A Routine System for Generation of Fertile Transgenic Rice Plants Using Biolistic Method

  • A routine system based on particle bombardment of embryogenic callus for recovery of fertile transgenic rice (Oryza sativa L.) plants was developed. Embryogenic callus was established within 2-3 months from calli derived from mature seeds of Korean rice cultivar, Nagdongbyeo. The callus was bombarded with the plasmid pRQ6 containing the $\beta$-glucuronidase gene (gusA) and hygromycin phosphotransferase gene (hph, conferring resistance to hygromycin B), both driven by CaMV 35S promoter. Placement of cells on an osmoticum-containing medium (0.2 M sorbitol and 0.2 M mannitol) 4 hrs prior to and 16 hrs after bombardment resulted in a statistically significant increase with 3.2-fold in transient expression frequency gusA. In five independent experiments, the average frequency of transformation showing GUS activities was $8.86\%$. A large number of morphologically normal, fertile transgenic rice plants were obtained. Integration of foreign gene into the genome of $R_0$ transgenic plants was confirmed by Southern blot analysis. GUS and HPT were detected in $R_1$ progeny and Mendelian segregation of these genes was observed in $R_1$ progeny.

  • Productivity of a rice plant is greatly influenced by salt stress. One of the ways to achieve tolerance to salinity is to transfer genes encoding protective enzymes from other organisms, such as microorganisms. The bacterial gene, mtlD, which encodes mannitol-1-phosphate dehydrogenase (Mtl-DH), was introduced to the cytosol of a rice plant by an imbibition technique to overproduce mannitol. The germination and survival rate of the imbibed rice seeds were markedly increased by transferring the mtlD gene when it was delivered in either a pBIN19 or pBmin binary vector. When a polymerase chain reaction was performed with the genomic DNAs of the imbibed rice leaves as a template and with mtlD-specific primers, several lines were shown to contain an exogenous mtlD DNA. However, a reverse transcription (RT)-PCR analysis revealed that not all of them showed an expression of this foreign gene. This paper demonstrates that the growth and germination of rice plants transiently transformed with the bacterial gene, mtlD, are enhanced and these enhancements may have resulted from the experssion of the mtlD gene. The imbibition method empolyed in this study fulfills the requirements for testing the function of such a putative gene in vivo prior to the production of a stable transgenic plant.

  • For two input ESD protection schemes utilizing the NMOS protection device or the lvtr_thyristor protection device, which is suitable for high-frequency CMOS ICs, we attempt an in-depth comparison study on the HBM ESD protection level in terms of lattice heating inside the protection devices and the peak voltage applied to the gate oxides in the input buffer through DC, mixed-mode transient, and AC analyses utilizing the 2-dimensional device simulator. For this purpose, we suggest a method for the equivalent circuit modeling of the input HBM test environment for the CMOS chip equipped with the input ESD protection circuit. And by executing mixed-mode simulations including up to four protection devices and analyzing the results for five different test modes, we attempt a detailed analysis on the problems which can be occurred in a real HBM test. In this procedure, we explain about the strength and weakness of the two protection schemes as an input protection circuit for high-frequency ICs, and suggest guidelines relating to the design of the protection devices.

  • This paper proposes a modified DCPC (Distributed Constrained Power Control, M-DCPC) algorithm that can improve the performance of a CDMA power control system. The control performance of the proposed method is verified using two performance measures : the SIR response of each mobile and the outage probability in a cell. As regards the SIR response, in simulations, the M-DCPC algorithm has shown a faster convergence and lower overshoot in transient time than the other power control algorithms when the desired SIR value was varying. For the outage probability. M-DCPC converged to a fixed outage rate faster than CSOPC while also maintaining the system capacity to make as high a connection as CSOPC. In particular, when the desired SIR was varying, CSOPC showed an abrupt outage probability increase during the desired SIR Increase, yet M-DCPC was unaffected.

A Comparative Study on the Thermal Conductivities and Viscosities of the Pure Water and Ethanol Carbon Nanofluids (순수 물과 에탄올 산화 탄소나노유체의 열전도도 및 점도 특성 비교 연구)

  • Nanofluids are advanced concept fluid that solid particles of nanometer size are stably dispersed in fluid likes water, ethylene glycol and others. They have higher thermal conductivities than base fluids. If using this characteristic, efficiencies of heat exchangers can be increased. Therefore in this study, we measured thermal conductivity and viscosity of carbon nanofluids. They were made to ultra sonic dispersed oxidized multi-walled carbon nanotubes(OMWCNTs) in distilled water and ethanol, respectively. The mixture ratios of OMWCNTs were from 0.0005 vol% ~ 0.1 vol%. Thermal conductivity and viscosity was measured by transient hot-wire method and rotational viscometer. The results of an experiment are as in the following: thermal conductivity of the 0.1 vol% pure-water nanofluid improved 7.98% ($10^{\circ}C$), 8.34% ($25^{\circ}C$), and 9.14% ($70^{\circ}C$), and its viscosity increased by 37.08% ($10^{\circ}C$), 33.96% ($25^{\circ}C$) and 21.64% ($70^{\circ}C$) than the base fluids. Thermal conductivity of the 0.1 vol% ethanol nanofluids improved 33.72% ($10^{\circ}C$), 33.14% ($25^{\circ}C$), and 32.25% ($70^{\circ}C$), and its viscosity increased by 35.12% ($10^{\circ}C$), 32.01% ($25^{\circ}C$) and 19.12% ($70^{\circ}C$) than the base fluids.

Silicone Implant-Based Paranasal Augmentation for Mild Midface Concavity

  • Background: Midface concavity is a relatively common facial feature in East Asian populations. Paranasal augmentation is becoming an increasingly popular procedure for patients with mild concavity and normal occlusion. In this study, we evaluate clinical outcomes following a series of paranasal augmentation. Methods: A retrospective review was performed for patients with Class I occlusion who had undergone bilateral paranasal augmentation using custom-made silicone implants, between October 2005 and September 2013. Patient charts were reviewed for demographic information, concomitant operations, and postoperative complications. Preoperative and postoperative (1-month) photographs were used to evaluate operative outcome. Results: The review identified a total of 93 patients meeting study criteria. Overall, aesthetic outcomes were satisfactory. Five-millimeter thick silicone implant was used in 81 cases, and the mean augmentation was 4.26 mm for this thickness. Among the 93 patients, 2 patients required immediate implant removal due to discomfort. An additional 3 patients experienced implant migration without any extrusion. Nine patients complained of transient paresthesia, which had resolved by 2 weeks. There were no cases of hematoma or infection. All patients reported improvement in their lateral profile and were pleased at follow-up. Complications that arose postoperatively included 9 cases of numbness in the upper lip and 3 cases of implant migration. All cases yielded satisfactory results without persisting complications. Sensations were fully restored postoperatively after 1 to 2 weeks. Conclusion: Paranasal augmentation with custom-made silicone implants is a simple, safe, and inexpensive method that can readily improve the lateral profile of a patient with normal occlusion. When combined with other aesthetic procedures, paranasal augmentation can synergistically improve outcome and lead to greater patient satisfaction.

Enhancement of $Ca^{2+}$ Spark Occurrence by Murrayafoline-A in Rat Ventricular Myocytes (Murrayafoline-A에 의한 심실 근육세포 $Ca^{2+}$ 스파크 발생의 증가)

  • Murrayafoline-A (1-methoxy-3-methylcarbazole) is a monomeric carbazole alkaloid found in Murraya euchrestifolia HAYATA and Glycosmis stenocarpa. We have recently shown that murrayafoline-A has positive inotropic effect in isolated rat ventricular myocytes. To know possible mechanisms for the positive inotropic effect of murrayafoline-A we examined the effects of murrayafoline-A on in situ behavior of cardiac $Ca^{2+}$ release units ('$Ca^{2+}$ sparks') and sarcoplasmic reticulum (SR) $Ca^{2+}$ loading using confocal $Ca^{2+}$ imaging method in single rat ventricular myocytes. Murrayafoline-A significantly increased the frequency (events/($10^3{\mu}m^2{\cdot}s$)) of $Ca^{2+}$ sparks in a concentration-dependent manner, with an $EC_{50}$ of $28{\pm}6.4{\mu}M$ and a maximal ~twofold change. The $Ca^{2+}$ content in the SR, measured as caffeine (10 mM)-induced $Ca^{2+}$ transient, was significantly increased by murrayafoline-A (${\approx}$116% and ${\approx}$123% of control at 25 and 100 ${\mu}M$, respectively). In addition, murrayafoline-A significantly increased the fractional $Ca^{2+}$ release, suggesting increase in the efficacy of $Ca^{2+}$ release at given SR $Ca^{2+}$ loading. These results suggest that murrayafoline-A may enhance contractility via increase in $Ca^{2+}$ release from the SR through the ryanodine receptors in ventricular myocytes.

Efficient simulation method for a gas inflow to the central molecular zone

  • We present hydrodynamic simulations of gas clouds that inflowing from the disk to a few hundred parsec region of the Milky Way. Realistic Galactic structures are included in our simulations by thousands of multipole expansions that describe 6.4 million stellar particles of a self-consistent Galaxy simulation (Baba, Saitoh & Wada, in prep.). We find that a hybrid multipole expansion model with two different basis sets and a thick disk correction well reproduces the overall structures of the Milky Way. We find that the nuclear ring evolves into 240 pc at T~1500 Myr, regardless of the initial size. For most of simulation runs, gas inflow rate to the nuclear region is equilibrated as ~0.02 Msun/yr, and thus accumulated gas mass and star formation activity is stabilized as $6{\times}10^7Msun$ and ~0.02M/yr, respectively. These stabilized values are in a good agreement with estimations for the CMZ. The nuclear ring is off-centered to the Galactic center by the lopsided central mass distribution of the Galaxy model, and thus an asymmetric mass distribution is arose accordingly. The lopsidedness also leads the nuclear ring to be tilted to the Galactic plane and to precess along the Galaxy rotation. In early evolutionary stage when gas clouds start to inflow and form the nuclear ring, the z-directional oscillations of the gas clouds results in the twisted, infinity-shaped nuclear ring. Since the infinity-shaped feature is transient only for first 100 Myr, the current infinity-shape observed in the CMZ may indicate that the CMZ forms quite recently.

An Efficient Control Strategy Based Multi Converter UPQC using with Fuzzy Logic Controller for Power Quality Problems

  • A custom power device provides an integrated solution to the present problems that are faced by the utilities and power distribution. In this paper, a new controller is designed which is connected to a multiconverter unified power quality conditioner (MC-UPQC) for improving the power quality issues adopted modified synchronous reference frame (MSRF) theory with Fuzzy logic control (FLC) technique. This newly designed controller is connected to a source in order to compensate voltage and current in two feeders. The expanded concept of UPQC is multi converter-UPQC; this system has a two-series voltage source inverter and one shunt voltage source inverter connected back to back. This configuration will helps mitigate any type of voltage / current fluctuations and power factor correction in power distribution network to improve power quality issues. In the proposed system the power can be conveyed from one feeder to another in order to mitigate the voltage sag, swell, interruption and transient response of the system. The control strategies of multi converter- UPQC are designed based on the modified synchronous reference frame theory with fuzzy logic controller. The fast dynamics response of dc link capacitor is achieved with the help of Fuzzy logic controller. Different types of fault conditions are taken and simulated for the analysis and the results are compared with the conventional method. The relevant simulation and compensation performance analysis of the proposed multi converter-UPQC with fuzzy logic controller is performed.

Compact Anode Design with the Heat Capacity Performance in Rotating Anode X-ray Tube for Digital Radiography

  • We studied the compact anode design to develop 100 kW rotating anode X-ray tube with large focal spot 1.2 mm, small focal spot 0.6 mm and tube voltage 150 kV for large hospital digital radiography using computer thermal simulation. The larger thermal radiation effect in a high vacuum can reduce the temperature of anode so the method to increase the surface area of anode is investigated. The anode has the multi-tier shape at the back side of TZM body of anode and also bigger diameter of anode. The number of multi-tiers was varied from 6 to 15 and the diameter of anode was also varied from ${\Phi}74$ to ${\Phi}82$. From ANSYS transient thermal simulation result, we could obtain $1056.4^{\circ}C$ anode maximum temperature when applying 100 kW input power at 0.1 second on target focal track which is less than $1091^{\circ}C$ of the conventional 75 kW X-ray tube with reduced anode weight by 15.5% than the conventional anode. The compact anode of reduced anode weight is able to improve the unwanted noise when the rotor is rotating at high-speed and also reduce the rotational torque which the cost effective stator-coil is possible. It is believed that the anode with 15 ea multi-tiers using ${\Phi}82$ can satisfy with the specification of the anode heat capacity. From the results of this paper, it has been confirmed that the proposed compact anode can be used as the anode of 100 kW rotating anode X-ray tube for digital radiography.
