Background: Green tobacco sickness (GTS), an occupational disease in tobacco harvesters, is a form of acute nicotine intoxication by nicotine absorption through the skin from the wet green tobacco plant. We carried out a questionnaire survey and measured cotinine concentration, the metabolic product of nicotine, to determine the prevalence, incidence, and risk factors of GTS in Korean tobacco harvesters. Methods: We measured cotinine concentrations, and administered a questionnaire survey to tobacco harvesters in Cheongsong-gun, Gyeongsangbuk-do, Korea. We repeatedly measured urine cotinine concentration five times with a questionnaire survey. Results: Cotinine concentration at dawn was significantly higher than that at other times; it was significantly lower during the nonharvesting period than during the harvesting period. However, little change in cotinine concentration was detected in the daytime during the harvesting period. Study participants included 20 men and 20 women. The prevalence of GTS was 37.5% and was significantly higher in women than in men (55.0% vs. 20.0%, p < 0.01). GTS incidence according to number of workdays was 3.4 occurrences/100 person days. Conclusion: In this study, nicotine exposure and metabolism were experimentally determined from the time of cotinine exposure, and biological monitoring was performed in each season. In the future, this information may be valuable for medical decision-making in GTS prevention.
Tobacco mosaic virus (TMV), TMV-common strain, causes severe damage on flue-cured tobacco in Korea. As the results, breeding programs were conducted to transfer the TMV resistance from NC567 to other cultivars. Advantage of F1, hybrid may not lie only in their heterotic value but also in directly combining of valuable dominant traits, e.g. for disease resistance, either in pairs or as groups already assembled in the two parents. A new TMV resistant flue-cured tobacco variety KF114 was developed from a cross as maternal parent MSNC567 and NC82 at the Suwon Experiment Station. KF114 was more vigorous growth characteristics in early stage than that of NC82, standard variety in Korea, and other characters were similar to those of NC82. It os high resistance to TMV, but bacterial wilt(Pseudomonas solanacerum) and black shank (Phytophthora parasitica var. nicotianae) is comparable to NC82. It should adapt well to the flue-cured production area and can reduce premature flowering under unfavorable weather conditions. Yield of KF114 is 1% to 5% higher, and nearly equal in value per KG compared with NC82. This variety, F1 hybrid, met acceptable standards for chemical and physical characteristics of cured leaf and high ranked good smoking taste by panel members in Korea Ginseng and Tobacco Research Institute.

Recent characterization of several genes involved in plant defense responses suggested that ubiquitin-mediated protein degradation has a role in these responses. We isolated two cDNAs (NtUBA1 and NtUBA2) encoding ubiquitin-activating enzyme (E1) from Nicotiana tabacum cv. BY-2. The open reading frames of both encoded 1080 amino acids, corresponding to molecular masses of 120 kDa. The E1s and corresponding transcripts were upregulated by infection with tobacco mosaic virus (TMV) and tomato mosaic virus (ToMV), and to a lesser extent by cucumber mosaic virus (CMV). Furthermore, they were also upregulated by wounding stress, and the plant hormones salicylic acid, jasmonic acid and the ethylene precursor, aminocyclopropane-1-carboxylic acid (ACC). Our findings support the idea that the ubiquitin-proteasome system plays a role in plant disease defenses.
This experiment was conducted to investigate the effect of growth medium in pots composed of upland soil(S), rice straw manure(M), carbonated rice hull(CRH), and their mixture on growth of tobacco cv. NC 82. The growth of shoot and root was vigorous in order of medium S+M+CRH>M>CRH>S. In M+S medium, root growth in the part of manure was superior to upland soil. But root growth of upland soil part in M+S plot was more vigorous than that in upland soil only. It is possibly due to be influenced by manure in M+S plot. Total length and weight of root, number of roots, and especially for development of adventitious root were closely related to shoot growth. Roots grown in upland soil part was brownish gray in color, while the roots in manure part was milky white. The milky white colored roots had longer life than others. It was concluded that root zone environments derived from several media in pots closely related to shoot growth and disease tolerance of tobacco plant.
Salicylic acid (SA) is involved in the establishment of systemic acquired resistance (SAR) in many plant including tobacco. Considering the important role of SA in disease resistance, biosynthetic and metabolic pathways of SA in tobacco have been studied extensively: The initial step for biosynthetic pathway of SA is conversion of phenylalanine to trans-cinnamic acid, followed by decarboxylation of trans-cinnamic acid to benzoic acid and ie subsequent ring hydroxylation at the C-2 position to form SA. In TMV inoculated tobacco, most of the newly synthesized SA is glucosylated or methylated. Methyl salicylate has been identified as a biologically active, volatile signal. In contrast, the two glucosylated forms accumulate in the vicinity of lesions and consist of SA glucoside, a major metabolite, and SA glucose ester, a relatively minor from. Two enzymes involved in SA biosynthesis and metabolism have been purified and characterized : benzoic acid 2-hydroxylase which catalyzes conversion of benzoic acid to SA; UDP-Glucose: SA 1-O-D glucosyltransferase which converts SA to SA glucose ester. Further studies of the biosynthetic and metabolic pathways of SA will help to elucidate the SAR signal transduction pathway and provide potential tools for the manipulation of disease resistance.
The aim of this study was to evaluate the association between the level urine cotinine and periodontal diseases in Korea adults. The date from the 2014, 2015 Korean National Health and Nutrition Survey were used, and 5,146 subjects over 30 years were included in the analysis. Periodontal disease was assessed using the Community Periodontal Index. Binary logistic regression model was used to estimate the odds ratio with 95% confidence intervals. There was a 2.08-fold (95% CI; 1.73-2.05) increased in the odds of periodontal disease for those with any ETS exposure compared with those with non-smokers following adjustment for sex, age, education, and income. The level of urine cotinine also showed a dose-dependent increase in extent of periodontal disease. Among persons in the Korea who had never used tobacco, those exposed to ETS were more likely to have periodontal disease than were those not exposed to ETS. In the future, voluntary compliance of smokers to measures to reduce ETS exposure should be encouraged.

A comprehensive survey of mungbean-growing areas was conducted to observe leaf spot disease caused by Alternaria alternata. Alternaria leaf spot symptoms were observed on the leaves. Diversity of 50 genotypes of mungbean was assessed against A. alternata and data on pathological traits was subjected to cluster analysis. The results showed that genotypes of mungbean were grouped into four clusters based on resistance parameters under the influence of disease. The principal component biplot demonstrated that all the disease-related parameters (% disease incidence, % disease intensity, lesion area, and % of infection) were strongly correlated with each other. Alt a 1 gene that is precisely found in Alternaria species and is responsible for virulence and pathogenicity. Alt a 1 gene was amplified using gene specific primers. The isolated pathogen produced similar symptoms when inoculated on mungbean and tobacco. The sequence analysis of the internal transcribed spacer (ITS) region, a 600 bp fragment amplified using specific primers, ITS1 and ITS2 showed 100% identity with A. alternata. Potato virus X (PVX) -based silencing vector expressing Alt a 1 gene was constructed to control this pathogen through RNA interference in tobacco. Out of 50 inoculated plants, 9 showed delayed onset of disease. Furthermore, to confirm our findings at molecular level semi-quantitative reverse transcriptase polymerase chain reaction was used. Both phenotypic and molecular investigation indicated that RNAi induced through the VIGS vector was efficacious in resisting the pathogen in the model host, Tobacco (Nicotiana tabacum). To the best of our knowledge, this study has been reported for the first time.
Resistance to Black shank of various varieties were investigated in field and incubator. The results were summarized as follows; 1) In occurrence of this disease, yellow tobacco varieties were ranged from 4 days to 12 days after inoculated. 2) Korean native varieties were ranged from 6 days to 20 days. 3) Orient tobacco takes from 8 days to 17 days in occurrence of this one. 4) Japanese varieties ranged from 4 days to 3 days in occurrence of this disease. 5) Dark air-curing tobacco was ranged from 5 days to 14 days. 6) Chinese variety was ranged from 6 days to 11 days. Each varieties of resistant to Black shank was as follows; 1) Yellow tobacco varieties; Cash, Oxford-1, Oxford-3, N.C-75, N.C-95, Golden-cure Delcrest. 2) Native varieties; Sinzongtchio, Muktchio Sosa-3 Mokgitchio. 3) Japanese varieties; Hatano Outaruma Maruha. 4) Chinese variety; Gkzuoe, Yung-zung, Tongzuoe. 5) Orient tobacco; Samsun, Baffra, Xanthix. 6) Dark air-curing varieties; B-Hw, Connecticut-Broad leaf, Connecticut-Havana, Tennessee-Black.
This experiment were conducted to investigate heterosis and combining ability for several mutant characters by analyzing dialled crosses of flue-cured tobacco. In a dialled cross of 3 flue-cured varieties and the mutant line 83H -5, the heterosis was somewhat higher in Fl than in F2. For growth character, the heterosis was 0.28-6.03% in plant height, leaf number, leaf shape index and yield, and was 43.2% for bacterial wilt disease index. The mutant line 83H-5 showed significantly negative GCA effect for plant height, leaf width and bacterial wilt disease index in Fl and F2, leaf length in F2, and positive GCA effect for total alkaloids, total nitrogen in Fl and days to flower in F2, respectively. Specific combining ability(SCA) in 83H-5 x Hicks was significant in negative effect for leaf length(F2), number of leaves(F2), leaf shape(F1, F2), bacterial wilt(F2) and alkaloids(F1), and in 83H-5 x NC 2326 in positive effect for leaf length(F1, F2) and leaf width(F2), and for 83H-5 x NC 82 in positive effect for plant height(F1, F2) and leaf width(F2), and for 83H-5 x NC 82 in Positive effect for Plant height(F1, F2), leaf length(F2) and yield(F1, F2).
