Screening and Characterization of a Novel Cellulase Gene from the Gut Microflora of Hermetia illucens Using Metagenomic Library

  • A metagenomic fosmid library was constructed using genomic DNA isolated from the gut microflora of Hermetia illucens, a black soldier fly. A cellulase-positive clone, with the CS10 gene, was identified by extensive Congo-red overlay screenings for cellulase activity from the fosmid library of 92,000 clones. The CS10 gene was composed of a 996 bp DNA sequence encoding the mature protein of 331 amino acids. The deduced amino acids of CS10 showed 72% sequence identity with the glycosyl hydrolase family 5 gene of Dysgonomonas mossii, displaying no significant sequence homology to already known cellulases. The purified CS10 protein presented a single band of cellulase activity with a molecular mass of approximately 40 kDa on the SDS-PAGE gel and zymogram. The purified CS10 protein exhibited optimal activity at $50^{\circ}C$ and pH 7.0, and the thermostability and pH stability of CS10 were preserved at the ranges of $20{\sim}50^{\circ}C$ and pH 4.0~10.0. CS10 exhibited little loss of cellulase activity against various chemical reagents such as 10% polar organic solvents, 1% non-ionic detergents, and 0.5 M denaturing agents. Moreover, the substrate specificity and the product patterns by thin-layer chromatography suggested that CS10 is an endo-${\beta}$-1,4-glucanase. From these biochemical properties of CS10, it is expected that the enzyme has the potential for application in industrial processes.

  • Chlorella has been reported to have certain beneficial physiological effects, including hypocholesterolemic, antihypertensive, antioxidative, and anti-tumor activities in animal and human studies. The aim of this study was to determine the anti-inflammatory activities of an 80% methanol extract(CE-Met), hexane fraction (CE-Hex), and ethyl acetate fraction (CE-EA) of Chlorella ellipsoidea in lipopolysaccharide (LPS)-stimulated RAW 264.7 murine macrophage. Treatment with various concentrations of the C. ellipsoidea extract resulted m a significant, dose-dependent reduction in nitric oxide (NO) production by LPS-induced macrophages. The C. ellipsoidea extract significantly inhibited LPS-induced NO production accompanied by an attenuation of IL-6 and TNF-$\alpha$ formation in macrophages. These results suggest potent inhibit이y effects on the production of inflammatory mediators by a C. ellipsoidea extract. Thus, C. ellipsoidea extract may be a potent anti-inflammatory agent for troubled skin.

인삼(人蔘)의 항산화(抗酸化) 성분(成分)에 관(關)한 연구(硏究) (Study on the Components of the Anti-oxidant Activity of Panax Ginseng)

  • 홍삼및 백삼에서 용매추출법(溶媒抽出法)에 의해 fr. 1과 fr. 2를 분리하여 양자의 차이점(差異點)을 몇가지 관점에서 조사하고 홍삼에서 강하게 나타나는 항산화(抗酸化) 활성물질(活性物質)을 단리(單離)하여 실험한 결과는 다음과 같다. 1. fr. 1과 fr. 2에서의 항산화(抗酸化) 활성(活性)은 공히 홍삼(紅蔘) 분획(分劃)이 백삼(白蔘) 분획(分劃)보다 강했다. 2. 홍삼의 fr. 2에 비해 fr. 1이 비활성(比活性)이 훨씬 강했다. 3. 홍삼의 fr. 1에서 단리(單離)한 Spot a는 강력한 항산화 활성을 가졌으로 indophenol용액을 탈색(脫色)시켰다. 4. 홍삼의 fr. 2에서 단리(單離)한 Spot b도 항산화활성물질(抗酸化活性物質)로서 남색의 indophenol용액을 홍색으로 변색시켰다. 5. 인삼제품으로부터 fr. 1과 fr. 2를 분리하여 TLC 또는 HPLC에 의해 그 패턴을 조사하며 원료삼(原料蔘)이 홍삼인지 백삼인지 식별(識別)할 수 있는 지표(指標)로 활용할 수 있을 것이다.

Cytochrome P450 2E1 Activity in a Korean Population

  • Cytochrome P450 2E1 (CYP2E1) is involved in the toxicity and carcinogenicity of a number of solvents and xenobiotics. Like the various types of oxidation pharmacogenetics, the activity of the enzyme shows a discernible interindividual and interethnic variation. However, no pharmacogenetic information on CYP2E1 polymorphism has been available from a Korean population. The aim of this study was to explore the pharmacogenetics of CYP2E1 polymorphism in a native Koreans after an oral 400 mg dose of chlorzoxazone administered to 128 subjects. Urine samples were collected during the subsequent 8-hour period and urinary concentrations of chlorzoxazone and 6-hydroxychlorzoxazone were determined by a high performance liquid chromatography with an ultraviolet detector. The limit of detection in the samples was found to be $0.5\;{\mu}g/ml$. The mean value of the 6-hydroxychlorzoxazone excreted in 8 hr urine expressed as the percentage was 48.2 13.8%. The frequency distribution of percentage of the administered dose excreted as the 6-hydroxy metabolite was unimodally distributed in the subjects studied. However, the values showed wide (7-fold) interindividual difference, ranged from 11.6% to 79.8% of the dose of chlorzoxazone. Thus, it was considered that the pharmacogenetic characteristics of CYP2E1 in a Korean population did not represent multimodal distribution in the 6-hydroxychlorzoxazone excreted in 8-hr urine expressed as the percentage. And the activity of the CYP2E1 in a Korean population seemed to be less compared with that of the Caucasian subjects.

다양한 용매에서 Soxhlet 추출기를 이용한 감귤 껍질에서 d-limonene 추출의 최적조건 연구 (Optimization of d-limonene Extraction from Tangerine Peel in Various Solvents by Using Soxhlet Extractor)

  • 감귤류의 과일 내에 포함된 것으로 알려진 d-limonene 성분은 정유 성분을 추출하여 분리하는 방법으로 얻을 수 있다. 연속 증류 추출법 중 하나인 Soxhlet 추출법을 사용하여 감귤 껍질의 정유 성분을 추출한 후, 역상 HPLC를 통해 정량분석을 하였다. 추출용매의 끓는점과 극성도가 수율에 주는 영향을 알아보기 위해 에탄올, n-헥산, 석유에테르 3가지 용매를 추출 용매로 사용하였다. $120^{\circ}C$에서 각각의 용매를 1, 2, 4시간 동안 추출했을 때의 시료를 얻어 총 9가지 시료를 분석하였다. 역상 HPLC에서 정량분석을 한 결과, 세 가지 용매 모두 2시간 동안 추출했을 때 d-limonene의 함유량이 가장 많았으며, 이 때 감귤 껍질 1 g당 함유량은 7.77 mg(에탄올), 0.49 mg(n-헥산), 0.28 mg(석유에테르)였다. 에탄올을 사용했을 때 수율이 가장 높은 것으로 보아 용매의 극성도가 끓는점보다 추출 효율에 더 큰 영향을 미치는 것을 알 수 있었다.

  • 有機클로로酸, 또는 에스타와 各種金屬(亞鉛, 硅素, 마그네슘, 錫)과의 여러 溶媒存在下(아세트나이트라일, 디옥센 및 토루엔)의 反應에서 얻은 反應生成物, 또는 反應系中의 金屬分布狀態를 (n,${\gamma}$)反應으로 製造한 各種放射性 金屬트레이서를 使用하여 決定하였다. 有機클로로 金屬 콤푸렉스의 溶解度는 親水性溶媒에서는 顯著하게 增加하였으며, 無極性溶媒에는 減少하였다. 이 事實은 回收된 金屬, 또는 回收된 金屬混合物의 水洗液中의 金屬分布狀態의 增加로서 確認되었다. 카보닐化合物 存在下에서 反應시켰 때에도 같은 事實을 나타내었다. 親水性溶媒中의 金屬콤푸렉스의 溶解度는 亞鉛, 硅素, 錫 및 마그네슘이 順序로 減少되었으며 無極性溶媒下에서는 硅素, 錫, 마그네슘 및 亞鉛의 順으로 減少되었다. 有機할로겐化合物과 金屬과의 反應生成物로 普通 豫期되는 有機金屬化合物은 全反應을 通하여 거의 없었다. 反應結果를 論議하였으며, 本反應過程에서 觀察한 溶媒의 影響에 關하여 論及하였다.

Selective Synthesis of a New Macropolycycle Containing One N-CH2-N Linkage and Its Reaction with Cu2+ and Ni2+ Ions in Methanol

  • The reaction of 3,14-dimethyl-2,6,13,17-tetraazatetracyclo[16.4.0$0^{1.18}.0^{7.12}$]docosane ($L^1$) with formaldehyde in warm methanol yielded 3,14-dimethyl-2,6,13,17-tetraazatetracyclo[16.4.$1^{2.6}.0^{1.18}.0^{7.12}$]tricosane ($L^7$) containing one 1,3-diazacyclohexane subunit. In methanol, $L^7$ readily reacts with $Cu^{2+}$ ion to form [$CuL^7(H_2O)$]$^{2+}$ which is extremely inert against methanolysis. In the solution containing $Ni^{2+}$ ion, however, $L^7$ reacts with methanol to yield [$NiL^3$]$^{2+}$ ($L^3$ = 2-(methoxymethyl)-5,16-dimethyl-2,6,13,17-tetraazatricyclo[16.4.$0^{1.18}.0^{7.12}$]- docosane), in which one N-$CH_2OCH_3$ pendant arm is appended. The copper(II) complex [$CuL^7(H_2O)$]- $(ClO_4)_2{\cdot}3H_2O\;(I{\cdot}3H_2O)$ has a severely distorted trigonal bipyramidal coordination geometry with a 4-5- 6-5 chelate ring sequence. The crystal structure of [$NiL^3$]$(PF_6)_2{\cdot}2H_2O$ (IIb) shows that the N-$CH_2OCH_3$ pendant arm is not coordinated to the metal ion in the solid state. In various solvents (S), however, the nickel(II) complex exists as a mixture of [$NiL^3$(S)]$^{2+}$, in which the N-$CH_2OCH_3$ group as well as S is coordinated to the metal ion, and [$NiL^3$]^{2+}.

A comparison of the characteristic properties between soybean (Glycine max [L.] Merrill) seeds with different seed coat colors

  • We profiled the health-promoting bioactive components in nine types of soybean seeds with different seed coat colors (yellow, green, brown, and black) and investigated the effects of different extraction solvents (methanol, ethanol, and water) on their antioxidant activities. The carotenoid and anthocyanin compositions varied greatly by seed color, and the phenolic acids, total phenol, and total flavonoid contents differed by genotype. The carotenoid content was relatively higher in soybean seeds with green and black seed coats than in those with a yellow seed coat while lutein was the most plentiful. The anthocyanin content was considerably higher in the soybean seed with the black seed coat. The results of the DPPH assay showed strong antioxidative activities in the methanol- and water-extracts compared to the ethanol-extract, irrespective of the seed coat colors. Moreover, the soybean seeds with the black seed coat exhibited the highest antioxidant activity among the samples, regardless of the extraction solvent used. Eighteen bioactive compounds were subjected to data-mining processes including principal component analysis and hierarchical clustering analysis. Multivariate analyses showed that brown and black seeds were distinct from the yellow and green seeds in terms of the levels of carotenoids and anthocyanins, respectively. These results help our understanding of the compositional differences in the bioactive components among soybean seeds of various colors, providing valuable information for future breeding programs that seek to enhance the levels of compounds with health benefits.

치자(Gardenia jasminoides Ellis) 과피의 용매별 추출물의 Flavonoid 함량 및 항산화 활성 비교 (Comparison of Flavonoid Content and Antioxidant Activities of Peel Extracts from Gardenia jasminoides Ellis by Various Solvents)

  • The purpose of this study was to measure the bioactivity and antioxidant activity of peel from Gardenia jasminoides fructus Ellis (GJE) in Namhae, Korea, following some established methods. CM (Chloroform:Methanol, 2:1, v/v), 70% ethanol, and n-butanol extracts were collected. Flavonoid content and value as a functional food ingredient of GJE peel was investigated through assessing antioxidant [DPPH (1,1'-diphenyl-2-picrylhydrazyl), ABTS (2,2-azino-bis-3-ethylbenzothiazoline-6-sulfonic acid)], and hydroxyl radical scavenging activities; superoxide dismutase like ability; ferrous ion-chelating capacity; and tannin content by solvent extraction. Solvent extract antioxidant activities significantly increased (p<0.05) at increasing concentrations (0.2, 0.4, 0.6 mg/mL). GJE peel extracts were less active than the positive control [ascorbic acid, BHA (butylated hydroxyanisole), and EDTA (ethylenediaminetetraacetic acid disodium salt dihydrate)]. Based on the results of this study, GJE peel could be used as a natural antioxidant source due to its high antioxidant activity and bioactive compound content.

황금(Scutellaria baicalensis)의 에틸 아세테이트 추출물의 항산화 및 항균 활성 (Antioxidant and Antibacterial Activities of Ethyl Acetate Extract from Scutellaria baicalensis)

  • This study aimed to determine the antioxidant and antibacterial effects of solvent extracts from Scutellaria baicalensis. The S. baicalensis was successively extracted with 5 kinds of solvents. The obtained samples were as follows; hexan fraction (IPK-1), chloroform fraction(IPK-2), ethyl acetate fraction(IPK-3), butanol fraction(IPK-4), and water fraction(IPK-5). These extracts were determined to process antibacterial effects using a paper disc method against 18 species of human pathogenic bacteria, antioxidant activity using 1,1-diphenyl-2-picrylhydrazyl(DPPH) method and inhibiting ability of linoleic acid autoxidation. Extracts of S. baicalensis has shown selective antibacterial effects on various strains of bacteria. Ethyl acetate fraction(IPK-3) showed the highest antimicrobial activity against selective bacteria, such as Staphylococcus aureus 285, S. aureus 503. In antioxidant activity test, free radical scavenger activity of IPK-3 fraction was 92% compared with BHA at the same concentration, while lipid peroxidation inhibition was 48%. These results suggest that IPK-3 fraction of S. baicalensis could be suitable in development of antimicrobial and antioxidant agents.
