes in seawater and atmospheric temperature, salinity, turbidity, and hydrogen ion concentration at the collection points. Results: The total isolation rates of V. vulnificus, V. cholera (non-pathogenic, non-O1, non-O139 serogroups), and V. parahaemolyticus from seawater specimens in 2016 were 14.2, 13.48, and 67.06%, respectively. Conclusions: The isolation rates of pathogenic vibrios genus showed a positive correlation with temperature of seawater and atmosphere but were negatively correlated with salinity and turbidity.

Propolis의 어류 병원성 세균에 대한 in vitro 항균 효과 (In vitro antimicrobial activity of Korean propolis against fish pathogenic bacteria)

  • The present study was to investigate in vitro antimicrobial effects of propolis against six different fish bacterial pathogens, Aeromonas hydrophila, Edwardsiella tarda, Vibrio vulnificus, V. parahaemolyticus, A. salmonicida subsp. masoucida, A. salmonicida subsp. salmonicida) using minimum inhibitory concentration (MIC) and minimum bacteriocidal concentration (MBC) tests. In the results, propolis exhibited antimicrobial activity against all bacteria used in the present study, but there was no marked difference in bacterial species except Vibrio species. Collectively, propolis was thought to be an usefulness antimicrobial substance for controlling bacterial diseases in the fish industry.

한국에서 분리된 Vibrio harveyi 외막단백질의 유전적 차이와 넙치(Paralichthys olivaceus)에 대한 OmpW의 면역원성 분석 (Genetic Variations of Outer Membrane Protein Genes of Vibrio harveyi Isolated in Korea and Immunogenicity of OmpW in Olive Flounder, Paralichthys olivaceus)

  • Vibrio harveyi is a pathogenic marine bacterium causing systemic symptoms resulting in mass mortalities in fishes and shrimps in aquaculture. Outer membrane proteins(OMPs) are related to the pathogenicity and thus good targets for diagnosis and vaccination for Gram negative bacteria. Recently vaccination strategies using the OMPs have been suggested to control vibriosis in several fish species. In this study, we have isolated V. harveyi from diseased marine fishes from different regions of Korea and investigated genetic variations of four OMP genes including OmpK, OmpU, OmpV and OmpW. Consequently, OmpK and U genes could be divided into 3 subgroups of type I, II, III and type A, B, C, respectively, without any correlation with geographical regions and species while OmpV and W were highly homologous. OmpW gene of V. harveyi FP4138 was fully sequenced and predicted the deduced amino acid sequence to form ${\beta}-barrel$ with hydrophobic channel. Indeed, the immunogenicity of recombinant OmpW produced in Escherichia coli was assessed by vaccinating flounder. As a result, the high antibody response with antibody titer of $4.2{\pm}0.7$ and protection with relative percent survival of 60% against artificial infection of V. harveyi were demonstrated. This result indicates that OmpW is a virulence related factor and it can be a vaccine candidate to prevent a high mortality caused by V. harveyi infection in olive flounder, Paralichthys olivaceus.

Antibacterial Effects of Natural Essential Oils from Ginger and Mustard against Vibrio Species Inoculated on Sliced Raw Flatfish

  • In order to extend the shelf life of sliced raw flatfish, the antimicrobial effects of natural essential oil from mustard and a mixture of ginger and mustard essential oils were tested at various temperatures. In addition, volatile components of the mixed essential oils were analyzed using gas chromatography and gas chromatography mass spectrometry. The viable cell counts of Vibrio parahaemolyticus treated with mixed essential oils from ginger and mustard was 0.7-1.3 log CFU/g lower than those of other treatments during storage at $5^{\circ}C$. During storage at $20^{\circ}C$, the viable cell counts of V. parahaemolyticus, V. vulnificus 01, and V. vulnificus 02 treated with the essential oils increased slightly from 6.53-6.64 log CFU/g at initial stages to 6.77-7.72 log CFU/g after 24-hr of storage, however they were 1.38-1.97 log CFU/g lower than those of the control group (8.74-9.10 log CFU/g). These results show that the growth of V. parahaemolyticus and V. vulnificus inoculated on sliced raw flatfish could be inhibited by treatment with natural essential oils from ginger and mustard at $5^{\circ}C$ of storage. However, the antibacterial effects of the essential oils on Vibrio species observed in this study were not sufficient to merit their use in sliced raw flatfish at temperatures exceeding $20^{\circ}C$.

2002-2003년도 복수증 양식산 넙치로부터 동정된 미생물상 (Microbial Flora in Ascitic Cultivated Olive Flounder (Paralichthys olivaceus) in Koe-je Island in Korea During 2002-2003.)

  • 본 연구는 2002년 6월부터 2003년 11월까지 복수증을 나타내는 양식넙치의 세균상과 virus를 동정한 것이다. 생화학적 성상을 기초로 한 표현형으로, 병어체의 복수, 간, 신장, 내장으로부터 Vibrio, Edwardsiella, 및 stonella 등 3개 속이 동정되었다. Vibrio속은 5종으로 우점이었고 V. harveyi (32균주), V. parahaemolyticus (23균주), V. alginolyticus (10균주), V. carcariae (2균주), V. metschnikovii (5균주)였다. 그밖에 Photobacterium damselae (6균주)와 장내세균인 Edwardsiella tarda (15균주)가 동정되었다. 감염어의 내부 장기로부터 해산어 병원성 바이러스로 알려진 marine birnavirus (MABV)가 분리되었다.
